| 1 | Genome size and sequence composition of moso bamboo: A comparative study显示文摘Moso bamboo (Phyllostachys pubescens) is one of the world's most important bamboo species. It has the largest area of all planted bamboo―over two-thirds of the total bamboo forest area―and the highest economic value in China. Moso bamboo is a tetraploid (4x=48) and a special member of the grasses family. Although several genomes have been sequenced or are being sequenced in the grasses family, we know little about the genome of the bambusoids (bamboos). In this study, the moso bamboo genome size was estimated to be about 2034 Mb by flow cytometry (FCM), using maize (cv. B73) and rice (cv. Nipponbare) as internal references. The rice genome has been sequenced and the maize genome is being sequenced. We found that the size of the moso bamboo genome was similar to that of maize but significantly larger than that of rice. To determine whether the bamboo genome had a high proportion of repeat elements, similar to that of the maize genome, approximately 1000 genome survey sequences (GSS) were generated. Sequence analysis showed that the proportion of repeat elements was 23.3% for the bamboo genome, which is significantly lower than that of the maize ge-nome (65.7%). The bamboo repeat elements were mainly Gypsy/DIRS1 and Ty1/Copia LTR retrotrans-posons (14.7%), with a few DNA transposons. However, more genomic sequences are needed to con-firm the above results due to several factors, such as the limitation of our GSS data. This study is the first to investigate sequence composition of the bamboo genome. | 2 | Differential expression of microRNAs in dorsal root ganglia after sciatic nerve injury显示文摘This study investigated the possible involvement of microRNAs in the regulation of genes that participate in peripheral neural regeneration.A microRNA microarray analysis was conducted and 23 microRNAs were identified whose expression was significantly changed in rat dorsal root ganglia after sciatic nerve transection.The expression of one of the downregulated microRNAs,microRNA-214,was validated using quantitative reverse transcriptase-PCR.MicroRNA-214 was predicted to target the 3′-untranslated region of Slit-Robo GTPase-activating protein 3.In situ hybridization verified that microRNA-214 was located in the cytoplasm of dorsal root ganglia primary neurons and was downregulated following sciatic nerve transection.Moreover,a combination of in situ hybridization and immunohistochemistry revealed that microRNA-214 and Slit-Robo GTPase-activating protein 3 were co-localized in dorsal root ganglion primary neurons.Western blot analysis suggested that Slit-Robo GTPase-activating protein 3 was upregulated in dorsal root ganglion neurons after sciatic nerve transection.These data demonstrate that microRNA-214 is located and differentially expressed in dorsal root ganglion primary neurons and may participate in regulating the gene expression of Slit-Robo GTPase-activating protein 3 after sciatic nerve transection. | Anjie Lu Zufa Huang Chaoyue Zhang Xianfang Zhang Jiuhong Zhao Haiying Zhang Quanpeng Zhang Song Wu Xinan Yi | 2014 | Neural Regeneration Research2014,9,10: | 5 |
| 3 | Phase Ⅱ study of apatinib in combination with oral vinorelbine in heavily pretreated HER2-negative metastatic breast cancer and clinical implications of monitoring ctDNA显示文摘Objective:Apatinib is an oral TKI targeting VEGFR-2.Single-agent apatinib treatment has been shown to produce an objective response in patients with pretreated m BC.Oral vinorelbine also holds promise as a treatment of choice in patients with m BC.This study aimed to investigate the efficacy and safety of the oral vinorelbine-apatinib combination in patients with pretreated m BC.In addition,we detected gene variants in ct DNA to explore the therapeutic implications.Methods:This study enrolled patients with HER2-negative m BC who were pretreated with anthracycline/taxanes.Patients were treated with apatinib at 500 mg/425 mg daily plus oral vinorelbine 60 mg/m2 on days 1,8,and 15 of every cycle(3 weeks).The primary endpoint was PFS.The secondary endpoints were ORR,CBR,OS,and safety.Patients eligible for ct DNA detection were evaluated before and during treatment.Results:Forty patients were enrolled.The median PFS was 5.2 months(95%CI,3.4–7.0 months),and the median OS was 17.4 months(95%CI,8.0–27.0 months).The ORR was 17.1%(6/35),and the CBR was 45.7%(16/35).The most common AEs included gastrointestinal reaction,myelosuppression,and hypertension.In 20 patients,ct DNA was detected at baseline and during treatment.A significant difference was found in PFS for undetected vs.detected baseline ct DNA(13.9 months vs.3.6 months,P=0.018).Conclusions:All-oral therapy with apatinib plus vinorelbine displayed objective efficacy in patients with heavily pretreated HER2-negative m BC,with acceptable and manageable toxicity profiles.Patients with no gene variant detected and lower variant allele frequencies in ct DNA at baseline showed longer PFS. | Anjie Zhu Peng Yuan Nanlin Hu Mingzhou Li Wenmiao Wang Xue Wang Jian Yue Jiayu Wang Yang Luo Fei Ma Pin Zhang Qing Li Binghe Xu Shanbo Cao Giuseppe Lippi Yoichi Naito Mohammed A.Osman Gustavo N.Marta Gianluca Franceschini Armando Orlandi | 2021 | Cancer Biology & Medicine2021,18,3: | 4 |
| 4 | Perspective on ultramicroporous carbon as sulphur host for Li-S batteries显示文摘Lithium-sulphur(Li-S)batteries are currently considered as next-generation battery technology.Sulphur is an attractive positive electrode for lithium metal batteries,mainly due to its high capacity(1675 m Ah g^(-1))and high specific energy(2600 Wh kg^(-1)).The electrochemical reaction of lithium with sulphur in non-aqueous electrolytes results in the formation of electrolyte soluble intermediate lithium-polysulphides.The dissolved polysulphides shuttle to the anode and get reduced at the anode resulting in Li metal corrosion.The solubility of polysulphide gradually reduces the amount of sulphur in the cathode,thereby limiting the cycle life of Li-S batteries.Several strategies have been proposed to improve the cycling stability of Li-S batteries.A unique approach to eliminate the polysulphide shuttle is to use ultramicroporous carbon(UMC)as a host for sulphur.The pore size of UMC which is below 7A,is the bottleneck for carbonate solvents to access sulphur/polysulphides confined in the pores,thereby preventing the polysulphide dissolution.This perspective article will emphasise the role of UMC host in directing the lithiation mechanism of sulphur and in inhibiting polysulphide dissolution,including the resulting parasitic reaction on the lithium anode.Further,the challenges that need to be addressed by UMC-S based Li-S batteries,and the strategies to realise high power density,high Coulombic efficiency,and resilient Li-S batteries will be discussed. | Helen Maria Joseph Maximilian Fichtner Anji Reddy Munnangi | 2021 | Journal of Energy Chemistry2021,30,8: | 2 |
| 5 | PREPARATION AND CHARACTERIZATION OF PVA BASED SOLID POLYMER ELECTROLYTES FOR ELECTROCHEMICAL CELL APPLICATIONS显示文摘Solid polymer electrolyte films containing poly(vinyl alcohol) (PVA) and magnesium nitrate (Mg(NO3)2) were prepared by solution casting technique and characterized by using XRD, FTIR, DSC and AC impedance spectroscopic analysis. The amorphous nature of the polymer electrolyte films has been confirmed by XRD. The complex formation between PVA and Mg salt has been confirmed by FTIR. The glass transition temperature decreases with increasing the Mg salt concentration. The AC impedance studies are performed to evaluate the ionic conductivity of the polymer electrolyte films in the range of 303 383 K, and the temperature dependence seems to obey the Arrhenius behavior. Transport number measurements show that the charge transport is mainly due to ions. Electrochemical cell of configuration Mg/(PVA + Mg(NO3)2) (70:30)/(I2 + C + electrolyte) has been fabricated. | 6 | Carboxymethytl Pachymaram Up-Regulates Dendritic Cell's Function in Hepatitis B Virus Transgenic Mice in vitro显示文摘The effect of carboxymethytl pachymaram ( CMP ) on the function of dendritic cells(DCs) derived from spleens of hepatitis B virus transgenic mice are studied in vitro. The phenotypes of DCs are tested by flow cytometry (FCM), cytokines measured by ELISA. The expression of DCs' phenotypes in HBV transgenic mice are low (CD80^+CD11c^+:59.12±11.53 vs 9.60±4.53, p<0.01; CD80+ MHC-Ⅱ^+: 44.86±12.31 vs 9.80±5.72, p<0.01, normal mice vs HBV transgenic mice), the ability of DCs stimulating T lymphocytes proliferation decreases (0.37±0.11 vs 0.20±0.11, p<0.05, normal mice vs HBV transgenic mice), levels of IL-12 and IFN-γ decrease whereas the level of IL-10 increases; CMP can enhance DCs' ability of stimulating T lymphocytes proliferation, facilitate the secretion of IL-12 and IFN-γ, inhibit the secretion of IL-10, thus up regulates DCs function.
