1Underexpression of LATS1 TSG in colorectal cancer is associated with promoter hypermethylation显示文摘AIM:To investigate large tumor suppressor 1 (LATS1 ) expression, promoter hypermethylation, and microsatellite instability in colorectal cancer (CRC).METHODS:RNA was isolated from tumor tissue of 142 CRC patients and 40 colon mucosal biopsies of healthy controls. After reverse transcription, quantitative polymerase chain reaction (PCR) was performed, and LATS1 expression was normalized to expression of the ACTB and RPL32 housekeeping genes. To analyze hypermethylation, genomic DNA was isolated from 44 tumor CRC biopsies, and methylation-specific PCR was performed. Microsatellite instability (MSI) status was checked with PCR using BAT26, BAT25, and BAT40 markers in the genomic DNA of 84 CRC patients, followed by denaturing gel electrophoresis. RESULTS:Decreased LATS1 expression was found in 127/142 (89.4%) CRC cases with the average ratio of the LATS1 level 10.33 ± 32.64 in CRC patients vs 32.85 ± 33.56 in healthy controls. The lowest expression was found in Dukes' B stage tumors and G1 (welldifferentiated) cells. Hypermethylation of the LATS1 promoter was present in 25/44 (57%) CRC cases analyzed. LATS1 promoter hypermethylation was strongly associated with decreased gene expression; methylated cases showed 162× lower expression of LATS1 than unmethylated cases. Although high-grade MSI (mutation in all three markers) was found in 14/84 (17%) cases and low-grade MSI (mutation in 1-2 markers) was found in 30/84 (36%) cases, we found no association with LATS1 expression. CONCLUSION:Decreased expression of LATS1 in CRC was associated with promoter hypermethylation, but not MSI status. 2Decreased serum essential and aromatic amino acids in patients with chronic pancreatitis显示文摘AIM:To investigate the influence of chronic pancreatitis(CP) on serum concentrations of amino acids.METHODS:Thirty-five male patients with alcoholic CP and 21 healthy male subjects were examined.Serum concentrations of amino acids were assayed by ionpair high-performance liquid chromatography with mass detection.RESULTS:Serum glutamate concentration was increased in CP patients as compared to controls.In contrast,serum concentrations of glutamine,histidine,tyrosine,proline,tryptophan and threonine were significantly decreased in CP patients.A trend towards decreasing concentrations of serum lysine,alanine,methionine and valine as well as for total serum amino acids was observed.The sum of aromatic and the sum of essential amino acid concentrations were significantly lower in CP patients than in controls.CONCLUSION:CP leads to decreased serum concentrations of several amino acids,such as essential and aromatic serum amino acids,most likely due to decreased exocrine function.Krystian Adrych Marian Smoczynski Magdalena Stojek Tomasz Sledzinski Ewa Slominska Elzbieta Goyke Ryszard Tomasz Smolenski Julian Swierczynski 2010World Journal of Gastroenterology2010,16,35:3
3Decreased serum platelet derived growth factor BB levels in acute and increased in chronic pancreatitis显示文摘AIM: To examine circulating growth factor concentrations in patients with acute pancreatitis(AP) and chronic pancreatitis(CP), and walled-off pancreatic necrosis(WOPN). METHODS: Forty patients with mild AP, 40 patients with alcoholic CP, 33 patients with WOPN and 40 healthy subjects were examined. Serum concentrations of platelet derived growth factor BB(PDGF-BB), transforming growth factor β-1(TGFβ-1), chemerin and high-mobility group box chromosomal protein 1(HMBG1) were assayed by enzyme linked immunosorbent assay.RESULTS: Patients with mild AP and those with WOPN had significantly lower serum levels of PDGF-BB compared to healthy subjects(4.0 ± 0.61 ng/mL vs 6.2 ± 0.76 ng/mL, P = 0.027, and 1.60 ± 0.31 ng/mL vs 6.2 ± 0.76 ng/mL, P < 0.001, respectively), while CP was associated with higher serum levels of PDGF-BB(12 ± 1.3 ng/mL vs 6.2 ± 0.76 ng/mL, P < 0.001). Circulating TGFβ-1 and chemerin levels were elevated in CP patients(57 ± 3.6 ng/mL vs 39 ± 3.6 ng/mL, P < 0.001 and 73 ± 7.2 ng/mL vs 48 ± 2.3 ng/mL, P < 0.001, respectively), but not in patients with AP and WOPN. No significant changes in serum HMBG1 levels were found either in patients with AP, WOPN or CP. CONCLUSION: The serum levels of some growth factors and cytokines differ significantly in AP, WOPN and CP. 12常规结肠镜检中显微镜下结肠炎的发生率显示文摘Lymphocytic colitis(LC)and collagenous colitis(CC),both known as microscopic colitis(MC),are uncommon entities with increasing incidence as more clinicians take biopsies from macroscopically normal colons and as pathologists use more rigorous diagnostic criteria to be confident of the diagnosis.Information on the incidence of this type of colitis is limited and based on reported cases.The purpose of this work is to estimate the incidence of LC and CC in reviewed routine colonoscopies.We reviewed 2815 colonoscopies performed at a tertiary referral center with an open-access service using restricted histological criteria in order to establish the frequency rate of LC and CC in routine colonoscopic biopsymaterial.Cases suspicious for MC were stained with Masson’s trichrome or Congo red stain and immunohistochemically for lymphocytes,where appropriate.Review of routine colonoscopic biopsies showed that MC is underreported in our colonoscopic material.Incidence rates of LC and CC(0.9 and 0.4,respectively)were based on morphological assessment of colonoscopic biopsies using stringent criteria together with clinical data and after differentiation with other lesions which can mimic MC.The 10.2%rate of this type of colitis in patients with chronic watery diarrhea indicates the necessity to consider these lesions in older individuals with diarrhea and normal endoscopical colonic mucosa.Jaskiewicz K.
