| 4 | Diversity of methanotrophs in Zoige wetland soils under both anaerobic and aerobic conditions显示文摘Zoige wetland is one of the most important methane emission centers in China. The oxidation of methane in the wetland affects global warming, soil ecology and atmospheric chemistry. Despite their global significance, microorganisms that consume methane in Zoige wetland remain poorly characterized. In this study, we investigated methanotrophs diversity in soil samples from both anaerobic site and aerobic site in Zoige wetland using pmoA gene as a molecular marker. The cloning library was constructed according to the pmoA sequences detected. Four clusters of methanotrophs were detected. The phylogenetic tree showed that all four clusters detected were affiliated to type I methanotrophs. Two novel clusters (cluster 1, cluster 2) were found to relate to none of the recognized genera of methanotrophs. These clusters have no cultured representatives and reveal an ecological adaptation of particular uncultured methanotrophs in Zoige wetland. Two clusters were belonging to Methylobacter and Methylococcus separately. Denaturing gradient gel electrophoresis gel bands pattern retrieved from these two samples revealed that the community compositions of anaerobic soil and aerobic soil were different from each other while anaerobic soil showed a higher metanotrophs diversity. Real-time PCR assays of the two samples demonstrated that aerobic soil sample in Zoige wetland was 1.5 times as much copy numbers as anaerobic soil. | 5 | Overexpression of SlOFP20 affects floral organ and pollen development显示文摘The OVATE gene was initially identified in tomato and serves as a key regulator of fruit shape.There are 31 OFP members in the tomato genome.However,their roles in tomato growth and reproductive development are largely unknown.Here,we cloned the OFP transcription factor SlOFP20.Tomato plants overexpressing SlOFP20 displayed several phenotypic defects,including an altered floral architecture and fruit shape and reduced male fertility.SlOFP20 overexpression altered the expression levels of some brassinosteroid(BR)-associated genes,implying that SlOFP20 may play a negative role in the BR response,similar to its ortholog OsOFP19 in rice.Moreover,the transcript accumulation of gibberellin(GA)-related genes was significantly affected in the transgenic lines.SlOFP20 may play an important role in the crosstalk between BR and GA.The pollen germination assay suggested that the pollen germination rate of SlOFP20-OE plants was distinctly lower than that of WT plants.In addition,the tomato pollen-associated genes SlCRK1,SlPMEI,LePRK3,SlPRALF,and LAT52 were all suppressed in the transgenic lines.Our data imply that SlOFP20 may affect floral organ and pollen development by modulating BR and GA signaling in tomato. | Shengen Zhou Zongli Hu Fenfen Li Shibing Tian Zhiguo Zhu Anzhou Li Guoping Chen | 2019 | Horticulture Research2019,6,1: | 4 |
| 6 | The characterization of auditory brainstem response(ABR) waveforms: A study in tree shrews(Tupaia belangeri)显示文摘To characterize the patterns of ABR waves in tree shrews, we must understand the hearing sensitivity and auditory function of healthy adult tree shrews. Fifteen tree shrews(30 ears) were stimulated with clicks and tone-pips at 11 different frequencies from 1 to 60 kHz. The ABR waves were recorded and analyzed. The ABR consisted of five to seven positive waves in the first 10 ms after a click stimulus, and the average hearing threshold of component III was 27.86 ± 3.78 dB SPL. Wave III was the largest and most clear. The ABR threshold was related to the tone-pip sitmulus by a 'U' shaped curve. The sensitive frequency was approximately 8 kHz in tree shrews. The latencies systematically decreased with increasing stimulus frequencies. The ABR amplitudes of wave III increased as the sound pressure level increased. | 7 | Profile of the culturable microbiome capable of producing acyl-homoserine lactone in the tobacco phyllosphere显示文摘Bacterial populations coexisting in the phyllosphere niche have important effects on plant health. Quorum sensing (QS) allows bacteria to communicate via diffusible signal molecules, but QS-dependent behaviors in phyllosphere bacterial populations are poorly understood. We investigate the dense and diverse N-acyl-homoserine lactone (AHL)-producing phyllosphere bacteria living on tobacco leaf surfaces via a culture-dependent method and 16S rRNA gene sequencing. Our results indicated that approximately 7.9%-11.7% of the culturable leaf-associated bacteria have the ability to produce AHL based on the assays using whole-cell biosensors. Sequencing of the 16S rRNA gene assigned the AHL-producing strains to two phylogenetic groups, with Gammaproteobacteria (93%) as the predominant group, followed by Alphaproteobacteria. All of the AHL-producing Alphaproteobacteria were affiliated with the genus Rhizobium, whereas the AHL-producing bacteria belonging to the Gammaproteobacteria mainly fell within the genera Pseudomonas, Acinetobacter, Citrobacter, Enterobacter, Pantoea and Serratia. The bioassays of supernatant extracts revealed that a portion of the strains have a remarkable AHL profile for AHL induction activity using the two different biosensors, and one compound in the active extract of a representative isolate, NTL223, corresponded to 3-oxo-hexanoyl-homoserine lactone. A large population size and diversity of bacteria capable of AHL-driven QS were found to cohabit on leaves, implying that cross-communication based AHL-type QS may be common in the phyllosphere. | 9 | In situ enhanced bioremediation of dichlorvos by a phyllosphere Flavobacterium strain显示文摘A bacterium capable of degrading dichlorvos was isolated from the rape phyllosphere and designated YD4. The strain was identified as Flavobacterium sp., based on its phenotypic features and 16S rRNA gene sequence. Strain YD4 was able to utilize dichlorvos as the sole source of phosphorus. In situ enhanced bioremedia- tion of dichlorvos by YD4 was hereafter studied. Chlorpyrifos and phoxim could also be degraded by this strain as the sole phosphorus source. A higher degradation rate of dichlorvos was observed after spraying YD4 onto the surface of rape leaves when compared to the sterilized- YD4 and water-treated samples. | 10 | Effects of tide-surge interactions on storm surges along the coast of the Bohai Sea, Yellow Sea, and East China Sea显示文摘A two-dimensional coupled tide-surge model was used to investigate the effects of tide-surge interactions on storm surges along the coast of the Bohai Sea, Yellow Sea, and East China Sea. In order to estimate the impacts of tide-surge interactions on storm surge elevations, Typhoon 7203 was assumed to arrive at 12 different times, with all other conditions remaining constant. This allowed simulation of tide and total water levels for 12 separate cases. Numerical simulation results for Yingkou, Huludao, Shijiusuo, and Lianyungang tidal stations were analyzed. Model results showed wide variations in storm surge elevations across the 12 cases. The largest difference between 12 extreme storm surge elevation values was of up to 58 cm and occurred at Yingkou tidal station. The results indicate that the effects of tide-surge interactions on storm surge elevations are very significant. | 12 | Three-dimensional numerical simulation of M_2 internal tides in the Luzon Strait显示文摘A three-dimensional isopycnic-coordinate internal tidal model is employed to investigate the generation,propagation, vertical structure and energy conversion of M2 internal tides in the Luzon Strait(LS) with mooring observations. Simulated results, especially the tidal current amplitudes, agree well with observations,demonstrating the reasonability and accuracy of the model. Results indicate that M2 internal tides mainly propagate into three directions horizontally, i.e., eastward towards the western Pacific Ocean, westward towards the Dongsha Island and southwestward towards the South China Sea Basin. In the horizontal direction, tidal current amplitudes decrease as distance increases away from the LS; in the vertical direction, they show an obvious decreasing tendency with depth. Between the double ridges of the LS, a clockwise gyre of M2 baroclinic energy flux appears, which is caused by reflections of M2 internal tides at supercritical topographies, and resonance of M2 internal tides happens along 19.5° and 21.5°N due to the heights and separation distance of the double ridges. | 16 | Construction of a dual fluorescence whole-cell biosensor to detect N-acyl homoserine lactones显示文摘Detection of N-acyl homoserine lactones(AHLs) is useful for understanding quorum sensing(QS) behaviors, including biofilm formation, virulence and metabolism. For detecting AHLs and indicating the host cells in situ, we constructed the plasmid pUCGMA2T1-4 to make a dual fluorescent wholecell biosensor based on the AhlI/R AHL system of Pseudomonas syringae pv. syringae B728a. The plasmid contains three components: constitutively expressed PnptII::gfp for indicating host cells, PahlI::mcherry that produces red fluorescence in response to AHL, and the ahlR gene that encodes an AHL regulatory protein. Meanwhile, two copies of T1-4(four tandem copies of a transcriptional terminator) were added into the plasmid to reduce background. The results showed that when the plasmid was placed into Escherichia coli, the dual fluorescence whole-cell biosensor was able to respond with red fluorescence within 6 hr to 5 × 10-8–1 × 10-5mol/L of 3OC6-HSL. Bright green fluorescence indicated the host cells. Furthermore, when the plasmid was transferred to wildtype Pseudomonas PhTA125(an AHL-producing bacterium), it also showed both green and red fluorescence.
