3Effect of manipulation on cartilage in rats with knee osteoarthritis based on the Rho-associated protein kinase/LIM kinase 1/Cofilin signaling pathways显示文摘OBJECTIVE:To investigate the effect of manipulation treatment on knee osteoarthritis rats and the effect on Rho-associated protein kinase(ROCK)/LIM-kinase1(LIMK1)/Cofilin signaling pathway.METHOD:Fifty Specific pathogen Free Sprague-Dawley rats were randomly divided into five groups(n=8 each):blank group,model group,manipulation group,celecoxib group,and manipulation combined with celecoxib group(MC group).The osteoarthritis model was established by injecting 0.2 m L 4%papain into the articular disc of the rats.After successfully establishing the model,we treated the manipulation group with pushing manipulation using one-finger-meditation to the Neixiyan(EX-LE4),Waixiyan(EX-LE5),Xuehai(SP10),Liangqiu(ST34),and Zusanli(ST36)acupoints for 10 min each time.Also,the celecoxib group was gavaged with 24 mg·kg^(-1)·d^(-1 )celecoxib,while the MC group was treated using both of these two methods.After four weeks,the cartilage of the right femur was removed for hematoxylin-eosin staining of the cartilage tissue.The expressions of interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)in serum were observed using the enzyme-linked immunosorbent assay.Besides,we detected the expressions of ROCK,LIMK1,Phospho-LIM-kinase1(Phospho-LIMK1),Cofilin,and Phospho-Cofilin by Western blot.RESULTS:Compared to the model group,the manipulation group,celecoxib group,and MC group all exhibited superior results concerning pathological morphologic changes of cartilage,as observed by hematoxylin-eosin staining and calculated using the Mankin score.Besides,in contrast to the blank group,the model group exhibited elevated serum levels of IL-1βand TNF-α(P<0.01),while the expression of ROCK,LIMK1,Phospho-LIMK1,Cofilin,and Phospho-Cofilin in cartilage were all higher(P<0.01).Also,the serum levels of IL-1βand TNF-αin each treatment group were lower(P<0.01)than in the model group.Moreover,there were lower expressions of ROCK,LIMK1,Phospho-LIMK1,Cofilin,and Phospho-Cofilin in cartilage in the manipulation group and the MC group(P<0.01).Compared with the model group,the expression of ROCK,LIMK1,PhosphoLIMK1,Cofilin,and Phospho-Cofilin in cartilage in the celecoxib group were not statistically different(P>0.05).CONCLUSION:In this study,we established that manipulation has a better curative effect than celecoxib.Manipulation inhibits the development of cytoskeleton damage in cartilage and slows articular degeneration by regulating the expression of related proteins in the cytoskeletal signaling pathway.GUO Xiao YANG Yunhao LIAO Dongmei PANG Fang YANG Zhixue ZHU Zhengwei LUO Ao TANG Chenglin 2022Journal of Traditional Chinese Medicine2022,42,2:1
4A high-precision multi-dimensional microspectroscopic technique for morphological and properties analysis of cancer cell显示文摘Raman and Brillouin scattering are sensitive approaches to detect chemical composition and mechanical elasticity pathology of cells in cancer development and their medical treatment researches.The application is,however,suffering from the lack of ability to synchronously acquire the scattering signals following three-dimensional(3D)cell morphology with reasonable spatial resolution and signal-to-noise ratio.Herein,we propose a divided-aperture laser differential confocal 3D Geometry-Raman-Brillouin microscopic detection technology,by which reflection,Raman,and Brillouin scattering signals are simultaneously in situ collected in real time with an axial focusing accuracy up to 1 nm,in the height range of 200μm.The divided aperture improves the anti-noise capability of the system,and the noise influence depth of Raman detection reduces by 35.4%,and the Brillouin extinction ratio increases by 22 dB.A high-precision multichannel microspectroscopic system containing these functions is developed,which is utilized to study gastric cancer tissue.As a result,a 25%reduction of collagen concentration,42%increase of DNA substances,17%and 9%decrease in viscosity and elasticity are finely resolved from the 3D mappings.These findings indicate that our system can be a powerful tool to study cancer development new therapies at the sub-cell level.Lirong Qiu Yunhao Su Ke-Mi Xu Han Cui Dezhi Zheng Yuanmin Zhu Lin Li Fang Li Weiqian Zhao 2023Light(Science & Applications)2023,12,6:0
5Spatio-temporal Distribution Characteristics and Environmental Impact Factors of Lung Cancer Mortality: A Case Study of Yuhui District in Bengbu City, China显示文摘Among cancers, lung cancer is the most common cause of death in China. For the prevention and control of lung cancer, it is necessary to investigate the spatial and temporal distribution of lung cancer mortality, as well as the changes in the trend and the affecting mechanism. Based on statistics and auto-correlation analysis, this paper studied the spatial and temporal distribution of lung cancer mortality in Yuhui District, Bengbu, Huaihe River Basin, from 2017 to 2020. In addition, Spearman’s Rank Correlation Assessment Model and Geographic Detector Model were used to examine the relationship between environmental factors and lung cancer mortality to identify impact factors and their mechanisms. The findings indicated that: 1) from the characteristics of temporal distribution, the number of lung cancer deaths exhibited a linear growth tendency, with the highest mortality in winter;2) from the characteristics of spatial distribution, lung cancer mortality showed a strong spatial agglomeration form, concentrating on two clustering areas, located in the old city and the central city of Bengbu, near the Huaihe River;3) from the point of view of the whole research area, there were 15 impact factors with significant correlation in the built and natural environment factors. The significant impacting factors in the built environment included land use, road traffic, spatial form and blue-green space, which could indirectly affect lung cancer mortality, while air pollution and temperature constituted the significant impacting factors in the natural environment;4) the influence of screened environmental factors on lung cancer mortality was different. Spatial stratified heterogeneity assessment, the interaction among environmental factors demonstrated statistical significance, it was found that the interaction between environmental factors in pairs had a significant enhancement effect on lung cancer mortality. 6Ruscogenin alleviates LPS-triggered pulmonary endothelial barrier dysfunction through targeting NMMHC IIA to modulate TLR4signaling显示文摘Pulmonary endothelial barrier dysfunction is a hallmark of clinical pulmonary edema and contributes to the development of acute lung injury(ALI).Here we reported that ruscogenin(RUS),an effective steroidal sapogenin of Radix Ophiopogon japonicus,attenuated lipopolysaccharides(LPS)-induced pulmonary endothelial barrier disruption through mediating non-muscle myosin heavy chain IIA(NMMHC IIA)-Toll-like receptor 4(TLR4)interactions.By in vivo and in vitro experiments,we observed that RUS administration significantly ameliorated LPS-triggered pulmonary endothelial barrier dysfunction and ALI.Moreover,we identified that RUS directly targeted NMMHC IIA on its N-terminal and head domain by serial affinity chromatography,molecular docking,biolayer interferometry,and microscale thermophoresis analyses.Downregulation of endothelial NMMHC IIA expression in vivo and in vitro abolished the protective effect of RUS.It was also observed that NMMHC IIA was dissociated from TLR4 and then activating TLR4 downstream Src/vascular endothelial cadherin(VE-cadherin)signaling in pulmonary vascular endothelial cells after LPS treatment,which could be restored by RUS.Collectively,these findings provide pharmacological evidence showing that RUS attenuates LPS-induced pulmonary endothelial barrier dysfunction by inhibiting TLR4/Src/VE-cadherin pathway through targeting NMMHC IIA and mediating NMMHC IIA-TLR4 interactions.Yunhao Wu Xiu Yu Yuwei Wang Yalin Huang Jiahui Tang Shuaishuai Gong Siyu Jiang Yuanli Xia Fang Li Boyang Yu Yuanyuan Zhang Junping Kou 2022Acta Pharmaceutica Sinica B2022,12,3:0

8Spatial planning for urban ventilation corridors by urban climatology显示文摘Ventilation corridors in cities can decrease air pollution and alleviate heat island problems but there remains a need to fully assess their effectiveness.Few urban managers have been able to take city-scale approaches to the construction of urban ventilation corridors.This study aimed to introduced the Ventilation Corridor Planning(VCP)model,which is a multi-criteria evaluation method combined with a geographical information system(GIS)to determine where the ventilated environment is most appropriate.Specifically,the VCP model took Bozhou,China as the research object and contained two scales,including mesoscale and local scale.In mesoscale scale,we got three outputs to build urban ventilation corridors,including 1)background wind environment,2)ventilation potential,3)heat island intensity.In local scale,we used traditional computational fluid dynamics(CFD)model to verify the impact of VCP criteria.The results revealed that compared with the traditional CFD model,the proposed VCP model has advantages in establishing a comprehensive evaluation standard.In addition,the application of VCP model in macro and micro also enhances the efficiency of ventilation corridor construction.Overall,this study introduced a effective modeling method to urban ventilation corridors planning,and provide a way to study the urban climate.Kangkang Gu Yunhao Fang Zhao Qian Zhen Sun Ai Wang 2020Ecosystem Health and Sustainability2020,6,1:0
