2Animal models for the study of hepatitis B virus infection显示文摘Even with an effective vaccine, an estimated 240 million people are chronically infected with hepatitis B virus(HBV) worldwide. Current antiviral therapies,including interferon and nucleot(s)ide analogues,rarely cure chronic hepatitis B. Animal models are very crucial for understanding the pathogenesis of chronic hepatitis B and developing new therapeutic drugs or strategies. HBV can only infect humans and chimpanzees, with the use of chimpanzees in HBV research strongly restricted. Thus, most advances in HBV research have been gained using mouse models with HBV replication or infection or models with HBV-related hepadnaviral infection. 3Parallel Reinforcement Learning-Based Energy Efficiency Improvement for a Cyber-Physical System显示文摘As a complex and critical cyber-physical system(CPS),the hybrid electric powertrain is significant to mitigate air pollution and improve fuel economy.Energy management strategy(EMS)is playing a key role to improve the energy efficiency of this CPS.This paper presents a novel bidirectional long shortterm memory(LSTM)network based parallel reinforcement learning(PRL)approach to construct EMS for a hybrid tracked vehicle(HTV).This method contains two levels.The high-level establishes a parallel system first,which includes a real powertrain system and an artificial system.Then,the synthesized data from this parallel system is trained by a bidirectional LSTM network.The lower-level determines the optimal EMS using the trained action state function in the model-free reinforcement learning(RL)framework.PRL is a fully data-driven and learning-enabled approach that does not depend on any prediction and predefined rules.Finally,real vehicle testing is implemented and relevant experiment data is collected and calibrated.Experimental results validate that the proposed EMS can achieve considerable energy efficiency improvement by comparing with the conventional RL approach and deep RL.Teng Liu Bin Tian Yunfeng Ai Fei-Yue Wang 2020IEEE/CAA Journal of Automatica Sinica2020,7,2:14

6Parallel Reinforcement Learning:A Framework and Case Study显示文摘In this paper, a new machine learning framework is developed for complex system control, called parallel reinforcement learning. To overcome data deficiency of current data-driven algorithms, a parallel system is built to improve complex learning system by self-guidance. Based on the Markov chain(MC) theory, we combine the transfer learning, predictive learning, deep learning and reinforcement learning to tackle the data and action processes and to express the knowledge. 7H2 strain attenuated live hepatitis A vaccines:protective efficacy in a hepatitis A outbreak显示文摘AIM To investigate the protective efficacy ofH2 strain attenuated live hepatitis A vaccines(H2-strain vaccines)in hepatitis A(HA)outbreaks.METHODS With the permission of theirparents,5551 pre-school and grade 1-3 primaryschool children were inoculated with 1 dose(106.5TCID50)of H2-strain vaccines in anonrandomized,controlled trial conducted inFucheng County,Hebei Province in May 1997.Another 6485 children in the same grades andcompatible in gender and age were enrolled ascontrols.Epidemiological and serological surveywas conducted to evaluate the protectiveefficacy of the vaccines.ELISA was used todetect serum IgM anti-HAV.RESULTS HA outbreak started in early May1998,peaked in the middle of the same month,and lasted about 80 days.Overall 302 HA caseswere found,192(53.58%)were 5-9 years old.One vaccinee and 25 control cases were found tohave hepatitis A,which account for 0.28%(1/356)and 5.92%(25/422)of all vaccinees andcontrols in the 14 villages,respectively.The protective efficacy of vaccines was 95.27%(95% Cl:85.83%-104.72%).In subjects testedfor anti-HAV IgM from 13 villages,1(0.40%)overt and 11(4.06%)asymptomatic HAV caseswere found in 271 vaccinees,but 21(6.69%)ofovert and asymptomatic ones were found in 314controls.CONCLUSION H2 strain vaccines were excellent in preventing overt hepatitis A, but not so effective in preventing asymptomatic hepatitis A virus infection. 10Meanders of the Jialing River in China: Morphology and formation显示文摘As a fundamental category of river system, comparisons in diverse regions and types with quantitative parameters on meander morphology will help to distinguish different characteristics and approach formations and evolutions. It also can provide scientific basis of geomorphology for their protections and exploitation of the meander resources. Based on the images of Google Earth, meanders’ parame- ters are proposed and then measured and calculated. The result suggests that incised meanders in the Jialing River are among the most complex, irregular and sinuous channels. Special river gradient, geological structures, strata and lithology, backwater effect of landform are key factors to shape me- anders of the Jialing River. The evolution of meander in the Jialing River obeys the principle of mini- mum-maximum energy dissipation. Easy acquisitions of remote sensing data make regional and global comparisons possible, and then the differences of formations can be explored. The morphology can probably provide an evidence of antecedent river and demonstrate the channel incisions and crustal uplifts. Meander core in Chinese is named after the isolated hills in the Jialing River. 11Adsorption/desorption of toluene on a hypercrosslinked polymeric resin in a highly humid gas stream显示文摘In many sources of volatile organic compounds (VOCs), large amounts of water vapor come from the air and the reactors. The relative humidity (RH) of exhaust gas is normally >60% and is supersaturated. Maintaining the property of adsorbent on VOCs in a highly humid gas stream is a serious industrial problem. In this study, the adsorption/desorption behavior of toluene in a micro-mesoporous polymeric resin was investigated in a highly humid environment to explore the influence of abound water vapor on resin adsorption and regeneration. This resin could selectively adsorb toluene at an RH of 80%, and its adsorption property was unaffected by the presence of water vapor. In the case of humidity saturation, the resin displayed a high adsorption capacity at a moisture content of <30%. Therefore, the polymer resin is an excellent water-resistant adsorbent of VOCs. In the regenerative experiment, the resin maintained its original adsorption capability after four adsorption/ desorption cycles of toluene purging with nitrogen gas at 120℃. 12Multifunctional dextran micelles as drug delivery carriers and magnetic resonance imaging probes显示文摘Multifunctional nanoparticles combining diagnostic and therapeutic agents into a single platform make cancer theranostics possible and have attracted wide interests in the field. In this study, a multifunctional nanocomposite based on dextran and superparamagnetic iron oxide nanoparticles(SPIO) was prepared for drug delivery and magnetic resonance imaging(MRI). Amphiphilic dextran was synthesized by grafting stearyl acid onto the carbohydrate backbone, and micelle was formed by the resulted amphiphilic dextran with low critical micelle concentration at 1.8 mg L-1. Doxorubicin(DOX) and a cluster of the manganese-doped iron oxide nanoparticles(Mn-SPIO)nanocrystals were then coencapsulated successfully inside the core of dextran micelles, resulting in nanocomposites with diameter at about 100 nm. Cell culture experiments demonstrated the potential of these Mn-SPIO/DOX nanocomposites as an effective multifunctional nanoplatform for the delivery of anticancer drug DOX with a loading content(DLC) of 16 %. Confocal laser scanning microscopyreveals that the Mn-SPIO/DOX had excellent internalization ability against MCF-7/Adr cells after 2-h labeling compared with free DOXáHCl. Under a 3.0-T MRI scanner, Mn-SPIO/DOX nanocomposite-labeled cells in gelatin phantom show much darker images than the control. Their transverse relaxation(T2) rate is also significantly higher than that of the control cells(33.9 versus 2.3 s-1). 13Robust fault detection for a class of nonlinear network control system with communication delay显示文摘To investigate a class of nonlinear network control system,a robust fault diagnosis method is presented based on the robust state observer.To access the objective that the designed robust filter is maximally tolerant to disturbances and sensitive to fault,the robustness and stability properties of the fault diagnosis scheme are established rigorously.Using the residual vector,a fault tolerant controller is established in order to guarantee the stability of the closed-loop system,and the controller law can be obtained by solving a set of linear matrix inequalities. 14Amphiphilic dextran/magnetite nanocomposites as magnetic resonance imaging probes显示文摘Superparamagnetic iron oxide (SPIO) nanoparticles are effective contrast agents for enhancement of magnetic resonance imaging at the tissue, cellular or even molecular levels. High quality SPIO nanoparticles can be synthesized in the organic phase but need to be transferred into water before any biomedical applications. In this study, amphiphilic poly(ε-caprolactone) grafted dextran (Dex-g-PCL) was used as carriers for particle encapsulation and stabilization in the aqueous phase. Multiple SPIO nanoparticles were self-assembled together with the help of Dex-g-PCL during phase transfer from chloroform to water, and diameters of Dex-g-PCL/SPIO nanocomposites were (64 ± 22) nm through dynamic light scattering measurement. These nanocomposites were superparamagnetic at 300 K with saturated magnetization of 88 emu/g Fe. In the magnetic field of 1.5 T, Dex-g-PCL/SPIO nanocomposites had a T2 relaxivity of 363 Fe mL·mol-1·s-1. This unique nanocomposite brought significant mouse liver contrast with signal intensity changes of -60% at 5 min after intravenous administration. However, uptake of Dex-g-PCL/SPIO nanocomposites in liver reticuloendothelial cells (Kupffer cells) did not immediately happen at shorter time points (<4 h) as verified by histology studies, and it was evident that more iron staining would be located in Kupffer cells 24 h after contrast agent administration. After 24 h and 10 d, the signal intensities (SI) gradually recovered, and SI changes were -44% and -31%, respectively. From our observation, the time window for enhanced-MRI could last at least 12 days and totally recovered after 16 days. 15Epidermal growth factor receptor mutation analysis in cytological specimens and responsiveness to gefitinib in advanced non-small cell lung cancer patients显示文摘Background: Epidermal growth factor receptor(EGFR) mutation is the key predictor of EGFR tyrosine kinase inhibitors(TKIs) efficacy in non-small cell lung cancer(NSCLC). We conducted this study to verify the feasibility of EGFR mutation analysis in cytological specimens and investigate the responsiveness to gefitinib treatment in patients carrying EGFR mutations.Methods: A total of 210 cytological specimens were collected for EGFR mutation detection by both direct sequencing and amplification refractory mutation system(ARMS). We analyzed EGFR mutation status by both methods and evaluated the responsiveness to gefitinib treatment in patients harboring EGFR mutations by overall response rate(ORR), disease control rate(DCR) and progression free survival(PFS).Results: Of all patients, EGFR mutation rate was 28.6%(60/210) by direct sequencing and 45.2%(95/210) by ARMS(P<0.001) respectively. Among the EGFR wild type patients tested by direct sequencing, 26.7% of them were positive by ARMS. For the 72 EGFR mutation positive patients treated with gefitinib, the ORR, DCR and median PFS were 69.4%, 90.2% and 9.3 months respectively. The patients whose EGFR mutation status was negative by direct sequencing but positive by ARMS had lower ORR(48.0% vs. 80.9%, P=0.004) and shorter median PFS(7.4 vs. 10.5 months, P=0.009) as compared with that of EGFR mutation positive patients by both detection methods. Conclusions: Our study verified the feasibility of EGFR analysis in cytological specimens in advanced NSCLC. ARMS is more sensitive than direct sequencing in EGFR mutation detection. EGFR Mutation status tested on cytological samples is applicable for predicting the response to gefitinib. 17Scaling behavior of magnitude clusters in aftershock sequence:An example of the Wenchuan Earthquake,China显示文摘The Gutenberg-Richter and Omori Laws, which are generally used to characterize the temporal distribution of aftershock, failed to reflect the statistic properties of climatic outbreak of aftershock energy. Based on a new concept of magnitude clusters describing the fluctuation of aftershock energy release of the Wenchuan Earthquake, we discovered that the pattern of the continuous high-magnitude aftershock follows a power-law rather than a non-Poisson distribution. This suggests that the after-shocks with high magnitudes are statistically clustered. We then divided the aftershock sequences into three sections and demonstrated that though the probability of strong outbreaks decreased with time, there exists a high possibility of the occurrence of isolated high-magnitude aftershocks in the future. Based on self-organized criticality theory, the mechanisms of the power-law pattern of magnitude clusters are discussed. 18Transplantation of tissue-engineered human corneal epithelium in limbal stem cell deficiency rabbit models显示文摘·AIM:To evaluate the biological functions of tissue- engineered human corneal epithelium (TE-HCEP) by corneal transplantation in limbal stem cell deficiency (LSCD) rabbit models.·METHODS:TE-HCEPs were reconstructed with DiI-labeled untransfected HCEP cells and denuded amniotic membrane (dAM) in air-liquid interface culture, and their morphology and structure were characterized by hematoxylin-eosin (HE) staining of paraffin-sections, immunohistochemistry and electron microscopy. LSCD models were established by mechanical and alcohol treatment of the left eyes of New Zealand white rabbits, and their eyes were transplanted with TE-HCEPs with dAM surface outside by lamellar keratoplasty (LKP). Corneal transparency, neovascularization, thickness, and epithelial integrality of both traumatic and post transplantation eyes were checked once a week by slit-lamp corneal microscopy, a corneal pachymeter, and periodic acid-Schiff (PAS) staining. At day 120 post surgery, the rabbits in each group were sacrificed and their corneas were examined by DiI label observation, HE staining, immunohistochemistry and electron microscopy.·RESULTS:After cultured for 5 days on dAM, HCEP cells, maintaining keratin 3 expression, reconstructed a 6-7 layer TE-HCEP with normal morphology and structure. The traumatic rabbit corneas, entirely opaque, conjunctivalized and with invaded blood vessels, were used as LSCD models for TE-HCEP transplantation. After transplantation, obvious edema was not found in TE-HCEP-transplanted corneas which became more and more transparent, the invaded blood vessels reduced gradually throughout the monitoring period. The corneas decreased to normal thickness on day 25, while those of dAM eyes were over 575μm in thickness during the monitoring period. A 4-5 layer of epithelium consisting of TE-HCEP originated cells attached tightly to the anterior surface of stroma was reconstructed 120 days after TE-HCEP transplantation, which was similar to the normal control eye in morphology and structure. In contrast, intense corneal edema, turbid, invaded blood vessels were found in dAM eyes, and no multilayer epithelium was found but only a few scattered conjunctiva-like cells appeared. ·CONCLUSION:The TE-HCEP, with similar morphology and structure to those of innate HCEP, could reconstruct a multilayer corneal epithelium with normal functions in restoring corneal transparency and thickness of LSCD rabbits after transplantation. 20Near-infrared fluorescent amphiphilic polycation wrapped magnetite nanoparticles as multimodality probes显示文摘Construction of multifunctional/multimodality nanoparticles for cancer diagnosis and therapy has become an attractive area of investigation. In this report, we designed a multimodality nanoprobe for cell labeling, and can be detectable by both magnetic resonance and near infrared (NIR) fluorescence imaging. Multiple hydrophobic superparamagnetic iron oxide (SPIO) nanocrystals are self-assembled into nanocomposites in water phase with the help of partially alkylated hyperbranched polycation, polyethylenimine (PEI), which already conjugated with the indocyanine dye Cy5.5 and can be used for cell imaging under NIR fluorescence imaging. The amphiphilic PEI/SPIO nanocomposites have a strong T 2 relaxivity. The iron uptake process in MCF-7/Adr displays a time dependent behavior. Confocal laser scanning microscopy reveals that the nanoprobes are internalized into the cytoplasm of MCF-7/Adr after 24 h labeling. Both MR and NIR fluorescence imaging showed strong image contrast against unlabeled cells. Under a clinical MRI scanner, labeled cells in gelatin phantom present much darker images than controlled ones. The T2 relaxation rate of the labeled cells is 98.2 s 1 , significantly higher than that of the control ones of 2.3 s 1 .
