| 1 | Antibiotic sensitivity pattern of bacterial pathogens in the intensive care unit of Fatmawati Hospital,Indonesia显示文摘Objective:To evaluate the sensitivity pattern of bacterial pathogens in the intensive care unit(ICU) of a tertiary care of Falmawati Hospital Jakarta Indonesia.Methods:A cross sectional retrospective study of bacterial pathogen was carried out on a total of 722 patients that were admitted to the ICU of Fatmawati Hospital Jakarta Indonesia during January 2009 to March 2010. All bacteria were identified by standard microbiologic methods,and(heir antibiotic susceptibility testing was performed using disk diffusion method.Results:Specimens were collected from 385 patients who were given antimicrobial treatment,of which 249(64.68%) were cultured positive and 136(35.32%) were negative.The most predominant isolate was Pseudomonas aeruginosa(P.aeruginosa)(26.5%) followed by Klebsiella pneumoniae(K.pneumoniae)(15.3%) and Staphylococcus epidermidis(14.9%).P.aeruginosa isolates showed high rate of resistance to cephalexin(95.3%),cefotaxime(64.1%),and ceftriaxone(60.9%).Amikacin was the most effective(84.4%) antibiotic against P.aeruginosa followed by imipenem(81.2%),and meropenem(75.0%).K.pneumoniae showed resistance to cephalexin(86.5%),ceftriaxone(75.7%),ceftazidime(73.0%),cefpirome(73.0%) and cefotaxime(67.9%),respectively.Conclusions:Most bacteria isolated from ICU of Fatmawati Hospital Jakarta Indonesia were resistant to the third generation of cephalosporins,and quinolone antibiotics.Regular surveillance of antibiotic susceptibility pallerns is very important for setting orders to guide the clinician in choosing empirical or directed therapy of infected patients. | Maksum Radji Siti Fauziah Nurgani Aribinuko | 2011 | Asian Pacific Journal of Tropical Biomedicine2011,1,1: | 7 |
| 2 | Microbiological assessment of indoor air of a teaching hospital in Nigeria显示文摘Objective:To investigate the quality of indoor air of different wards and units of Olabisi Onabanjo University Teaching Hospital, Sagamu, to ascertain their contribution to infection rate in the hospital.Methods:The microbial quality of indoor air of nine wards/units of Olabisi Onabanjo University Teaching Hospital, Sagamu, Nigeria was conducted. Sedimentation technique using open Petri-dishes containing different culture media was employed and samplings were done twice daily, one in the morning shortly after cleaning and before influx of people/patients into the wards/units and the other in the evening when a lot of activities would have taken place in these wards. Isolates were identified according to standard methods.Results:Results showed that there was a statistically significant difference(氈2= 6.0167) in the bacteria population of the different sampling time whereas it was not so for fungi population(氈2= 0.2857). Male medical ward(MMW) and male surgical general(MSG) recorded the highest bacterial and fungal growth while the operating theatre(OT) was almost free of microbial burden. The bacteria isolates were Staphylococcus aureus, Klebsiellasp., Bacillus cereus, Bacillus subtilis, Streptococcus pyogenes andSerratia marscenceswhile the fungi isolates includedAspergillus flavus, Penicilliumsp.,Fusariumsp.,Candida albicansandAlternariasp.Staphylococcus aureuswas the predominantly isolated bacterium whilePenicilliumsp. was the most isolated fungus.Conclusions:Though most of the microbial isolates were potential and or opportunistic pathogens, there was no correlation between the isolates in this study and the surveillance report of nosocomial infection during the period of study, hence the contribution of the indoor air cannot be established. | 3 | Antibacterial activity of Lawsonia inermis Linn(Henna) against Pseudomonas aeruginosa显示文摘Objective:To investigate the antibacterial activity of henna(Lawsonia inermis Linn) obtained from different regions of Oman against a wide array of micro-organisms.Methods:fresh henna samples were obtained from different regions of Oman as leaves and seeds,100 g fresh and dry leaves and SO g of fresh and dry seeds were separately soaked in 500 mL of ethanol for three days,respectively,with frequent agitation.The mixture was filtered,and the crude extract was collected.The crude extract was then heated,at 48 ℃ in a water bath to evaporate its liquid content.The dry crude henna extract was then tested for its antibacterial activity using well-diffusion antibiotic susceptibility technique.Henna extracts were investigated for their antibacterial activity at different concentrations against a wide array of different micro-organisms including a laboratory standard bacterial strain of Pseudomonas aeruginosa(NCTC 10662)(A aeruginosa) and eleven fresh clinical isolates of P.aeruginosa obtained from patients attending the Sultan Qaboos University Hospital(SQUH).2-Hydroxy-p-Nathoqinone-Tech(2-HPNT, MW=174.16,C_(10)H_40_3) was included as control(at 50%concentration) along with the henna samples tested.Results:Henna samples demonstrated antibacterial activity against all isolates but the highest susceptibility was against P.aeruginosa with henna samples obtained from Al-sharqyia region.Conclusions:Omani henna from Al-sharqyia region demonstrates high in vitro anti-P. aeruginosa activity compared with many henna samples from different regions of Oman. | Habbal O Hasson SS El-Hag AH Al-Mahrooqi Z Al-Hashmi N Al-Bimani Z MS Al-Balushi Al-Jabri AA | 2011 | Asian Pacific Journal of Tropical Biomedicine2011,1,3: | 3 |

| 5 | Assessment of in vitro sensitivity of Plasmodium vivax fresh isolates显示文摘Objective:To compare the applicability of the SYBK Grcen-Ⅰ assay with the standard schizont maturalion assay,for determination of sensitivity of Plasmodium vivax(P.vivax) to chloroquine and a new antifolale WR 99210.Methods:The study was conducted at Mae Tao Clinic for migrant workers,Tak Province during April 2009 to July 2010.A total of 64 blood samples(1 mL blood collected into sodium heparinized plastic tube) were collected from patients with monoinfection with P.vivax malaria prior to treatment with standard regimen of a 3-day chloroquine. In vitro sensitivity of P.vivax isolates was evaluated by schizont maturation inhibition and SYBR Green-Ⅰ assays.Results:A total of 30 out of 64 blood samples collected from patients with P.vivax malaria were successfully analyzed using both the microscopic schizont maturation inhibition and SYBR Green-I assays.The failure rates of the schizont maturation inhibition assay(50%) and the SYBR Green-I assay(54%) were similar(P=0.51).The median IC_(10)s,IC_(50)s and IC_(90)s of both chloroquine and WR99210 were not significantly different from the clinical isolates of P.vivax tested.Based on the cut-off of 100 nM,the prevalences of chloroquine resistance determined by schizont maturation inhibition and SYBR Green-I assays were 19 and 11 isolates,respectively.The strength of agreement between the two methods was very poor for both chloroquine and WR992I0.Conclusions:On the basis of this condition and its superior sensitivity,the microscopic method appears better than the SYUK Green-I Green assay for assessing in vitro sensitivity of fresh P.vivax isolates to antimalarial drugs. | Poonuch Muhamad Wanna Chacharoenkul Kanchana Rungsihirunrat Ronnatrai Ruengweerayut Kesara NaBangchang | 2011 | Asian Pacific Journal of Tropical Biomedicine2011,1,1: | 2 |
| 6 | Positional cloning of PmCH1357 reveals the origin and allelic variation of the Pm2 gene for powdery mildew resistance in wheat显示文摘Powdery mildew, caused by the biotrophic fungus Blumeria graminis f. sp. tritici(Bgt), is a prevalent disease in common wheat(Triticum aestivum L.) and causes serious yield losses worldwide. We used a map-based approach to clone the major broad-spectrum powdery mildew resistance gene Pm CH1357 from wheat breeding line CH1357. Pm CH1357 was mapped to a 526 kb region containing only Traes CS5 D01 G044600. The Traes CS5 D01 G044600 sequence of the susceptibility allele in Taichung 29(TC29) was identical to that in Chinese Spring, whereas the sequence of the resistance allele in CH1357 was identical to Pm2a previously cloned from the germplasm Ulka/*8Cc. The susceptibility allele in TC29 contained a 7 bp deletion in exon 1, resulting in loss of 856 of the 1277 amino acids in the predicted nucleotide-binding domain leucine-rich repeat containing Pm2a protein.Pm CH1357/Pm2a sequence was also isolated from the Chinese wheat landraces and cultivars that were previously reported to possess the resistance gene Pm2b, Pm2c,PmLX66, or PmND399. The Pm CH1357/Pm2a resistance allele was present in 10 of 495 accessions in core germplasm and contemporary cultivars from China and the USA. | 7 | Succinylated whey protein isolate as a sustained-release excipient of puerarin derivative oral tablets:Preparation,optimization and pharmacokinetics显示文摘This work was done to investigate succinylated commercial whey protein isolate(S-WPI)as an oral sustained-release delivery carrier for puerarin 5(PR-5). The succinylation condi-tions were established for S-WPIs by optimization of single factor study and Box–Beehnkendesign. The effect of succinylation degree on S-WPIs solubility was evaluated. Physicochem-ical properties of S-WPIs dried by different three methods on their flow ability, particle size,morphology and in vitro release behavior were studied. After preparing PR-5 sustained re-lease protein tablets with S-WPIs as the carrier by direct powder compression method, thedrug release were studied in vitro and the oral pharmacokinetics and bioavailability wasevaluated using in vivo dog model. It was observed that concentration of substrate has asignificant effect on succinylation. Release behavior in vitro showed spry dried S-WPIs with100% succinylation rate and 30% drug loading would be applied to the preparation of PR-5 sustained-release protein tablets based on the swelling mechanism(protein loss). Comparedwith PR-5 conventional tablet with oral administration, T max value of PR-5 sustained-releaseprotein tablets was approximately 1.58 fold greater than those of the conventional tablets asfurther evidenced by the significantly prolonged MRT and T 1/2. | 8 | Degradation of Phenolic Compounds in Coal Gasification Wastewater by Biofilm Reactor with Isolated Klebsiella sp.显示文摘This study was conducted to evaluate the degradation of phenolic compounds by one strain isolated from coal gasification wastewater( CGW). 16S rRNA gene sequences homology and phylogenetic analysis showed that the isolate is belonged to the genus Klebsiella sp. The effect of different phenolic compounds on the isolate was investigated by determining OD600and phenoloxidase activity,of which the results showed that the isolate can utilize phenol,4-methyl phenol,3,5-dimethyl phenol and resorcinol as carbon resources. The biofilm reactor( formed by the isolate) can resist the influent concentration of phenolic compounds as high as750 mg /L when fed with synthetic CGW and incubated at optimum conditions. The capacity of improving the biodegradability of CGW through degrading phenolic compounds was testified with fed the biofilm reactor with real CGW.
