| 1 | Characterization of root architecture in an applied core collection for phosphorus efficiency of soybean germplasm显示文摘In the present study, we constructed an “ap- plied core collection” for phosphorus (P) efficiency of soy- bean germplasm using a GIS-assisted approach. Systematic characterization and comparative analysis of root architec- ture were conducted to evaluate the relationship between root architecture and P efficiency and its possible evolution- ary pattern. Our results found that: i) root architecture was closely related to P efficiency in soybean. Shallow root archi- tecture had better spatial configuration in the P-rich culti- vated soil layer hence higher P efficiency and soybean yield; ii) there was a possible co-evolutionary pattern among shoot type, root architecture and P efficiency. The bush cultivated soybean had a shallow root architecture and high P efficiency, the climbing wild soybean had a deep root architecture and low P efficiency, while the root architecture and P efficiency of semi-wild soybean were intermediate between cultivated and wild soybean; iii) P availability regulated root architec- ture. Soybean roots became shallower with P addition to the topsoil, indicating that the co-evolutionary relationship be- tween root architecture and P efficiency might be attributed to the long-term effects of topsoil fertilization. | 2 | Expression of fragile histidine triad in primary hepatocellular carcinoma and its relation with cell proliferation and apoptosis显示文摘AIM: To evaluate the expression of fragile histidine triad (FHIT) gene protein, product of a candidate tumor suppressor,and to investigate the relationship between FHIT, cell apoptosis and proliferation, and pathological features of primary hepatocellular carcinoma (HCC).METHODS: Forty-seven HCC and ten normal liver specimens were collected during surgical operation between 2001and 2003. FHIT and proliferating cell nuclear antigen (PCNA)expression were detected by immunohistochemistry, and apoptotic level was evaluated by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) assay on the tissue sections.RESULTS: All normal liver tissues showed a strong expression of FHIT, whereas 28 of 47 (59.6%) carcinomas showed a significant loss or absence of FHIT expression (P = 0.001).The proportion of reduced FHIT expression in those carcinomas at stages Ⅲ-Ⅳ (70.6%) and in those with extrahepatic metastasis (86.7%) showed an increasing trend compared with those at stages Ⅰ-Ⅱ (30.8%, P= 0.013) and those without metastasis (46.9%, P = 0.010) respectively. Apoptotic incidence in advanced TNM stage carcinoma and those with positive FHIT expression was higher than that in early stage carcinoma (P = 0.030) and in those with negative FHIT expression (P = 0.044) respectively. The proliferating potential of hepatocellular carcinoma was associated with FHIT expression (P = 0.016) and the aggressive feature (P = 0.019). Kaplan-Meier analysis demonstrated that the survival time of these 47 patients correlated with TNM stage,FHIT expression and metastasis.CONCLUSION: There is marked loss or absence of FHIT expression, as well as abnormal apoptosis-proliferation balance in HCC. | 3 | Expression of p57^(kip2) and its relationship with clinicopathology, PCNA and p53 in primary hepatocellular carcinoma显示文摘AIM: To investigate the expression of p57kip2 and its relationship with dinicopathology, PCNA and p53 in primary hepatocellular carcinoma (HCC).METHODS: Expression of p57kip2, PCNA and p53 in tumor tissues from 32 patients with HCC and 10 liver tissues of normal persons was detected with Elivision immunohistochemical technique.RESULTS: The p57kip2 protein positive-expression rate in HCC was 56.25%, lower than that in normal tissues (100%, P<0.05). The reduced expression of p57kip2 protein correlated significantly with moderate or low differentiation of tumor cells (P = 0.007 <0.05), high clinical stage (P = 0.041 <0.05) and poor prognosis (P = 0.036 <0.05),but did not correlate significantly with metastasis, tumor size, level of AFP and age (P>0.05). The PCNA positiveexpression rate was 56.25%, which was correlated significantly with the expression of p57kip2 (P= 0.025<0.05).The p53 positive-expression rate was 46.88%, which was not correlated significantly with the expression of p57kip2(P>0.05).CONCLUSION: There is a marked loss or absence of p57kip2 expression and high expression of PCNA in HCC,which are involved in carcinogenesis and development of HCC. | 4 | Expression and altered subcellular localization of the cyclin-dependent kinase inhibitor p27^(kip1) in hepatocellular carcinoma显示文摘AIM: To investigate p27 expression in hepatocellular carcinoma (HCC), adjacent nontumoral and normal liver tissues, and to verify whether the subcellular localization of p27 was altered in HCC.METHODS: The level of p27 in tumoral, nontumoral, and normal liver tissues were assessed by immunohistochemical (IHC) analysis. Parallel immunostaining was done for proliferating cell nuclear antigen (PCNA) to evaluate cell proliferation.RESULTS: The labeling index (LI) of p27 in tumoral lesions was significantly lower than that in adjacent nontumoral lesions (t=2.444, P=0.017) and normal controls (t=-2.268,P=0.029). The LI of p27 significantly decreased in patients with massive type (t=-2.227, P=0.037) and infiltration (t=2.197, P=0.036). The prognosis of patients with higher p27 LI was longer than that of patients with lower p27 LI (P=0.0247, log-rank test). The LI of PCNA was significantly higher in HCC than that in adjacent nontumoral lesions (t=2.092, P=0.041) and normal controls (t=-3.533, P=0.002).There was no significant correlation between p27 expression and cell proliferation in tumor samples. The level of p27 in the cytoplasmic fraction was higher in tumoral and nontumoral liver tissues, and was associated with clinical stage (t=-2.520, P=0.029) and the degree of invasion (t=2.640, P=0.019). Survival analysis showed that p27 was an independent prognosis marker for HCC patients.CONCLUSION: These results suggest that p27 underexpressing in patients with HCC is closely associated with infiltration, metastasis, and prognosis. | 5 | Cell proliferation, apoptosis and the related regulators p27, p53 expression in hepatocellular carcinoma显示文摘AIM: To investigate the expression of cell apoptosis,proliferation and the related regulators p27, p53 in hepatocellular carcinoma (HCC).METHODS: The expression of p27, p53, proliferating cell nuclear antigen (PCNA) and apoptosis in 47 HCC specimens and 42 surrounding non-cancerous tissues were detected by the immunohistochemistry and terminal deoxy-nudeotidyl transferase-mediated nick end labeling (TUNEL) technique.Meanwhile, the clinical significance of them was analyzed combining with the clinicopathological factors and followup data.RESULTS: (1) The average proliferating index and apoptotic index in HCC were significantly higher than that in adjacent liver tissues. The proliferating index was associated with extrahepatic metastasis. The apoptotic index was significantly lower in TNM stage Ⅰ-Ⅱ than in stage Ⅲ-Ⅳ. The proliferating index of groups with p53-/p27+ was significantly lower than that in group with p53+/p27- (P = 0.030); (2) The level of p27 in the cytoplasmic fraction was higher in non-tumoral liver tissues and was associated with clinical stage; (3) Survival analysis showed advanced stage (P = 0.031) and with extrahepatic metastasis (P = 0.045) was significantly associated with shorter survival. In addition, the prognosis of patients with p53-/p27+ was longer than that of patients with p53+/p27- (P = 0.0356).CONCLUSION: The p53 mutation and decreased p27 expression might be involved in the imbalance of proliferation and apoptosis in HCC. Cytoplasmic displacement might lead to the inactivation of p27 protein in HCC cells and acts early during carcinogenesis of HCC. | 6 | Expression and clinical significance of TAp73α, p53, PCNA and apoptosis in hepatocellular carcinoma显示文摘AIM: To study the prognostic role of TAp73α, p53, proliferating cell nuclear antigen (PCNA) and apoptosis inpatients with hepatocellular carcinoma (HCC) atter surgical tumor ablation.METHODS: Forty-seven human resected HCC tissues and 42 adjacent non-cancerous tissues were studied with 10 normal liver tissues as control group. TAp73α, p53, and PCNA were detected with Elivision immunohistochemistry.Terminal deoxynucleotidyl transferase (TdT)-mediatedd-UTP-biotin nick-end labeling (TUNEL) method wasused to detect the apoptosis cells. All clinical and pathological materials were analyzed by SPSS10.0statistical package.RESULTS: TAp73α overexpressed in HCC tissues (36.2%) when compared with adjacent non-cancerous tissues (2.38%, P<0.005) and normal liver tissues (0, P<0.01).Mutant type p53 (mt-p53) overexpressed in HCC tissues (38.3%) when contracted with adjacent non-cancerous tissues (16.7%, P<0.05) and normal liver tissues (0, P<0.01). Proliferation index (PI) level in HCC tissues was significantly higher than that in adjacent non-cancerous tissues (30.34%±4.46% vs 27.88%±5.89%, t, P= 0.028). Apoptosis index (AI) level in HCC tissues was higher than that in adjacent non-cancerous tissues (8.62%±2.28% vs 7.38%±2.61%, t, P = 0.019). Expression of TAp73α was associated with lymph node metastasis and mt-p53, with r = 0.407 and 0.265, respectively. Expression of mtp53 was associated with Edmondson's stage and AFP, with r = 0.295 and -0.357, respectively. In Kaplan-Meier univariant analysis, TAp73α, AFP, TNM stage, portal vein invasion, liver membrane invasion and HBsAg correlated with prognosis (log rank, P = 0.039, 0.012, 0.002, 0.000,0.014, 0.007, respectively). Multivariant Cox regression analysis showed that TAp73α, AFP, TNM stage, portal vein invasion, liver membrane invasion and age were independent factors of prognosis. CONCLUSION: These results suggest that TAp73α can be used as a prognostic indicator of patients with HCC undergoing surgical tumor ablation. | 8 | Biodegradation of indole by a newly isolated Cupriavidus sp. SHE显示文摘Indole, a typical nitrogen heterocyclic aromatic pollutant, is extensively spread in industrial wastewater. Microbial degradation has been proven to be a feasible approach to remove indole, whereas the microbial resources are fairly limited. A bacterial strain designated as SHE was isolated and found to be an efficient indole degrader. It was identified as Cupriavidus sp. according to 16 SrRNA gene analysis. Strain SHE could utilize indole as the sole carbon source and almost completely degrade 100 mg/L of indole within 24 hr. It still harbored relatively high indole degradation capacity within p H 4–9 and temperature 25°C–35°C. Experiments also showed that some heavy metals such as Mn2+, Pb2+and Co2+did not pose severe inhibition on indole degradation. Based on high performance liquid chromatography–mass spectrum analysis, isatin was identified as a minor intermediate during the process of indole biodegradation. A major yellow product with m/z 265.0605(C15H8N2O3) was generated and accumulated, suggesting a novel indole conversion pathway existed. Genome analysis of strain SHE indicated that there existed a rich set of oxidoreductases, which might be the key reason for the efficient degradation of indole. | 9 | Specific targeted killing of ErbB2 positive breast cancer by retrovirus-mediated Immunocaspase-3 secreting T cells显示文摘In this study, lmmunocaspase-3 gene was transfected into Jurkat T lymphocytes and the targeted proapoptotic protein Immunocaspase-3 was stably secreted.Its entry to ErbB2 positive SKBr3 breast carcinoma cell line was observed by indirect immunofluorescence staining.Growth of SKBr3 cells was significantly inhibited when they were cultured with medium containing Immunocaspase-3.Next, lmmunocaspase-3 gene was cloned into retrovirus vector pLNCX, which was then transfected into PA317 cells to package. Packaged cells producing high titer pseudoviruses were acquired and the pseudoviruses were harvested to infect PBMCs, which had been stimulated to division. The latter were selected and administered to nude mice bearing SKBr3 tumors through tail vein. The results showed that the treatment contributed to an inhibition of tumor growth and prolonged the lifetime of nude mice bearing SKBr3 tumor.The efficiency of inhibition of tumor reached 73.25%, and the average lifetime of treated nude mice was 80.95% longerthan that of control group. Immunohistochemical examination revealed the exclusive distribution of Immunocaspase-3 proteins only in the tumor tissue samples; and TUNEL assay confirmed the occurrence of apoptosis in tumor calls. | 10 | Economic evaluation of cervical cancer screening strategies in urban China显示文摘Objective:This study evaluated the feasibility of different cervical cancer screening strategies in urban China.Methods:A Markov model was constructed to simulate a hypothetical cohort of 100,000 females aged 30-59 years in a 20-year period.Screening strategies included liquid-based cytology(LBC)every three years,human papillomavirus(HPV)DNA testing every three and five years,respectively,and a combination of HPV DNA testing and LBC(HPV+LBC)every three and five years,respectively.Model outcomes included cumulative incidence over 20 years,cumulative risk of cervical cancer,costs,life year saved(LYS),quality-adjusted life years(QALYs)and benefits.The cost-effectiveness ratios(CERs),incremental cost-effectiveness ratios(ICERs),costutility ratios(CURs),and benefit-cost ratios(BCRs)were used as outcomes in the health economic evaluation analysis.Univariate sensitivity analyses were performed to examine the stability of the results.Results:The cumulative incidence of the five screening strategies ranged from 833.02 to 1,158.07 cases per100,000 females.HPV DNA testing was most effective in reducing the cumulative risk of cervical cancer,saving life years and QALYs and gaining benefits.The CERs of HPV DNA testing every three and five years,and LBC every three years were considered to be very cost-effective if they were below China's GDP per capita.The CERs of HPV+LBC were considered to be cost-effective if they were below three times GDP per capita.The incremental cost-effectiveness analysis showed that HPV DNA testing every three and five years,LBC every three years and HPV+LBC every five years were dominant strategies.Conclusions:The findings of this study indicated that HPV DNA testing every five years or LBC every three years should be recommended in urban China. | Li Ma Yuying Wang Xiaohong Gao Yi Dai Yu Zhang Zhaojing Wang Xiaoxia Wang Limin Wang Jing Jiang Xinhua Jing Chunxia Yang Fanghui Zhao Jinghe Lang Youlin Qiao | 2019 | Chinese Journal of Cancer Research2019,31,6: | 4 |
| 11 | Erosive Wear and Wear Mechanism of in situ TiC_P/Fe Composites显示文摘The base structure of in situ TiCp/Fe composites fabricated under industrial condition was changed by differentheat treatments. Erosive wear tests were carried out and the results were compared with that of wear-resistant whitecast iron. The results suggest that the wear resistance of the in situ TiCP/Fe composite is higher than that ofwear-resistant white cast iron under the sand erosive wear condition. The wear mechanism of the wear-resistantwhite cast iron was a cycle process that base surface was worn and carbides were exposed, then carbides was brokenand wear pits appeared. While the wear mechanism of in situ TiCP/Fe composite was a cycle process that basesurface was worn and TiC grains were exposed and dropped. The wear resistance of in situ TiCP/Fe composite waslower than that of wear-resistant white cast iron under the slurry erosive wear condition. Under such circumstance,the material was not only undergone erosive wear but also electrochemistry erosion due to the contact with waterin the medium. | 12 | Hardening Effect on Machined Surface for Precise Hard Cutting Process with Consideration of Tool Wear显示文摘During hard cutting process there is severe thermodynamic coupling effect between cutting tool and workpiece, which causes quenching effect on finished surfaces under certain conditions. However, material phase transformation mechanism of heat treatment in cutting process is different from the one in traditional process, which leads to changes of the formation mechanism of damaged layer on machined workpiece surface. This paper researches on the generation mechanism of damaged layer on machined surface in the process of PCBN tool hard cutting hardened steel Cr12 Mo V. Rules of temperature change on machined surface and subsurface are got by means of finite element simulation. In phase transformation temperature experiments rapid transformation instrument is employed, and the effect of quenching under cutting conditions on generation of damaged layer is revealed. Based on that, the phase transformation points of temperature under cutting conditions are determined. By experiment, the effects of cutting speed and tool wear on white layer thickness in damaged layer are revealed. The temperature distribution law of third deformation zone is got by establishing the numerical prediction model, and thickness of white layer in damaged layer is predicted, taking the tool wear effect into consideration. The experimental results show that the model prediction is accurate, and the establishment of prediction model provides a reference for wise selection of parameters in precise hard cutting process. Researches have shown that attention parameters λ and parameters B, C directly influence on the recognition results, but there is no general research theory to control these parameters in the recognition process. We abstractly analyze these parameters in this paper, and purpose a novel parameters optimization method based on simulated annealing algorithm. SA algorithm has good optimization performance and is used to search the global optimized solution of these parameters. | 15 | Distributed convex optimization for nonlinear multi-agent systems disturbed by a second-order stationary process over a digraph显示文摘In this paper, we investigate the distributed convex optimization problem for a class of nonlinear multi-agent systems disturbed by random noise over a directed graph. The target problem involves designing a continuous-time algorithm to minimize the sum of all local cost functions associated with each agent. The target noise is considered as a second-order stationary process under mild assumptions. The noise-to-state exponential stability for the multi-agent system based on random differential equations is analyzed using a random field method. Sufficient conditions corresponding to the second moment relative to the optimal solution in the form of matrix inequalities are established. Then, the grid search method is employed to determine the best system parameters such that the second moment of the estimation error has the minimum value. In addition, the obtained results are applied to solve the average consensus problem in the presence of a stationary process.
