| 2 | Alleviated mucosal and neuronal damage in a rat model of Crohn's disease显示文摘AIM:To establish a rat model suitable to investigate the repetitive relapsing inflammations(RRI)characteristic to Crohn’s disease.METHODS:Colitis was induced by 2,4,6-trinitrobenzenesulfonic acid(TNBS).RRI were mimicked by repeating administrations of TNBS.Tissue samples were taken from control,once,twice and three times treated rats from the inflamed and adjacent non-inflamed colonic segments at different timepoints during the acute intestinal inflammation.The means of the ulcerated area were measured to evaluate the macroscopic mu-cosal damage.The density of myenteric neurons was determined on whole mounts by Hu C/Hu D immunohistochemistry.Heme oxygenase-1(HO-1)expression was evaluated by molecular biological techniques.RESULTS:TNBS-treated rats displayed severe colitis,but the mortality was negligible,and an increase of body weight was characteristic throughout the experimental period.The widespread loss of myenteric neurons,and marked but transient HO-1 up-regulation were demonstrated after the first TNBS administration.After repeated doses the length of the recovery time and extent of the ulcerous colonic segments were markedly decreased,and the neuronal loss was on a smaller scale and was limited to the inflamed area.HO-1 m RNA level was notably greater than after a single dose and overexpression was sustained throughout the timepoints examined.Nevertheless,the HO-1protein up-regulation after the second TNBS treatment proved to be transient.Following the third treatment HO-1 protein expression could not be detected.CONCLUSION:Experimentally provoked RRI may exert a protective preconditioning effect against the mucosal and neuronal damage.The persistent up-regulation of HO-1 m RNA expression may correlate with this. | Petra Talapka Lajos István Nagy Alexandra Pál Marietta Zita Poles Anikó Berkó Mária Bagyánszki László Géza Puskás éva Fekete Nikolett Bódi | 2014 | World Journal of Gastroenterology2014,20,44: | 3 |
| 3 | Direct synthesis of La-Mg-Ni-Co type hydrogen storage alloys from oxide mixtures显示文摘(La_(1-x)Mg_x)_2(Ni_(0.8)Co_(0.2))_7(x = 0.125, 0.25, 0.5) alloys were synthesized from the sintered mixture of La_2O_3+ Ni O + Co O + Mg O in the molten CaCl_2 electrolyte at 750 °C and the electrochemical hydrogen storage capacities of the synthesized alloys were measured. Non-hygroscopic LaNiO_3 phase formed during sintering(at 1200 °C for 2 h) as a result of the reaction of hygroscopic La_2O_3 with NiO. Another sinter product was Mg_(0.4)Ni_(0.6)O phase. Both mixed oxide sinter products facilitated the La-Ni and Mg-Ni phase formations. X-ray diffraction peaks indicated that the first stable phase appeared in the alloy structure was LaNi_5 which formed upon reduction of La_2NiO_4 phase. Increase in Mg content caused formation of La_(1.5)Mg_(0.5)Ni_7 phase in the alloy structure and the presence of this phase improved the hydrogen storage performance of the electrodes. | 4 | Structural and molecular features of intestinal strictures in rats with Crohn's-like disease显示文摘AIM: To develop a new rat model we wanted to gain a better understanding of stricture formation in Crohn's disease(CD).METHODS: Chronic colitis was induced locally by the administration of 2,4,6-trinitrobenzenesulfonic acid(TNBS). The relapsing inflammation characteristic to CD was mimicked by repeated TNBS treatments. Animals were randomly divided into control, once, twice and three times TNBS-treated groups. Control animals received an enema of saline. Tissue samples were taken from the strictured colonic segments and also adjacent proximally and distally to its 60, 90 or 120 d after the last TNBS or saline administrations. The frequency and macroscopic extent of the strictures were measured on digital photographs. The structural features of strictured gut wall were studied by light- and electron microscopy. Inflammation related alterations in TGF-beta 2 and 3, matrix metalloproteinases 9(MMP9) and TIMP1 m RNA and protein expression were determined by quantitative real-time PCR and western blot analysis. The quantitative distribution of caspase 9 was determined by post-embedding immunohistochemistry.RESULTS: Intestinal strictures first appeared 60 d after TNBS treatments and the frequency of them increased up to day 120. From day 90 an intact lamina epithelialis, reversible thickening of lamina muscularis mucosae and irreversible thickening of the muscularis externa were demonstrated in the strictured colonic segments. Nevertheless the morphological signs of apoptosis were frequently seen and excess extracellular matrix deposition was recorded between smooth muscle cells(SMCs). Enhanced caspase 9 expression on day 90 in the SMCs and on day 120 also in myenteric neurons indicated the induction of apoptosis. The m RNA expression profile of TGF-betas after repeated TNBS doses was characteristic to CD, TGF-beta 2, but not TGF-beta 3 was up-regulated. Overexpression of MMP9 and down-regulation of TIMP1 were demonstrated. | 5 | Rare presentation of primary (AL) amyloidosis as gastrointestinal hemorrhage without systemic involvement显示文摘We are reporting a rare case of a patient with primary(AL) amyloidosis presenting with an acute non-varicealupper gastrointestinal hemorrhage in the absence ofother systemic involvement. The case report involves a58-year-old woman with significant cardiac history andhereditary blood disorder who came in complaining ofabdominal pain and coffee-ground emesis for two days.Computed tomography(CT) scan of the abdomen andpelvis with contrast revealed segmental wall thickeningof the proximal jejunum with hyperdense, heterog-enous luminal content. Similar findings were evident inthe left lower small bowel region, suspicious for smallbowel hematoma and the possibility of intraluminalclots. Esophagogastroduodenoscopy performed postresuscitation showed punctate, erythematous lesionsthroughout the stomach as well as regions of smallbowel mucosa that appeared scalloped, ulcerated, andhemorrhaged on contact. Despite initial treatment for immunostain-positive focal cytomegalovirus gastritis, follow-up esophagogastroduodenoscopy after two months continued to demonstrate friable and irregular duodenal mucosa hinting at a different underlying etiol-ogy. Pathology reports from analyses of biopsy samples highlighted infiltration and expansion of the lamina pro-pria and submucosa. Subsequent staining with congo red/crystal violet and appropriate subtyping established the diagnosis of AL(kappa)-type amyloidosis. The sig-nificance of this case lies in the fact that our patient did not have the typically seen diagnostic systemic involvements-namely of heart and kidneys-usually seen in primary(AL) amyloidosis patients. | 6 | Discrete twinning dynamics and size-dependent dislocation-to twin transition in body-centred cubic tungsten显示文摘Body-centred cubic(BCC) metals are known to have unstable intrinsic stacking faults and high resistance to deformation twinning, which can strongly influence their twinning behaviour. Though twinning mechanisms of BCC metals have been investigated for more than 60 years, the atomistic level dynamics of twinning remains under debate, especially regarding its impact on competition between twinning and slip. Here, we investigate the atomistic level dynamics of twinning in BCC tungsten(W) nanowires using in situ nanomechanical testing. Quantitative experimental studies directly visualize that deformation twins in W nanowires have a minimum size of six-layers and grow in increments of approximately three-layers at a time, in contrast to the layer-by-layer growth of deformation twins in face-centred cubic metals. These unique twinning dynamics induces a strong competition with ordinary dislocation slip,as exhibited by a size-dependent dislocation-to-twin transition in W nanowires, with a transition size of ~40 nm. | 7 | Status and ethno-medicinal usage of invasive plants in traditional health care practices: a case study from northeastern Bangladesh显示文摘Species diversity and ethno-medicinal usage of 39 species of invasive plants were explored in and around two protected areas, Khadimnagar National Park (KNP) and Rema-Kalenga Wildlife Sanctu- ary (RKWS), of northeastern Bangladesh,. Status of invasive plants were investigated in 60 sample plots with 5 different habitat types, including forest, roadside, homestead, fallow land and others (ponds, canals, water logged areas, agricultural land, etc.). Data about the usage of invasive plants in traditional health care were collected through interviewing 110 households. Among the five habitat types, fallow land (28 species) pos- sessed the highest number of species, followed by roadside (25 species), forest (23 species), homestead (22 species) and others (13 species). Based on the survey, invasive plants of study areas were also categorized into five degrees of invasiveness e.g., naturalized, introduced, possibly invasive, moderately invasive and highly invasive. Additionally, there is the linear trend between degrees of invasiveness and use percentage of invasive plants. Total 39 species of invasive plants belonging to 29 fami- lies were recorded, which are generally used to treat 37 diseases, ranging from simple headache to highly complicated eye and heart diseases. The majority of the species used by the local inhabitants were herbs (16 spe- cies), followed by some shrubs (11 species), climbers (5 species), trees (5 species) and grasses (2 species).
