| 1 | High temperature ceramic thermal insulation material显示文摘Flexible and lightweight thermal insulation materials with hierarchical microstructures are ubiquitous in thermal management and protection systems.Ceramic aerogels promise high-temperature thermal insulation but lack mechanical robustness,while the fibrous materials with excellent mechanical elasticity display modest thermal insulation.Here we describe flexible hierarchical superhydrophobic ceramic insulation nanocomposites through the densified architectured hierarchical nanostructures,radiative insulation coating,and interfacial cross-linking among composites.The lightweight flexible ceramic nanocomposites exhibit a density of 0.13 g/cm^(3),high-temperature fire resistance with thermal conductivity of 0.024 W/(m·K),and super-hydrophobicity with the water contact angle of 152°.The mechanical robustness and high-temperature thermal insulation of ceramic nanocomposites,together with its soundproof performance,shed light on the low-cost flexible insulation materials manufacturing with scalability for high-temperature thermal insulation applications under high mechanical loading conditions. | Lu An Jason N.Armstrong Yong Hu Yulong Huang Zheng Li Donghui Zhao Jesse Sokolow Zipeng Guo Chi Zhou Shenqiang Ren | 2022 | Nano Research2022,15,7: | 1 |
| 2 | Modified methods for isolation of pancreatic stellate cells from human and rodent pancreas显示文摘Primary cultures of pancreatic stellate cells(PSCs) remain an important basis for in vitro study.However,effective methods for isolating abundant PSCs are currently lacking.We report on a novel approach to isolating PSCs from normal rat pancreases and human pancreatic ductal adenocarcinoma(PDAC) tissue.After anaesthesia and laparotomy of the rat,a blunt cannula was inserted into the pancreatic duct through the anti-mesentery side of the duodenum,and the pancreas was slowly infused with an enzyme solution until all lobules were fully dispersed.The pancreas was then pre-incubated,finely minced and incubated to procure a cell suspension.PSCs were obtained after the cell suspension was filtered,washed and subject to gradient centrifugation with Nycodenz solution.Fresh human PDAC tissue was finely minced into 1×1×l mm^3 cubes with sharp blades.Tissue blocks were placed at the bottom of a culture plate with fresh plasma(EDTA-anti-coagulated plasma from the same patient,mixed with CaCL) sprinkled around the sample.After culture for 5-10 days under appropriate conditions,activated PSCs were harvested.An intraductal perfusion of an enzyme solution simplified the procedure of isolation of rat PSCs,as compared with the multiple injections technique,and a modified outgrowth method significantly shortened the outgrowth time of the activated cells.Our modification in PSC isolation methods significantly increased the isolation efficiency and shortened the culture period,thus facilitating future PSC-related research. | Liangtao Zhao Baobao Cai Zipeng Lu Lei Tian Song Guo Pengfei Wu Dong Qian Qingcheng Xu Kuirong Jiang Yi Miao | 2016 | The Journal of Biomedical Research2016,30,6: | 1 |
| 3 | Function identification of Md TIR1 in apple root growth benefited from the predicted Md PPI network显示文摘Protein–protein interaction(PPI)network analysis is an effective method to identify key proteins during plant development,especially in species for which basic molecular research is lacking,such as apple(Malus domestica).Here,an MdPPI network containing 30806 PPIs was inferred in apple and its quality and reliability were rigorously verified.Subsequently,a rootgrowth subnetwork was extracted to screen for critical proteins in root growth.Because hormone-related proteins occupied the largest proportion of critical proteins,a hormonerelated sub-subnetwork was further extracted from the root-growth subnetwork.Among these proteins,auxin-related M.domestica TRANSPORT INHIBITOR RESISTANT 1(MdTIR1)served as the central,high-degree node,implying that this protein exerts essential roles in root growth.Furthermore,transgenic apple roots overexpressing an MdTIR1 transgene displayed increased primary root elongation.Expression analysis showed that MdTIR1 significantly upregulated auxin-responsive genes in apple roots,indicating that it mediates root growth in an auxin-dependent manner.Further experimental validation revealed that MdTIR1 interacted with and accelerated the degradation of MdIAA28,MdIAA43,andMdIAA46.Thus,MdTIR1-mediated degradation of MdIAAs is critical in auxin signal transduction and root growth regulation in apple.Moreover,our network analysis and high-degree node screening provide a novel research technique for more generally characterizing molecular mechanisms. | Lin Liu Zipeng Yu Yang Xu Cheng Guo Lei Zhang Changai Wu Guodong Yang Jinguang Huang Kang Yan Huairui Shu Chengchao Zheng Shizhong Zhang | 2021 | Journal of Integrative Plant Biology2021,63,4: | 0 |
| 4 | Selecting small molecule DNA aptamers with significant conformational changes for constructing transcriptional switches and biosensors显示文摘Small molecule aptamers discovered by traditional selection methods usually lack conformational changes upon target binding.This limits the use of aptamers as molecular probes for small molecule detection and regulatory elements of genetic circuits.Here,we report a new method called capture and in vitro transcription-systematic evolution of ligands by exponential enrichment(CIVT-SELEX)to select DNA aptamers that can not only bind to small molecule ligands but also undergo significant conformational changes.Through this method,we select a structure-switching aptamer of uridine-5′-diphosphate(UDP).Taking advantage of its conformational changes,we first construct a UDP-responsive transcriptional switch by inserting the aptamer in a genetic circuit and demonstrate that it can respond to the addition of UDP and regulate the transcription of downstream genes.We also build a UDP aptamer-based biosensor that can be used for active glycosyltransferase screening.We believe this method can provide a universal platform for selecting small molecule aptamers with conformational changes and expand the use of aptamers in small molecule detection and genetic regulation. | Shaobin Guo Jingjing Lin Lujie Lin Wen Xu Yan Guo Zipeng Xu Chunhua Lu Xianai Shi Lanlan Chen Huanghao Yang | 2023 | Science China Chemistry2023,66,5: | 0 |
