3Establishment of a rat model of severe spontaneous intracerebral hemorrhage显示文摘Background:Severe intracerebral hemorrhage(ICH)is the most devastating subtype of stroke resulting in high mortality and disability.At present,the development of targeted treatments to minimize the high morbidity and mortality is limited partly due to the lack of a severe ICH animal model.In this study,we aimed to establish an accurate severe ICH model in rats and examine the pathological and physiological changes associated with ICH.Methods:A rat model of severe ICH model was established by intrastriatal injection of autologous blood using different blood volumes(ICH 100μL group,ICH 130μL group,ICH 160μL group,ICH 170μL group,and ICH 180μL group).The mortality was assessed during the 28-day post-ICH period.Short-and long-term neurological deficits were evaluated using the Longa method,foot fault,falling latency,and Morris water maze tests.Brain water content,hematoma volume,hemoglobin content,and magnetic resonance imaging were assessed to determine the extent of brain injury.Immunofluorescence staining was conducted to examine microglial activation and neuronal apoptosis.Hematoxylin and eosin(H&E)staining,lung water content,and western blotting were used to assess lung injury following ICH.Results:The mortality of ICH rats increased significantly with an increase in autologous blood injection.The 28-day mortality in the 100μL,130μL,160μL,170μL,and 180μL ICH groups were 5%,20%,40%,75%,and 100%,respectively.A significantly higher 28-day mortality was observed in the ICH 160μL group compared to the ICH 100μL group.The ICH 160μL group exhibited significantly increased neurological deficits,brain edema,hematoma volume,and hemoglobin content compared to the sham group.Compared with the sham operation group,the activation of microglia and neuronal death in ICH 160μL rats increased.The use of H&E staining and western blotting demonstrated that disruption of the intra-alveolar structure,alveolar edema,and infiltration of inflammatory cells and cytokines into the lung tissue were more severe in the ICH 160μL group than the sham group.Conclusions:A severe ICH model in rats was successfully established using an injection of autologous blood at a volume of 160μL.This model may provide a valuable tool to examine the pathological mechanisms and potential therapeutic interventions of severe ICH.Shuixiang Deng Shengjie Feng Yuewen Xin Yu He Yao Wang Mi Tian Ye Gong 2024Journal of Intensive Medicine2024,4,1:0
4Lower activity time constraint on the ore-controlling Erdaogou fault in the Qingchengzi orefield of the Liaodong Peninsula,NE China显示文摘The Erdaogou fault,also known as the 101 fault,comprises the most important NE strike component in the ore-controlling system in the Qingchengzi orefield,Liaodong Peninsula.Due to the poor outcrop conditions in the field,the Erdaogou fault lacks activity time constraint.We demonstrate the constraint activity time of the fault since we observe a lamprophyre that was cut through by the Erdaogou fault in the Taoyuan area,central to the Qingchengzi orefield.Zircon grains from the lamprophyre dyke exhibit typical oscillatory zoning and yield weighted mean U–Pb age of 223.8±1.1 Ma(MSWD=1.5).The lower activity time limit of the Erdaogou fault is thus first constrained as 224 Ma or so and is correlated with the crystallization age of the Xinling and Shuangdinggou plutons within the orefield.Taking previous mineralization studies into consideration,a Triassic tectonic-magmatism-mineralization model is approved in the Qingchengzi orefield.Yan Zhao Yuewen Feng Peng Zhang Linlin Kou Zhongzhu Yang Shenghui Li Wei Wang 2022Acta Geochimica2022,41,2:0
5Uncovering pharmacological mechanisms of Phellinus linteus on focal segmental glomeruloscleosis rats through tandem mass tag-based quantitative proteomic analysis,network pharmacology analysis and experimental validation显示文摘OBJECTIVE:To explore the underlying molecular mechanism of Phellinus linteus(P.linteus).METHODS:We used a tandem mass tag-based quantitative proteomic method to determine the differentially expressed proteins.Network pharmacology analysis was used to analysis the main components of P.linteus and construct the compound-target network.Western blotting and quantitative real-time polymerase chain reaction(qRT-PCR)were used to validate the analyses results.RESULTS:The expression levels of thrombospondin-1(TSP-1)and transforming growth factor(TGF)-β1/Smad3 signaling pathway proteins were significantly upregulated in focal segmental glomeruloscleosis(FSGS)rats.The P.linteus reduced the expression levels of TSP-1 and TGF-β1 signaling pathway proteins.Network pharmacology analysis revealed that protocatechualdehyde was the main active component.Subsequent in vivo and in vitro experiments validated the results of proteomic and network pharmacology analyses.CONCLUSIONS:Our results suggested that P.linteus may inhibit renal sclerosis by inhibiting TSP-1-activated TGF-β1 signaling and may have potential applications in the treatment of FSGS.WAN Feng YANG Ruchun TANG Yuewen 2023Journal of Traditional Chinese Medicine2023,43,4:0
