1Repurposing metformin for the treatment of gastrointestinal cancer显示文摘Diabetes mellitus type 2 and cancer share many risk factors.The pleiotropic insulin-dependent and insulin-independent effects of metformin might inhibit pathways that are frequently amplified in neoplastic tissue.Particularly,modulation of inflammation,metabolism,and cell cycle arrest are potential therapeutic cancer targets utilized by metformin to boost the anti-cancer effects of chemotherapy.Studies in vitro and in vivo models have demonstrated the potential of metformin as a chemo-and radiosensitizer,besides its chemopreventive and direct therapeutic activity in digestive system(DS)tumors.Hence,these aspects have been considered in many cancer clinical trials.Case-control and cohort studies and associated meta-analyses have evaluated DS cancer risk and metformin usage,especially in colorectal cancer,pancreatic cancer,and hepatocellular carcinoma.Most clinical studies have demonstrated the protective role of metformin in the risk for DS cancers and survival rates.On the other hand,the ability of metformin to enhance the actions of chemotherapy for gastric and biliary cancers is yet to be investigated.This article reviews the current findings on the anti-cancer mechanisms of metformin and its apparatus from pre-clinical and ongoing studies in DS malignancies.Ademar Dantas Cunha Júnior Arinilda Campos Bragagnoli Felipe Osório Costa JoséBarreto Campello Carvalheira 2021World Journal of Gastroenterology2021,27,17:5
2A sensitive electrochemical detection of metronidazole in synthetic serum and urine samples using low-cost screen-printed electrodes modified with reduced graphene oxide and C60显示文摘Monitoring the concentration of antibiotics in body fluids is essential to optimizing the therapy and minimizing the risk of bacteria resistance,which can be made with electrochemical sensors tailored with appropriate materials.In this paper,we report on sensors made with screen-printed electrodes(SPE)coated with fullerene(C60),reduced graphene oxide(rGO)and Nafion(NF)(C60-rGO-NF/SPE)to determine the antibiotic metronidazole(MTZ).Under optimized conditions,the C60-rGO-NF/SPE sensor exhibited a linear response in square wave voltammetry for MTZ concentrations from 2.5×10^(-7) to 34×10^(-6) mol/L,with a detection limit of 2.1×10^(-7) mol/L.This sensor was also capable of detecting MTZ in serum and urine,with recovery between 94%and 100%,which are similar to those of the standard chromatographic method(HPLC-UV).Because the C60-rGO-NF/SPE sensor is amenable to mass production and allows for MTZ determination with simple principles of detection,it fulfills the requirements of therapeutic drug monitoring programs.Elsa Maria Materon Ademar Wong Tayane Aguiar Freitas Ronaldo Censi Faria Osvaldo N.Oliveira Jr. 2021Journal of Pharmaceutical Analysis2021,11,5:1

9Analysis of a New Composite Material for Watercraft Manufacturing显示文摘In this paper, we investigate the properties of an alternative material for use in marine engineering, namely a rigid and light sandwich-structured composite made of expanded polystyrene and fiberglass. Not only does this material have an improved section modulus, but it is also inexpensive, light, easy to manipulate, and commercially available in various sizes. Using a computer program based on the finite element method, we calculated the hogging and sagging stresses and strains acting on a prismatic boat model composed of this material, and determined the minimum sizes and maximum permissible stresses to avoid deformation. 10Unlocking the in vitro anti-Trypanosoma cruzi activity of halophyte plants from the southern Portugal显示文摘Objective:To evaluate the in vitro anti-Trypanosoma cruzi(T.cruzi) activity of organic extracts prepared from halophyte species collected in the southern coast of Portugal(Algarve),and chemically characterize the most active samples.Methods:Acetone,dichloromethane and methanol extracts were prepared from 31 halophyte species and tested in vitro against trypomastigotes and intracellular amastigotes of the Tulahuen strain of T.cruzi.The most active extract was fractionated by preparative HPLC-DAD,affording 11 fractions.The most selective fraction was fully characterized by 1H-NMR.Results:From 94 samples tested,one was active,namely the root dichloromethane extract of Juncus acutus(IC50 < 20 μg/mL).This extract was fractionated by HPLC,affording 11 fractions,one of them containing only a pure compound(juncunol),and tested for anti-parasitic activity.Fraction 8(IC50 = 4.1 μg/mL) was the most active,and was further characterized by 1H-NMR.The major compounds were phenanthrenes,9,10-dihydrophenanthrenes and benzocoumarins.Conclusion:Our results suggest that the compounds identified in fraction 8 are likely responsible for the observed anti parasitic activity.Further research is in progress aiming to isolate and identify the specific active molecules.To the best of our knowledge,this is the first report on the in vitro anti T.cruzi activity of halophyte species.Marta Oliveira Policarpo Ademar Sales Junior Maria Joao Rodrigues Marina DellaGreca Luisa Barreira Silvane Maria Fonseca Murta Alvaro Jose Romanha Luisa Custodio 2016Asian Pacific Journal of Tropical Medicine2016,9,8:1

13一氧化氮合成酶、环氧化酶-2、硝基酪氨酸及血管生成与软骨肉瘤预后的相关性研究显示文摘背景:免疫组织化学标记物有助于肿瘤诊断及预后评估。通过对硝基酪氨酸分析及采用一氧化氮合成酶1(NOS1)、一氧化氮合成酶2(NOS2)、一氧化氮合成酶3(NOS3)检测分析可了解一氧化氮(NO)与肿瘤的相关性。NO与超氧阴离子反应产生的过氧亚硝酸对基因有毒性作用,过氧亚硝酸传递一个硝基基团到酪氨酸的苯环上形成硝基酪氨酸。细胞内硝基酪氨酸的积累说明过氧亚硝酸持续生成。NO刺激所生成的环氧化酶-2(COX-2)与肿瘤内新生血管水平相关,研究表明新生血管影响肿瘤预后,免疫组化标记CD34可表示微血管生成,从而被用于预测肿瘤预后。本研究检测上述标记物在软骨肉瘤中的表达并分析与软骨肉瘤分级和预后的相关性。方法:101例软骨肉瘤患者肿瘤组织样本采用甲醛固定,组织样本采用免疫组织化学染色,指标为:NOS1、NOS2、NOS3、COX-2、硝基酪氨酸和CD34。5例正常软骨样本作为对照。研究分析患者特征、手术变量、肿瘤分级与这些指标的相关性。使用Kaplan—Meier多变量统计学方法分析生存曲线及生存率。结果:硝基酪氨酸、COX-2和CD34与组织学分级有明显相关性(p=0.022,p=0.014,p=0.028),但与总体预后无相关性(p=0.064,p=0.143,p=0.581)。NOS2阳性表达常预示较高的局部复发率(p=0.038)。NOS1、NOS2阳性表达预示较低的生存率(p=0.007,p〈0.001)。通过多变量分析处理,NOS2的表达被证实可作为局部复发率的独立预后判断因素;NOS1、NOS2的表达作为因变量,它们单独或复合阳性表达常与较低的总生存率有关(p=0.046,p=0.004)[危险率,3.17(95%置信区间,1.0-9.8)和5.58(95%置信区间,1.7-18.0)]。Suely Akiko Nakagawa Ademar Lopes Andre Lopes de Carvalho Benedito Mauro Rossi Isabela Werneck da Cunha Fernando Augusto Soares Wu Tu Chung Luciola Assuncao 刘沛宜(译) 张伟滨(译) 2010中华骨科杂志2010,30,12:0

15Poly(Vinyl Alcohol)(PVAl)and Poly(N-2-Vinyl-Pyrrolidone)Hydrogels Nanostructured by Laponite Clay for Drug Delivery显示文摘Hydrogels for wound dressings are usually developed for contact surfaces where mechanical properties are conveniently required.In this sense nanocomposite hydrogels based on PVAl(poly(vinyl alcohol))and PVP(poly(N-2-vinil-pirrolidone))containing 0.5-1.5 wt%of the synthetic laponite RD clay were prepared by a gamma radiation process and compared with similar membranes composed separately of PVP or PVAl.This study aimed to evaluate the effect of clay on the properties and the differences of the polymer blend instead of a unique polymer.The morphology of the hydrogels was evaluated by spectrometric techniques using XRD(X-ray diffraction),SEM(scanning electron microscopy),swelling assay,and FTIR(infrared spectroscopy).The swelling kinetics at 22℃and the mechanical properties by a tensile test comprised the structural properties that were assessed.The results showed PVA/PVP network depends directly on the clay concentration in the nanocomposite hydrogels.The blend PVP/PVAl proved to have potentially efficient mechanical properties for drug delivery in the treatment of wounds.Maria José.A.Oliveira Vinicius J.Santos Lucas Freitas de Freitas Ademar B.Lugão Duclerc F.Parra 2022材料科学与工程(中英文B版)2022,12,3:0
16Chemical speciation of copper and manganese in solution of a copper-contaminated soil and young grapevine growth with amendment application显示文摘Copper(Cu)-based fungicide application to control foliar diseases in grapevine(Vitis vinifera L.)can increase soil Cu availability.Brazilian soils have high natural Cu and manganese(Mn)concentrations,increasing the potential for toxicity to grapevine plants.Application of amendment substances can reduce the concentrations of toxic chemical species of Cu and Mn in soil solution,especially for the soils grown with young plants.We evaluated the chemical speciation of Cu and Mn in soil solution and young grapevine growth using a contaminated soil with amendment application.Grapevine seedlings of Paulsen 1103rootstock were grown for 12 months on an old vineyard(>30 years of cultivation)soil without(control)and with vermicompost or limestone application.Grapevine dry matter,root morphological characteristics,and plant tissue Cu and Mn concentrations were evaluated.The cation and anion concentrations,dissolved organic carbon,p H,and chemical speciation of Cu and Mn in soil solution from different soil layers were analyzed.Vermicompost application did not reduce Cu and Mn phytotoxicity,and grapevine plants died after winter pruning.High soluble Mn concentration in soil(5.56 mg L-1in 0–45 cm soil layer)and Mn concentration in plant tissue at pruning(380.8 mg kg-1)might be the primary cause of plant death.Limestone application reduced Cu2+and Mn2+chemical species in soil solution,thereby increasing plant growth.The Cu2+and Mn2+species were negatively correlated(P<0.05)with grapevine shoot production at pruning(r=-0.92 and-0.97,respectively)and at the end of the trial(r=-0.73 and-0.51,respectively).Limestone application increased grapevine shoot and root dry matter production by 36%and 41%,respectively,while also presenting the highest root length and volume in the 5–10 cm soil layer.Limestone application is a viable and profitable alternative for reducing soil Cu and Mn availability and their phytotoxic effects.Daniela Basso FACCO Edicarla TRENTIN Gerson Laerson DRESCHER Rodrigo Knevitz HAMMERSCHMITT Carlos Alberto CERETTA Leandro Souza DA SILVA Gustavo BRUNETTO Paulo Ademar Avelar FERREIRA 2023Pedosphere2023,33,3:0
