1单肺通气在新生儿胸腔镜手术中的应用显示文摘目的探讨单肺通气在新生儿胸腔镜手术中的应用效果。方法择期行胸腔镜下食道重建术的先天性食道闭锁患儿60例,ASAⅡ或Ⅲ级,男31例,女29例,出生1-28d。随机均分为气管内插管组(A组)和支气管内插管组(B组)。麻醉诱导后,A组患儿在喉镜明视下完成气管插管,B组患儿在超纤细支气管内窥镜(Φ2.4mm)引导下完成普通气管导管的支气管内插管,其中左位主动脉弓者行左主支气管插管,反之,右位主动脉弓者行右主支气管插管,并于侧卧位后再次检查确认气管导管位置。记录两组患儿气管插管后5min(T_0)和人工气胸后5min(T_1)、10min(T_2)、30min(T_3)的HR、MAP、SpO_2、PETCO_2、RR、Pinsp、PEEP,并于上述各时点行动脉血气分析测得儿动脉血pH值、PaO_2、PaCO_2;评估两组患儿人工气胸后5min(T_1)、10min(T_2)的肺萎陷和手术野情况。结果 T_1-T_2时B组患儿SpO_2、PaO_2、肺萎陷和手术野评分均明显高于A组(P<0.05)。T_3时两组患儿SpO_2和PaO_2差异无统计学意义。T_0-T_3时两组患儿HR、MAP、PETCO_2、RR、Pinsp、PEEP、动脉血pH值、PaCO_2差异均无统计学意义。结论单肺通气用于新生儿胸腔镜手术,不仅安全可行,还可有效改善氧合,提高肺萎陷和手术野暴露程度。胡华琨 李强 PENG Xihua XIAO Yu CHENG Liqun SHEN Shihui 2017江西医药2017,52,1:8
2A consistency improving method in the analytic hierarchy process based on directed circuit analysis显示文摘Test of consistency is critical for the analytic hierarchy process(AHP) methodology. When a pairwise comparison matrix(PCM) fails the consistency test, the decision maker(DM) needs to make revisions. The state of the art focuses on changing a single entry or creating a new matrix based on the original inconsistent matrix so that the modified matrix can satisfy the consistency requirement. However, we have noticed that the reason that causes inconsistency is not only numerical inconsistency, but also logical inconsistency, which may play a more important role in the whole inconsistency. Therefore, to realize satisfactory consistency, first of all, we should change some entries that form a directed circuit to make the matrix logically consistent, and then adjust other entries within acceptable deviations to make the matrix numerically consistent while preserving most of the original comparison information. In this paper, we firstly present some definitions and theories, based on which two effective methods are provided to identify directed circuits. Four optimization models are proposed to adjust the original inconsistent matrix. 3Effects of different aperture-sized type I collagen/silk fibroin scaffolds on the proliferation and differentiation of human dental pulp cells显示文摘This study aimed at evaluate the effects of different aperture-sized type I collagen/silk fibroin(CSF)scaffolds on the proliferation and differentiation of human dental pulp cells(HDPCs).The CSF scaffolds were designed with 3D mapping software Solidworks.Three different aperture-sized scaffolds(CSF1-CSF3)were prepared by low-temperature deposition 3D printing technology.The morphology was observed by scanning electron microscope(SEM)and optical coherence tomography.The porosity,hydrophilicity and mechanical capacity of the scaffold were detected,respectively.HDPCs(third passage,1105 cells)were seeded into each scaffold and investigated by SEM,CCK-8,alkaline phosphatase(ALP)activity and HE staining.The CSF scaffolds had porous structures with macropores and micropores.The macropore size of CSF1 to CSF3 was 421627 lm,579636 lm and 707643 lm,respectively.The porosity was 69.862.2%,80.162.8%and 86.563.3%,respectively.All these scaffolds enhanced the adhesion and proliferation of HDPCs.The ALP activity in the CSF1 group was higher than that in the CSF3 groups(P<0.01).HE staining showed HDPCs grew in multilayer within the scaffolds.CSF scaffolds significantly improved the adhesion and ALP activity of HDPCs.CSF scaffolds were promising candidates in dentine-pulp complex regeneration.Shihui Jiang Zhaoxia Yu Lanrui Zhang Guanhua Wang Xiaohua Dai Xiaoli Lian Yan Yan Linpu Zhang Yue Wang Ruixin Li Huiru Zou 2021Regenerative Biomaterials2021,8,4:2
4Assessment of the curving performance of heavy haul trains under braking conditions显示文摘To study the curving performance of trains,ID and 3D dynamic models of trains were built using numerical methods.The ID model was composed of 210 simple wagons,each allowed only longitudinal motion;whereas the 3D model included three complicated wagons for which longitudinal,lateral,and vertical degrees of freedom were considered.Combined with the calculated results from the ID model under braking conditions,the behavior of draft gears and brake shoes were added to the3 D model.The assessment of the curving performance of trains was focused on making comparisons between idling and braking conditions.The results indicated the following:when a train brakes on a curved track,the wheel-rail lateral force and derailment factor are greater than under idling conditions.Because the yawing movement of the wheelset is limited by brake shoes,the zone of wheel contact along the wheel tread is wider than under idling conditions.Furthermore,as the curvature becomes tighter,the traction ratio shows a nonlinear increasing trend,whether under idling or braking conditions.By increasing the brake shoe pressure,train steering becomes more difficult.Liangliang Yang Yu Kang Shihui Luo Maohai Fu 2015Journal of Modern Transportation2015,23,3:2

9Coordinated regulation of BACH1 and mitochondrial metabolism through tumor-targeted self-assembled nanoparticles for effective triple negative breast cancer combination therapy显示文摘The poor prognosis of triple negative breast cancer(TNBC)results from a lack of approved targeted therapies coupled with aggressive proliferation and metastasis,which is associated with high recurrence and short overall survival.Here we developed a strategy by employing tumor-targeted selfassembled nanoparticles to coordinately regulate BACH1(BTB domain and CNC homology 1)and mitochondrial metabolism.The BACH1 inhibitor hemin and mitochondria function inhibitor berberine derivative(BD)were used to prepare nanoparticles(BH NPs)followed by the modification of chondroitin sulfate(CS)on the surface of BH NPs to achieve tumor targeting(CS/BH NPs).CS/BH NPs were found to be able to inhibit tumor migration and invasion by significantly decreasing the amounts of tumor cell metabolites,glycolysis and metastasis-associated proteins,which were related to the inhibition of BACH1 function.Meanwhile,decreased mitochondrial membrane potential,activated caspase 3/9 and increased ROS production demonstrated coordinated regulation of BACH1 and mitochondrial metabolism.In a xenograft mice model of breast cancer,CS/BH NPs significantly inhibited tumor growth and metastasis due to the synergetic effect of hemin and BD without showing obvious toxicities for major organs.In sum,the results of efficacy and safety experiments suggest potential clinical significance of the prepared self-assembled CS/BH nanoparticles for the treatment of TNBC.Xuan Yang Yalong Wang Junke Zhao Hehui Rong Yujun Chen Mengting Xiong Xiaoxing Ye Shihui Yu Haiyan Hu 2022Acta Pharmaceutica Sinica B2022,12,10:1

11Mechanisms of secondary damage to the thalamic nucleus and substantia nigra in an adult hypertensive rat model following middle cerebral artery occlusion显示文摘BACKGROUND: Following ischemia, apoptosis is observed at the ipsilateral ventroposterior thalamic nucleus and substantia nigra, which are distant from, but connected to, the ischemic cerebral cortex, in animals with normotension. However, secondary brain damage in hypertension has not been clearly investigated. OBJECTIVE: The present study determined whether neuronal apoptosis is associated with neuronal loss in the ipsilateral ventroposterior thalamic nucleus and substantia nigra following cortical ischemia in adult hypertensive rats. Results should provide options for determining a time window for anti-apoptotic therapy. DESIGN, TIME AND SETTING: All experimental procedures in this randomized, controlled trial were conducted at the Neurological Laboratory of the First Affiliated Hospital of Sun Yat-sen University of China between October 2006 and July 2008. MATERIALS: Monoclonal primary antibodies specific to mouse anti-rat microtubule-associated protein 2 and glial fibrillary acidic protein were respectively purchased from Sigma Chemical, USA and BD Pharmingen, USA. Terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate-biotin nick end-labeling (TUNEL) detection kits were purchased from Roche Applied Science, Switzerland and Upstate, USA, respectively. METHODS: A total of 64 male, Sprague Dawley rats, aged 60-90 days, were equally and randomly divided into middle cerebral artery occlusion and sham surgery groups. Renovascular hypertension was established in both groups by renal artery occlusion. Right distal middle cerebral artery occlusion was performed by electrocoagulation in the middle cerebral artery occlusion group. MAIN OUTCOME MEASURES: Microtubule-associated protein 2 and glial fibrillary acidic protein were detected by immunohistochemistry, and apoptotic cells were observed by TUNEL assay. The stainings were separately detected in the ipsilateral ventroposterior thalamic nucleus and substantia nigra. RESULTS: During the 4 weeks following distal middle cerebral artery occlusion in renovascular hypertensive rats, microtubule-associated protein 2 expression gradually, but significantly, decreased (P < 0.05). Expression of glial fibrillary acidic protein increased significantly in the ipsilateral ventroposterior thalamic nucleus and substantia nigra (P < 0.05) and reached a peak at 4 weeks. In addition, number of apoptotic cells was significantly increased in both areas compared with the sham controls (P < 0.05), with a peak at 2 weeks. CONCLUSION: Results suggested that neuronal loss in the ipsilateral ventroposterior thalamic nucleus and substantia nigra following distal middle cerebral artery occlusion in hypertensive rats could be a secondary event resulting from apoptosis. 17Interleukin-12 supports in vitro self-renewal of long-term hematopoietic stem cells显示文摘Hematopoietic stem cells(HSCs)self-renew or differentiate through division.Cytokines are essential for inducing HSC division,but the optimal cytokine combination to control self-renewal of HSC in vitro remains unclear.In this study,we compared the effects of interleukin-12(IL-12)and thrombopoietin(TPO)in combination with stem cell factor(SCF)on in vitro self-renewal of HSCs.Single-cell assays were used to overcome the heterogeneity issue of HSCs,and serum-free conditions were newly established to permit reproduction of data.In single-cell cultures,CD150^(+)CD48^(-)CD41^(-)CD34^(-)c-Kit^(+)Sca-1^(+)lineage^(-)SCs divided significantly more slowly in the presence of SCF+IL-12 compared with cells in the presence of SCF+TPO.Serial transplantation of cells from bulk and clonal cultures revealed that TPO was more effective than IL-12 at supporting in vitro self-renewal of short-term(<6 months)HSCs,resulting in a monophasic reconstitution wave formation,whereas IL-12 was more effective than TPO at supporting the in vitro selfrenewal of long-term(>6 months)HSCs,resulting in a biphasic reconstitution wave formation.The control of division rate in HSCs appeared to be crucial for preventing the loss of self-renewal potential from their in vitro culture.Shanshan Zhang Maiko Morita Zhao Wang Jun Ooehara Sen Zhang Miner Xie Haitao Bai Wenying Yu Xiaofang Wang Fang Dong Jinhong Wang Shihui Ma Satoshi Yamazaki Hideo Ema 2019Blood Science2019,1,1:0

20An amelogenin-based peptide hydrogel promoted the odontogenic differentiation of human dental pulp cells显示文摘Amelogenin can induce odontogenic differentiation of human dental pulp cells(HDPCs),which has great potential and advantages in dentine-pulp complex regeneration.However,the unstability of amelogenin limits its further application.This study constructed amelogenin self-assembling peptide hydrogels(L-gel or D-gel)by heating-cooling technique,investigated the effects of these hydrogels on the odontogenic differentiation of HDPCs and explored the underneath mechanism.The critical aggregation concentration,conformation,morphology,mechanical property and biological stability of the hydrogels were characterized,respectively.The effects of the hydrogels on the odontogenic differentiation of HDPCs were evaluated via alkaline phosphatase activity measurement,quantitative reverse transcription polymerase chain reaction,western blot,Alizarin red staining and scanning electron microscope.The mechanism was explored via signaling pathway experiments.Results showed that both the L-gel and D-gel stimulated the odontogenic differentiation of HDPCs on both Day 7 and Day 14,while the D-gel showed the highest enhancement effects.Meanwhile,the D-gel promoted calcium accumulation and mineralized matrix deposition on Day 21.The D-gel activated MAPK-ERK1/2 pathways in HDPCs and induced the odontogenic differentiation via ERK1/2 and transforming growth factor/smad pathways.Overall,our study demonstrated that the amelogenin peptide hydrogel stimulated the odontogenic differentiation and enhanced mineralization,which held big potential in the dentine-pulp complex regeneration.Xinxin Li Zhaoxia Yu Shihui Jiang Xiaohua Dai Guanhua Wang Yue Wang Zhimou Yang Jie Gao Huiru Zou 2022Regenerative Biomaterials2022,9,1:0
