1Structural design and parameter determination for fluted-roller fertilizer applicator显示文摘The instability of the motion layer of fertilizer particle influences the precision and the accuracy of the amount of fertilizer application in the working process of fluted-roller fertilizer applicator.The characteristics of the motion layer were investigated,and a systematic scheme was proposed for designing the structures of fertilizer-filling cavity,fertilizer-filling surface and fertilizer-delivery cavity.The key parameters of the structure were studied by discrete element method.It was figured out that to ensure the smooth operation of fertilizer applicator,the fertilizer-filling angle was 105°,fertilizer-contact angle should be large than 100°,and the fertilizer-resistance angle should be smaller than 35°.Moreover,the stability of fertilizer motion morphology was investigated to verify the design.The results showed that when the roll rotational speed was larger than 120 deg/s,compared with the conventional design the CV of the amount of fertilizer application was reduced from 1.5%-3.5%to 0.5%-1.0%,and the accuracy of the fertilizer discharge was improved with the square of the correlation coefficient between the amount of fertilizer application and the roll rotational speed was larger than 0.999.It was deduced from the results that the precision and accuracy of the amount of fertilizer application are improved via eliminating the driving layer and enhancing the stable filling and conveying of the forced layer.Shan Zeng Yipeng Tan Yu Wang Xiwen Luo Lamei Yao Dengpan Huang Zewen Mo 2020International Journal of Agricultural and Biological Engineering2020,13,2:4
2SRSF1 plays a critical role in invariant natural killer T cell development and function显示文摘Invariant natural killer T(iNKT)cells are highly conserved innate-like T lymphocytes that originate from CD4^(+)CD8^(+)double-positive(DP)thymocytes.Here,we report that serine/arginine splicing factor 1(SRSF1)intrinsically regulates iNKT cell development by directly targeting Myb and balancing the abundance of short and long isoforms.Conditional ablation of SRSF1 in DP cells led to a substantially diminished iNKT cell pool due to defects in proliferation,survival,and TCRαrearrangement.The transition from stage 0 to stage 1 of iNKT cells was substantially blocked,and the iNKT2 subset was notably diminished in SRSF1-deficient mice.SRSF1 deficiency resulted in aberrant expression of a series of regulators that are tightly correlated with iNKT cell development and iNKT2 differentiation,including Myb,PLZF,Gata3,ICOS,and CD5.In particular,we found that SRSF1 directly binds and regulates pre-mRNA alternative splicing of Myb and that the expression of the short isoform of Myb is substantially reduced in SRSF1-deficient DP and iNKT cells.Strikingly,ectopic expression of the Myb short isoform partially rectified the defects caused by ablation of SRSF1.Furthermore,we confirmed that the SRSF1-deficient mice exhibited resistance to acute liver injury uponα-GalCer and Con A induction.Our findings thus uncovered a previously unknown role of SRSF1 as an essential post-transcriptional regulator in iNKT cell development and functional differentiation,providing new clinical insights into iNKT-correlated disease.Jingjing Liu Menghao You Yingpeng Yao Ce Ji Zhao Wang Fang Wang Di Wang Zhihong Qi Guotao Yu Zhen Sun Wenhui Guo Juanjuan Liu Shumin Li Yipeng Jin Tianyan Zhao Hai-Hui Xue Yuanchao Xue Shuyang Yu 2021Cellular & Molecular Immunology2021,18,11:3
3Effect of Carbonation and Drying-Wetting Cycles on Chloride Diffusion Behavior of Coral Aggregate Seawater Concrete显示文摘Based on seawater immersion,drying-wetting cycles,carbonation and drying-wetting cycles for coral aggregate sea-water concrete(CASC)with different strength grades,the effect of carbonation and drying-wetting cycles on chloride diffusion be-havior of CASC is studied.The results show that the free surface chloride concentration(Cs),free chloride diffusion coefficient(Df)and time-dependent index(m)of CASC in the drying-wetting cycles is obviously higher than that in seawater immersion.The Df and m of CASC of carbonation and drying-wetting cycles is higher than that in the drying-wetting cycles.Carbonation increases the Df and m of CASC,which is against CASC to resist chloride corrosion.The corrosion possibility of CASC structures in different ex-posed areas is as follows:splash zone(carbonation and drying-wetting cycles)>tidal zone(drying-wetting cycles)>underwater zone(seawater immersion).Besides,the chloride diffusion rate of C65-CASC is 17.8%-63.4%higher than that of C65-ordinary aggre-gate concrete(OAC)in seawater immersion(underwater zone).Therefore,anti-corrosion measures should be adopted to improve the service life of CASC structure in the oceanic environment.DA Bo LI Yipeng YU Hongfa MA Haiyan CHEN Haoyu DOU Xuemei WU Zhangyu 2022Journal of Ocean University of China2022,21,1:2
4Novel Cathelicidins with Potent Antimicrobial,Biofilm Inhibitory,and Anti-inflammatory Activities from the Frog Fejervarya multistriata显示文摘Antimicrobial peptides(AMPs),a class of gene-encoded peptides,are the first line of immune system to defense microbial invasions in multicellular organisms.Cathelicidins are an important family of AMPs that have been identified exclusively in vertebrates.However,up to now,cathelicidins from amphibians are poorly understood.In the present study,we reported the identification and characterization of two novel cathelicidins(FM-CATH1 and FMCATH2) from the frog Fejervarya multistriata.The c DNA sequences encoding FM-CATHs were successfully cloned from the constructed lung c DNA library of F.multistriata.Both of the c DNA sequences encoding FM-CATHs are 447 bp in length,and the deduced mature peptides of FM-CATHs are composed of 34 residues.Structural analysis indicated that FM-CATH1 and FM-CATH2 mainly assume amphipathic alpha-helical conformations.Antimicrobial and bacterial killing kinetic analysis indicated that both FM-CATH1 and FM-CATH2 possess potent,broad-spectrum and rapid antimicrobial potency.And cytoplasmic membrane permeabilization analysis indicated that FM-CATH1 and FMCATH2 kill bacteria by inducing the permeabilization of bacterial membrane.Besides direct antimicrobial activities,FM-CATHs also exhibited significant inhibitory effect on the formation of bacterial biofilms at low concentrations below 1×MIC.Furthermore,FM-CATH1 and FM-CATH2 exhibited potent anti-inflammatory activities by inhibiting LPS-induced transcription and production of pro-inflammatory cytokines TNF-α,IL-1β,and IL-6 in mouse peritoneal macrophages.Meanwhile,FM-CATHs showed relatively low cytotoxic activity against mammalian normal and tumor cell lines,and low hemolytic activity against human erythrocytes.In summary,the identification of FM-CATHs provides novel clues for our understanding of the roles of cathelicidins in amphibian immune systems.The potent antimicrobial,biofilm inhibitory,anti-inflammatory activities,and low cytotoxicity of FM-CATHs imply their great potential in novel antibiotics development.Aili WANG Yan CHEN Haining YU Yipeng WANG 2017Asian Herpetological Research2017,18,3:2
5Rice FLOURY ENDOSPERM22, encoding a pentatricopeptide repeat protein, is involved in both mitochondrial RNA splicing and editing and is crucial for endosperm development显示文摘Most of the reported P-type pentatricopeptide repeat(PPR) proteins play roles in organelle RNA stabilization and splicing. However, P-type PPRs involved in both RNA splicing and editing have rarely been reported, and their underlying mechanism remains largely unknown. Here, we report a rice floury endosperm22(flo22) mutant with delayed amyloplast development in endosperm cells. Map-based cloning and complementation tests demonstrated that FLO22 encodes a mitochondrion-localized P-type PPR protein.Mutation of FLO22 resulting in defective transsplicing of mitochondrial nad1 intron 1 and perhaps causing instability of mature transcripts affected assembly and activity of complex Ⅰ, and mitochondrial morphology and function. RNA-seq analysis showed that expression levels of many genes involved in starch and sucrose metabolism were significantly down-regulated in the flo22mutant compared with the wild type, whereas genes related to oxidative phosphorylation and the tricarboxylic acid cycle were significantly upregulated. In addition to involvement in splicing as a P-type PPR protein, we found that FLO22 interacted with DYW3, a DYW-type PPR protein, and they may function synergistically in mitochondrial RNA editing. 6Landform classification based on optimal texture feature extraction from DEM data in Shandong Hilly Area, China显示文摘Texture and its analysis methods are crucial for image feature extraction and classification. Digital elevation model (DEM) is the most important data source of digital terrain analysis and landform classification, and considerable research values are gained from texture feature extraction and analysis from DEM data. In this research, on the basis of optimal texture feature extraction, the hilly area in Shandong, China, was selected as the study area, and DEM data with a resolution of 500 m were used as the experimental data for landform classification. First, second-order texture measures and texture image were extracted from DEM data by using a gray level cooccurrence matrix (GLCM). Second, the variation characteristics of each texture measure were analyzed, and the optimal feature parameters, such as direction, gray level, and texture window, were determined. Meanwhile, the texture feature value, combined with maximum information, was calculated, and the multiband texture image was obtained by resolving three optimal texture measure images. Finally, a support vector machine (SVM) method was adopted to classify landforms on the basis of the multiband texture image. Results indicated that the texture features of DEM data can be sufficiently represented and measured via the quantitative GLCM method. However, the feature parameters during the texture feature value calculation required further optimization. 8Gd_(2)O_(3) nanoparticles modified g-C_(3)N_(4) with enhanced photocatalysis activity for degradation of organic pollutants显示文摘Gd_(2) O_(3) nanoparticles modified g-C_(3) N_(4) photocatalytic composites were synthesized by a simple one-step hydrothermal method.The structure,morphology,optical properties of the prepared photocatalyst were characterized by X-ray diffraction(XRD),field emission scanning electron microscopy(FESEM),field emission transmission electron microscopy(FETEM) and X-ray photoelectron spectroscopy(XPS).The result demonstrates that gadolinium is mainly dispersed on the surface of g-C_(3) N_(4) in the form of Gd_(2) O_(3),and does not destroy the lattice structure of g-C_(3) N_(4).Besides,the gadolinium can cause the red shift of the absorption edge of light,narrow the band gap,and increase the separation efficiency of the photogenerated electron and hole of g-C_(3) N_(4).Especially,the specific surface area of g-C_(3) N_(4) can be significantly increased.Furthermore,g-C_(3) N_(4)/Gd-0.05 displays the highest photodegradation performance when it is used for degradation of methyl orange(MO),methylene blue(MB) and Rhodamine B(RhB).The photodegradation rate of g-C_(3) N_(4)/Gd-0.05 composites is 72.4% for MO,95.5% for RhB,100% for MB after120 min under visible light(λ> 420 nm) irradiation.Narrow band gap promotes the separation of photogenerated electron and hole,which enhances the photocatalytic activity of g-C_(3) N_(4).It is noted that g-C_(3) N_(4)/Gd-0.05 exhibits excellent photocatalytic stability by the photocurrent and the cyclic photodegradation of MO.Yipeng Zhou Fanshan Zeng Chaoyang Sun Jun Wu Yu Xie Fayun Zhang Senlin Rao Fahui Wang Jinbing Zhang Jinsheng Zhao Shiqian Li 2021Journal of Rare Earths2021,39,11:1
9Super enhancers targeting ZBTB16 in osteogenesis protect against osteoporosis显示文摘As the major cell precursors in osteogenesis, mesenchymal stem cells(MSCs) are indispensable for bone homeostasis and development. However, the primary mechanisms regulating osteogenic differentiation are controversial. Composed of multiple constituent enhancers, super enhancers(SEs) are powerful cis-regulatory elements that identify genes that ensure sequential differentiation. The present study demonstrated that SEs were indispensable for MSC osteogenesis and involved in osteoporosis development. Through integrated analysis, we identified the most common SE-targeted and osteoporosis-related osteogenic gene,ZBTB16. ZBTB16, positively regulated by SEs, promoted MSC osteogenesis but was expressed at lower levels in osteoporosis.Mechanistically, SEs recruited bromodomain containing 4(BRD4) at the site of ZBTB16, which then bound to RNA polymerase IIassociated protein 2(RPAP2) that transported RNA polymerase Ⅱ(POL Ⅱ) into the nucleus. The subsequent synergistic regulation of POL Ⅱ carboxyterminal domain(CTD) phosphorylation by BRD4 and RPAP2 initiated ZBTB16 transcriptional elongation, which facilitated MSC osteogenesis via the key osteogenic transcription factor SP7. 12Tuning the concentration of surface/bulk oxygen vacancies in CeO_(2) nanorods to promote highly efficient photodegradation of organic dyes显示文摘To enhance the photodegradation ability of CeO_(2) for organic dyes,an effective strategy is to introduce oxygen vacancies(Vo).In general,the introduced Vo are simultaneously present both on the surface and in the bulk of CeO_(2).The surface oxygen vacancies(Vo-s) can decrease the band gap,thus enhancing light absorption to produce more photogenerated e-for photodegradation.However,the bulk oxygen vacancies(Vo-b) will inhibit photocatalytic activity by increasing the recombination of photogene rated e-and Vo-b.Therefore,regulating the concentrations of Vo-s to Vo-b is a breakthrough for achieving the best utilization of photogene rated e-during photodegradation.We used an easy hydrothermal method to achieve tunable concentrations of Vo-s to Vo-b in CeO_(2) nanorods.The optimized CeO_(2) presents a 70.2%removal of rhodamine B after 120 min of ultraviolet-visible light irradiation,and a superior photodegradation performance of multiple organics.This tuning strategy for Vo also provides guidance for developing other advanced metal-oxide semiconductor photocatalysts for the photodegradation of organic dyes.Zhen Shen Yipeng Zhou Yue Guo Jie Zhao Jianhua Song Yu Xie Yun Ling Wei Zhang 2021Chinese Chemical Letters2021,32,8:0
13Design and experimental study of the fertilizer applicator with vertical spiral fluted rollers显示文摘To improve the uniformity and accuracy of fertilizing amount for fertilizer applicators with spiral fluted roller,this paper experimentally studied the influence of major structural parameters on the fertilizing performance.Through two sets of orthogonal tests,it is found that the helix angle and installation angle of spiral fluted rollers are the main factors,which affect the uniformity of fertilizer discharge and the linear relationship between fertilizing amount and opening,respectively.Based on these findings,the experiment and analysis were carried out to determine the optimal helix angle and installation angle for the spiral fluted roller The experimental results of fertilizing performance show that:when the helix angle is 45°,it able to achieve satisfactory uniformity of fertilizer discharge and linear relationship between fertilizing amount and rotational speed;when the installation angle is 40°,it enhances the accuracy of fertilizer discharge with good linear relationship between fertilizing amount and opening.Compared with the fertilizing performance of traditional fertilizer applicators with horizontally installed straight fluted roller,in the aspect of uniformity,the optimized fertilizer applicators reduce the variation coefficient of fertilizing amount within 0.2 s at low speed(10-30 r/min)from 5.1%-52.5%to 4.2%-14.7%;in the aspect of accuracy;and increase the correlation coefficient square R2 between fertilizing amount and opening from 0.93-0.97 to no less than 0.996,and the regression intercept in the fitting equation is reduced from larger than 10.0 to less than 1.0.Yu Wang Yipeng Tan Silong Wei Mingming Liao Ying Zang Shan Zeng 2023International Journal of Agricultural and Biological Engineering2023,16,5:0
14TiO_(2) nanofiber-supported copper nanoparticle catalysts for highly efficient methane conversion to C1 oxygenates under mild conditions显示文摘The selective oxidation of methane under mild conditions remains the“Holy Grail of Catalysis”.The key to activating methane and inhibiting over-oxidation of target oxygenates lies in designing active centers.Copper nanoparticles were loaded onto TiO_(2) nanofibers using the photo-deposition method.The resulting catalysts were found to effectively convert methane into C1 oxygenated products under mild conditions.Compared with previously reported catalysts,it delivers a superior performance of up to 2510.7 mmol·g_(Cu)^(-1)·h^(-1) productivity with a selectivity of around 100%at 80℃for 5 min.Microstructure characterizations and density functional theory(DFT)calculations indicate that TiO_(2) in the mixed phase of anatase and rutile significantly increases the Cu^(+)/CuO ratio of the supported Cu species,and this ratio is linearly related to the formation rate of oxygen-containing species.The CuI site promotes the generation of active O species from H_(2)O_(2) dissociation on Cu_(2)O(111).These active O species reduce the energy barrier for breaking the C-H bond of CH_(4),thus boosting the catalytic activity.The methane conversion mechanism was proposed as a methyl radical pathway to form CH_(3)OH and CH_(3)OOH,and then the generated CH_(3)OH is further oxidized to HOCH_(2)OOH.Wencui Li Yu Ren Zean Xie Yipeng Wang Hang Zhang Dianxiang Peng Hengfang Shen Hongfei Shi Jiaxin Cai Peng Wang Tongxin Zhang Zhen Zhao 2024Nano Research2024,17,5:0
15A telomere-to-telomere reference genome provides genetic insight into the pentacyclic triterpenoid biosynthesis in Chaenomeles speciosa显示文摘Chaenomeles speciosa(2n=34),a medicinal and edible plant in the Rosaceae,is commonly used in traditional Chinese medicine.To date,the lack of genomic sequence and genetic studies has impeded efforts to improve its medicinal value.Herein,we report the use of an integrative approach involving PacBio HiFi(third-generation)sequencing and Hi-C scaffolding to assemble a high-quality telomere-to-telomere genome of C.speciosa.The genome comprised 650.4 Mb with a contig N50 of 35.5 Mb.Of these,632.3 Mb were anchored to 17 pseudo-chromosomes,in which 12,4,and 1 pseudo-chromosomes were represented by a single contig,two contigs,and four contigs,respectively.Eleven pseudo-chromosomes had telomere repeats at both ends,and four had telomere repeats at a single end.Repetitive sequences accounted for 49.5%of the genome,while a total of 45515 protein-coding genes have been annotated.The genome size of C.speciosa was relatively similar to that of Malus domestica.Expanded or contracted gene families were identified and investigated for their association with different plant metabolisms or biological processes.In particular,functional annotation characterized gene families that were associated with the biosynthetic pathway of oleanolic and ursolic acids,two abundant pentacyclic triterpenoids in the fruits of C.speciosa.Taken together,this telomere-to-telomere and chromosome-level genome of C.speciosa not only provides a valuable resource to enhance understanding of the biosynthesis of medicinal compounds in tissues,but also promotes understanding of the evolution of the Rosaceae.Shaofang He Duanyang Weng Yipeng Zhang Qiusheng Kong Keyue Wang Naliang Jing Fengfeng Li Yuebin Ge Hui Xiong Lei Wu De-Yu Xie Shengqiu Feng Xiaqing Yu Xuekui Wang Shaohua Shu Zhinan Mei 2023Horticulture Research2023,10,10:0
16More tree growth reduction due to consecutive drought and its legacy effect for a semiarid larch plantation in Northwest China显示文摘Extreme climate has increasingly led to negative impacts on forest ecosystems globally,especially in semiarid areas where forest ecosystems are more vulnerable.However,it is poorly understood how tree growth is affected by different drought events.In 2006–2009,the larch plantations in the semiarid areas of Northwest China were negatively affected by four consecutive dry years,which was a very rare phenomenon that may occur frequently under future climate warming.In this study,we analyzed the effect of these consecutive dry years on tree growth based on the data of the tree rings in the dominant layer of the forest canopy on a larch plantation.We found that the tree-ring width index(RWI)in dry years was lower than that in normal years,and it experienced a rapidly decreasing trend from 2006 to 2009(slope=-0.139 year^(-1),r=-0.94)due to water supply deficits in those dry years.Drought induced legacy effects of tree growth reduction,and consecutive dry years corresponded with greater growth reductions and legacy effects.Growth reductions and legacy effects were significantly stronger in the third and fourth consecutive dry years than that of single dry year(p<0.05),which might have been due to the cumulative stress caused by consecutive dry years.Our results showed that larch trees experienced greater tree growth reduction due to consecutive dry years and their legacy effect,and the trees had lower recovery rates after consecutive dry years.Our results highlight that consecutive dry years pose a new threat to plantations under climate warming,and thus,the effect of climate extremes on tree growth should be considered in growth models in semiarid areas.Yanfang Wan Pengtao Yu Yanhui Wang Jiamei Li Yushi Bai Yipeng Yu Bingbing Liu Xiaocha Wei 2024Journal of Forestry Research2024,35,2:0
17Atmospheric Pressure-Aware Seamless 3-D Localization and Navigation for Mobile Internet Devices显示文摘This paper describes a seamless three-dimensional (3-D) localization and navigation system for smartphones.The smartphone includes an atmospheric pressure sensor to measure the user's altitude that is combined with the outdoor Global Positioning System (GPS) and indoor WiFi-APs localization systems in a seamless 3-D localization system.The smartphone software also provides seamless navigation services by updating map information for both indoor and outdoor locations through the mobile Internet.The indoor floor information calculated from the altitude information is used to project localization anchor nodes,e.g.,WiFi-AP,on different floors onto the user's floor with an indoor 3-D localization algorithm using projection distances based on a Received Signal Strength (RSS) algorithm.Tests show that the 3-D method reduces systematic errors and achieves much higher accuracy than the traditional two-dimensional localization method.Nengqiang He Jinhai Huo Yuhan Dong Yipeng Li Yang Yu Yong Ren 2012Tsinghua Science and Technology2012,17,2:0

19Geographically Distinct Expression Profile of Host Defense Peptides in the Skin of the Chinese Odorous Frog, Odorrana margaretae显示文摘Odorrana margaretae(Anura: Ranidae) is widely distributed in the southern provinces of China. Previously, 72 antimicrobial peptides(AMPs) belonging to 21 families were identified from the skin of O. margaretae, which were captured in the Hunan province. In the present study, five O. margaretae frogs were captured from the Guizhou province and a total of 28 cDNAs encoding 17 host defense peptides(HDPs) belonging to 14 families were cloned from the skin cDNA library of O. margaretae. Among the 17 HDPs, only one(brevinin-1-Omar5) had been characterized. The distinct HDP expression profiles for O. margaretae in the previous and present study may be attributed to the environmental differences between the sampling locations and the genetic divergence among O. margaretae populations. Besides, 11 of the 17 HDPs identified in the present study were novel for ranids. In order to understand their roles in host defense reactions, three HDPs(odorranain-H-OM1, odorranain-M-OM and ranatuerin-2-OM), which possess low sequence similarity with the known amphibian HDPs, were selected for further chemical synthesis and functional analysis. Odorranain-H-OM1 showed direct antimicrobial activity against bacteria and fungi. Odorranain-M-OM exhibited concentration-dependent anti-oxidant activity. Ranatuerin-2-OM showed lectin-like activity and could strongly hemagglu-tinate human intact erythrocytes with or without the presence of Ca2+.
