| 1 | QTL-Seq Identified a Major QTL for Grain Length and Weight in Rice Using Near Isogenic F_2 Population显示文摘Mapping and isolation of quantitative trait loci(QTLs)or genes controlling grain size or weight is very important to uncover the molecular mechanisms of seed development and crop breeding. To identify the QTLs controlling grain size and weight,we developed a near isogenic line F_2(NIL-F_2)population,which was derived from a residual heterozygous plant in an F_7 generation of recombinant inbred line(RIL).With the completion of more than 30×whole genome re-sequencing of the parents, two DNA bulks for large and small grains,a total of 58.94 Gb clean nucleotide data were generated.A total of455 262 single nucleotide polymorphisms(SNPs)between the parents were identified to perform bulked QTL-seq.A candidate genomic region containing SNPs strongly associated with grain length and weight was identified from 15 to 20 Mb on chromosome 5.We designated the major QTL in the candidate region as q TGW5.3.Then,q TGW5.3 was further validated with PCR-based conventional QTL mapping method through developing simple sequence repeat and Insertion/Deletion markers in the F_2 population.Furthermore, recombinants and the progeny tests delimited the candidate region of q TGW5.3 to 1.13 Mb,flanked by HX5009(15.15 Mb)and HX5003(16.28 Mb).A set of NILs,selected from the F_2 population, was developed to evaluate the genetic effect of q TGW5.3.Significant QTL effects were detected on grain length, grain width and 1000-grain weight of H12-29 allele with 1.14 mm,-0.11 mm and 3.11 g,which explained 99.64%,95.51%and 97.32%of the phenotypic variations, respectively. | QIN Yaobin CHENG Peng CHENG Yichen FENG Yue HUANG Derun HUANG Tingxu SONG Xianjun YING Jiezheng | 2018 | Rice science2018,25,3: | 2 |
| 2 | Novel evidence from Taxus fuana forests for niche-neutral process assembling community显示文摘Background:Understanding the mechanisms underlying community assembly is helpful for conservation and restoration of communities, particularly those that contain rare and endangered species like Taxus fuana, which are endemic to the Western Himalayas. The niche (limiting similarity) vs. neutral (randomness) assembly of the T.fuana forest community in Gyirong County, Tibet, China, was investigated. The net relatedness index (NRI) was calculated using a phylogenetic tree. The phylogenetic characteristics of the community and its relationships with environment were analyzed. Results:The value of the mean NRI at the community level was less than-1.96, indicating that the phylogenetic structure was overdispersed;whereas majority of the NRIs at the tree, shrub, and herb layers were within-1.96 to1.96, indicating random dispersion. Environmental factors accounted for 44.38%, 46.52%, 24.04%, and 14.07%of the variation at the community level, tree, shrub, and herb layer, respectively. The phylogenetic structure at the community level and tree layer were significantly influenced by both topographic and soil factors, while shrub and herb layers tended to be affected by a single environmental factor. Conclusions:Community assembly of the T. fuana forest was simultaneously affected by niche and neutral processes, and their variations were closely related to the environment. Neutral process dominated community assembly in the shrub and herb layers. However, the interaction of limiting similarity and randomness played a dominant role at the community level and tree layer;and contributed to maintenance of biodiversity stability. The synergy of multiple environmental factors had a more obvious influence on community assembly than individual environmental factors, especially at the community level. These findings would help to understand the conservation of rare and endangered tree species, such as T. fuana, in the native community;and highlight the importance of random and non-random processes in assembly and biodiversity maintenance of alpine plant communities. | Tianxiang Li Li Xu Feng Wang Weijun Zhang Junpeng Duan Xiaolu Shen-Tu Yaobin Song Runguo Zang Ming Dong | 2022 | Forest Ecosystems2022,9,3: | 1 |
| 3 | Molecular-level proton acceptor boosts oxygen evolution catalysis to enable efficient industrial-scale water splitting显示文摘Industrial water splitting has long been suppressed by the sluggish kinetics of the oxygen evolution reaction(OER),which requires a catalyst to be efficient.Herein,we propose a molecular-level proton acceptor strategy to produce an efficient OER catalyst that can boost industrial-scale water splitting.Molecular-level phosphate(-PO_(4))group is introduced to modify the surface of PrBa_(0.5)Ca_(0.5)Co_(2)O_(5)+δ(PBCC).The achieved catalyst(PO_(4)-PBCC)exhibits significantly enhanced catalytic performance in alkaline media.Based on the X-ray absorption spectroscopy results and density functional theory(DFT)calculations, the PO_(4)on the surface, which is regarded as the Lewis base,is the key factor to overcome the kinetic limitation of the proton transfer process during the OER.The use of the catalyst in a membrane electrode assembly(MEA)is further evaluated for industrial-scale water splitting, and it only needs a low voltage of 1.66 V to achieve a large current density of 1 A cm^(-2).This work provides a new molecular-level strategy to develop highly efficient OER electrocatalysts for industrial applications. | Yaobin Wang Qian Lu Xinlei Ge Feng Li Le Chen Zhihui Zhang Zhengping Fu Yalin Lu Yang Song Yunfei Bu | 2024 | Green Energy & Environment2024,9,2: | 0 |
| 4 | Single-nucleus transcriptomics reveals a gatekeeper role for FoxP1 in primate cardiac aging显示文摘Aging poses a major risk factor for cardiovascular diseases, the leading cause of death in the aged population. However, the cell type-specific changes underlying cardiac aging are far from being clear. Here,we performed single-nucleus RNA-sequencing analysis of left ventricles from young and aged cynomolgus monkeys to define cell composition changes and transcriptomic alterations across different cell types associated with age.We found that aged cardiomyocytes underwent a dramatic loss in cell numbers and profound fluctuations in transcriptional profles.Via transcription regulatory network analysis,we identified FOxP1,a core transcription factor in organ development,as a key downregulated factor in aged cardiomyocytes, concomitant with the dysregulation of FoxP1 target genes associated with heart function and cardiac diseases.Consistently, the deficiency of FOxP1 led to hypertrophic and senescent phenotypes in human embryonic stem cell-derived cardiomyocytes.Altogether, our findings depict the celiular and molecular landscape of ventricular aging at the single-cell resolution, and identify drivers for primate cardiac aging and potential targets for intervention against cardiac aging and associated diseases. | Yiyuan Zhang Yandong Zheng Si Wang Yanling Fan Yanxia Ye Yaobin Jing Zunpeng Liu Shanshan Yang Muzhao Xiong Kuan Yang Jinghao Hu Shanshan Che Qun Chu Moshi Song Guang-Hui Liu Weiqi Zhang Shuai Ma Jing Qu | 2023 | Protein & Cell2023,14,4: | 0 |
