1Osteopontin as potential biomarker and therapeutic target in gastric and liver cancers显示文摘Gastric cancer and liver cancer are among the most common malignancies and the leading causes of death worldwide,due to late detection and high recurrence rates.Today,these cancers have a heavy socioeconomic burden,for which a full understanding of their pathophysiological features is warranted to search for promising biomarkers and therapeutic targets.Osteopontin (OPN) is overexpressed in most patients with gastric and liver cancers.Over the past decade,emerging evidence has revealed a correlation of OPN level and clinicopathological features and prognosis in gastric and liver cancers,indicating its potential as an independent prognostic indicator in such patients.Functional studies have verified the potential of OPN knockdown as a therapeutic approach in vitro and in vivo .Furthermore,OPN mediates multifaceted roles in the interaction between cancer cells and the tumor microenvironment,in which many details need further exploration.OPN signaling results in various functions,including prevention of apoptosis,modulation of angiogenesis,malfunction of tumor-associated macrophages,degradation of extracellular matrix,activation of phosphoinositide 3-kinase-Akt and nuclear factor-κB pathways,which lead to tumor formation and progression,particularly in gastric and liver cancers.This editorial aims to review recent findings on alteration in OPN expression and its clinicopathological associations with tumor progression,its potential as a therapeutic target,and putative mechanisms in gastric and liver cancers.Better understanding of the implications of OPN in tumorigenesis might facilitate development of therapeutic regimens to benefit patients with these deadly malignancies.Dong-Xing Cao Zhi-Jie Li Xiao-Ou Jiang Yick Liang Lum Ester Khin Nikki P Lee Guo-Hao Wu John M Luk 2012World Journal of Gastroenterology2012,18,30:29
2甘草次酸靶向肝星状细胞治疗肝纤维化的体内研究显示文摘目的观察以6-磷酸甘露糖修饰的白蛋白(M6P26-HSA)作为特异性载体,将甘草次酸(GA) 靶向释放到肝星状细胞治疗肝纤维化的效果。方法用125I记由M6P26-HSA和GA在体外合成新的偶合物GA-HSA-M6P26,观察其在体内的器官分布情况,用双重免疫组织化学的方法观察星状细胞对GA- HSA-M6P26的选择性摄取;选用Sirius红染色观察GA-HSA-M6P26对肝纤维化时胶原沉积的影响,用定量聚合酶链反应检测GA-HSA-M6P26对Ⅰ型前胶原mRNA表达的影响。结果静脉注射后10min,GA-HSA- M6P26选择性地分布于肝脏,摄取高峰可达(5 5.093±5.404)%。双重免疫组织化学染色证实GA-HSA- M6P26主要被星状细胞选择性摄取,GA-HSA-M6P26治疗后肝脏胶原沉积明显减少,Ⅰ型前胶原和α-平滑肌肌动蛋白mRNA表达明显降低。结论GA-HSA-M6P26可以选择性地分布于肝脏星状细胞,有显著的抗肝纤维化作用。张其胜 John M Luk 张健 田庚元 2005中华肝脏病杂志2005,13,9:22
3Role of LPS/CD14/TLR4-mediated inflammation in necrotizing enterocolitis:Pathogenesis and therapeutic implications显示文摘AIM:To establish the roles of lipopolysaccharide (LPS)/CD14/toll-like receptor 4 (TLR4)-mediated inflammation in a rat model of human necrotizing enterocolitis (NEC).METHODS: Six pairs of intestinal samples from human NEC were collected before and after recovery for histological and molecular analysis of inflammatory cytokines and signaling components. In the rat NEC model, we isolated 10-cm jejunum segments and divided them into six groups (n=6) for sham operation, treatment with LPS, bowel distension, combined bowel distension and LPS stimulation, and two therapeutic groups. The potential eff icacy of a recombinant CD18 peptide and a monoclonal CD14 antibody was evaluated in the latter two groups. The serum and tissue levels of several inflammatory mediators were quantified by real-time polymerase chain reaction, ELISA and immunoblotting.RESULTS: Human acute phase NEC tissues displayed significant increases (P<0.05) in levels of TLR4, CD14, myeloid differentiation protein (MD)-2, tumor necrosis factor (TNF)-α and nuclear factor-κB when compared to those after recovery. The histological and inflammatory picture of human NEC was reproduced in rats that were treated with combined bowel distension and LPS, but not in the sham-operated and other control rats. Serum levels of interleukin-6 and TNF-α were also elevated. The NEC pathology was attenuated by treating the NEC rats with a monoclonal CD14 antibody or an LPS-neutralizing peptide.CONCLUSION:LPS and distension are required to produce the histological and inflammatory features of NEC. 4超顺磁性氧化铁标记大鼠骨髓基质细胞经门静脉移植MRI活体示踪的研究显示文摘目的观察1.5T场强MRI联合动物专用线圈是否可以活体示踪经门静脉移植的纳米级超顺磁性氧化铁颗粒标记的骨髓基质细胞(bonemarrowstromalcellsBMSCs),为介入性门静脉骨髓基质细胞移植治疗终末期肝脏疾病的研究提供进一步的依据。方法供体大鼠5只,梯度密度离心分离BMSCs,纳米级超顺磁性氧化铁颗粒和脂质体转染BMSCs,体外经普鲁士蓝染色和HE染色确定细胞标记率。受体大鼠15只,分为5组,分别为对照组和纳米级超顺磁性氧化铁颗粒标记的骨髓基质细胞经门静脉移植入正常大鼠肝脏后2h、3d、7d及2周组。1.5T场强MRI联合动物专用线圈行T1W、T2W和T2序列扫描,观察肝脏信号改变情况,与对照组比较,并且与组织切片对照。结果纳米级超顺磁性氧化铁颗粒和脂质体转染BMSCs,细胞标记率>95%。经门静脉移植入正常大鼠肝脏后,T2序列扫描显示经标记的BMSCs在肝内显示弥漫性的结节性低信号影,移植后2h到2周均可见到细胞在受体肝脏内存在,组织学切片显示信号缺失部位与铁颗粒标记细胞相一致。结论纳米级超顺磁性铁氧体颗粒标记的大鼠BMSCs经门静脉移植后可以通过1.5T场强行MRI活体示踪,为临床干细胞移植的应用提供可行的示踪方法。王平 王建华 颜志平 胡美玉 徐鹏举 周梅玲 严福华 FAN Sheung-tat LUK John-m 2006中华放射学杂志2006,40,2:16
5血液透析病人生存质量质性研究显示文摘[目的]探讨血液透析病人的生存质量及其影响因素。[方法]采用质性研究中现象学研究法,对15例血液透析病人进行半结构式深入访谈,并运用Colaizzi分析法对资料进行分析。[结果]5个层面8个主题被提炼,生理层面:生理不适的困扰;精神、心理层面:焦虑(等待肾源及对未来的不确定感),抑郁、失落、沮丧(各种消极的情绪影响生存质量);经济层面:沉重的经济负担降低了生存质量;社会层面:无法履行社会职能,家庭支持是面对困难的力量;病人需求层面:完善的社会支持系统及医疗保健制度,相关健康教育知识的普及。[结论]开展以病人为中心的护患交流,提供个性化的护理措施,促进病人生理、心理、社会功能方面的适应,提高病人的生存质量。喻惠丹 Weety Suet-Ching Luk 李斯俭 2008护理研究(中旬版)2008,22,8:14
6Dickkopfs and Wnt/β-catenin signalling in liver cancer显示文摘Liver cancer is the fifth and seventh most common cause of cancer in men and women,respectively.Wnt/β-catenin signalling has emerged as a critical player in both the development of normal liver as well as an oncogenic driver in hepatocellular carcinoma(HCC).Based on the current understanding,this article summarizes the possible mechanisms for the aberrant activation of this pathway with specific focus on HCC.Furthermore,we will discuss the role of dickkopfs(DKKs)in regulating Wnt/β-catenin signalling,which is poorly understood and understudied.DKKs are a family of secreted proteins that comprise at least four members,namely DKK1-DKK4,which act as inhibitors of Wnt/β-catenin signalling.Nevertheless,not all members antagonize Wnt/β-catenin signalling.Their functional significance in hepatocarcinogenesis remains to be further characterized for which these studies should provide new insights into the regulatory role of DKKs in Wnt/β-catenin signalling in hepatic carcinogenesis.Because of the important oncogenic roles,there are an increasing number of therapeutic molecules targetingβ-catenin and the Wnt/β-catenin pathway for potential therapy of HCC.Sarwat Fatima Nikki P Lee John M Luk 2011World Journal of Clinical Oncology2011,2,8:11
7Hazard-evaluation-oriented Moving Horizon Parallel Steering Control for Driver-Automation Collaboration During Automated Driving显示文摘Prompted by emerging developments in connected and automated vehicles, parallel steering control, one aspect of parallel driving, has become highly important for intelligent vehicles for easing the burden and ensuring the safety of human drivers. This paper presents a parallel steering control framework for an intelligent vehicle using moving horizon optimization.The framework considers lateral stability, collision avoidance and actuator saturation and describes them as constraints, which can blend the operation of a human driver and a parallel steering controller effectively. Moreover, the road hazard and the steering operation error are employed to evaluate the operational hazardous of an intelligent vehicle. Under the hazard evaluation,the intelligent vehicle will be mainly operated by the human driver when the vehicle operates in a safe and stable manner.The automated steering driving objective will play an active role and regulate the steering operations of the intelligent vehicle based on the hazard evaluation. To verify the effectiveness of the proposed hazard-evaluation-oriented moving horizon parallel steering control approach, various validations are conducted, and the results are compared with a parallel steering scheme that does not consider automated driving situations. 9Full-space Cloud of Random Points with a Scrambling Metasurface显示文摘With the rapid progress in computer science,including artificial intelligence,big data and cloud computing,full-space spot generation can be pivotal to many practical applications,such as facial recognition,motion detection,augmented reality,etc.These opportunities may be achieved by using diffractive optical elements(DOEs)or light detection and ranging(LIDAR).However,DOEs suffer from intrinsic limitations,such as demanding depth-controlled fabrication techniques,large thicknesses(more than the wavelength),Lambertian operation only in half space,etc.LIDAR nevertheless relies on complex and bulky scanning systems,which hinders the miniaturization of the spot generator.Here,inspired by a Lambertian scatterer,we report a Hermitian-conjugate metasurface scrambling the incident light to a cloud of random points in full space with compressed information density,functioning in both transmission and reflection spaces.Over 4044 random spots are experimentally observed in the entire space,covering angles at nearly 90°.Our scrambling metasurface is made of amorphous silicon with a uniform subwavelength height,a nearly continuous phase coverage,a lightweight,flexible design,and low-heat dissipation.Thus,it may be mass produced by and integrated into existing semiconductor foundry designs.Our work opens important directions for emerging 3D recognition sensors,such as motion sensing,facial recognition,and other applications.Zile Li Qi Dai Muhammad Q.Mehmood Guangwei Hu Boris Luk’yanchuk Jin Tao Chenglong Hao Inki Kim Heonyeong Jeong Guoxing Zheng Shaohua Yu Andrea Alù Junsuk Rho Cheng-Wei Qiu 2018Light(Science & Applications)2018,7,1:6
10Comparing the removal of polycyclic aromatic hydrocarbons in soil after different bioremediation approaches in relation to the extracellular enzyme activities显示文摘A 120-day experiment was conducted to compare the removal of polycyclic aromatic hydrocarbons(PAHs) from agricultural soil after natural attenuation(NA), phytoremediation(P), mycoremediation(M), and plant-assisted mycoremediation(PAM) approaches in relation to the extracellular enzyme activities in soil. The NA treatment removed the total soil PAH content negligibly. The P treatment using maize(Zea mays) enhanced only the removal of low and medium molecular PAHs. The Pleurotus ostreatus cultivated on 30–50 mm wood chip substrate used in M treatment was the most successful in the removal of majority PAHs. Therefore,significantly(p < 0.05) highest total PAH removal by 541.4 μg/kg dw(dry weight)(36%) from all tested M treatments was observed. When using the same fungal substrate together with maize in PAM treatment, the total PAH removal was not statistically different from the previous M treatment. However, the maize-assisted mycoremediation treatment significantly boosted fungal biomass, microbial and manganese peroxidase activity in soil which strongly correlated with the removal of total PAHs. The higher PAH removal in that PAM treatment could be reflected in the following post-harvest time. 11Cryopreservation for delayed circulating tumor cell isolation is a valid strategy for prognostic association of circulating tumor cells in gastroesophageal cancer显示文摘AIM To demonstrate the feasibility of cryopreservation of peripheral blood mononuclear cells(PBMCs) for prognostic circulating tumor cell(CTC) detection in gastroesophageal cancer.METHODS Using 7.5 m L blood samples collected in EDTA tubes from patients with gastroesopheagal adenocarcinoma, CTCs were isolated by epithelial cell adhesion molecule based immunomagnetic capture using the Iso Flux platform. Paired specimens taken during the same blood draw(n = 15) were used to compare number of CTCs isolated from fresh and cryopreserved PBMCs. Blood samples were processed within 24 h to recover the PBMC fraction, with PBMCs used for fresh analysis immediately processed for CTC isolation. Cryopreservation of PBMCs lasted from 2 wk to 25.2 mo(median 14.6 mo). CTCs isolated from pre-treatment cryopreserved PBMCs(n = 43) were examined for associations with clinicopathological variables and survival outcomes.RESULTS While there was a significant trend to a decrease in CTC numbers associated with cryopreserved specimens(mean number of CTCs 34.4 vs 51.5, P = 0.04), this was predominately in samples with a total CTC count of > 50, with low CTC count samples less affected(P = 0.06). There was no significant association between the duration of cryopreservation and number of CTCs. In cryopreserved PBMCs from patient samples prior to treatment, a high CTC count(> 17) was associated with poorer overall survival(OS)(n = 43, HR = 4.4, 95%CI: 1.7-11.7, P = 0.0013). In multivariate analysis, after controlling for sex, age, stage, ECOG performance status, and primary tumor location, a high CTC count remained significantly associated with a poorer OS(HR = 3.7, 95%CI: 1.2-12.4, P = 0.03). 12Effect of aerobic and anaerobic exercises on glycemic control in type 1 diabetic youths显示文摘AIM:To evaluate the long-term effect of aerobic and/or anaerobic exercise on glycemic control in youths with type 1 diabetes.METHODS:Literature review was performed in spring and summer 2014 using Pub Med/MEDLINE,Google Scholar,Scopus,and Science Direct with the following terms:aerobic,anaerobic,high-intensity,resistance,exercise/training,combined with glycemic/metabolic control,glycated haemoglobin A1c(Hb A1c) and type 1diabetes.Only peer-reviewed articles in English were included published in the last 15 years.It was selected from 1999 to 2014.Glycemic control was measured with Hb A1 c.Studies with an intervention lasting at least 12 wk were included if the Hb A1 c was measured before and after the intervention.RESULTS:A total of nine articles were found,and they were published between the years of 2002-2011.The sample size was 401 diabetic youths(166 males and 235 females) with an age range of 10-19 years except one study,in which the age range was 13-30 years.Study participants were from Australia,Tunisia,Lithuania,Taiwan,Turkey,Brazilia,Belgium,Egypt and France.Four studies were aerobic-based,four were combined aerobic and anaerobic programs,and one compared aerobic exercise to anaerobic one.Available studies had insufficient evidence that any type of exercise or combined training would clearly improve the glycemic control in type 1 diabetic youth.Only three(two aerobic-based and one combined) studies could provide a significant positive change in glycemic control.CONCLUSION:The regular physical exercise has several other valuable physiological and health benefits that justify the inclusion of exercise in pediatric diabetes treatment and care.Andrea Lukács László Barkai 2015World Journal of Diabetes2015,6,3:5

15Hepatic tight junctions:From viral entry to cancer metastasis显示文摘The tight junction (TJ) is a critical cellular component for maintenance of tissue integrity, cellular interactions and cell-cell communications, and physiologically functions as the 'great wall' against external agents and the surrounding hostile environment. During the host-pathogen evolution, viruses somehow found the key to unlock the gate for their entry into cells and to exploit and exhaust the host cells. In the liver, an array of TJ molecules is localized along the bile canaliculi forming the blood-biliary barrier, where they play pivotal roles in paracellular permeability, bile secretion, and cell polarity. In pathology, certain hepatic TJ molecules mediate virus entry causing hepatitis infection; deregulation and functional abnormality of the TJ have also been implicated in triggering liver cancer development and metastasis. 18Effects of Organic Matter-Rich Amendments on Selenium Mobility in Soils显示文摘Soil organic matter (SOM) plays an important role in the Se dynamics in soil. The potential effects of vermicompost and digestate as important sources of SOM on selenium (Se) mobility were assessed in this study. Three soils differing in their physicochemical parameters, fluvisol, chernozem, and luvisol, were chosen, and three types of vermicomposts based on various bio-waste materials as digestate (vermicompost 1), kitchen waste with woodchips (vermicompost 2), and garden bio-waste (vermicompost 3) were used due to their high organic matter content. Additionally, digestate samples alone were applied. To evaluate the potential effect of vermicompost application on sorption characteristics of soils, batch sorption experiments were performed. The results showed a predominant effect on Se species in the soils, where selenite sorbed more intensively compared to selenate, regardless of the soil and ameliorative material applied. In the control, the soil sorption ability of selenite tended to decrease in the order: fluvisol > luvisol > chernozem. However, these differences were not significant. Moreover, the effects of the ameliorative materials depended on both soil and amendment used. In fluvisol, all the amendment applications resulted in a decrease in distribution coefficient ( K d values) of Se, whereas in chernozem, this effect was observed only for the digestate-based vermicompost 1. Increasing K d levels were reported in luvisol treated with digestate;the application of garden bio-waste-based vermicompost 3 tended to decrease the K d values. Further studies are required on long-term effects of these amendments on Se mobility in soils and the role of individual organic matter fractions in this context.Petra SMAZíKOVá Luká?
