| 1 | Characterization of codon usage bias in the dUTPase gene of duck enteritis virus显示文摘A comparative analysis of the codon usage bias in the newly discovered dUTPase gene (Assigned Accession No.:DQ486149) of the duck enteritis virus (DEV) and the dUTPase gene of 32 reference herpesviruses was performed.The results indicated that the DEV dUT-Pase gene encodes a protein of 477 amino acids, which includes five conserved motifs with a 3-1-2-4-5 arrangement.The codon adap-tation index (CAI), efiective number of codons (ENC), and GC3S values indicated synonymous codon usage bias in the dUTPase gene of herpesviruses, and this synonymous bias was correlated with host evolution.The codon usage patterns of the DEV dUTPase gene were phylogenetically conserved and similar to that of the dUTPase genes of the avian alphaherpesvirus.Although codon usage in each micro-organism was difierent, there were no strain-specific difierences among them.Sixty-one codons in the predicted polypeptide, with a strong bias towards A and T at the third codon position, were used.Comparison of the codon usage in the dUTPase gene of difierent organisms revealed that there were 19 codons showing distinct codon usage difierences between the DEV and Escherichia coli dUTPase genes;16 between the DEV and yeast dUTPase genes;and 15 between the DEV and human dUTPase genes.Analysis of variance (ANOVA) showed significant difierences between the DEV and yeast dUTPase genes (r = 0.536, P < 0.01).The extent of codon usage bias in the DEV dUTPase gene was highly correlated with the gene expression level, therefore the results may provide useful information for gene classification and functional studies. | Lichan Zhao Anchun Cheng Mingshu Wang Guiping Yuan Mingsheng Cai | 2008 | Progress in Natural Science:Materials International2008,18,9: | 4 |
| 2 | Host cell apoptosis induced by infection with duck swollen head hemorrhagic disease virus显示文摘This study aimed to examine the host cell apoptosis in the tissues of Peking ducks infected with duck swollen head hemorrhagic disease virus(DSHDV).The dynamic changes associated with apoptosis occurring in the internal tissues were evaluated at different time points postinoculation(PI) by performing hematoxylin and eosin(HE) staining,followed by light microscopy,terminal deoxynucleotidyl transferase dUTP nick-end labeling(TUNEL) assay,and transmission electron microscopy(TEM).The results showed that DSHDV infection could induce apoptosis in host cells,including those of the bursa of Fabricius(BF),thymus,spleen,liver,intestinal tract,kidney,and esophagus.The apoptotic index(AI) values increased with time from 2 to 72 h PI,and the highest values were recorded at 72 h PI.Further,cell death due to classic necrosis was observed in the dying or deceased ducks after 72 h PI.In conclusion,host cell apoptosis can be induced by DSHDV and may play an important role in the pathogenesis of duck viral swollen head hemorrhagic disease(DVSHD). | Chuanfeng Li Anchun Cheng Mingshu Wang Chanjuan Shen Na Zhang Yi Zhou Dekang Zhu Qihui Luo Renyong Jia Xiaoyue Chen | 2009 | Progress in Natural Science:Materials International2009,19,1: | 3 |

Results showed that nerve growth factor expression decreased, but inducible nitric oxide synthase expression increased, in the mesencephalon and diencephalon, as well as visual- and auditory- related nervous tissues. | 4 | Dynamic changes of apoptosis in duck embryo fibroblasts induced by new type Gosling viral enteritis virus显示文摘The monolayer duck embryo fibroblast (DEF) cells were experimentally infected with new type Gosling viral enteritis virus (NGVEV) and the dynamic changes of apoptosis were detected at different time points after NGVEV infection by transmission electron microscopy (TEM),DNA agarose gel electrophoresis and Annexin V-FITC/PI stained fluorescence-activated cell sorter (FACS). The result shows that NGVEV can induce infected cells undergoing apoptosis and changing regularly. A series of characteristic apoptotic morphological changes including shrinkage of the cells,chromatin condensation and margination,as well as formation of apoptotic bodies,were observed by TEM. The typical ladder pattern of DNA fragmentation was demonstrated by agarose gel electrophoresis. Soybean isoflavone, a type of non-steroid estrogen withsimilar molecular structure and function to estradiol, exhibits estrogen-like characteristics.OBJECTIVE: To investigate the effects of various doses of soybean isoflavone on expression ofERα, NGF, IL-2, and AR in the cerebellum of ovariectomized rat, and to determine whether there is adose-dependent effect.DESIGN, TIME AND SETTING: Controlled trial at the cellular and molecular level. The study wasperformed at the Experimental Animal Engineering Center, College of Veterinary Medicine, SichuanAgricultural University from July 2006 to May 2008.MATERIALS: Soybean isoflavone, comprised of daidzin, genistein and isoflavone, was provided byTaiyuan Yuantai Biochemical Industry, China. The ERα, NGF, IL-2, and AR in situ hybridization kit,rabbit anti-rat ERα, NGF, IL-2, and AR monoclonal antibodies, and SABC kit were purchased fromWuhan Boster Biological Technology, China.METHODS: A total of 50 female, Sprague Dawley rats, aged 3 months, were randomly assigned to5 groups, with 10 animals in each group. With the exception of the sham-operation group (abdominalcavity opening alone), all rats underwent bilateral ovariectomy. At 14 days after surgery, rats in thehigh-, middle-, and low-dose soybean isoflavone groups were subcutaneously injected with 1.5, 1.0,and 0.5 mg/kg soybean isoflavone, respectively, every 2 days for 6 consecutive weeks. Rats in thesham-operation and ovariectomized groups were subcutaneously injected with absolute alcohol(0.5 mL/kg).MAIN OUTCOME MEASURES: Expression levels and distribution of ERα, NGF, IL-2, and AR in thecerebellum were detected by immunohistochemistry and in situ hybridization.RESULTS: Compared with the sham-operation group, immunoreactive products and hybridizationsignals of ERα, NGF, IL-2, and AR were significantly decreased in the cerebellar cortex and nuclei ofovariectomized rats (P< 0.05 or P< 0.01), but increased following soybean isoflavone treatment. Inparticular, levels of the high-dose soybean isoflavone group were almost restored to levels of thesham-operation group (P > 0.05). The immunoreactive products were primarily located in thecytoplasm and neurites, and rarely in the cell membrane and nuclei. However, the hybridizationsignals were predominantly located in the nuclei, but rarely in the cytoplasm, cell membrane, orneurites.CONCLUSION: Soybean isofiavone upregulated ERα, NGF, IL-2, and AR protein and geneexpression in a dose-dependent manner, and played an important role in sustaining and protectingstructure and function of cerebellar neurons.
