1Dysregulation of innate immunity in ulcerative colitis patients who fail anti-tumor necrosis factor therapy显示文摘AIM To study the innate immune function in ulcerative colitis(UC) patients who fail to respond to anti-tumor necrosis factor(TNF) therapy.METHODS Effects of anti-TNF therapy, inflammation and medications on innate immune function were assessed by measuring peripheral blood mononuclear cell(PBMC) cytokine expression from 18 inflammatory bowel disease patients pre- and 3 mo post-anti-TNF therapy. Toll-like receptor(TLR) expression and cytokine production post TLR stimulation was assessed in UC 'responders'(n = 12) and 'non-responders'(n = 12) and compared to healthy controls(n = 12). Erythrocyte sedimentation rate(ESR) and C-reactive protein(CRP) levels were measured in blood to assess disease severity/activity and inflammation. Pro-inflammatory(TNF, IL-1β, IL-6), immuno-regulatory(IL-10), Th1(IL-12, IFNγ) and Th2(IL-9, IL-13, IL-17A) cytokine expression was measured with enzyme-linked immunosorbent assay while TLR cellular composition and intracellular signalling was assessed with FACS.RESULTS Prior to anti-TNF therapy, responders and nonresponders had similar level of disease severity and activity. PBMC's ability to respond to TLR stimulation was not affected by TNF therapy, patient's severity of the disease and inflammation or their medication use. At baseline, non-responders had elevated innate but not adaptive immune responses compared to responders(P < 0.05). Following TLR stimulation, nonresponders had consistently reduced innate cytokine responses to all TLRs compared to healthy controls(P < 0.01) and diminished TNF(P < 0.001) and IL-1β(P < 0.01) production compared to responders. This innate immune dysfunction was associated with reduced number of circulating plasmacytoid dendritic cells(p DCs)(P < 0.01) but increased number of CD4+ regulatory T cells(Tregs)(P = 0.03) as well as intracellular accumulation of IRAK4 in non-responders following TLR-2,-4 and-7 activation(P < 0.001). CONCLUSION Reduced innate immunity in non-responders may explain reduced efficacy to anti-TNF therapy. 3The current status of nitrogen fertiliser use efficiency and future research directions for the Australian cotton industry显示文摘Fifty years of sustained investment in research and development has left the Australian cotton industry well placed to manage nitrogen(N) fertiliser. The average production in the Australian cotton industry today is greater than two tonnes of lint per hectare due to improved plant genetics and crop management. However, this average yield is well below the yield that would be expected from the amount of N fertiliser used. It is clear from the recent studies that across all growing regions, conversion of fertiliser N into lint is not uniformly occurring at application rates greater than 200-240 kg·hm^(-2) of N. This indicates that factors other than N availability are limiting yield, and that the observed nitrogen fertiliser use efficiency(NFUE) values may be caused by subsoil constraints such as sodicity and compaction. There is a need to investigate the impact of subsoil constraints on yield and NFUE.Gains in NFUE will be made through improved N fertiliser application timing, better targeting the amount of fertiliser applied for the expected yield, and improved soil N management. There is also a need to improve the ability and confidence of growers to estimate the contribution of soil N mineralisation to the crop N budget. Many Australian studies including data that could theoretically be collated in a meta-analysis suggest relative NFUE values as a function of irrigation technique; however, with the extensive list of uncontrolled variables and few studies using non-furrow irrigation, this would be a poor substitute for a single field-based study directly measuring their efficacies. In irrigated cotton, a re-examination of optimal NFUE is due because of the availability of new varieties and the potential management and long-term soil resilience implications of the continued removal of mineralised soil N suggested by high NFUE values. 4Deep Scalogram Representations for Acoustic Scene Classification显示文摘Spectrogram representations of acoustic scenes have achieved competitive performance for acoustic scene classification. Yet, the spectrogram alone does not take into account a substantial amount of time-frequency information. In this study, we present an approach for exploring the benefits of deep scalogram representations, extracted in segments from an audio stream. The approach presented firstly transforms the segmented acoustic scenes into bump and morse scalograms, as well as spectrograms; secondly, the spectrograms or scalograms are sent into pre-trained convolutional neural networks; thirdly,the features extracted from a subsequent fully connected layer are fed into(bidirectional) gated recurrent neural networks, which are followed by a single highway layer and a softmax layer;finally, predictions from these three systems are fused by a margin sampling value strategy. We then evaluate the proposed approach using the acoustic scene classification data set of 2017 IEEE AASP Challenge on Detection and Classification of Acoustic Scenes and Events(DCASE). On the evaluation set, an accuracy of 64.0 % from bidirectional gated recurrent neural networks is obtained when fusing the spectrogram and the bump scalogram, which is an improvement on the 61.0 % baseline result provided by the DCASE 2017 organisers. 6Modification and characterization of fibers of three sandy willow shrub species显示文摘Periploca sepium, Salix psammophila and Tamarix spp. make up an important part of the local vegetation in the sandland of China. This paper described a novel method on modification of the three shrub fibers as oil sorbents. Natural and modified fibers were chemically studied by non-destructive methods, infrared spectroscopy (IR) and cross-polarization nuclear magnetic resonance (CP-MAS 13C-NMR). The results show that during the reaction with acetic anhydride under mild conditions, the use of N-bromoscciniamide (NBS) catalyst resulted in a high weight percent gain (WPG). The acetylation of P. sepium fiber gave rise to the highest WPG (20.0%) compared with S. psammophila and Tamarix spp. fiber.
