1Procyanidins from grape seeds protect against phorbol ester-induced oxidative cellular and genotoxic damage显示文摘AIM: To evaluate the inhibitory effects of Vitis vinifera procyanidins (PAs) on carcinogen-induced oxidative stress. METHODS: The single cell gel electrophoresis technique (comet assay) was employed to detect DNA damage induced by the carcinogen phorbol-12-myristate-13-acetate (PMA), The release of hydrogen peroxidase from polymorphonuclear leukocytes (PMNs) was assayed by the horseradish peroxidase-mediated oxidation of phenol red. The microplate assay was used to detect the presence of oxidative products by means of 2',7'-dichlorofluorescindiacetate (DCFH-DA). The superoxide dismutase (SOD) activity of liver mitochondria was assayed, based on the ability of SOD to inhibit the generation of superoxidate anions by the xanthine-xanthine oxidase system. The malondialdehyde (MDA) level was determined by the thiobarbituric acid (TBA) assay. RESULTS: DNA of NIH3T3 cells was significantly damaged after addition of PMA. The length of the comet tail was observed ,while in normal cells the comet tail could not be observed. PAs showed significant protective effects on carcinogen PMA-induced DNA damage. Through assessment of DCFH-DA oxidation, PAs were shown to inhibit the PMA-induced release of hydrogen peroxide by PMNs, and to inhibit respiratory burst activity in NIH3T3 mouse fibroblasts. Ex vivo study showed that serum from rats administered with PAs displayed similar effects in a dose-dependent manner. In addition, PAs suppressed liver mitochondrial lipid peroxidation induced by PMA. PAs protected the activity of SOD and decreased the level of MDA in liver mitochondria damaged by PMA. 2Use of suppression subtractive hybridization strategy for cloning and identifying specifically expressed genes of renal cell carcinoma显示文摘Using suppression subtractive hybridization, a renal cell carcinoma (RCC) cDNA subtractive library which only contains differently expressed cDNAs between human RCC and normal kidney has been constructed. 200 clones were picked out randomly to perform enzyme digest analysis, a part of them underwent sequence analysis and Northern blot to identify RCC specially expressed genes. Results showed that 190 clones contain 50-400 bp inserts respectively. Sequence analysis was performed in 10 clones. All the 10 sequences were unknown before and derived from 6 unique novel genes among which the cDNA insert RCC18 has five copies. Northern blot analysis showed that RCC18 cDNA expressed highly in RCC, but there was no signal detected in normal kidney, and the full length of RCC18 was about 2.5 kb. The constructed cDNA subtractive library of human RCC is a highly efficient one and lays the solid foundation for large-scale screening and cloning new and specific oncogenes or tumor suppressor genes of RCC. 6Engineered biocontainable RNA virus vectors for non-transgenic genome editing across crop species and genotypes显示文摘CRISPR/Cas genome-editing tools provide unprecedented opportunities for basic plant biology research and crop breeding.However,the lack of robust delivery methods has limited the widespread adoption of these revolutionary technologies in plant science.Here,we report an efficient,non-transgenic CRISPR/Cas delivery platform based on the engineered tomato spotted wilt virus(TSWV),an RNA virus with a host range of over 1000 plant species.We eliminated viral elements essential for insect transmission to liberate genome space for accommodating large genetic cargoes without sacrificing the ability to infect plant hosts.The resulting non-insect-transmissible viral vectors enabled effective and stable in planta delivery of Cas12a and Cas9 nucleases as well as adenine and cytosine base editors.In systemically infected plant tissues,the deconstructed TSWV-derived vectors induced efficient somatic gene mutations and base conversions in multiple crop species with little genotype dependency.Plants with heritable,bi-allelic mutations could be readily regenerated by culturing the virus-infected tissues in vitro without antibiotic selection.Moreover,we showed that antiviral treatment with ribavirin during tissue culture cleared the viral vectors in 100%of regenerated plants and further augmented the recovery of heritable mutations.Because many plants are recalcitrant to stable transformation,the viral delivery system developed in this work provides a promising tool to overcome gene delivery bottlenecks for genome editing in various crop species and elite varieties.Qian Liu Chenglu Zhao Kai Sun Yinlu Deng Zhenghe Li 2023Molecular Plant2023,16,3:2
7High-efficiency Extraction of Bitumen from Oil Sands Using Mixture of Ionic Liquid [Emim][BF_(4)] and Dichloromethane显示文摘An ionic liquid(IL),1-ethyl-3-methyl imidazolium tetrafluoroborate([Emim][BF_(4)]),was used to enhance bitumen recovery from oil sands by dichloromethane solvent extraction.A multiphase system could be formed by simply mixing the components at ambient temperature,consisting of sands and clays,mixtures of ionic liquid and dichloromethane,and concentrated bitumen layer.The results demonstrated that[Emim][BF_(4)]increased the bitumen recovery up to 92%.Much less clay fines were found in the recovered bitumen than those formed by using dichloromethane solvent extraction alone,and the dichloromethane residue was not detected in the spent sands.We proposed that[Emim][BF_(4)]had an ability to reduce the adhesion of bitumen to sand,resulting in an improved separation efficiency.Furthermore,[Emim][BF_(4)]could facilitate the transfer of the extracted bitumen to the surface interface,and then the bitumen was auto-partitioned to a separate immiscible phase for ease of harvesting.This technology circumvented the issue of high consumption of distillation energy due to separation of bitumen phase and low boiling point of dichloromethane.[Emim][BF_(4)]and dichloromethane could be readily recycled through the system and used repeatedly.After ten cycles,the bitumen recovery remained above 88%.Initial scale-up work suggested that this approach would form the basis for a viable large-scale process.Sun Yinlu Sun Yizhuo Jia Yushan Yuan Wei Song Ximing 2021China Petroleum Processing & Petrochemical Technology2021,23,2:2

12PKB negatively modulates TGF-β responsiveness in prostate carcinoma PC-3 cells through its interaction with Smad3显示文摘Most prostate cancers are insensitive to growth-inhibitory effect of TGF-β, while PI3K-PKB signaling is highly activated in prostate cancers. We investigated whether the PI3K-PKB signaling con- tributes to TGF-β insensitivity in PTEN-null prostate cancer PC-3 cells. Cell growth analysis showed that inhibition of PI3K-PKB pathway by LY294002 en- hanced growth inhibition and cell cycle arrest induced by TGF-β. Furthermore, activation of PI3K-PKB pathway by insulin or overexpression of PKB de- creased the transcriptional activity of TGF-β, as measured by the TGF-β/Smad3-responsive CAGA- luciferase reporter, while inhibition of PI3K-PKB pathway by introducing PTEN, inactive PKB mutant or using LY294002 promoted TGF-β-induced expres- sion of CAGA-luciferase. Co-immunoprecipitation studies further demonstrated that Smad3 interacted with PKB through its linker region and MH2 domain. This interaction was facilitated by insulin and disrupted by TGF-β signaling activation. 17Lingual flange protrusion:diagnostic marker for metastatic liver cancer显示文摘OBJECTIVE: To investigate the diagnostic significance of lingual flange protrusion for liver metastasis of patients with malignant neoplasia.METHODS: The data of 191 patients with malignant neoplasia were analyzed. All photos of patients' tongue image were recorded and lingual flange protrusion was the positive standard. χ~2 test for paired data and Kappa test were used to determine the diagnostic value of lingual flange protrusion for metastatic liver cancer. Mann-Whitney U test was used to compare the levels of liver serolog-ical markers. The area under receiver operating characteristic curve(ROC) and logistic regression model were used to analyze the predictive values of lingual flange protrusion, alkaline phosphatase(ALP), and lactate dehydrogenase(LDH) levels.RESULTS: Patients with lingual flange protrusion had a higher risk of liver metastasis than those without it(P < 0.001). There was no significant difference in diagnosis of liver metastasis between lingual flange protrusion and traditional diagnostic criteria(P = 0.541). Kappa was 0.738(P < 0.001). Lingual flange protrusion was significantly correlated with increased serum ALP and LDH levels(P < 0.01).Comparison of ROC curves showed that the diagnostic value of lingual flange protrusion is better than ALP, LDH and the combination of ALP and LDH(P < 0.01). Furthermore, the combined diagnostic values of lingual flange protrusion and ALP,lingual flange protrusion and LDH, and lingual flange protrusion, ALP and LDH are not better than lingual flange protrusion alone(P > 0.05).CONCLUSION: Lingual flange protrusion is a potential diagnostic marker for liver metastasis of patients with malignant neoplasia.Ma Yinlu Yang Xue Zhu Cuihong Cao Rui Wu Xiongzhi 2017Journal of Traditional Chinese Medicine2017,37,6:0
