2Meningeal lymphatic vessels regulate brain tumor drainage and immunity显示文摘Recent studies have shown that meningeal lymphatic vessels(MLVs),which are located both dorsally and basally beneath the skull,provide a route for draining macromolecules and trafficking immune cells from the central nervous system(CNS)into cervical lymph nodes(CLNs),and thus represent a potential therapeutic target for treating neurodegenerative and neuroinflammatory diseases.However,the roles of MLVs in brain tumor drainage and immunity remain unexplored.Here we show that dorsal MLVs undergo extensive remodeling in mice with intracranial gliomas or metastatic melanomas.RNA-seq analysis of MLV endothelial cells revealed changes in the gene sets involved in lymphatic remodeling,fluid drainage,as well as inflammatory and immunological responses.Disruption of dorsal MLVs alone impaired intratumor fluid drainage and the dissemination of brain tumor cells to deep CLNs(dCLNs).Notably,the dendritic cell(DC)trafficking from intracranial tumor tissues to dCLNs decreased in mice with defective dorsal MLVs,and increased in mice with enhanced dorsal meningeal lymphangiogenesis.Strikingly,disruption of dorsal MLVs alone,without affecting basal MLVs or nasal LVs,significantly reduced the efficacy of combined anti-PD-1/CTLA-4 checkpoint therapy in striatal tumor models.Furthermore,mice bearing tumors overexpressing VEGF-C displayed a better response to anti-PD-1/CTLA-4 combination therapy,and this was abolished by CCL21/CCR7 blockade,suggesting that VEGF-C potentiates checkpoint therapy via the CCL21/CCR7 pathway.Together,the results of our study not only demonstrate the functional aspects of MLVs as classic lymphatic vasculature,but also highlight that they are essential in generating an efficient immune response against brain tumors.Xueting Hu Qiuping Deng Lu Ma Qingqing Li Yidong Chen Yuhan Liao Fan Zhou Chen Zhang Linlin Shao Jun Feng Tubao He Weihai Ning Yan Kong Yingqing Huo Aibin He Bing Liu Jingjing Zhang Ralf Adams Yulong He Fuchou Tang Xiuwu Bian Jincai Luo 2020Cell Research2020,30,3:11
3MicroRNAs are involved in the development and progression of gastric cancer显示文摘MicroRNAs (miRNAs) are recognized as an essential component of the RNA family, exerting multiple and intricate biological functions, particularly in the process of tumorigenesis, proliferation, and metastatic progression. MiRNAs are altered in gastric cancer (GC), showing activity as both tumor suppressors and oncogenes, although their true roles have not been fully understood. This review will focus upon the recent advances of miRNA studies related to the regulatory mechanisms of gastric tumor cell proliferation, apoptosis, and cell cycle. We hope to provide an in-depth insight into the mechanistic role of miRNAs in GC development and progression. In particular, we summarize the latest studies relevant to miRNAs’ impact upon the epithelial- mesenchymal transition, tumor microenvironment, and chemoresistance in GC cells. 4Simultaneous removal of ammonia and N-nitrosamine precursors from high ammonia water by zeolite and powdered activated carbon显示文摘When adding sufficient chlorine to achieve breakpoint chlorination to source water containing high concentration of ammonia during drinking water treatment, high concentrations of disinfection by-products(DBPs) may form. If N-nitrosamine precursors are present, highly toxic N-nitrosamines, primarily N-nitrosodimethylamine(NDMA), may also form. Removing their precursors before disinfection should be a more effective way to minimize these DBPs formation. In this study, zeolites and activated carbon were examined for ammonia and N-nitrosamine precursor removal when incorporated into drinking water treatment processes.The test results indicate that Mordenite zeolite can remove ammonia and five of seven N-nitrosamine precursors efficiently by single step adsorption test. The practical applicability was evaluated by simulation of typical drinking water treatment processes using six-gang stirring system. The Mordenite zeolite was applied at the steps of lime softening, alum coagulation, and alum coagulation with powdered activated carbon(PAC) sorption. While the lime softening process resulted in poor zeolite performance, alum coagulation did not impact ammonia and N-nitrosamine precursor removal. During alum coagulation, more than67% ammonia and 70%–100% N-nitrosamine precursors were removed by Mordenite zeolite(except 3-(dimethylaminomethyl)indole(DMAI) and 4-dimethylaminoantipyrine(DMAP)). PAC effectively removed DMAI and DMAP when added during alum coagulation. 5Potential predictors for mental stress-induced myocardial ischemia in patients with coronary artery disease显示文摘Background: Mental stress-induced myocardial ischemia (MSIMI) is closely associated with adverse cardiac events in patients with coronary artery disease (CAD) and we aimed to determine whether biomarkers and blood pressure could be potential predictors of MSIMI. Methods: This study enrolled 82 patients with documented CAD between June 1, 2017 and November 9, 2017. Patient blood samples were obtained at resting period and at the end of mental arithmetic. Then, patients were assigned to MSIMI positive group and MSIMI negative group. The main statistical methods included linear regression, receiver operating characteristic (ROC) curves, and logistic regression. Results: Patients with CAD with MSIMI had significantly greater median resting N-terminal pro-brain natriuretic peptide (NT-proBNP, 141.02 [45.85–202.76] pg/mL vs. 57.95 [27.06–117.64] pg/mL;Z =-2.23, P = 0.03) and mean systolic blood pressure (SBP)(145.56 ± 16.87 mmHg vs. 134.92 ± 18.16 mmHg, Z =-2.13, P = 0.04) when compared with those without MSIMI. After 5-min mental stress task, those who developed MSIMI presented higher elevation of median post-stressor high sensitivity cardiac troponin I (hs-cTnI, 0.020 [0.009–0.100] ng/mL vs. 0.009 [0.009–0.010] ng/mL;Z =-2.45, P = 0.01), post-stressor NT-proBNP (138.96 [39.93–201.56] pg/mL vs. 61.55 [25.66–86.50] pg/mL;Z =-2.15, P = 0.03) compared with those without MSIMI. Using the ROC curves, and after the adjustment for basic characteristics, the multiple logistic regression analysis showed that patients presenting a post-stressor hs-cTnI ≥ 0.015 ng/mL had seven-fold increase in the risk of developing MSIMI (odds ratio [OR]: 7.09;95% confidence interval [CI]: 1.65–30.48;P = 0.009), a rest NT-proBNP ≥ 80.51 pg/mL had nearly eight-fold increase (OR: 7.85;95% CI: 1.51–40.82;P = 0.014), a post-stressor NT-proBNP ≥ 98.80 pg/mL had 35-fold increase (OR: 34.96;95% CI: 3.72– 328.50;P = 0.002), a rest SBP ≥ 129.50 mmHg had 11-fold increase (OR: 11.42;95% CI: 1.21–108.17;P = 0.034). 6Differential gene expression in nontransgenic and transgenic“M.26”apple overexpressing a peach CBF gene during the transition from eco-dormancy to bud break显示文摘The CBF signal pathway is responsible for a significant portion of plant responses to low temperature and freezing.Overexpression of CBF genes in model organisms such as Arabidopsis thaliana enhances abiotic stress tolerance but also reduces growth.In addition to these effects,overexpression of the peach(Prunus persica[L.]Batsch)CBF1 gene in transgenic apple(Malus x domestica Borkh.)line T166 also results in early entry into and late exit from dormancy.Although the regulation of dormancy-induction and dormancy-release occur while the CBF regulon is operative in perennial,woody plants,how overexpression of CBF1 affects these dormancy-related changes in gene expression is incompletely understood.The objective of the present study was to characterize global changes in gene expression in peach CBF1-overexpressing and non-transformed apple bark tissues at different states of dormancy via RNA-seq.RNAseq bioinformatics data was confirmed by RT-qPCR on a number of genes.Results indicate that the greatest number of significantly differentially expressed genes(DEGs)occurred in April when dormancy release and bud break normally occur but are delayed in Line T166.Genes involved in storage and inactivation of auxin,GA,and cytokinin were generally upregulated in T166 in April,while those for biosynthesis,uptake or signal transduction were generally downregulated in T166.Genes for cell division and cambial growth were also downregulated in T166 relative to the non-transformed line.These data suggest that overexpression of the peach CBF1 gene impacts growth hormone homeostasis and as a result the activation of growth in the spring,and most likely growth cessation in the fall as well.Timothy Artlip Adam McDermaid Qin Ma Michael Wisniewski 2019Horticulture Research2019,6,1:6
7Metformin and cancer immunity显示文摘The immune system plays an essential and central role in tumor cell differentiation,proliferation,angiogenesis,apoptosis,invasion,and metastasis.Over the past decade,cancer therapy has rapidly evolved from traditional approaches,such as surgery,chemotherapy,and radiotherapy,to revolutionary new treatment options with immunotherapy.This new era of cancer treatment options has now been clinically tested and applied to many forms of human malignancies,often with quite dramatic results.As we develop more effective combinations of cancer treatment,several agents have been recently investigated,putatively identified as antica ncer age nts,or imm uno stimulatory molecules.One such age nt is metformin,originally developed as a fairly standard first-line therapy for patients with type-2 diabetes mellitus(T2DM).Given the underlying mechanisms of action,researchers began to exami ne the alter native function s and possible utility of metformin,finding that the can cer risk in patie nts with T2DM was reduced.It appears that metformin,at least in part,has an antitumor effect through activation of the 5'adenosine monophosphate-activated protein kinase(AMPK)signaling pathway.Moreover,numerous studies have demonstrated that metformin interferes with key immun opathological mecha nisms that are in volved in the pathological processes or associated with maligna nt progression.Such in sights may shed light on further analyzi ng whether metformi n enhances the effectiveness of the immuno therapy and overcomes the immunotherapy resista nee in the patients.Herein,we provide a comprehe nsive review of the literature exami ning the impact of metformin upon the host immune system and cancer immunity.Ruixia Ma Bin Yi Adam I.Riker Yaguang Xi 2020Acta Pharmacologica Sinica2020,41,11:3
8Gastric food retention at endoscopy is associated with severity of liver cirrhosis显示文摘BACKGROUND Gastrointestinal symptoms are prevalent in patients with cirrhosis.Cirrhotic patients have a known predilection to delayed gastric emptying compared to those without cirrhosis.However,the contributing factors have not been fully elucidated.Retained gastric food on esophagogastroduodenoscopy(EGD)has been used as a surrogate marker for delayed gastric emptying with reasonably high specificity.Therefore,we hypothesize that the frequency of retained gastric food contents at EGD will be higher in a cirrhotic population compared to a control population without liver disease.Additionally,we hypothesize that increased frequency of gastric food contents will be associated with increased severity of cirrhosis.AIM To determine the relative frequency of delayed gastric emptying among cirrhotics as compared to non-cirrhotics and to identify associated factors.METHODSWe performed a retrospective case-control study of cirrhotic subjects whounderwent EGD at an academic medical center between 2000 and 2015. Threehundred sixty-four patients with confirmed cirrhosis, who underwent a total of1044 EGDs for the indication of esophageal variceal screening or surveillance,were identified. During the same period, 519 control patients without liverdisease, who underwent a total of 881 EGDs for the indication of anemia, wereidentified. The presence of retained food on EGD was used as a surrogate fordelayed gastric emptying. The relative frequency of delayed gastric emptyingamong cirrhotics was compared to non-cirrhotics. Characteristics of patients withand without retained food on EGD were compared using univariable andmultivariable logistic regression analysis to identify associated factors.RESULTSOverall, 40 (4.5%) patients had evidence of retained food on EGD. Cirrhotics weremore likely to have retained food on EGD than non-cirrhotics (9.1% vs 1.4%, P <0.001). Characteristics associated with retained food on univariable analysisincluded age less than 60 years (12.6% vs 5.2%, P = 0.015), opioid use (P = 0.004),Child-Pugh class C (24.1% Child-Pugh class C vs 6.4% Child-Pugh class A, P =0.007), and lower platelet count (P = 0.027).
