| 1 | Potentiating CD8^(+) T cell antitumor activity by inhibiting PCSK9 to promote LDLR-mediated TCR recycling and signaling显示文摘Metabolic regulation has been proven to play a critical role in T cell antitumor immunity.However,cholesterol metabolism as a key component of this regulation remains largely unexplored.Herein,we found that the low-density lipoprotein receptor(LDLR),which has been previously identified as a transporter for cholesterol,plays a pivotal role in regulating CD8+T cell antitumor activity.Besides the involvement of cholesterol uptake which is mediated by LDLR in T cell priming and clonal expansion,we also found a non-canonical function of LDLR in CD8+T cells:LDLR interacts with the T-cell receptor(TCR)complex and regulates TCR recycling and signaling,thus facilitating the effector function of cytotoxic T-lymphocytes(CTLs).Furthermore,we found that the tumor microenvironment(TME)downregulates CD8+T cell LDLR level and TCR signaling via tumor cell-derived proprotein convertase subtilisin/kexin type 9(PCSK9)which binds to LDLR and prevents the recycling of LDLR and TCR to the plasma membrane thus inhibits the effector function of CTLs.Moreover,genetic deletion or pharmacological inhibition of PCSK9 in tumor cells can enhance the antitumor activity of CD8+T cells by alleviating the suppressive effect on CD8+T cells and consequently inhibit tumor progression.While previously established as a hypercholesterolemia target,this study highlights PCSK9/LDLR as a potential target for cancer immunotherapy as well. | Juanjuan Yuan Ting Cai Xiaojun Zheng Yangzi Ren Jingwen Qi Xiaofei Lu Huihui Chen Huizhen Lin Zijie Chen Mengnan Liu Shangwen He Qijun Chen Siyang Feng Yingjun Wu Zhenhai Zhang Yanqing Ding Wei Yang | 2021 | Protein & Cell2021,12,4: | 12 |
| 2 | Reduced function and disassembled microtubules of cultured cardiomyocytes in spaceflight显示文摘Lack of gravity during spaceflight has profound effects on cardiovascular system, but little is known about how the cardiomyocytes respond to microgravity. In the present study, the effects of spaceflight on the structure and function of cultured cardiomyocytes were reported. The primary cultures of neo- natal rat cardiomyocytes were carried on Shenzhou-6 spacecraft and activated at 4 h in orbit. 8 samples were fixed respectively at 4, 48 and 96 h after launching for immunofluorescence of cytoskeleton, and 2 samples remained unfixed to analyze contractile and secretory functions of the cultures. Ground sam- ples were treated in our laboratory in parallel. After 115 h spaceflight, video recordings displayed that the number of spontaneous beating sites in flown samples decreased significantly, and the cells in the beating aggregate contracted in fast frequency without synchrony. Radioimmunoassay of the medium showed that the atrial natriuretic peptide secreted from flown cells reduced by 59.6%. Confocal images demonstrated the time-dependant disassembly of mirotubules versus unchanged distribution and or- ganization of microfilaments. | 3 | Historical trends in the anthropogenic heavy metal levels in the tidal flat sediments of Lianyungang, China显示文摘The sedimentation of metals can preserve the historical record of contaminant input from local and regional sources and provide information on the historical changes in regionalwater and sediment quality. We report the 210Pb activities and the heavy metal (Cd, Cr, Cu, Mn, Pb and Zn)depth profiles from sediment cores retrieved in 2010. The mean sedimentation rates of 0.85-1.5 cm/yr aredetermined by 210 Pbdating. The sediments in the tidal flat have recorded heavy metaldeposition and thus allow the establishment of a connection between the temporal evolution of the heavy metal pollution and the historical changes in the economicdevelopment of Lianyungang. The enrichment factors (EF) are calculated to estimate the level of contamination stored in these sediments. The results show that in the studied sites, Cr and Cudisplay low EF values and are mainly from lithogenic origin. For the other studied trace metals, a great variability in the sedimentary record is observed. Significant anthropogenic enrichment over the last 50 years is revealed at the tidal flat that receives fluvial inputs. Zinc is the elementwith the highest EF values, followed by the order of Pb > Cd > Mn > Cu and Cr. The temporal variations of the heavy metals peakduring the late 1980s to the early2000s and show adecreasing trend afterward. | 4 | Theoretical and simulation research of hydrodynamic instabilities in inertial-confinement fusion implosions显示文摘Inertial fusion energy(IFE)has been considered a promising,nearly inexhaustible source of sustainable carbon-free power for the world's energy future.It has long been recognized that the control of hydrodynamic instabilities is of critical importance for ignition and high-gain in the inertial-confinement fusion(ICF)hot-spot ignition scheme.In this mini-review,we summarize the progress of theoretical and simulation research of hydrodynamic instabilities in the ICF central hot-spot implosion in our group over the past decade.In order to obtain sufficient understanding of the growth of hydrodynamic instabilities in ICF,we first decompose the problem into different stages according to the implosion physics processes.The decomposed essential physics processes that are associated with ICF implosions,such as Rayleigh-Taylor instability(RTI),Richtmyer-Meshkov instability(RMI),Kelvin-Helmholtz instability(KHI),convergent geometry effects,as well as perturbation feed-through are reviewed.Analytical models in planar,cylindrical,and spherical geometries have been established to study different physical aspects,including density-gradient,interface-coupling,geometry,and convergent effects.The influence of ablation in the presence of preheating on the RTI has been extensively studied by numerical simulations.The KHI considering the ablation effect has been discussed in detail for the first time.A series of single-mode ablative RTI experiments has been performed on the Shenguang-Ⅱlaser facility.The theoretical and simulation research provides us the physical insights of linear and weakly nonlinear growths,and nonlinear evolutions of the hydrodynamic instabilities in ICF implosions,which has directly supported the research of ICF ignition target design.The ICF hot-spot ignition implosion design that uses several controlling features,based on our current understanding of hydrodynamic instabilities,to address shell implosion stability,has been briefly described,several of which are novel. | LiFeng Wang WenHua Ye XianTu He JunFeng Wu ZhengFeng Fan Chuang Xue HongYu Guo WenYong Miao YongTeng Yuan JiaQin Dong Guo Jia Jing Zhang YingJun Li Jie Liu Min Wang YongKun Ding WeiYan Zhang | 2017 | Science China(Physics,Mechanics & Astronomy)2017,60,5: | 4 |
