| 1 | Soybean Root System Architecture Trait Study through Genotypic,Phenotypic,and Shape-Based Clusters显示文摘We report a root system architecture(RSA)traits examination of a larger scale soybean accession set to study trait genetic diversity.Suffering from the limitation of scale,scope,and susceptibility to measurement variation,RSA traits are tedious to phenotype.Combining 35,448 SNPs with an imaging phenotyping platform,292 accessions(replications=14)were studied for RSA traits to decipher the genetic diversity.Based on literature search for root shape and morphology parameters,we used an ideotypebased approach to develop informative root(iRoot)categories using root traits.The RSA traits displayed genetic variability for root shape,length,number,mass,and angle.Soybean accessions clustered into eight genotype-and phenotype-based clusters and displayed similarity.Genotype-based clusters correlated with geographical origins.SNP profiles indicated that much of US origin genotypes lack genetic diversity for RSA traits,while diverse accession could infuse useful genetic variation for these traits.Shape-based clusters were created by integrating convolution neural net and Fourier transformation methods,enabling trait cataloging for breeding and research applications.The combination of genetic and phenotypic analyses in conjunction with machine learning and mathematical models provides opportunities for targeted root trait breeding efforts to maximize the beneficial genetic diversity for future genetic gains. | Kevin G.Falk Talukder Zaki Jubery Jamie A.O’Rourke Arti Singh Soumik Sarkar Baskar Ganapathysubramanian Asheesh K.Singh | 2020 | Plant Phenomics2020,2,1: | 3 |
| 2 | Review: laser ignition for aerospace propulsion显示文摘Renewed interest in the use of high-speed ramjets and scramjets and more efficient lean burning engines has led to many subsequent developments in the field of laser ignition for aerospace use and application.Demands for newer,more advanced forms of ignition,are increasing as individuals strive to meet regulations that seek to reduce the level of pollutants in the atmosphere,such as CH_(x),NO_(x),and SO_(2).Many aviation gas turbine manufacturers are interested in increasing combustion efficiency in engines,all the while reducing the aforementioned pollutants.There is also a desire for a new generation of aircraft and spacecraft,utilizing technologies such as scramjet propulsion,which will never realize their fullest potential without the use of advanced ignition processes.These scenarios are all limited by the use of conventional spark ignition methods,thus leading to the desire to find new,alternative methods of ignition.This paper aims to provide the reader an overview of advanced ignition methods,with an emphasis on laser ignition and its applications to aerospace propulsion.A comprehensive review of advanced ignition systems in aerospace applications is performed.This includes studies on gas turbine applications,ramjet and scramjet systems,and space and rocket applications.A brief overview of ignition and laser ignition phenomena is also provided in earlier sections of the report.Throughout the reading,research papers,which were presented at the 2nd Laser Ignition Conference in April 2014,are mentioned to indicate the vast array of projects that are currenty being pursued. | Steven A.O’Briant Sreenath B.Gupta Subith S.Vasu | 2016 | Propulsion and Power Research2016,5,1: | 3 |
| 3 | Update of Meat Standards Australia and the cuts based grading scheme for beef and sheepmeat显示文摘Changing markets and evolving consumer demand present new challenges for the beef and sheep industries. In response, the industry has been investing in innovations to deliver new products and management systems to consumers. One such innovation is the Meat Standards Australia(MSA) system. This system is a Total Quality Management System, aimed at delivering an eating quality guarantee to consumers, and through this adding value to the entire supply chain. At present, it is well developed for beef and still evolving for sheepmeat. MSA has identified Critical Control Points(CCPs) in the production, pre-slaughter, processing and value-adding aspects of the supply chain that impact on consumer palatability through the large-scale taste testing of meat by untrained consumers. These CCPs are used as either(1) mandatory criteria determining eligibility for grading, and(2) inputs in a model predicting the palatability of individual combinations of muscle and different cooking methods. Through the prediction of palatability, MSA increases consumer satisfaction and is used to provide assurance for branded products and new marketing innovations in Australia and internationally. This has added significant value to the Australian beef industry, with several retail examples demonstrating consumer willingness to pay more for premium quality beef and sheepmeat products based on the MSA grading scores. This price differential at retail allows the value of the carcass to be calculated based on the eating quality as well as the volume produced, thereby delivering a financial reward for farmers producing high quality carcasses. The continuous quality scale of MSA allows producers to realise the financial gain of incremental improvements in quality, as well as the precise economic weights associated with traits such as marbling, ossification score, or breed. The use of MSA in this fashion has underpinned a new and innovative supply chain where the pricing is transparent and allows producers to make informed decisions to modify both quality and yield traits. To date, the MSA system for beef has proved to be effective in predicting beef palatability not only in Australia but also in many other countries(France, Poland, Ireland, Northern Ireland, Japan, South Korea, New-Zealand, the USA and South Africa). In Europe, results of the Pro Safe Beef and Pro Opti Beef projects as well as other national projects demonstrate the potential to develop an MSA-like international grading system for the supply chain in the EU, despite thediverse cultures and complex beef production systems within the member states. | 4 | Designing chimeric enzymes inspired by fungal cellulosomes显示文摘Cellulosomes are synthesized by anaerobic bacteria and fungi to degrade lignocellulose via synergistic action of multiple enzymes fused to a protein scaffold.Through templating key protein domains(cohesin and dockerin),designer cellulosomes have been engineered from bacterial motifs to alter the activity,stability,and degradation efficiency of enzyme complexes.Recently a parts list for fungal cellulosomes from the anaerobic fungi(Neocallimastigomycota)was determined,which revealed sequence divergent fungal cohesin,dockerin,and scaffoldin domains that could be used to expand the available toolbox to synthesize designer cellulosomes.In this work,multi-domain carbohydrate active enzymes(CAZymes)from 3 cellulosome-producing fungi were analyzed to inform the design of chimeric proteins for synthetic cellulosomes inspired by anaerobic fungi.In particular,Piromyces finnis was used as a structural template for chimeric carbohydrate active enzymes.Recombinant enzymes with retained properties were engineered by combining thermophilic glycosyl hydrolase domains from Thermotoga maritima with dockerin domains from Piromyces finnis.By preserving the protein domain order from P.finnis,chimeric enzymes retained catalytic activity at temperatures over 80°C and were able to associate with cellulosomes purified from anaerobic fungi.Fungal cellulosomes harbor a wide diversity of glycoside hydrolases,each representing templates for the design of chimeric enzymes.By conserving dockerin domain position within the primary structure of each protein,the activity of both the catalytic domain and dockerin domain was retained in enzyme chimeras.Taken further,the domain positioning inferred from native fungal cellulosome proteins can be used to engineer multi-domain proteins with non-native favorable properties,such as thermostability. | Sean P.Gilmore Stephen P.Lillington Charles H.Haitjema Randall de Groot Michelle A.O'Malley | 2020 | Synthetic and Systems Biotechnology2020,5,1: | 2 |
