2肠膜蛋白粉对早期断奶仔猪生产性能及消化道的影响显示文摘90头断奶仔猪(26~29日龄)根据体重随机分为6个处理 ,每个处理3个初始体重水平。试验前3周分别在基础日粮中添加如下6种成分 :0 %AP950(血浆蛋白粉) +0 %DPS(DriedPorcineSaluble ,肠膜蛋白粉)(对照)、2.5 %AP950、2.5 %DPS50、3.5 %DPS50、2.5 %AP950 +2.5 %DPS50和5 %DPS30 ;试验后两周 ,所有处理饲喂普通市售生长猪日粮。各处理日粮中的蛋白质、代谢能、赖氨酸、乳糖和钠浓度均在相同或相近水平上。在试验的第14天 ,从每个处理中随机抽取3头(共18头)试猪屠宰测定其肠绒毛高度、腺窝深度和消化道内胰蛋白酶、糜蛋白酶、淀粉酶、脂肪酶的活性。结果表明 :1.与对照组相比 ,试验前3周日粮中添加2.5 %AP950、2.5 %DPS50、3.5 %DPS50、2.5 %DPS50 +2.5 %AP950和5 %DPS30分别提高日采食量37.0 %、35.9 %、34.8%、37.4 %和41.9 % ;提高日增重69.5 %、52.7 %、81.7 %、61.8 %和82.4 % ;改善饲料转化率18.9 %、11.2 %、25.7 %、15.0 %和21.8 % ;降低腹泻指数42.2 %、20.4 %、40.9 %、34.8 %和37.8 %。2.试验后两周各处理间差异不显著 ,但添加2.5 %DPS50 +2.5 %AP950处理的日采食量和日增重较低。3.对照组酶活性低于其它各组 ,但各处理间消化道酶活性差异不显著。4.高欣 马秋刚 计成 Gu Shao chen Gerry Keller 2001动物营养学报2001,13,2:32
3Evolution of transanal total mesorectal excision for rectal cancer:From top to bottom显示文摘The gold standard for curative treatment of locally advanced rectal cancer involves radical resection with a total mesorectal excision(TME). TME is the most effective treatment strategy to reduce local recurrence and improve survival outcomes regardless of the surgical platform used. However, there are associated morbidities, functional consequences, and quality of life(QoL) issues associated with TME; these risks must be considered during the modern-day multidisciplinary treatment for rectal cancer. This has led to the development of new surgical techniques to improve patient, oncologic, and QoL outcomes. In this work, we review the evolution of TME to the transanal total mesorectal excision(TaTME) through more traditional minimally invasive platforms. The review the development, safety and feasibility, proposed benefits and risks of the procedure, implementation and education models, and future direction for research and implementation of the TaTME in colorectal surgery. 4Down.regulation of E.cadherin enhances prostate cancer chemoresistance via Notch signaling显示文摘Background:The chemoresistance of prostate cancer(PCa)is invariably associated with the aggressiveness and metastasis of this disease.New emerging evidence indicates that the epithelial-to-mesenchymal transition(EMT)may play pivotal roles in the development of chemoresistance and metastasis.As a hallmark of EMT,E-cadherin is suggested to be a key marker in the development of chemoresistance.However,the molecular mechanisms underlying PCa chemoresistance remain unclear.The current study aimed to explore the association between EMT and chemoresistance in PCa as well as whether changing the expression of E-cadherin would affect PCa chemoresistance.Methods:Parental PC3 and DU145 cells and their chemoresistant PC3-Tx R and DU145-Tx R cells were analyzed.PC3-Tx R and DU145-Tx R cells were transfected with E-cadherin-expressing lentivirus to overexpress E-cadherin;PC3 and DU145 cells were transfected with small interfering RNA to silence E-cadherin.Changes of EMT phenotype-related markers and signaling pathways were assessed by Western blotting and quantitative real-time polymerase chain reaction.Tumor cell migration,invasion,and colony formation were then evaluated by wound healing,transwell,and colony formation assays,respectively.The drug sensitivity was evaluated using MTS assay.Results:Chemoresistant PC3-Tx R and DU145-Tx R cells exhibited an invasive and metastatic phenotype that associated with EMT,including the down-regulation of E-cadherin and up-regulation of Vimentin,Snail,and N-cadherin,comparing with that of parental PC3 and DU145 cells.When E-cadherin was overexpressed in PC3-Tx R and DU145-Tx R cells,the expression of Vimentin and Claudin-1 was down-regulated,and tumor cell migration and invasion were inhibited.In particular,the sensitivity to paclitaxel was reactivated in E-cadherin-overexpressing PC3-Tx R and DU145-Tx R cells.When E-cadherin expression was silenced in parental PC3 and DU145 cells,the expression of Vimentin and Snail was up-regulated,and,particularly,the sensitivity to paclitaxel was decreased.Interestingly,Notch-1 expression was up-regulated in PC3-Tx R and DU145-Tx R cells,whereas the E-cadherin expression was down-regulated in these cells comparing with their parental cells.The use ofγ-secretase inhibitor,a Notch signaling pathway inhibitor,significantly increased the sensitivity of chemoresistant cells to paclitaxel.Conclusion:The down-regulation of E-cadherin enhances PCa chemoresistance via Notch signaling,and inhibiting the Notch signaling pathway may reverse PCa chemoresistance.Wenchu Wang Lihui Wang Atsushi Mizokami Junlin Shi Chunlin Zou Jinlu Dai Evan T. 5经肛全直肠系膜切除术治疗直肠癌的优点和局限显示文摘在美国,结直肠癌的发病率和死亡率分别位于恶性肿瘤的第4位和第2位,其中直肠癌约占结直肠癌的1/3。对于可切除直肠癌而言,全直肠系膜切除术(total mesorectal excision,TME)仍然是手术的金标准。但骨盆狭窄、新辅助治疗后肿瘤触摸不清、照明及术野显露困难、远切缘确定困难及吻合器无法放置等解剖、技术和患者方面的因素,会使开腹及腹腔镜TME手术困难。在此环境下,"自下而上"的TME手术——经肛全直肠系膜切除手术(taTME)应运而生。taTME手术结合了腹腔镜及机器人手术的微创化、高清内镜平台的可视化及功能化以及经肛经腹直肠癌TME手术的优势。2010—2013年期间,Lacy等研究者陆续系列报道了taTME手术的实施情况,将该术式推向了高潮。 taTME手术采取改良截石位。手术分为腹部手术和会阴手术两个部分。腹部手术具体步骤可由术者偏好调整,但总体包括游离左侧结肠和脾曲、高位结扎肠系膜下血管和标准的TME手术。会阴部手术先在肿瘤远端采用荷包缝合封闭肠腔,在进行直肠冲洗后,放入操作平台,环形切开肠壁,"自下而上"沿直肠系膜无血管层面进行游离,直至与腹部组会合。标本可以经肛或经造口部位取出。可采取手工缝合或吻合器吻合完成消化道重建。taTME手术的优点和局限如下。 1. taTME手术优点:taTME手术采用经肛进入盆腔的入路方式,即使在低位局部进展期直肠癌、肥胖、骨盆狭窄等操作难度大的手术中,依然可以获得满意的视野显露。同时,由于在肿瘤远端荷包缝合肠壁,故对肿瘤远端切缘的判断更加准确。taTME手术相对简便地完成远端直肠系膜的游离切除,可能更有利于确保手术标本切缘的安全性和淋巴清扫的彻底性,并能得到更高质量的TME手术切除标本,使直肠癌患者的生存获益。国际登记机构的结果显示,taTME术后系膜完整率为96%,环周切缘和远端切缘阳性率分别为2.4%和0.3%。与腹腔镜手术相比,taTME手术时间更短,术后并发症发生率更低。taTME术后吻合口漏发生率和病死率分别为6.7%和2.6%。 2. taTME手术局限:尽管taTME手术有上述优点,现有研究也提示,taTME术后短期肿瘤学安全性满意,但这毕竟是一种新术式,尚缺乏长期随访结果,需要更大样本的对照试验来探究其临床、肿瘤学和功能学结果。此外,对于一个新术式,在实践中会遇见一些新的并发症。特别是"自下而上"的路径,要求外科医生从不同的视角重新认识原来的解剖结构,如直肠后间隙的游离、血管神经束的保护等。此外,也需要重视和预防术中输尿管损伤、盆壁自主神经损伤、骶前静脉损伤和术后盆腔脓肿的发生。上述损伤尤其容易发生在系膜或前列腺层面上,因此,对于有肿瘤史、盆腔放疗史、前列腺切除史或前列腺肥大的患者尤其需要谨慎。必须指出的是,梗阻性直肠肿瘤、急诊手术和T4期肿瘤是taTME的禁忌证。 综上所述,taTME有利于骨盆狭窄的"高难度"低位直肠癌患者的手术切除和标本质量,但作为一种全新的术式,还需要进一步研究其临床、肿瘤学和功能学结果。Deborah S Keller Scott R Steele 楼征 张卫 2018中华胃肠外科杂志2018,21,3:16

7Review of current diagnostic methods and advances in Helicobacter pylori diagnostics in the era of next generation sequencing显示文摘Helicobacter pylori(H.pylori)infection is highly prevalent in the human population and may lead to severe gastrointestinal pathology including gastric and duodenal ulcers,mucosa associated tissue lymphoma and gastric adenocarcinoma.In recent years,an alarming increase in antimicrobial resistance and subsequently failing empiric H.pylori eradication therapies have been noted worldwide,also in many European countries.Therefore,rapid and accurate determination of H.pylori’s antibiotic susceptibility prior to the administration of eradication regimens becomes ever more important.Traditionally,detection of H.pylori and its antimicrobial resistance is done by culture and phenotypic drug susceptibility testing that are cumbersome with a long turn-around-time.Recent advances in diagnostics provide new tools,like real-time polymerase chain reaction(PCR)and line probe assays,to diagnose H.pylori infection and antimicrobial resistance to certain antibiotics,directly from clinical specimens.Moreover,high-throughput whole genome sequencing technologies allow the rapid analysis of the pathogen’s genome,thereby allowing identification of resistance mutations and associated antibiotic resistance.In the first part of this review,we will give an overview on currently available diagnostic methods for detection of H.pylori and its drug resistance and their implementation in H.pylori management.The second part of the review focusses on the use of next generation sequencing technology in H.pylori research.To this end,we conducted a literature search for original research articles in English using the terms“Helicobacter”,“transcriptomic”,“transcriptome”,“next generation sequencing”and“whole genome sequencing”.This review is aimed to bridge the gap between current diagnostic practice(histology,rapid urease test,H.pylori culture,PCR and line probe assays)and new sequencing technologies and their potential implementation in diagnostic laboratory settings in order to complement the currently recommended H.pylori management guidelines and subsequently improve public health.Daniel Pohl Peter M Keller Valentine Bordier Karoline Wagner 2019World Journal of Gastroenterology2019,25,32:15

9华北克拉通北缘(怀来—苏尼特右旗)地壳结构显示文摘2009年,中国地质科学院地质研究所与美国俄克拉荷马大学合作实施了一条长453km的深地震反射、宽角反射与折射、三分量反射地震联合探测剖面.剖面南起怀来盆地,向北依次穿过燕山造山带西缘、内蒙地轴、白乃庙弧带、温都尔庙杂岩带,到达索伦缝合带.其中,宽角反射与折射剖面采用8个0.5~1.5t炸药震源激发,使用300套Texan单分量数字检波器接收,获得了高质量的地震资料.通过资料分析和处理,识别出沉积层及结晶基底的折射波(Pg)、来自上地壳底界面的反射波(Pcp),中地壳底界面的反射波(Plp),莫霍界面的反射波(Pmp)及上地幔顶部的折射波(Pn)等5个震相.分别采用Hole有限差分层析成像和Rayinvr算法对华北克拉通北缘及中亚造山带南部进行了上地壳P波速度结构成像和全地壳二维射线追踪反演成像.结果显示:(1)中亚造山带地壳厚度~40km,变化平缓,低于全球平均造山带地壳平均厚度,可能为造山后区域伸展的结果.阴山—燕山带附近莫霍明显加深,推测其为燕山期造山过程形成的山根,但该山根很可能在后期被改造.(2)测线中部地壳上部速度较高,对应地表大面积花岗岩出露,而下地壳速度较低,速度梯度低,呈通道状,推测其可能曾为古亚洲洋向南俯冲消亡的主动陆缘,并在碰撞后演变为伸展环境下岩浆侵入的通道.(3)华北克拉通北缘与中亚造山带显示出不同速度变化特征,前者变化相对缓而后者则变化剧烈,二者的分界出现在赤峰—白云鄂博断裂附近.李文辉 高锐 Keller Randy 李秋生 侯贺晟 李英康 张世红 2014地球物理学报2014,57,2:13
10综合地球物理资料揭示青藏高原东缘龙日坝断裂带构造属性和大地构造意义显示文摘龙日坝断裂带位于松潘-甘孜地块的东北部,距离其东侧龙门山断裂带大约200km.龙日坝断裂带具有右旋逆冲活动特征,呈北东向展布,与龙门山断裂带基本平行.这条新发现的陆内走滑断裂带由于其特殊的地理位置,目前已引起了广泛的关注.本文利用重力数据的处理和解释,并结合SinoProbe-02深反射地震剖面数据进行由段到面的综合研究,揭示了扬子板块西部边界的伸展和龙日坝断裂带的构造属性.结果显示扬子板块西边界呈不规则分布,而龙日坝断裂带的形成与扬子板块的不规则分布有关.扬子西边界的北段已越过龙门山断裂带延伸至龙日坝断裂带附近,南段则逐渐沿龙门山断裂带分布.龙日坝断裂带代表了扬子板块结晶基底延伸的最西端.印度-欧亚板块碰撞造成了青藏高原物质北东-东向运移.至松潘地块东缘,由于受扬子板块结晶基底的阻挡,在龙日坝断裂带附近产生了应力差而形成了龙日坝断裂带.本文的研究结果将有助于我们更好的了解青藏高原东缘对印度板块与欧亚板块陆陆碰撞所产生的构造响应,以及揭示青藏高原东缘地震灾害发生机制.郭晓玉 高锐 G.Randy Keller 沙爱军 徐啸 王海燕 李文辉 2014地球物理学进展2014,0,5:13

12Genome-wide MicroRNA Expression Profiles in COPD: Early Predictors for Cancer Development显示文摘Chronic obstructive pulmonary disease(COPD) significantly increases the risk of developing cancer. Biomarker studies frequently follow a case-control set-up in which patients diagnosed with a disease are compared to controls. Longitudinal cohort studies such as the COPD-centered German COPD and SYstemic consequences-COmorbidities NETwork(COSYCONET) study provide the patient and biomaterial base for discovering predictive molecular markers. We asked whether microRNA(miRNA) profiles in blood collected from COPD patients prior to a tumor diagnosis could support an early diagnosis of tumor development independent of the tumor type. From 2741 participants of COSYCONET diagnosed with COPD, we selected 534 individuals including 33 patients who developed cancer during the follow-up period of 54 months and 501 patients who did not develop cancer, but had similar age, gender and smoking history. Genome-wide miRNA profiles were generated and evaluated using machine learning techniques. For patients developing cancer we identified nine miRNAs with significantly decreased abundance(two-tailed unpaired t-test adjusted for multiple testing P < 0.05), including members of the miR-320 family. The identified mi RNAs regulate different cancer-related pathways including the MAPK pathway(P = 2.3 x 10^(-5)). We also observed the impact of confounding factors on the generated miRNA profiles, underlining the value of our matched analysis. For selected miRNAs, qRT-PCR analysis was applied to validate the results. 13Review of 500 single incision laparoscopic colorectalsurgery cases-Lessons learned显示文摘Single incision laparoscopic surgery(SILS) is a minimally invasive platform with specific benefits over traditional multiport laparoscopic surgery. The safety and feasibility of SILS has been proven, and the applications continue to grow with experience. After 500 cases at a high-volume, single-institution, we were able to standardize instrumentation and operative steps, as well as develop adaptations in technique to help overcome technical and ergonomic challenges. These technical adaptations have allowed the successful application of SILS to technically difficult patient populations, such as pelvic cases, inflammatory bowel disease cases, and high body mass index patients. The safety and feasibility of SILS for both benign and malignant colorectal disease has been proven. SILS provides the potential for improved cosmesis,postoperative pain,recovery time,and quality of life at the drawback of higher technical skill required. 15Probiotic metabolites from Bacillus coagulans GanedenBC30^(TM) support maturation of antigen-presenting cells in vitro显示文摘AIM:To study the effects of probiotic metabolites on maturation stage of antigen-presenting immune cells.METHODS:Ganeden Bacillus coagulans 30(GBC30) bacterial cultures in log phase were used to isolate the secreted metabolite(MET) fraction.A second fraction was made to generate a crude cell-wall-enriched fraction,by centrifugation and lysis,followed by washing.A preparation of MET was subjected to size exclusion centrifugation,generating three fractions:< 3 kDa,3-30 kDa,and 30-200 kDa and activities were tested in comparison to crude MET and cell wall in primary cultures of human peripheral blood mononuclear cell(PBMC) as a source of antigen-presenting mononuclear phagocytes.The maturation status of mononuclear phagocytes was evaluated by staining with monoclonal antibodies towards CD14,CD16,CD80 and CD86 and analyzed by flow cytometry.RESULTS:Treatment of PBMC with MET supported maturation of mononuclear phagocytes toward both macrophage and dendritic cell phenotypes.The biological activity unique to the metabolites included a reduction of CD14+ CD16+ pro-inflammatory cells,and this property was associated with the high molecular weight metabolite fraction.Changes were also seen for the dendritic cell maturation markers CD80 and CD86.On CD14dim cells,an increase in both CD80 and CD86 expression was seen,in contrast to a selective increase in CD86 expression on CD14bright cells.The co-expression of CD80 and CD86 indicates effective antigen presentation to T cells and support of T helper cell differentiation.The selective expression of CD86 in the absence of CD80 points to a role in generating T regulatory cells.CONCLUSION:The data show that a primary mechanism of action of GBC30 metabolites involves support of more mature phenotypes of antigen-presenting cells,important for immunological decision-making.Kathleen F Benson Kimberlee A Redman Steve G Carter David Keller Sean Farmer John R Endres Gitte S Jensen 2012World Journal of Gastroenterology2012,18,16:8

17Immune mediators in the tumor microenvironment of prostate cancer显示文摘Prostate cancer tissue is composed of both cancer cells and host cells.The milieu of host components that compose the tumor is termed the tumor microenvironment(TME).Host cells can be those derived from the tissue in which the tumor originates(e.g.,fibroblasts and endothelial cells)or those recruited,through chemotactic or other factors,to the tumor(e.g.,circulating immune cells).Some immune cells are key players in the TME and represent a large proportion of non-tumor cells found within the tumor.Immune cells can have both anti-tumor and pro-tumor activity.In addition,crosstalk between prostate cancer cells and immune cells affects immune cell functions.In this review,we focus on immune cells and cytokines that contribute to tumor progression.We discuss T-regulatory and T helper17 cells and macrophages as key modulators in prostate cancer progression.In addition,we discuss the roles of interleukin-6 and receptor activator of nuclear factor kappa-B ligand in modulating prostate cancer progression.This review highlights the concept that immune cells and cytokines offer a potentially promising target for prostate cancer therapy.Jinlu Dai Yi Lu Hernan Roca Jill M. 18龙门山断裂带隆起造山独特性探讨显示文摘龙门山断裂带位于四川盆地西缘,青藏高原东部,为四川盆地与松潘—甘孜地块的接触构造边界。龙门山地区海拔从东侧100 km外四川盆地的500 m突升至3 000 m高度,明显地标注了青藏高原的东部边界,其隆升机制也引起了国内外地质工作者的广泛兴趣,并且提出了多种隆升机制模型。在本次研究中,我们利用SinoProbe-02深反射地震剖面数据对龙门山地区的隆升机制进行研究,从而进一步探讨龙门山地区隆起造山的独特性,并讨论其与传统意义中的造山带的区别,认为龙门山断裂造山带为板块内部构造活动引起岩石圈隆起所形成的。本文的研究结果将使我们更深刻地了解龙门山地区的构造活动特点,并且有助于了解青藏高原东缘对印度-欧亚板块碰撞的构造响应。郭晓玉 高锐 Keller G R 沙爱军 徐啸 王海燕 李文辉 2014地质科学2014,49,4:6

20概率配点法在流域非点源污染不确定性分析中的应用显示文摘农业非点源污染的形势十分严峻,流域水环境模型虽能为非点源污染管理提供决策支撑,但显著的不确定性制约着模型的应用效果。系统的不确定性分析十分重要,而传统的基于Monte Carlo模拟的不确定性分析方法存在着计算成本高昂的缺点。因此,尝试将概率配点法(Probabilistic Collocation Method)应用于流域非点源污染的不确定性分析。研究以美国Newport Bay流域的二嗪磷农药非点源污染为案例,利用WARMF模型进行污染模拟。结果表明,在许多情况下,概率配点法可以通过很小的计算成本获得与传统Monte Carlo模拟方法相近的不确定性分析结果,体现出该方法在非点源污染研究中应用的优势。此外,还揭示了管理因素对于非点源污染模拟及其不确定性分析的影响作用。王伟明 郑一 Arturo Keller 2010农业环境科学学报2010,29,9:6
