| 1 | Isolation and oral immunogenicity assessment of porcine epidemic diarrhea virus NH-TA2020 strain:One of the predominant strains circulating in China from 2017 to 2021显示文摘Porcine epidemic diarrhea(PED)caused by porcine epidemic diarrhea virus(PEDV)is one of the most devastating diseases in the global pig industry due to its high mortality rate in piglets.Maternal vaccines can effectively enhance the gut-mammary gland-secretory IgA axis to boost lactogenic immunity and passive protection of nursing piglets against PEDV challenge.From 2017 to 2021,we collected 882 diarrhea samples from 303 farms in China to investigate the epidemiology of PEDV.The result showed that about 52.15%(158/303)of the farms were positive for PEDV with an overall detection rate of 63.95%(564/882)of the samples.The S1 fragments of S gene from 104 strains were sequenced for the phylogenetic analysis.A total of 71 PEDV strains(68.27%)sequenced in this study were clustered into the predominant G2c subgroup,while the newly-defined G2d strains(9.62%)were identified in three provinces of China.The NH-TA2020 strain of G2c subgroup was isolated and cultured,and its infection to piglets caused watery diarrhea within 24 h,indicating its strong pathogenicity.Oral administration of NH-TA2020 strain to pregnant gilts stimulated high levels of IgA antibody in colostrum.The piglets fed by the gilts above were challenged with NH-TA2020 strain or CH-HeB-RY-2020 strain from G2d subgroup,and the clinical symptoms and virus shedding were significantly reduced compared to the mock group.Our findings suggest that G2c subgroup is the predominant branch circulating in China from 2017 to 2021.Oral administration of NH-TA2020 enhances maternal IgA and lactogenic immune responses,which confer protection against the homologous and emerging G2d PEDV strains challenges in neonates. | Xiaowen Li Yang Li Jiapei Huang Yali Yao Wenying Zhao Yunjing Zhang Jie Qing Jing Ren Zhong Yan Zewei Wang Xiaofang Hu Duli Kang Hongqiang Liu Zhichun Yan | 2022 | Virologica Sinica2022,37,5: | 6 |
| 2 | Copy number and zygosity determination of transgenic rapeseed by droplet digital PCR显示文摘Establishing an accurate and rapid method for copy number and zygosity determination can accelerate genetic engineering research process. In this study, droplet digital PCR (ddPCR), an emerging DNA absolute quantification technology, was used to identify single-copy homozygous transgenic lines from a batch of T0 transgenic rapeseed harboring 11 exogenous elements. Copy number of exogenous gene was evaluated in T0 generation based on calculated ratio between transgene and reference CruA gene, single-copy transformants were selected for selfing followed by subsequent zygosity analysis. Single-copy homozygous transgenic plants were successfully screened out in T1 generation by ddPCR.Segregation analysis with T2 seedlings verified that identification results of ddPCR were accurate and reliable. | 5 | The CRL3^(BTBD9) E3 ubiquitin ligase complex targets TNFAIP1 for degradation to suppress cancer cell migration显示文摘Tumor necrosis factor alpha-induced protein 1(TNFAIP1)modulates a plethora of important biological processes,including tumorigenesis and cancer cell migration.However,the regulatory mechanism of TNFAIP1 degradation remains largely elusive.In the present study,with a label-free quantitative proteomic approach,TNFAIP1 was identified as a novel ubiquitin target of the Cullin-RING E3 ubiquitin ligase(CRL)complex.More importantly,Cul3-ROC1(CRL3),a subfamily of CRLs,was identified to specifically interact with TNFAIP1 and promote its polyubiquitination and degradation.Mechanistically,BTBD9,a specific adaptor component of CRL3 complex,was further defined to bind and promote the ubiquitination and degradation of TNFAIP1 in cells.As such,downregulation of BTBD9 promoted lung cancer cell migration by upregulating the expression of TNFAIP1,whereas TNFAIP1 deletion abrogated this effect.Finally,bioinformatics and clinical sample analyses revealed that BTBD9 was downregulated while TNFAIP1 was overexpressed in human lung cancer,which was associated with poor overall survival of patients.Taken together,these findings reveal a previously unrecognized mechanism by which the CRL3^(BTBD9) ubiquitin ligase controls TNFAIP1 degradation to regulate cancer cell migration. | Lihui Li Wenjuan Zhang Yue Liu Xiaojun Liu Lili Cai Jihui Kang Yunjing Zhang Wenlian Chen Changsheng Dong Yanmei Zhang Mingsong Wang Wenyi Wei Lijun Jia | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 2 |

| 12 | MIR205HG facilitates carcinogenesis of lung squamous cell carcinoma in vitro revealed by long noncoding RNA profiling显示文摘As a subtype of non-small-cell lung cancer,lung squamous cell carcinoma(LUSC)accounts for one-fifth of all lung cancers.Unfortunately,no specific targetable aberration has yet been identified.Hence,it is of huge urgency and potential to identify aberrantly regulated genes in LUSC.Here,five pairs of LUSC samples and their corresponding adjacent tissues were subject to whole transcriptome sequencing.Our results showed that CTD-2562J17.6 and FENDRR were significantly downregulated while MIR205HG,LNC_000378,RP11-116G8.5,RP3-523K23.2,and RP5_968D22.1 were significantly upregulated in all five LUSC samples.Importantly,MIR205HG was upregulated in LUSC clinical samples as well as in LUSC cell lines.Interestingly,our results demonstrated that the expression level of MIR205HG is positively correlated with the malignancy.In addition,MIR205HG is required for LUSC cell growth and cell migration.Most importantly,our results showed that MIR205HG prohibits LUSC apoptosis via regulating Bcl-2 and Bax.Taken together,our data shed lights on the IncRNA regulatory nexus that controls the carcinogenesis of LUSC and provided potential novel diagnostic markers and therapeutic targets for LUSC. | Yan Chang Xinying Xue Chunsun Li Wei Zhao Yongfu Ma Fei Xu Zhen Wu Yu Dai Yunjing Li Yang Liu Liang’an Chen | 2020 | Acta Biochimica et Biophysica Sinica2020,52,4: | 1 |

| 19 | Microsphere-assisted,nanospot,non-destructive metrology for semiconductor devices显示文摘As smaller structures are being increasingly adopted in the semiconductor industry,the performance of memory and logic devices is being continuously improved with innovative 3D integration schemes as well as shrinking and stacking strategies.Owing to the increasing complexity of the design architectures,optical metrology techniques including spectroscopic ellipsometry(SE)and reflectometry have been widely used for efficient process development and yield ramp-up due to the capability of 3D structure measurements.However,there has been an increasing demand for a significant reduction in the physical spot diameter used in the SE technique;the spot diameter should be at least 10 times smaller than the cell dimension(~30×40μm2)of typical dynamic random-access memory to be able to measure in-cell critical dimension(CD)variations.To this end,this study demonstrates a novel spectrum measurement system that utilizes the microsphere-assisted super-resolution effect,achieving extremely small spot spectral metrology by reducing the spot diameter to~210 nm,while maintaining a sufficiently high signal-to-noise ratio.In addition,a geometric model is introduced for the microsphere-based spectral metrology system that can calculate the virtual image plane magnification and depth of focus,providing the optimal distance between the objective lens,microsphere,and sample to achieve the best possible imaging quality.The proof of concept was fully verified through both simulations and experiments for various samples.Thus,owing to its ultra-small spot metrology capability,this technique has great potential for solving the current metrology challenge of monitoring in-cell CD variations in advanced logic and memory devices. | Soonyang Kwon Jangryul Park Kwangrak Kim Yunje Cho Myungjun Lee | 2022 | Light(Science & Applications)2022,11,2: | 1 |
