Efficacy, safety and immunogenicity of hexavalent rotavirus vaccine in Chinese infants显示文摘A randomized,double-blind,placebo-controlled multicenter trial was conducted in healthy Chinese infants to assess the efficacy and safety of a hexavalent live human-bovine reassortant rotavirus vaccine(HRV)against rotavirus gastroenteritis(RVGE).A total of 6400 participants aged 6-12 weeks were enrolled and randomly assigned to either HRV(n?3200)or placebo(n?3200)group.All the subjects received three oral doses of vaccine four weeks apart. The vaccine efficacy(VE)against RVGE caused by rotavirus serotypes contained in HRV was evaluated from 14 days after three doses of administration up until the end of the second rotavirus season.VE against severe RVGE,VE against RVGE hospitalization caused by serotypes contained in HRV,and VE against RVGE,severe RVGE,and RVGE hospitalization caused by natural infection of any serotype of rotavirus were also investigated. All adverse events(AEs)were collected for 30 days after each dose.Serious AEs(SAEs)and intussusception cases were collected during the entire study. Our data showed that VE against RVGE caused by serotypes contained in HRV was 69.21%(95%CI:53.31-79.69).VE against severe RVGE and RVGE hospitalization caused by serotypes contained in HRV were 91.36%(95%CI:78.45-96.53)and 89.21%(95%CI:64.51-96.72)respectively.VE against RVGE,severe RVGE,and RVGE hospitalization caused by natural infection of any serotype of rotavirus were 62.88%(95%CI:49.11-72.92),85.51%(95%CI:72.74-92.30)and 83.68%(95%CI:61.34-93.11).Incidences of AEs from the first dose to one month post the third dose in HRV and placebo groups were comparable. There was no significant difference in incidences of SAEs in HRV and placebo groups. This study shows that this hexavalent reassortant rotavirus vaccine is an effective,well-tolerated,and safe vaccine for Chinese infants. | Zhiwei Wu Qingliang Li Yan Liu Huakun Lv Zhaojun Mo Fangjun Li Qingchuan Yu Fei Jin Wei Chen Yong Zhang Teng Huang Xiaosong Hu Wei Xia Jiamei Gao Haisong Zhou Xuan Bai Yueyue Liu Zhenzhen Liang Zhijun Jiang Yingping Chen Jiuwei Zhang Jialiang Du Biao Yang Bo Xing Yantao Xing Ben Dong Qinghai Yang Chen Shi Tingdong Yan Bo Ruan Haiyun Shi Xingliang Fan Dongyang Feng Weigang Lv Dong Zhang Xiangchu Kong Liuyifan Zhou Dinghong Que Hong Chen Zhongbing Chen Xiang Guo Weiwei Zhou Cong Wu Qingrong Zhou Yuqing Liu Jian Qiao Ying Wang Xinguo Li Kai Duan Yuliang Zhao Gelin Xu Xiaoming Yang | 2022 | Virologica Sinica2022,37,5: | 2
Decoupling engineering of formamidinium–cesium perovskites for efficient photovoltaics显示文摘Although pure formamidinium iodide perovskite(FAPbI3)possesses an optimal gap for photovoltaics, their poor phase stability limits the long-term operational stability of the devices.A promising approach to enhance their phase stability is to incorporate cesium into FAPbI3.However,state-of-the-art formamidinium–cesium(FA–Cs)iodide perovskites demonstrate much worse efficiency compared with FAPbI3,limited by the different crystallization dynamics of formamidinium and cesium, which result in poor composition homogeneity and high trap densities. We develop a novel strategy of crystallization decoupling processes of formamidinium and cesium via a sequential cesium incorporation approach. As such,we obtain highly reproducible, highly efficient and stable solar cells based on FA_(1–x)Cs_(x)PbI_(3)(x=0.05–0.16)films with uniform composition distribution in the nanoscale and low defect densities. We also revealed a new stabilization mechanism for Cs doping to stabilize FAPbI3,i.e.the incorporation of Cs into FAPbI3 significantly reduces the electron–phonon coupling strength to suppress ionic migration, thereby improving the stability of FA–Cs-based devices. | Haoran Chen Yong Wang Yingping Fan Yuetian Chen Yanfeng Miao Zhixiao Qin Xingtao Wang Xiaomin Liu Kaicheng Zhu Feng Gao Yixin Zhao | 2022 | National Science Review2022,9,10: | 1
Understanding the scale-up of fermentation processes from the viewpoint of the flow field in bioreactors and the physiological response of strains显示文摘The production capability of a fermentation process is predominately determined by individual strains, which ultimately affected ultimately by interactions between the scale-dependent flow field developed within bioreactors and the physiological response of these strains.Interpreting these complicated interactions is key for better understanding the scale-up of the fermentation process. We review these two aspects and address progress in strategies for scaling up fermentation processes.A perspective on how to incorporate the multiomics big data into the scale-up strategy is presented to improve the design and operation of industrial fermentation processes. | Jianye Xia Guan Wang Meng Fan Min Chen Zeyu Wang Yingping Zhuang | 2021 | Chinese Journal of Chemical Engineering2021,34,2: | 1
Effectiveness of two different acupuncture strategies in patients with vulvodynia Study protocol for a pilot pragmatic controlled trial显示文摘Background: Vulvodynia, or vulvar pain, is a common condition in women; however, there are few evidence-based clinical trials evaluating nonpharmacological therapies for this condition. Acupuncture is one complementary and integrative medicine therapy used by some patients with vulvodynia. This study evaluates two different acupuncture strategies for the treatment of vulvodynia and aims to evaluate whether either of the acupuncture protocols reduces vulvar pain, pain duration or pain with intercourse. The study also examines how long the effect of acupuncture lasts in women with vulvodynia.Methods/design: The study is designed as a randomized controlled trial, focused on two acupuncture protocols. Fifty-one patients who have had vulvodynia for more than 3 months will be recruited.Among them, 34 patients will be randomized into Groups 1 a and 1 b; those who are unwilling to receive acupuncture will be recruited into the standard care group(Group 2). Patients in Group 1 a will have acupuncture focused on the points in the pudendal nerve distribution area, while patients in Group 1 b will receive acupuncture focused on traditional(distal) meridian points. Patients in Group 2 will receive routine conventional treatments, such as using pain medications, local injections and physical therapies or other nonsurgical procedures. Acupuncture will last 45 min per session, once or twice a week for6 weeks. The primary outcome measurement will be objective pain intensity, using the cotton swab test. The secondary outcome measurement will be subjective patient self-reported pain intensity, which will be conducted before cotton swab test. Pain intensities will be measured by an 11-point Numeric Pain Rating Scale. Pain duration and pain score during intercourse are recorded. Local muscle tension, tenderness and trigger points(Ashi points) are also recorded. All measurements will be recorded at baseline(before the treatment), at the end of each week during treatment and at the end of the 6 weeks.Follow-up will be done 6 weeks following the last treatment.Discussion: Results of this trial will provide preliminary data on whether acupuncture provides better outcomes than nonacupuncture treatments, i.e., standard care, and whether acupuncture focused on the points in pudendal nerve distribution, near the pain area, has better results than traditional acupuncture focused on distal meridian points for vulvodynia. | Arthur Yin Fan Sarah F.Alemi Yingping H.Zhu Sudaba Rahimi Hui Wei Haihe Tian Deguang He Changzhen Gong Guanhu Yang Chong He Hui Ouyang | 2018 | Journal of Integrative Medicine2018,16,6: | 1
Archaeal diversity in the seawater of Changjiang River estuary reveals its adaptability to bottom seawaters显示文摘Archaea regulate the biogeochemical processes of ocean systems. The Changjiang(Yangtze)River estuary(CRE) is a complex and dynamic area strongly affected by seawaters and ocean currents. In this study, the planktonic archaeal communities in the surface and bottom seawaters of the CRE were investigated.Significant differences in the archaeal community composition were found between the surface and bottom seawaters(P<0.001).Marine Group Ⅱ(MG-Ⅱ) was dominant in the surface layers, and Nitrosopumilales was enriched in the bottom layers.Marine Group Ⅲ(MG-Ⅲ) was more abundant in the bottom layers than in the surface ones(P<0.001).These results were completely different from previous findings in the CRE seawater.Instead of dissolved oxygen(DO),temperature and salinity were the most vital environmental variations in the distribution of the archacal communities.According to the predicted metabolic pathways, the following functional subcategories were enriched in the hypoxic condition:replication and repair, membrane transport, glycan biosynthesis and metabolism, amino acid metabolism, metabolism of cofactors and vitamins, and xenobiotics biodegradation and metabolism(P<0.001),which indicated the strong adaptability of archaea to the harsh environment in the bottom seawater. These findings expand the understanding on archaeal structure and functions in the surface and bottom seawaters, including the hypoxic zones in the CRE,and may contribute to further works of the archaeal community in the CRE. | Yan HUANG Wu QU Yingping FAN Jianxin WANG | 2022 | Journal of Oceanology and Limnology2022,40,3: | 1

Continuous-flow enzymatic synthesis of chiral lactones in a three-dimensional microfluidic reactor显示文摘A new continuous-flow process for the enzymatic synthesis of optically pureγ-lactones,which are used as flavors and fragrances in the food and cosmetic industries, was developed in a three-dimensional microfluidic reactor. The microchannels(175 mm in length,0.9 mm in depth, and 1.72 mL in volume)were carved precisely inside a single borosilicate glass(90 mm×75 mm×12 mm)with ultrafast femtosecond laser micromachining. The flow field analysis and reaction simulation showed that the mixing of substrates and enzymes was enhanced, allowing the adjustment of residence time in a wide window.SmCR_(V4),a carbonyl reductase with excellent catalytic activity and enantioselectivity towardγ/δ-keto acids, was employed for the asymmetric synthesis of various chiral lactones.30 mmol/L(R)-γ-decalactone(3g)can be obtained in 26 s with a space-time yield(STY)up to 16,877 g L^(-1)d^(-1),which is 14.4 times higher than the highest STY of batch reaction reported previously. This continuous-flow process was applied to the synthesis of 6 chiral lactones. In addition, the scaled-up synthesis of 3g was carried out in 6 cascade microreactors continuously for 6 h,demonstrating the feasibility and stability of the 3D continuous-flow process in enzymatic synthesis of optically pure compounds. | Xuelei Deng Meng Fan Miao Wu Xiaoyan Zhang Ya Cheng Jianye Xia Yingping Zhuang Weiping Zhu Xuhong Qian Yunpeng Bai | 2024 | Chinese Chemical Letters2024,35,3: | 0
Nuclear peripheral chromatin-lamin B1 interaction is required for global integrity of chromatin architecture and dynamics in human cells显示文摘The eukaryotic genome is folded into higher-order conformation accompanied with constrained dynamics for coordinated genome functions. However, the molecular machinery underlying these hierarchically organized three-dimensional(3D)chromatin architecture and dynamics remains poorly understood. Here by combining imaging and sequencing,we studied the role of lamin B1 in chromatin architecture and dynamics. We found that lamin B1 depletion leads to detachment of lamina-associated domains(LADs)from the nuclear periphery accompanied with global chromatin redistribution and decompaction.Consequently, the interchromosomal as well as inter-compartment interactions are increased, but the structure of topologically associating domains(TADs)is not affected.Using live-cell genomic loci tracking,we further proved that depletion of lamin B1 leads to increased chromatin dynamics, owing to chromatin decompaction and redistribution toward nucleoplasm.Taken together, our data suggest that lamin B1 and chromatin interactions at the nuclear periphery promote LAD maintenance, chromatin compaction, genomic compartmentalization into chromosome territories and A/B compartments and confine chromatin dynamics, supporting their crucial roles in chromatin higher-order structure and chromatin dynamics. | Lei Chang Mengfan Li Shipeng Shao Chen Li Shanshan Ai Boxin Xue Yingping Hou Yiwen Zhang Ruifeng Li Xiaoying Fan Aibin He Cheng Li Yujie Sun | 2022 | Protein & Cell2022,13,4: | 0
CTCF organizes inter-A compartment interactions through RYBP-dependent phase separation显示文摘Chromatin is spatially organized into three-dimensional structures at different levels including A/B compartments, topologically associating domains and loops. The canonical CTCF-mediated loop extrusion model can explain the formation of loops. However, the organization mechanisms underlying long-range chromatin interactions such as interactions between A–A compartments are still poorly understood. Here we show that different from the canonical loop extrusion model, RYBP-mediated phase separation of CTCF organizes inter-A compartment interactions. Based on this model, we designed and verified an induced CTCF phase separation system in embryonic stem cells (ESCs), which facilitated inter-A compartment interactions, improved self-renewal of ESCs and inhibited their differentiation toward neural progenitor cells. These findings support a novel and non-canonical role of CTCF in organizing long-range chromatin interactions via phase separation. | Chao Wei Lumeng Jia Xiaona Huang Jin Tan Mulan Wang Jing Niu Yingping Hou Jun Sun Pengguihang Zeng Jia Wang Li Qing Lin Ma Xinyi Liu Xiuxiao Tang Fenjie Li Shaoshuai Jiang Jingxin Liu Tingting Li Lili Fan Yujie Sun Juntao Gao Cheng Li Junjun Ding | 2022 | Cell Research2022,32,8: | 0
Diurnal changes in bacterial communities in oxic surface and hypoxic middle seawater layers of the Changjiang River Estuary显示文摘The Changjiang River Estuary(CRE)in the East China Sea suffers from seasonal hypoxia in summer. The vertical distributions and seasonal changes of microbial communities in the CRE were well documented. However, little is known about the diurnal changes of bacterial communities in the hypoxic zone of the CRE.Here,16 S rRNA gene analysis was used to explore the changes of bacterial communities in the oxic surface and hypoxic middle seawater layers during 24 h in the CRE.Significant differences between the hypoxic and oxic layers were observed:the phyla Cyanobacteria,Bacteroidetes and Acidimicrobiia were enriched in the oxic layer, whereas the phylum SAR406 and the class Deltaproteobacteria were more abundant in the hypoxic layer. In addition, some subtle diurnal variations of the bacterial relative abundance were found in both two layers. The relative abundance of Synechococcus increased at night, and this change was more obvious in the hypoxic layer. The similar trend was also found in some phototrophic and several heterotrophic bacteria, such as Rhodobacteraceae,OM60 and Flavobacteriaceae.Their relative abundances peaked at 16:00 in the oxic layer, while the relative abundances peaked at around 7:00 and decreased until 13:00 in the hypoxic layer.Together, the results of the present study suggest that some photosynthetic bacteria and several heterotrophic bacteria have similar diurnal variations implying the light and physicochemical heterogeneity in the course of a day are important for bacterial diurnal changes in the CRE. | Yan Huang Lei Yuan Yingping Fan Habib U Rehman Jakhrani Jianxin Wang | 2021 | Acta Oceanologica Sinica2021,40,4: | 0
