1CRISPR/Cas9-mediated multiple gene editing in Brassica oleracea var.capitata using the endogenous tRNA-processing system显示文摘Cabbage(Brassica oleracea var.capitata)is a biennial plant with strong self-incompatibility and an obligate requirement for prolonged vernalization by exposure to low temperatures to induce flowering.These characteristics significantly increase the difficulty of exploiting novel germplasm induced by physical or chemical mutagens.In this study,we report a CRISPR/Cas9 gene-editing system based on endogenous tRNA processing to induce high efficiency and inheritable mutagenesis in cabbage.Using the phytoene desaturase gene BoPDS,the S-receptor kinase gene BoSRK,and the male-sterility-associated gene BoMS1 as the target genes,multisite and multiple gene mutations were achieved using a construct with tandemly arrayed tRNA-sgRNA architecture to express multiple sgRNAs.The BoSRK3 gene mutation suppressed self-incompatibility completely,converting the self-incompatible line into a self-compatible line.In addition,the BoMS1 gene mutation produced a completely male-sterile mutant,which was highly cross compatible with its nonmutant isoline at the flowering stage as a result of a simultaneous BoSRK3 gene mutation,enabling the economic propagation of the male-sterile line through bee-mediated cross-pollination.Interestingly,higher site mutation efficiency was detected when a guide sequence was inserted into a location in the tandemly arrayed tRNA-sgRNA architecture that was distal from the upstream Pol III promoter.In addition,mutation sites were also detected in the paralogous genes of the BoPDS and BoSRK genes that had fully consistent sequences or base mismatches but beyond the“seed”region in the spacer sequence compared with the target sgRNAs.Collectively,our results demonstrate that the CRISPR/Cas9 system,coupled with an endogenous tRNA-processing system,is an efficient tool to improve cabbage traits.Cunfa Ma Chenzeng Zhu Min Zheng Mengci Liu Dejun Zhang Baoli Liu Qinfei Li Jun Si Xuesong Ren Hongyuan Song 2019Horticulture Research2019,6,1:8
2Dynamical mechanism of the stratospheric quasibiennial oscillation impact on the South China Sea Summer Monsoon显示文摘The National Center for the Atmospheric Research (NCAR) middle atmospheric model is used to study the effects of the quasi-biennial oscillation in the stratosphere (QBO) on the tropopause and uppe troposphere, and the relationship between the QBO and South China Sea Summer Monsoon (SCSSM is explored through NCEP (the National Centers for Environmental Prediction)/NCAR, ECMWF (Euro pean Centre for Medium-Range Weather Forecasts) monthly mean wind data and in situ sounding data The simulations show that the QBO-induced residual circulations propagate downwards, and affect the tropopause and upper troposphere during the periods of mid-late QBO phase and phase transition Meanwhile, diagnostic analyses indicate that anomalous circulation similar to SCSSM circulation is generated to strengthen the SCSSM during the easterly phase and anomalous Hadley-like circulation weakens the SCSSM during the westerly. 3A case of rapid growing colonic NK/T cell lymphoma complicated by Crohn's disease显示文摘A 37-year-old man developed abdominal pain and bloody diarrhea 11 months before admission. The colonoscopy revealed multifocal ulcers in the colon. Histology showed active chronic inflammation. Although anti-tuberculosis medication was effective, his symptoms repeated 2 months later. The subsequent colonoscopy revealed more extensive irregular ulcers than before, and he was clinically suspected with intestinal malignant lymphoma. He underwent subtotal colectomy and was histologically suggested Crohn's disease, then 5-aminosalicylic and a combination of prednisone and azathioprine were administered in succession postoperatively, but they achieved minimal relief of symptoms for a period of 7 months. The third colonoscopy showed a large irregular ulcer in the ileocolon stomas, and primary colonic NK/T cell lymphoma was diagnosed through histological and immunophenotypic studies. 6Unraveling the stabilization mechanism of solid electrolyte interface on ZnSe by rGO in sodium ion battery显示文摘Transition metal selenides have been widely studied as anode materials of sodium ion batteries(SIBs),however,the investigation of solid-electrolyte-interface(SEI)on these materials,which is critical to the electrochemical performance of SIBs,remains at its infancy.Here in this paper,ZnSe@C nanoparticles were prepared from ZIF-8 and the SEI layers on these electrodes with and without reduced graphene oxide(rGO)layers were examined in details by X-ray photoelectron spectroscopies at varied charged/discharged states.It is observed that fast and complicated electrolyte decomposition reactions on ZnSe@C leads to quite thick SEI film and intercalation of solvated sodium ions through such thick SEI film results in slow ion diffusion kinetics and unstable electrode structure.However,the presence of rGO could efficiently suppress the decomposition of electrolyte,thus thin and stable SEI film was formed.ZnSe@C electrodes wrapped by rGO demonstrates enhanced interfacial charge transfer kinetics and high electrochemical performance,a capacity retention of 96.4%,after 1000 cycles at 5 A/g.This study might offer a simple avenue for the designing high performance anode materials through manipulation of SEI film.Shuang Men Hui Zheng Dejun Ma Xiaolian Huang Xiongwu Kang 2021Journal of Energy Chemistry2021,30,3:1
7Characteristics of source rocks and hydrocarbon resource prospects of Permian Linxi Formation in northern Songliao Basin显示文摘In order to explore the oil and gas resource prospects in the Carboniferous–Permian strata in northern Songliao Basin,geological survey boreholes(HFD 1 and HFD 2)were drilled in the area,and thick dark mudstone and slate of the Upper Permian Linxi Formation were encountered.Source rock geochemistry analysis of the samples show that the organic matter abundance of the Upper Permian Linxi Formation source rock in the north of Songliao Basin is high,which belongs to medium to good source rock.The organic matter belongs to type Ⅱ,and it is in the evolution stage of highly mature to over mature.The Pr/Ph ratios of the source rocks range from 0.16 to 0.93,with an average of 0.53.The phytane predominance is obvious,and indicates a strong reduction to reduction sedimentary environment,which is conducive to the preservation of organic matter.Pr/nC_(17),Ph/nC_(18) and C_(27)–C_(28)–C_(29) regular steranes indicate that the organic matter was derived from a mixture of vascular plants and aquatic organisms such as algae,and is mainly contributed by phytoplankton.Through comprehensive analysis,it is considered that the source rocks of the Upper Permian Linxi Formation in northern Songliao Basin have entered the gas generation stage and have shale gas exploration prospects.ZHANG Haihua GENG Shufeng ZHANG Jian SUN Lei CHEN Shuwang SU Fei ZHENG Yuejuan ZHANG Dejun BIAN Xiongfei 2022Global Geology2022,25,3:1

9Versatile nanocomposite augments high-intensity focused ultrasound for high-efficacy sonodynamic therapy of glioma显示文摘High-intensity focused ultrasound(HIFU),with inherent advantages of improved ultrasonic depth and low off-target damage,holds the promising capability for glioma treatment,but the relatively long therapeutic time and potential physical complications may hamper its clinical application.Herein,a bovine serum albumin(BSA)-based nanoplatform with in situ growth of MnO_(2) was synthesized,and Protoporphyrin IX(PpIX)was further anchored to obtain a versatile PpIX@MnO_(2)@BSA nanoplatform(denoted as BMP).By employing HIFU as the exogenous irradiation source,a high-efficacy sonodynamic therapy(SDT)is developed,in which the excited BMP enables the production of tumoricidal reactive oxygen species(ROS).The inherent tumor microenvironment(TME)-responsive property of MnO_(2) endows BMP with specific T1-weighted magnetic resonance imaging(MRI)by releasing Mn2+,and the simultaneously generated O_(2) facilitates hypoxia alleviation as well as ^(1)O_(2) generation.Compared with HIFU therapy alone,suppression of glioma growth and improved survival benefits are achieved through the designed TMEresponsive nanocomposite under HIFU exposure.The high-efficacy SDT strategy combining BMP and HIFU demonstrated favorable TME-responsive T1-weighted MRI,hypoxic environment alleviation,and anti-tumor capability,providing a perspective paradigm for MRI-guided glioma treatment.Yingyan Zheng Dejun She Huihui Huang Lin Lin Sunhui Chen Yiping Lu Li Liu Zhiqing Pang Bo Yin 2022Nano Research2022,15,10:0
10Compressive Behaviour and Failure Mechanisms of GFRP Composite at High Strain Rates显示文摘Experimental investigations into the compressive behavior of glass fiber reinforced polymer(GFRP)composite at high strain rates were carried out using a split Hopkinson pressure bar(SHPB)setup.The GFRP laminates were made from E-glass fibers and epoxy resins by vacuum assisted compression molding machine.The results of the compressive tests indicated that the mechanical behavior of the GFRP composite depends highly on the strain rate.The compressive peak stress,toughness and Young's modulus of the GFRP composite increased with the increase of strain rate,while the strain level at the initial stages of damage was shortened with the increase of strain rate.In addition,the dynamic deformation behavior and failure process of the specimens were observed directly by using a high-speed camera.Following the experiments,the fracture morphologies and damage modes were examined by scanning electron microscopy(SEM)to explore the possible failure mechanisms of the specimens.The results showed that multiple failure mechanisms appeared,such as matrix crack,fiber-matrix debonding,fiber failure and shear fracture.Dejun Yin Jian Zheng Chao Xiong Junhui Yin Huiyong Deng Xiaobo Su 2019Journal of Beijing Institute of Technology2019,28,1:0
11Three dimensional simulation of internal wave attractors in the Luzon Strait显示文摘Internal waves propagate along wave beams that are inclined with respect to the horizontal plane. It is conjectured that the internal waves generated in the Luzon Strait may be confined between the double ridges in the strait and concentrate to a closed trajectory, the so-called internal wave attractor, due to the reflection of wave beams from the lateral boundaries, sea surface and bottom. This work carried out two experiments using a three dimensional non-hydrostatic general circulation model, MITgcm, to investigate the possibility that the ridges in the Luzon Strait allows for internal wave attractors. Baroclinic current in both of the experiments demonstrate the forming of ring-like patterns in some section around 20° and 21°N, indicating that the development of the internal wave attractors are allowed in the Luzon Strait. 12Excess PbI_(2)evolution for triple-cation based perovskite solar cells with 21.9%efficiency显示文摘The triple cation mixed perovskites(Cs FAMA)are known as one of the most efficient candidates for perovskite solar cells(PSCs).It is found that the power conversion efficiency(PCE)of triple-cation based devices would increase with the test time extending,and the maximum efficiency is normally obtained after several days aging storage.Here,the relationship between enhanced device performance,excess PbI_(2)and its evolution in triple cation perovskite films of initial days was systematically explored.The Cs FAMA-PSCs are prepared by two-step methods under two environmental conditions,including in the glove box and the ambient air(30%humidity).After 7 days testing,the maximum PCE of PSCs under two conditions dramatically increased 12.4%and 12.2%,reached 21.68%and 21.89%,respectively.At initial days,the XRD peak intensities of perovskite phase gradually decreased and those corresponding to PbI_(2)increased.Along with time-resolved photoluminescence(TRPL)and kelvin probe force microscopy(KPFM),it was found that the defects were passivated with the evolution of PbI_(2).This work reveals the excess PbI_(2)and its evolution in perovskite film,which can further supplement the understanding of PbI_(2)defect passivation.Zhu Ma Dejun Huang Qianyu Liu Guangyuan Yan Zheng Xiao Dong Chen Jiaxuan Zhao Yan Xiang Changtao Peng Haijin Li Meng Zhang Wenfeng Zhang Lianfeng Duan Yuelong Huang 2022Journal of Energy Chemistry2022,31,3:0
13Growth of ablative Rayleigh-Taylor instability induced by time-varying heat-flux perturbation显示文摘The evolution of ablative Rayleigh–Taylor instability(ARTI)induced by single-mode stationary and time-varying perturbations in heat flux is studied numerically in two dimensions.Compared with the stationary case,time-varying heat-flux perturbation mitigates ARTI growth because of the enhanced thermal smoothing induced by the wave-like traveling heat flux.A resonance is found to form when the phase velocity of the heat-flux perturbation matches the average sound speed in the ablation region.In the resonant regime,the coherent density and temperature fluctuations enhance the electron thermal conduction in the ablation region and lead to larger ablation pressure and effective acceleration,which consequently yield higher linear growth rate and saturated bubble velocity.The enhanced effective acceleration offers increased implosion velocity but can also compromise the integrity of inertial confinement fusion shells by causing faster ARTI growth.Yang Liu De-Hua Zhang Jing-Fei Xin Yudong Pu Jun Li Tao Tao Dejun Sun Rui Yan Jian Zheng 2024Matter and Radiation at Extremes2024,9,1:0
14Combined linkage and association mapping reveals two major QTL for stripe rust adult plant resistance in Shaanmai 155 and their haplotype variation in common wheat germplasm显示文摘The development and deployment of diverse resistance sources in new wheat cultivars underpin the durable control of stripe rust.In the present study,two loci for adult plant resistance(APR),QYr SM155.1 and QYr SM155.2,were identified in the Chinese wheat breeding line Shaanmai 155.QYr SM155.1 was mapped to a 3.0-c M interval between the single-nucleotide polymorphism(SNP)markers AX-109583610 and AX-110907562 on chromosome arm 2 BL.QYr SM155.2 was mapped to a 2.1-c M interval flanked by the SNP markers AX-110378556 and AX-86173526 on chromosome arm 7 AS.A genome-wide association study was used to identify markers associated with APR in a panel of 411 spring wheat lines.Thirteen and 11 SNPs were significantly associated with QYr SM155.1 and QYr SM155.2,respectively,corresponding to physical intervals of 653.75–655.52 Mb on 2 BL and 81.63–83.93 Mb on7 AS.To characterize the haplotype variation and the distribution of these QTL,haplotype analysis was performed based on these SNPs in an independent panel of 1101 worldwide wheat accessions.Three major haplotypes(2 B_h1,2 B_h2,and 2 B_h3)for QYr SM155.1 and four major haplotypes(7 A_h1,7 A_h2,7 A_h3,and 7 A_h4)for QYr SM155.2 were identified.Accessions individually harboring QYr SM155.1_h1 and QYr SM155.2_h1 haplotypes and their combination displayed resistance.Additional assays of 1306 current Chinese cultivars and breeding lines using markers flanking QYr SM155.1 and QYr SM155.2 indicated that the resistance haplotypes of the two QTL were present in respectively 1.45%and 14.16%of lines.Increasing resistance haplotype frequencies at these two loci using marker-assisted selection should benefit wheat production in China.Cai’e Zhou Dan Liu Xu Zhang Qimeng Wu Shengjie Liu Qingdong Zeng Qilin Wang Changfa Wang Chunlian Li Ravi PSingh Sridhar Bhavani Zhensheng Kang Dejun Han Weijun Zheng Jianhui Wu 2022The Crop Journal2022,10,3:0
