1Heparin improves organ microcirculatory disturbances in caerulein-induced acute pancreatitis in rats显示文摘AIM: Microcirculatory disturbances are important early pathophysiological events in various organs during acute pancreatitis. The aim of the study was to evaluate changes in rnicroperfusion of the pancreas, liver, kidney, stomach,colon, skeletal muscle, and to investigate the influence of heparin on the organ rnicrocirculation in caerulein-induced experimental acute pancreatitis.METHODS: Acute pancreatitis was induced by 4 intraperitoneal injections of caerulein (Cn) (15 μg/kg). The organ microcirculation was measured by laser Doppler flowrnetry. Serum interleukin 6 and hernatocrit levels were analysed.RESULTS: Acute pancreatitis resulted in a significant drop of microperfusion in all examined organs. Heparin administration (2×2.5 mg/kg) improved the rnicrocirculation in pancreas (36.9±4% vs75.9±10%), liver (56.6±6% vs 75.2±16%), kidney (45.1±6% vs79.3±5%), stomach (65.2±8% vs78.1±19%), colon (69.8±6% vs 102.5±19%),and skeletal muscle (59.2±6% vs 77.9±13%). 2Serum and urine metabolomic fingerprinting in diagnostics of inflammatory bowel diseases显示文摘AIM:To evaluate the utility of serum and urine metabolomic analysis in diagnosing and monitoring of inflammatory bowel diseases(IBD).METHODS:Serum and urine samples were collected from 24 patients with ulcerative colitis(UC),19 patients with the Crohn’s disease(CD)and 17 healthy controls.The activity of UC was assessed with the Simple Clinical Colitis Activity Index,while the activity of CD was determined using the Harvey-Bradshaw Index.The analysis of serum and urine samples was performed using proton nuclear magnetic resonance(NMR)spectroscopy.All spectra were exported to Matlab for preprocessing which resulted in two data matrixes for serum and urine.Prior to the chemometric analysis,both data sets were unit variance scaled.The differences in metabolite fingerprints were assessed using partial least-squaresdiscriminant analysis(PLS-DA).Receiver operating characteristic curves and area under curves were used to evaluate the quality and prediction performance of the obtained PLS-DA models.Metabolites responsible for separation in models were tested using STATISTICA10 with the Mann-Whitney-Wilcoxon test and the Student’s t test(α=0.05).RESULTS:The comparison between the group of patients with active IBD and the group with IBD in remission provided good PLS-DA models(P value 0.002for serum and 0.003 for urine).The metabolites that allowed to distinguish these groups were:N-acetylated compounds and phenylalanine(up-regulated in serum),low-density lipoproteins and very low-density lipoproteins(decreased in serum)as well as glycine(increased in urine)and acetoacetate(decreased in urine).The significant differences in metabolomic profiles were also found between the group of patients with active IBD and healthy control subjects providing the PLS-DA models with a very good separation(P value<0.001 for serum and 0.003 for urine).The metabolites that were found to be the strongest biomarkers included in this case:leucine,isoleucine,3-hydroxybutyric acid,N-acetylated compounds,acetoacetate,glycine,phenylalanine and lactate(increased in serum),creatine,dimethyl sulfone,histidine,choline and its derivatives(decreased in serum),as well as citrate,hippurate,trigonelline,taurine,succinate and 2-hydroxyisobutyrate(decreased in urine).No clear separation in PLS-DA models was found between CD and UC patients based on the analysis of serum and urine samples,although one metabolite(formate)in univariate statistical analysis was significantly lower in serum of patients with active CD,and two metabolites(alanine and N-acetylated compounds)were significantly higher in serum of patients with CD when comparing jointly patients in the remission and active phase of the diseases.Contrary to the results obtained from the serum samples,the analysis of urine samples allowed to distinguish patients with IBD in remission from healthy control subjects.The metabolites of importance included in this case up-regulated acetoacetate and down-regulated citrate,hippurate,taurine,succinate,glycine,alanine and formate.CONCLUSION:NMR-based metabolomic fingerprinting of serum and urine has the potential to be a useful tool in distinguishing patients with active IBD from those in remission.Tomasz Dawiskiba Stanislaw Deja Agata Mulak Adam Zabek Ewa Jawień Dorota Pawelka Miroslaw Banasik Agnieszka Mastalerz-Migas Waldemar Balcerzak Krzysztof Kaliszewski Jan Skóra Piotr Bar Krzysztof Korta Kornel Pormańczuk Przemyslaw Szyber Adam Litarski Piotr Mlynarz 2014World Journal of Gastroenterology2014,20,1:15
3Proteolytic-antiproteolytic balance and its regulation in carcinogenesis显示文摘Cancer development is essentially a tissue remodeling process in which normal tissue is substituted with cancer tissue. A crucial role in this process is attributed toproteolytic degradation of the extracellular matrix (ECM).Degradation of ECM is initiated by proteases, secreted by different cell types, participating in tumor cell invasion and increased expression or activity of every known class of proteases (metallo-, serine-, aspartyl-, and cysteine)has been linked to malignancy and invasion of tumor cells.Proteolytic enzymes can act directly by degrading ECM or indirectly by activating other proteases, which then degrade the ECM. They act in a determined order, resulting from the order of their activation. When proteases exert their action on other proteases, the end result is a cascade leading to proteolysis. Presumable order of events in this complicated cascade is that aspartyl protease (cathepsin D) activates cysteine proteases (e.g. cathepsin B) that can activate pro-uPA. Then active uPA can convert plasminogen into plasmin. Cathepsin B as well as plasmin are capable of degrading several components of tumor stroma and may activate zymogens of matrix metalloproteinases, the main family of ECM degrading proteases. The activities of these proteases are regulated by a complex array of activators,inhibitors and cellular receptors. Tn physiological conditions the balance exists between proteases and their inhibitors.Proteolytic-antiproteolytic balance may be of major significance in the cancer development. One of the reasons for such a situation is enhanced generation of free radicals observed in many pathological states. Free radicals react with main cellular components like proteins and lipids and in this way modify proteolytic-antiproteolytic balance and enable penetration damaging cellular membrane. 4Mucosal bacterial microflora and mucus layer thickness in adolescents with inflammatory bowel disease显示文摘AIM:To assess the mucosa-associated bacterial microflora and mucus layer in adolescents with inflammatory bowel disease(IBD) .METHODS:Sixty-one adolescents(mean age 15 years,SD ± 4.13) were included in the study.Intestinal biopsies from inflamed and non-inflamed mucosa of IBD patients and from controls with functional abdominal pain were cultured under aerobic and anaerobic conditions.The number of microbes belonging to the same group was calculated per weight of collected tissue.The mucus thickness in frozen samples was measured under a fluorescent microscope.RESULTS:The ratios of different bacterial groups in inflamed and non-inflamed mucosa of IBD patients and controls were specific for particular diseases.Streptococcus spp.were predominant in the inflamed mucosa of Crohn's disease(CD) patients(80% of all bacteria) ,and Lactobacillus spp.were predominant in ulcerative colitis patients(90%) .The differences were statistically significant(P = 0.01-0.001) .Lower number of bifidobacteria was observed in the whole IBD group.A relation was also found between clinical and endoscopic severity and decreased numbers of Lactobacillus and,to a lesser extent,of Streptococcus in biopsies from CD patients.The mucus layer in the inflamed sites was significantly thinner as compared to controls(P = 0.0033) and to non-inflamed areas in IBD patients(P = 0.031) .CONCLUSION:The significantly thinner mucosa of IBD patients showed a predominance of some aerobes specific for particular diseases,their numbers decreased in relation to higher clinical and endoscopic activity of the disease.Krzysztof Fyderek Magdalena Strus Kinga Kowalska-Duplaga Tomasz Gosiewski Andrzej Wedrychowicz Urszula Jedynak-Wasowicz Malgorzata Sladek Stanislaw Pieczarkowski Pawel Adamski Piotr Kochan Piotr B Heczko 2009World Journal of Gastroenterology2009,15,42:14
5Lipid peroxidation and antioxidant status in colorectal cancer显示文摘AIM: Reactive oxygen species (ROS) can induce carcinogenesis via DNA injury. Both enzymatic and non-enzymatic parameters participate in cell protection against harmful influence of oxidative stress. The aim of the present study was to assess the levels of final lipid peroxidation products like malondialdehyde (MDA) and 4-hydroxy-2-nonenal (4-HNE) in primary colorectal cancer. Moreover, we analysed the activity of main antioxidative enzymes, superoxide dismutase (Cu, Zn-SOD),catalase (CAT), glutathione peroxidase (GSH-Px) and glutathione reductase (GSSRG-R) and the level of nonenzymatic antioxidants (glutathione, vitamins C and E).METHODS: Investigations were conducted in 81 primary colorectal cancers. As a control, the same amount of sample was collected from macroscopically unchanged colon regions of the most distant location to the cancer.Homogenisation of specimens provided 10% homogenates for our evaluations. Activity of antioxidant enzymes and level of glutathione were determined by spectrophotometry.HPLC revealed levels of vitamins C and E and served as a method to detect terminal products of lipid peroxidation in colorectal cancer.RESULTS: Our studies demonstrated a statistically significant increase in the level of lipid peroxidation products (MDA-Adc.muc.-2.65±0.48 nmol/g, Adc. G3-2.15±0.44 nmol/g, clinical Ⅳ stage 4.04±0.47 nmol/g, P<0.001 and 4-HNE-Adc.muc.-0.44±0.07 nmol/g, Adc. G3-0.44±0.10 nmol/g, clinical Ⅳstage 0.52±0.11 nmol/g, P<0.001) as well as increase of Cu,Zn-SOD (Adc.muc.-363±72 U/g, Adc. G3-318±48 U/g,clinical Ⅳ stage 421±58 U/g, P<0.001), GSH-Px (Adc.muc.-2143±623 U/g, Adc. G3-2005±591 U/g, clinical Ⅳ stage 2467±368 U/g, P<0.001) and GSSG-R (Adc. muc.-880±194 U/g,Adc. G3-795±228 U/g, dinical Ⅳ stage 951±243 U/g, P<0.001)in primary tumour comparison with normal colon (MDA1.39±0.15 nmol/g, HNE-0.29±0.03 nmol/g, Cu, Zn-SOD-117±25 U/g, GSH-Px-1723±189 U/g, GSSG-R-625±112 U/g)especially in mucinous and G3-grade adenocarcinomas as well as clinical Ⅳ stage of colorectal cancer. We also observed a decrease of CAT activity (Adc.muc. -40±14 U/g,clinical Ⅳ stage 33±18 U/g vs84±17 U/g, P<0.001) as well as a decreased level of reduced glutathione (clinical Ⅳ stage 150±48 nmol/g vs 167±15 nmol/g, P<0.05) and vitamins C and E (vit. C-clinical Ⅳ stage 325±92 nmol/g vs 513±64 nmol/g, P<0.001; vit. 6Intentional ingestions of foreign objects among prisoners: A review显示文摘The intentional ingestion of foreign objects(IIFO) is described more commonly in prison populations than in the general population, with an estimated annual incidence of 1 in 1900 inmates in our state correctional facilities. Incidents often involve ingestion of small metal objects(e.g., paperclips, razor blades) or other commonly available items like pens or eating utensils. Despite ingestion of relatively sharp objects, most episodes can be clinically managed with either observation or endoscopy. Surgery should be reserved for those with signs or symptoms of gastrointestinal perforation or obstruction. For those with a history of IIFO, efforts should focus on prevention of recurrenceas subsequent episodes are associated with higher morbidity, significant healthcare and security costs. The pattern of IIFO is often repetitive, with escalation both in frequency of ingestions and in number of items ingested. Little is known about successful prevention strategies, but efforts to monitor patients and provide psychiatric care are potential best-practice strategies. 8Pioglitazone, a specific ligand of peroxisome proliferator-activated receptor-gamma, protects pancreas against acute cerulein-induced pancreatitis显示文摘ABM: To determine the effect of pioglitazone, a specific peroxisome proliferator-activated receptor-γ(PPARγ) ligand, on the development of acute pancreatitis (AP) and on the expression of heat shock protein 70 (HSP70) in the pancreas. METHODS: AP was induced in rats by subcutaneous infusion of cerulein for 5 h. Pancreatic blood flow was measured by laser Doppler flowmetry. Plasma lipase activity, interleukin-1β (IL-1β) and IL-10 were determined, Pancreatic weight and histology were evaluated and pancreatic DNA synthesis and blood flow as well as pancreatic mRNA for IL-1β and HSP70 were assessed in rats treated with pioglitazone alone or in combination with cerulein. RESULTS: Pioglitazone administered (10-100 mg/kg i.g.) 30 min before cerulein, attenuated dose-dependently the pancreatic tissue damage in cerulein-induced pancreatitis (CIP) as demonstrated by the improvement of pancreatic histology, reduction in plasma lipase activity, plasma concentration of pro-inflammatory IL-1β and its gene expression in the pancreas and attenuation of the pancreatitis-evoked fall in pancreatic blood flow. CIP increased pancreatic HSP70 mRNA and protein expression in the pancreas and this effect was enhanced by pioglitazone treatment. CONCLUSION: Pioglitazone attenuates CIP and the beneficial effect of this pioglitazone is multifactorial probably due to its anti-inflammatory activities, to the suppression of IL-1β and to the over-expression of HSP70. 9Evaluation of serum catnepsin B and D in relation to diniGopatnological staging of colorectal cancer显示文摘AIM: Proteolytic degradation of the extracellular matrix facilitates cancer invasion and promotes metastasis. The study aims at evaluation of preoperative and postoperative serum cathepsins B and D levels in correlation with selected anatomoclinical features of colorectal cancer.METHODS: Blood samples were collected from 63colorectal cancer patients before curative operation of the tumor 10 d later. Blood that was obtained from 20healthy volunteers, served as a control. The activity of cathepsin B was measured with Bz-DL-arginine-pNA as a substrate at pH 6.0, while cathepsin D activity was determined with urea-denatured hemoglobin (pH 4.0).RESULTS: The preoperative and postoperative activities of cathepsin B were significantly (P<0.00001) lower in serum of colorectal cancer patients than in control group.However, postoperative values of this protease were significantly increased in comparison with preoperative ones (P = 0.031). Activity of cathepsin D appeared to be significantly higher in colorectal cancer sera (P<0.00001)compared with controls. No statistically significant differences between preoperative and postoperative activity of cathepsin D were noted (P = 0.09). We revealed a strong linkage of cathepsins' levels with lymph node status and pT stage of colorectal cancer.CONCLUSION: Blood serum activities of cathepsin B and D depend on the time of sampling, tumor size and lymph node involvement. Significantly, increased activity of cathepsin D could indicate a malignant condition of the large intestine. 11Gas-dynamic phenomena caused by rock mass tremors and rock bursts显示文摘Similar to coal, rock and gas ejections, rock mass tremors and rock bursts are among the most serious natural hazards accompanying the underground extraction of coal. Gas-dynamic phenomena caused by rock mass tremors and rock bursts observed as transient states of air parameters in mining headings,are usually generated as a result of a change in the geometry of headings and the release of considerable amounts of gases. Particular significance is attributed to transient states caused by disasters, which are often accompanied by rapid incidents, presenting threats to the life and health of the underground crew.In Polish mining there are known examples of transient states of air parameters recorded during gasdynamic phenomena, e.g. tremors and rock bursts. 12Connexin 26 correlates with Bcl-xL and Bax proteins expression in colorectal cancer显示文摘AIM: To evaluate of Cx26 in correlation with Bcl-xL and Bax proteins in colorectal cancer.METHODS: Immunohistochemical staining using specific antibodies was performed to evaluate the protein expression of Cx26, Bax and Bcl-xL in 152 colorectal cancer samples and the correlations among studied proteins as well as the relationships between the expression of Cx26,Bax, Bcl-xL and clinicopathological features were analyzed.RESULTS: Both normal epithelial cells and carcinoma cells expressed Cx26, Bax and Bcl-xL, but Cx26 in cancer cells showed aberrant, mainly cytoplasmic staining. Expression of Cx26, Bax and Bcl-xL was observed in 55.9%, 55.5%and 72.4% of evaluated colorectal cancers respectively.We found the positive correlation between Cx26 and Bax expression (r = 0.561, P<0.0001), Cx26 and Bcl-xL (r = 0.409, P<0.0001) as well as between Bax and Bcl-xL (r = 0.486, P<0.0001). Association of Cx26, Bax and Bcl-xL expression with histological G2 grade of tumors was noted (P<0.005, P<0.001 and P<0.002 respectively).CONCLUSION: Cytoplasmic presence of Cx26 and its association with apoptotic markers could indicate a distinct role from physiological functions of Cx26 in cancer cells and it could suggest that connexins might be a target point for modulations of apoptosis with therapeutic implications.Luiza Kanczuga-Koda Stanislaw Sulkowski Mariusz Koda Elzbieta Skrzydlewska Mariola Sulkowska 2005World Journal of Gastroenterology2005,11,10:5
13Laparoscopy-assisted percutaneous endoscopic gastrostomy enables enteral nutrition even in patients with distorted anatomy显示文摘AIM:To analyzed whether laparoscopy-assisted percutaneous endoscopic gastrostomy(PEG)could be a valuable option for patients with complicated anatomy.METHODS:A retrospective analysis of twelve patients(seven females,five males;six children,six young adults;mean age 19.2 years)with cerebral palsy,spastic quadriparesis,severe kyphoscoliosis and interposed organs and who required enteral nutrition(EN)due to starvation was performed.For all patients,standard PEG placement was impossible due to distorted anatomy.All the patients qualified for the laparoscopyassisted PEG procedure.RESULTS:In all twelve patients,the laparoscopy-assisted PEG was successful,and EN was introduced four to six hours after the PEG placement.There were no complications in the perioperative period,either technical or metabolic.All the patients were discharged from the hospital and were then effectively fed using bolus methods.CONCLUSION:Laparoscopy-assisted PEG should become the method of choice for gastrostomy tube placement and subsequent EN if PEG placement cannot be performed safely.Adam Hermanowicz Ewa Matuszczak Marta Komarowska Elzbieta Jarocka-Cyrta Jerzy Wojnar Wojciech Debek Konrad Matysiak Stanislaw Klek 2013World Journal of Gastroenterology2013,19,43:5

18高压输电线路雷击闪络的实验室研究(英文)显示文摘This paper presents an investigation on the attractive width of high voltage transmission lines to lightning strikes.In order to design the optimal lightning protection,the estimated number of lightning flashes on the line, which is based on its attractive width,needs to be determined.The investigation was performed using experiments with model tests at the Mississippi State University High Voltage Laboratory.For laboratory experiments,a total of 2,100 negative and positive switching impulse voltages were applied to transmission line models from a conducting rod,which represented a lightning downward leader.Different tested models of transmission lines on a scale of 1:100 were used.The effects of overhead ground wires,phase conductors,tower structures,and the magnitude and polarity of lightning strokes were also studied.The attractive width increased gradually with the height of overhead ground wires and towers as well as the magnitude of the lightning stroke current.Impulse polarity had an impact on the attractive width,and the attractive width for negative polarity was larger than that for positive polarity.The taller tower had more effect on flash distribution to transmission lines than the shorter one.The experimental results agree with the actual transmission line observations published in literature.The new expressions for the attractive width of transmission lines,based on the experimental results,were established.The accurate estimation of the attractive width can help electric power utilities plan transmission systems reliably and economically.The detailed description of the background problem,proposed method,experimental results,and analysis are presented in this paper.Thongchai Disyadej Stanislaw Grzybowski 2011高电压技术2011,37,11:3
19Investigation of the immune effects of Scutellaria baicalensis on blood leukocytes and selected organs of the chicken's lymphatic system显示文摘Background: The health of chickens and the welfare of poultry industry are central to the efforts of addressing global food security. Therefore, it is essential to study chicken immunology to maintain and improve its health and to find novel and sustainable solutions. This paper presents a study on investigation of the effect of Scutellaria baicalensis root(SBR) on the immune response of broiler chicken, especially on lymphocytes and heterophils reactivity, regarding their contribution to the development of immunity of the chickens.Methods: The 121-day-old Hubbard Hi-Y male broiler hybrids were randomly assigned to four treatment groups,three SBR supplemented groups(0.5, 1.0, and 1.5% of SBR) and one control group. Each treatment was replicated five times with six birds per replicate pen in a battery brooder. Blood was collected after 3^(rd) and 6^(th)wk of the experiment, and hemoglobin and hematocrit values were determined, as well as total leukocyte count and differential count were performed. Nitroblue tetrazolium test and phagocytosis assay as nonspecific immune parameters and humoral immune responses to the antigenic challenge by sheep red blood cells were performed.Moreover, the ability of peripheral blood lymphocytes to form radial segmentation(RS) of their nuclei was analyzed.Body weight and relative weight of spleen, liver, and bursa of Fabricius were recorded.Results: Results showed that mean heterophile/lymphocyte ratio increased in the SBR groups compared to the control group and the blood of the chickens showed lymphocytic depletion. The results also demonstrated that the relative weight of bursa of Fabricius and spleen in groups fed with SBR significantly decreased compared to the control group. This study also showed that the addition of SBR significantly inhibited the formation of RS of nuclei compared to some cytotoxic substances.Conclusion: We found that SBR supplementation should be carefully evaluated when given to poultry. The excess intake of SBR supplementation may cause immunologic inhibition and may negatively affect the development of immune organs.
