3Geochemical and isotopic characteristics of volcanic rocks from the northern East China Sea shelf margin and the Okinawa Trough显示文摘Volcanic rocks both from the northern East China Sea (NECS) shelf margin and the northern Okinawa Trough are subalkaline less aluminous,and lower in High Field Strength Elements (HFSE).These rocks are higher in Large Ion Lithophile Elements (LILE),thorium and uranium contents,positive lead anomalies,negative Nb-Ta anomalies,and enrichment in Light Rare Earth Elements (LREE).Basalts from the NECS shelf margin are akin to Indian Ocean Mid-Ocean Ridge Basalt (MORB),and rhyolites from the northern Okinawa Trough have the highest 207 Pb/ 204 Pb and 208 Pb/ 204 Pb ratios.The NECS shelf margin basalts have lower 87 Sr/ 86 Sr ratios,ε N d and σ 18 O than the northern Okinawa Trough silicic rocks.According to 40 K– 40 Ar isotopic ages of basalts from the NECS shelf margin,rifting of the Okinawa Trough may have been active since at least 3.65–3.86 Ma.The origin of the NECS shelf margin basalt can be explained by the interaction of melt derived from Indian Ocean MORB-like mantle with enriched subcontinental lithosphere.The basalts from both sides of the Okinawa Trough may have a similar origin during the initial rifting of the Okinawa Trough,and the formation of basaltic magmas closely relates to the thinning of continental crust.The source of the formation of the northern Okinawa Trough silicic rocks was different from that of the middle Okinawa Trough,which could have been generated by the interaction of basaltic melt with an enriched crustal component.From the Ryukyu island arc to East China,the Cenozoic basalts have apparently increasing trends of MgO contents and ratios of LREE to Heavy Rare Earth Elements (HREE),suggesting that the trace element variabilities of basalts may have been influenced by the subduction of the Philippine Sea plate,and that the effects of subduction of the Philippine Sea plate on the chemical composition of basaltic melts have had a decreasing effect from the Ryukyu island arc to East China.ZENG Zhigang YU Shaoxiong WANG Xiaoyuan FU Yongtao YIN Xuebo ZHANG Guoliang WANG Xiaomei CHEN Shuai 2010Acta Oceanologica Sinica2010,29,4:19
4Ti/(Ti,Cr)N/CrN multilayer coated 316L stainless steel by arc ion plating as bipolar plates for proton exchange membrane fuel cells显示文摘Arc ion plating(AIP) is applied to form Ti/(Ti,Cr)N/Cr N multilayer coating on the surface of 316 L stainless steel(SS316L) as bipolar plates for proton exchange membrane fuel cells(PEMFCs). The characterizations of the coating are analyzed by scanning electron microscopy(SEM) and X-ray diffraction(XRD). Interfacial contact resistance(ICR) between the coated sample and carbon paper is 4.9 m cm^2 under 150 N/cm^2,which is much lower than that of the SS316 L substrate. Potentiodynamic and potentiostatic tests are performed in the simulated PEMFC working conditions to investigate the corrosion behaviors of the coated sample. Superior anticorrosion performance is observed for the coated sample, whose corrosion current density is 0.12 μA/cm2. Surface morphology results after corrosion tests indicate that the substrate is well protected by the multilayer coating. 5Overexpression of a dominant-negative mutant of SIRT1 in mouse heart causes cardiomyocyte apoptosis and early-onset heart failure显示文摘SIRT1,a mammalian ortholog of yeast silent information regulator 2(Sir2),is an NAD+-dependent protein deacetylase that plays a critical role in the regulation of vascular function.The current study aims to investigate the functional significance of deacetylase activity of SIRT1 in heart.Here we show that the early postnatal hearts expressed the highest level of SIRT1deacetylase activity compared to adult and aged hearts.We generated transgenic mice with cardiac-specific expression of a dominant-negative form of the human SIRT1(SIRT1H363Y),which represses endogenous SIRT1 activity.The transgenic mice displayed dilated atrial and ventricular chambers,and died early in the postnatal period.Pathological,echocardiographic and molecular phenotype confirmed the presence of dilated cardiomyopathy.Terminal deoxynucleotidyl transferase-mediated dUTP nick-end-labeling analysis revealed a greater abundance of apoptotic nuclei in the hearts of transgenic mice.Furthermore,we show that cardiomyocyte apoptosis caused by suppression of SIRT1 activity is,at least in part,due to increased p53acetylation and upregulated Bax expression.These results indicate that dominant negative form of SIRT1(SIRT1H363Y)overexpression in mouse hearts causes cardiomyocyte apoptosis and early-onset heart failure,suggesting a critical role of SIRT1 in preserving normal cardiac development during the early postnatal period.MU WenLi ZHANG QingJun TANG XiaoQiang FU WenYan ZHENG Wei LU YunBiao LI HongLiang WEI YuSheng LI Li SHE ZhiGang CHEN HouZao LIU DePei 2014Science China(Life Sciences)2014,57,9:12
6Sources and characteristics of fine particles over the Yellow Sea and Bohai Sea using online single particle aerosol mass spectrometer显示文摘Marine aerosols over the East China Seas are heavily polluted by continental sources. During the Chinese Comprehensive Ocean Experiment in November 2012, size and mass spectra of individual atmospheric particles in the size range from 0.2 to 2.0 μm were measured on board by a single particle aerosol mass spectrometer(SPAMS). The average hourly particle number(PN)was around 4560 ± 3240 in the South Yellow Sea(SYS), 2900 ± 3970 in the North Yellow Sea(NYS), and 1700 ± 2220 in the Bohai Sea(BS). PN in NYS and BS varied greatly over 3 orders of magnitude, while that in SYS varied slightly. The size distributions were fitted with two log-normal modes. Accumulation mode dominated in NYS and BS, especially during episodic periods. Coarse mode particles played an important role in SYS. Particles were classified using an adaptive resonance theory based neural network algorithm(ART-2a). Six particle types were identified with secondary-containing, aged sea-salt, soot-like, biomass burning, fresh sea-salt,and lead-containing particles accounting for 32%, 21%, 18%, 16%, 4%, and 3% of total PN,respectively. Aerosols in BS were relatively enriched in particles from anthropogenic sources compared to SYS, probably due to emissions from more developed upwind regions and indicating stronger influence of continental outflow on marine environment. Variation of source types depended mainly on origins of transported air masses. This study examined rapid changes in PN, size distribution and source types of fine particles in marine atmospheres. 7Decolorization on Indigo Dyeing Wastewater by Laccase from Coriolus versicolor显示文摘[Object]The study aimed to discuss the decolorization on indigo dyeing wastewater by laccase from Coriolus versicolor.[Method]Firstly,the effects of temperature,pH,indigo concentration,HBT concentration,laccase dosage on the decolorization of indigo dyeing wastewater by laccase /HBT,and then the synergism of laccase and acid cellulase was analyzed.[Result]Using ABTS as the substrate,the kinetic parameters,K m and V max,were 0.318 mmol/L and 0.035 5 mmol/(L·min) respectively.The decolorization rate of indigo reached 96.5% when the laccase acted on indigo for 40 min with HBT as an introducer at temperature 50 ℃,pH =4.5,indigo concentration 100 mg /L,HBT concentration 0.1% and laccase dosage 100 IU /L.Due to the synergism of laccase and acid cellulase during the bio-finishing of blue jeans,the backstaining degree of blue jeans reduced by 85% when the amount of laccase added was 15 000 IU /kg.Menawhile,the synergism of the laccase and acid cellulase decreased indigo concentration in wastewater by 83.8%.[Conclusion] The laccase from Coriolus versicolor had a good prospect in the bio-finishing of blue jeans and the decolorization of indigo dyeing wastewater.Enli Gao Weijia Zhong Xiaolan Fu Fushan Chen Zhigang Ye 2013Meteorological and Environmental Research2013,4,1:10
8Simulation and Experimental Investigation of Jetting Penetrator Charge at Large Stand-off Distance显示文摘In order to study and apply the penetration performance of jetting penetrator charge at long stand-off distance, three jetting penetrator charges(JPC), including spherical cone liner, truncated wide-angle liner and spherical segment liner, are designed. The numerical simulation analysis of the formation, elongation and penetration processes of rod-like jet is conducted by using LS-DYNA software. And the penetrating test is carried out at long stand-off distance. The test results show that the rod-like jet formed by the optimized spherical segment liner can pierce through a 90mm thick 45# steel target at 20 charge diameters(CD) stand-off distance when the charge detonation mode is a central point initiation, and the penetration depth can be up to 1.6CD. 9Numerical study on Rayleigh-Taylor effect on cylindrically converging Richtmyer-Meshkov instability显示文摘Evolution of a two-dimensional air/SF6 single-mode interface is numerically investigated by an upwind CE/SE method under a cylindrically converging circumstance. The Rayleigh-Taylor effect caused by the flow deceleration on the phase inversion(RTPI)is highlighted. The RTPI was firstly observed in our previous experiment, but the related mechanism remains unclear. By isolating the three-dimensional effect, it is found here that the initial amplitude(a0), the azimuthal mode number(k0) and the re-shocking moment are the three major parameters which determine the RTPI occurrence. In the variable space of(k0, a0), a critical a0 for the RTPI occurrence is solved for each k0, and there exists a threshold value of k0 below which the RTPI will not occur no matter what a0 is. There exists a special k0 corresponding to the largest critical a0, and the reduction rule of critical a0 with k0 can be well described by an exponential decay function. The results show that the occurrence of the RTPI requires a small a0 which should be less than a critical value, a large k0 which should exceed a threshold, and a right impinging moment of the re-shock which should be later than the RTPI occurrence. Finally, the effects of the incident shock strength, the density ratio and the initial position of the interface on the threshold value of k0 and on the maximum critical a0 are examined. 11Adipocyte-derived SFRP5 inhibits breast cancer cells migration and invasion through Wnt and epithelial-mesenchymal transition signaling pathways显示文摘Objective:Obesity is closely associated with metastasis in breast cancer patients.Secreted frizzled-related protein 5(SFRP5),one of the novel adipokines with anti-inflammatory properties,is associated with obesity.This study aims to study the role of SFRP5 in the crosstalk between obesity and breast cancer metastasis and identify the underlying mechanism.Methods:3T3-L1 pre-adipocytes were differentiated to mature adipocytes and a hypertrophic adipocyte model was induced with palmitic acid(PA).Cell motility was measured in MDA-MB-231 and MCF-7 breast cancer cells co-cultured with adipocytes conditioned medium(CM)with or without SFRP5 protein.Wnt and epithelialmesenchymal transition(EMT)signal pathways were investigated by western blot.Circulating SFRP5 level in 218 breast cancer patients and the association with clinicopathologic characteristics of breast cancer were further determined.Online databases ENCORI and PREDICT Plus were used to exam the link between SFRP5 and prognosis.Results:Reduced SFRP5 level was detected in the hypertrophic adipocyte model.Recombinant SFRP5 protein inhibited MDA-MB-231 and MCF-7 cells invasion and migration induced by PA-treated adipocyte CM,and SFRP5 inhibition by specific antibody reversed the effect of SFRP5.Furthermore,SFRP5 significantly inhibited Wnt and downstream EMT in breast cancer cells.Low circulating SFRP5 level correlated with body mass index(BMI),lymph node(LN)metastasis,TNM stage and high Ki67 expression in breast cancer patients.Increased SFRP5 level was associated with favorable predicted survival.Kaplan-Meier curves showed high SFRP5 level in tumor tissue was associated with better outcome of breast cancer patients.Conclusions:Our findings demonstrated SFRP5 is a vital adipokine that mediates the crosslink between obesity and the metastatic potential of breast cancer.Promotion of SFRP5 expression in the adipose microenvironment may represent a novel approach for preventing breast cancer metastasis.Wenzhong Zhou Chunmiao Ye Liang Li Liyuan Liu Fei Wang Lixiang Yu Fei Zhou Yujuan Xiang Yongjiu Wang Gengshen Yin Zhongbing Ma Qinye Fu Qiang Zhang Dezong Gao Shuya Huang Zhigang Yu 2020Chinese Journal of Cancer Research2020,32,3:5
12Chemical formation and source apportionment of PM_(2.5) at an urban site at the southern foot of the Taihang mountains显示文摘The region along the Taihang Mountains in the North China Plain(NCP) is characterized by serious fine particle pollution. To clarify the formation mechanism and controlling factors, an observational study was conducted to investigate the physical and chemical properties of the fine particulate matter in Jiaozuo city, China. Mass concentrations of the water-soluble ions(WSIs) in PM_(2.5) and gaseous pollutant precursors were measured on an hourly basis from December 1, 2017, to February 27, 2018. The positive matrix factorization(PMF) method and the FLEXible PARTicle(FLEXPART) model were employed to identify the sources of PM_(2.5). The results showed that the average mass concentration of PM_(2.5) was 111 μg/m^(3) during the observation period. Among the major WSIs, sulfate, nitrate, and ammonium(SNA) constituted 62% of the total PM_(2.5) mass, and NO_(3)^(-) ranked the highest with an average contribution of 24.6%. NH_(4)^(+) was abundant in most cases in Jiaozuo. According to chemical balance analysis, SO_(2)-4, NO_(3)^(-), and Cl^(-) might be present in the form of(NH_4)_(2)SO_4, NH_4NO_3, NH_4Cl, and KCl. The liquid-phase oxidation of SO_(2) and NO_(2) was severe during the haze period. The relative humidity and pH were the key factors influencing SO_(4)^(2-) formation. We found that NO_(3)^(-) mainly stemmed from homogeneous gas-phase reactions in the daytime and originated from the hydrolysis of N_(2)O_5 in the nighttime, which was inconsistent with previous studies. 13Magnetic resonance imaging tracing of transplanted bone marrow mesenchymal stem cells in a rat model of cardiac arrest-induced global brain ischemia显示文摘<正>BACKGROUND:Numerous studies have shown that magnetic resonance imaging(MRI) can detect survival and migration of super paramagnetic iron oxide-labeled stem cells in models of focal cerebral infarction. OBJECTIVE:To observe distribution of bone marrow mesenchymal stem cells(BMSCs) in a rat model of global brain ischemia following cardiac arrest and resuscitation,and to investigate the feasibility of tracing iron oxide-labeled BMSCs using non-invasive MRI. DESIGN,TIME AND SETTING:The randomized,controlled,molecular imaging study was performed at the Linbaixin Medical Research Center,Second Affiliated Hospital,Sun Yat-sen University,and the Institute of Cardiopulmonary Cerebral Resuscitation,Sun Yat-sen University, China from October 2006 to February 2009. MATERIALS:A total of 40 clean,Sprague Dawley rats,aged 6 weeks and of either gender,were supplied by the Experimental Animal Center,Sun Yat-sen University,China,for isolation of BMSCs. Feridex(iron oxide),Gyroscan Inetra 1.5T MRI system,and cardiopulmonary resuscitation device were used in this study. METHODS:A total of 30 healthy,male Sprague Dawley rats,aged 6 months,were used to induce ventricular fibrillation using alternating current.After 8 minutes,the rats underwent 6-minute chest compression and mechanical ventilation,followed by electric defibrillation,to establish rat models of global brain ischemia due to cardiac arrest and resuscitation.A total of 24 successful models were randomly assigned to Feridex-labeled and non-labeled groups(n=12 for each group).At 2 hours after resuscitation,5×10~6 Feridex-labeled BMSCs,with protamine sulfate as a carrier,and 5×10~6 non-labeled BMSCs were respectively transplanted into both groups of rats through the right carotid artery(cells were harvested in 1 mL phosphate buffered saline). MAIN OUTCOME MEASURES:Feridex-labeled BMSCs were observed by Prussian blue staining and electron microscopy.Signal intensity,celluar viability,and proliferative capacity of BMSCs were measured using MRI,Trypan blue test,and MTT assay,respectively.Distribution of transplanted cells was observed in rats utilizing MRI and Prussian blue staining prior to and 1,3,7,and 14 days after transplantation. RESULTS:Prussian blue staining displayed many blue granules in the Feridex-labeled BMSCs. High density of iron granules was observed in the cytoplasm under electron microscopy.According to MRI results,and compared with the non-labeled group,the signal intensity was decreased in the Feridex-labeled group(P<0.05).The decrease was most significant in the 50μg/mL Feridex-labeled group(P<0.01).There were no significant differences in celluar viability and proliferation of BMSCs between the Feridex-labeled and non-labeled groups after 1 week(P>0.05). Low-signal lesions were detected in the rat hippocampus and temporal cortex at 3 days after transplantation.The low-signal lesions were still detectable at 14 days,and positively stained cells were observed in the hippocampus and temporal cortex using Prussian blue staining.There were no significant differences in signal intensity in the non-labeled group. 14Physical-biogeochemical interactions and potential effects on phytoplankton and Ulva prolifera in the coastal waters off Qingdao(Yellow Sea, China)显示文摘In recent years, the spectacular massive green tide of Ulva prolifera has become a recurrent phenomenon appearing every summer in the coastal waters off Qingdao(Yellow Sea, China), attracting the attention of scientists and local government. Based on multidisciplinary data collected during summer and winter, this study focuses on the hydrological characteristics and regional biogeochemical processes in coastal waters off Qingdao.The results show that the boundary of the Yellow Sea Cold Water Mass(YSCWM) can reach the Qingdao coastal region in summer and is locally raised to the upper layers to form coastal upwelling beyond tidal mixing and favorable wind. The regional summer upwelling off the Qingdao coast effectively enriches the nutrient concentrations in the upper water column and thus promotes growth of phytoplankton but reduces the dissolved oxygen(DO) concentration and pH value in the bottom. The regional summer upwelling off Qingdao coast may facilitate the growth and regional blooming of the U. prolifera that migrate to this region with the southerly wind.Additionally, the effects of the front on the aggregation of U. prolifera may be significant. In winter, the Yellow Sea Warm Current(YSWC) extends and spreads along the offshore region off the Subei Shoal towards the Qingdao coastal sea. This tongue-shaped warm water meets the cold coastal water off Qingdao, which leads to the formation of a physical front. As a consequence, remarkable fronts of nutrient and chlorophyll a(Chl a) also form between the shoreward warm water and the cold coastal water. 16Spatiotemporal variability of physical-biogeochemical processes and intrinsic correlations in the semi-enclosed South Yellow Sea显示文摘Investigating the spatiotemporal variability of biogeochemical processes and ecological responses under multiple physical controls in shelf seas is of great importance for obtaining an in-depth understanding of marine ecosystem.Based on a compiled data set of historical observations and remote sensing data,the spatiotemporal variability and heterogeneity of physical-biogeochemical processes in the semi-enclosed South Yellow Sea(SYS)are investigated,and the intrinsic connectivity among different subregions and the associated mechanisms are examined.The results show that the seasonal alternation between southward transport in cold seasons and upwelling-induced vertical delivery in warm seasons is the primary physical control of the biogeochemical processes and primary production off Shidao and in the area adjacent to the Haizhou Bay.The northeastward expansion of coastal waters in the Subei Shoal constitutes an important physical driver for the offshore transport of Ulva prolifera in summer.Stratification significantly affects the biogeochemical processes in the Yellow Sea Cold Water Mass(YSCWM)-dominated area during warm seasons,and nutrients can accumulate in bottom waters from spring to autumn,making the Yellow Sea Cold Water Mass(YSCWM)be an important nutrient pool.Upwelling around the YSCWM boundary in the stratified season leads to consistency among the high chlorophyll a(Chl a)area,high primary productivity region and low-temperature upwelling zone.During cold seasons,the interactions of the southward cold waters in the western nearshore area and the northward warm waters in the central region lead to an“S”-shaped front in the SYS.In summer,upwelling can extract nutrients from the YSCWM;thus,the biogeochemical-ecological processes inside the cold-water mass and in the frontal zone are well connected via upwelling,and three typical physical-biogeochemical coupling regions are generated,namely,the Shidao coast,the area beyond the Haizhou Bay and the area off the Subei Shoal.This work refines and integrates studies on regional oceanography in the SYS and provides a comprehensive and systematic framework of physical-biogeochemical-ecological processes.Qinsheng Wei Baodong Wang Mingzhu Fu Junchuan Sun Qingzhen Yao Ming Xin Zhigang Yu 2020Acta Oceanologica Sinica2020,39,10:3

18M2-specific reduction of CD1d switches NKT cell-mediated immune responses and triggers metaflammation in adipose tissue显示文摘Metaflammation is responsible for several metabolic syndromes,such as type 2 diabetes.However,the mechanisms by which metabolic disorders trigger metaflammation remain unclear.We identified a cell type-specific downregulation of CD1d expression in M2 macrophages during the progression of obesity prior to the onset of inflammation in visceral adipose tissues.A reduction in CD1d expression influenced the ability of M2 macrophages to present antigens and caused a change in antigen-presenting cells from M2 macrophages to M1 macrophages.With CD1d conditional knockout(KO)mice,we further demonstrated that natural killer T(NKT)cell activation by M2 macrophages inhibited metaflammation and insulin resistance by promoting Th2 responses and M2 polarization in visceral adipose tissues of obese mice,whereas NKT cell activation by M1 macrophages exacerbated metaflammation and insulin resistance by promoting Th1 responses and inhibiting M2 polarization.Our results suggest that an M2-specific reduction of CD1d is an initiating event that switches NKT cell-mediated immune responses and disrupts the immune balance in visceral adipose tissues in obese mice.Huimin Zhang Rufeng Xue Shasha Zhu Sicheng Fu Zuolong Chen Rongbin Zhou Zhigang Tian Li Bai 2018Cellular & Molecular Immunology2018,15,5:3

20ZmcrtRB3 Encodes a Carotenoid Hydroxylase that Affects the Accumulation of α-carotene in Maize Kernel显示文摘α-carotene is one of the important components of pro-vitamin A,which is able to be converted into vitamin A in the human body.One maize(Zea mays L.) ortholog of carotenoid hydroxylases in Arabidopsis thaliana,ZmcrtRB3,was cloned and its role in carotenoid hydrolyzations was addressed.ZmcrtRB3 was mapped in a quantitative trait locus(QTL) cluster for carotenoid-related traits on chromosome 2(bin 2.03) in a recombinant inbred line(RIL) population derived from By804 and B73.Candidate-gene association analysis identified 18 polymorphic sites in ZmcrtRB3 significantly associated with one or more carotenoid-related traits in 126 diverse yellow maize inbred lines.These results indicate that the enzyme ZmcrtRB3 plays a role in hydrolyzing both α-and β-carotenes,while polymorphisms in ZmcrtRB3 contributed more variation in α-carotene than that in β-carotene.Two single nucleotide polymorphisms(SNPs),SNP1343 in 5 untranslated region and SNP2172 in the second intron,consistently had effects on α-carotene content and composition with explained phenotypic variations ranging from 8.7% to 34.8%.There was 1.7-to 3.7-fold change between the inferior and superior haplotype for α-carotene content and composition.Thus,SNP1343 and SNP2172 are potential polymorphic sites to develop functional markers for applying marker-assisted selection in the improvement of pro-vitamin A carotenoids in maize kernels.Yi Zhou Yingjia Han Zhigang Li Yang Fu Zhiyuan Fu Shutu Xu Jiansheng Li Jianbing Yan Xiaohong Yang 2012Journal of Integrative Plant Biology2012,54,4:2
