| 1 | Jurassic Hornblende Gabbros in Dongga,Eastern Gangdese,Tibet:Partial Melting of Mantle Wedge and Implications for Crustal Growth显示文摘The Gangdese batholith, more than 2500 km in length, is composed mainly of JurassicMiocene igneous rocks. This batholith is one of the most important constituents of the Tibetan orogenesis and provides an ideal place for study of Neo-Tethyan ocean geodynamic evolution and plateau uplift. Recent studies on the Gangdese Jurassic felsic magmatism highlight its juvenile source. However, important aspects concerning the genesis of the juvenile magmatism and related deep geodynamic evolution are still unclear. Here, we report detailed petrological, geochronological, geochemical, whole-rock Sr-Nd isotopic, and in situ Sr-Hf isotopic data for a recently identified hornblende gabbro in the Dongga area, southern Lhasa sub-block. This hornblende gabbro is dominated by hornblende and plagioclase, dated at Early Jurassic(ca. 180–190 Ma), and characterized by a narrow compositional range in SiO_2(49.38wt%–52.27wt%), MgO(4.08wt%–7.00wt%), FeO(10.43wt%–11.77wt%), Na_2O(2.58wt%–3.51wt%), and K_2O(0.48wt%–1.53wt%). It has depleted isotopic signatures, with whole-rock(^(87)Sr/^(86)Sr)i ratios of 0.7033–0.7043, ε_(Nd)(t) values of +4.90 to +6.99, in situ plagioclase(^(87)Sr/^(86)Sr)i ratios of 0.7034–0.7042, and zircon ε_(Hf)(t) of +12.2 to +16.8. Our results integrated with published data suggest a model of Gangdese juvenile crustal growth by a subduction-related water-enriched mantle wedge. The hydrous partial melting of the lithosphere mantle was triggered by the dehydration of a Neo-Tethyan oceanic slab. | 2 | SERK Receptor-like Kinases Control Division Patterns of Vascular Precursors and Ground Tissue Stem Cells during Embryo Development in Arabidopsis显示文摘During embryo development, the vascular precursors and ground tissue stem cells divide to renew them-selves and produce the vascular tissue, endodermal cells, and cortical cells. However, the molecular mech-anisms regulating division of these stem cells have remained largely elusive. In this study, we show that loss of function of SOMATIC EMBRYOGENESIS RECEPTOR-UKE KINASE (SERK) genes results in aberrant em-bryo development. Fewer cortical, endodermal, and vascular cells are generated in the embryos of serk1 serk2bak1 triple mutants. WUSCHEL-RELATED HOMBOBOX5 (WOXS) is ectopically expressed in vascular cells of serkl serk2 bak1 embryos. The first transverse division of vascular precursors in mid-globular em-bryos and second asymmetric division of ground tissue stem cells in early-heart embryos are abnormally altered to a longitudinal division. The embryo defects can be partially rescued by constitutively activated mitogen-activated protein kinase (MAPK) kinase kinase YODA (YDA) and MAPK kinase MKK5. | 3 | Methionine deficiency and its hydroxy analogue influence chicken intestinal 3-dimensional organoid development显示文摘Methionine and its hydroxy analogue(MHA)have been shown to benefit mouse intestinal regeneration.The intestinal organoid is a good model that directly reflects the impact of certain nutrients or chemicals on intestinal development.Here,we aimed to establish a chicken intestinal organoid culture method first and then use the model to explore the influence of methionine deficiency and MHA on intestinal organoid development.The results showed that 125-mm cell strainer exhibited the highest efficiency for chicken embryo crypt harvesting.We found that transforming growth factor-b inhibitor(A8301)supplementation promoted enterocyte differentiation at the expense of the proliferation of intestinal stem cells(ISC).The mitogen-activated protein kinase p38 inhibitor(SB202190)promoted intestinal organoid formation and enterocyte differentiation but suppressed the differentiation of enteroendocrine cells,goblet cells and Paneth cells.However,the suppression of enteroendocrine cell and Paneth cell differentiation by SB202190 was alleviated at the presence of A8301.The glycogen synthase kinase 3 inhibitor(CHIR99021),valproic acid(VPA)alone and their combination promoted chicken intestinal organoid formation and enterocyte differentiation at the expense of the expression of Paneth cells and goblet cells.Chicken serum significantly improved organoid formation,especially in the presence of A8301,SB202190,CHIR99021,and VPA,but inhibited the differentiation of Paneth cells and enteroendocrine cells.Chicken serum at a concentration of 0.25%meets the requirement of chicken intestinal organoid development,and the beneficial effect of chicken serum on chicken intestinal organoid culture could not be replaced by fetal bovine serum and insulin-like growth factor-1.Moreover,commercial mouse organoid culture medium supplemented with A8301,SB202190,CHIR99021,VPA,and chicken serum promotes chicken organoid budding.Based on the chicken intestinal organoid model,we found that methionine deficiency mimicked by cycloleucine suppressed organoid formation and organoid size,and this effect was reinforced with increased cycloleucine concentrations.Methionine hydroxy analogue promoted regeneration of ISC but decreased cell differentiation compared with the results obtained with L-methionine.In conclusion,our results provide a potentially excellent guideline for chicken intestinal organoid culture and insights into methionine function in crypt development. | Youli Wang Qihang Hou Yuqin Wu Yanwei Xu Yan Liu Jing Chen Lingling Xu Yuming Guo Shuai Gao Jianmin Yuan | 2022 | Animal Nutrition2022,,1: | 4 |
| 4 | Edema and neuronal apoptosis in the hippocampus and cortex of elderly rats following transient cerebral ischemia/reperfusion injury显示文摘BACKGROUND: Previous studies of cerebral ischemia have used young animals, with an ischemictime greater than 5 minutes (safe time limit). Despite an increased understanding of neuronalapoptosis, it remains uncertain whether brief cerebral ischemic events of 5 minutes or less damagebrain tissue in elderly rodents.OBJECTIVE: To investigate the effects of transient cerebral ischemia (5 minutes)/reperfusion injuryon brain cortical and hippocampal edema, aquaporin-4 (AQP-4) expression, and neuronal apoptosisin aged rats, and to compare ischemic sensitivity between cortex and hippocampus.DESIGN, TIME AND SETTING: A randomized, controlled, animal experiment was performed at theInstitute of Cerebrovascular Disease, Qingdao University Medical School from April 2008 to March2009.MATERIALS: Rabbit anti-AQP-4 polyclonal antibody, TUNEL kit, and SABC immunohistochemistrykit were purchased from Wuhan Boster Bioengineering, China.METHODS: A total of 160 healthy, male, aged 19-21 months, Wistar rats were randomly assignedto 4 groups: sham-surgery, and ischemia 1-, 3-, and 5-minute groups, with 40 rats in each group.The global cerebral ischemia model was established using the Pusinelli four-vessel occlusion, andthe three cerebral ischemia groups were subdivided into reperfusion 12-hour, 1-, 2-, 3-, and 7-daysubgroups, with 8 rats in each subgroup. The sham-surgery group was subjected to exposure of thefirst cervical bilateral alar foramina and bilateral common carotid arteries.MAIN OUTCOME MEASURES: The dry-wet weight assay was used to measure brain water contentand histopathology of the cortex and hippocampus was observed following hematoxylin-eosinstaining. In addition, cortical and hippocampal AQP-4 expression was detected by streptavidin-biotincomplex immunohistochemistry, and neuronal apoptosis was detected by the TUNEL method.RESULTS: There was no significant difference in brain water content or AQP-4 expression in thecortex and hippocampus between ischemia 1- and 3-minute groups and the sham-surgery group orbrain water content or AQP-4 expression in the cortex between ischemia 5-minute group andsham-surgery group (P> 0.05). However, brain water content and AQP-4 expression in thehippocampus after 5 minutes of cerebral ischemia were significantly increased compared with thesham-surgery group (P < 0.05 or P < 0.01 ). Several TUNEL-positive cells were observed in thecortex and hippocampus of the sham-surgery group and ischemia 1-minute group, as well as in thecortex of the ischemia 3-minute group. In addition, the number of apoptotic neurons in thehippocampus of ischemia 3-minute group and in the cortex and hippocampus of ischemia 5-minutegroup was significantly increased (P < 0.05 or P < 0.01). Neuronal apoptosis was increased after 12hours of ischemia/reperfusion, and it reached a peak by 2 days (P < 0.01).CONCLUSION: Transient cerebral ischemia (5 minutes) resulted in increased hippocampal edema,AQP-4 expression, and neuronal apoptosis. | 5 | Comparative population genomics dissects the genetic basis of seven domestication traits in jujube显示文摘Jujube(Ziziphus jujuba Mill.)is an important perennial fruit tree with a range of interesting horticultural traits.It was domesticated from wild jujube(Ziziphus acidojujuba),but the genomic variation dynamics and genetic changes underlying its horticultural traits during domestication are poorly understood.Here,we report a comprehensive genome variation map based on the resequencing of 350 accessions,including wild,semi-wild and cultivated jujube plants,at a>15×depth.Using the combination of a genome-wide association study(GWAS)and selective sweep analysis,we identified several candidate genes potentially involved in regulating seven domestication traits in jujube.For fruit shape and kernel shape,we integrated the GWAS approach with transcriptome profiling data,expression analysis and the transgenic validation of a candidate gene to identify a causal gene,ZjFS3,which encodes an ethyleneresponsive transcription factor.Similarly,we identified a candidate gene for bearing-shoot length and the number of leaves per bearing shoot and two candidate genes for the seed-setting rate using GWAS.In the selective sweep analysis,we also discovered several putative genes for the presence of prickles on bearing shoots and the postharvest shelf life of fleshy fruits.This study outlines the genetic basis of jujube domestication and evolution and provides a rich genomic resource for mining other horticulturally important genes in jujube. | Mingxin Guo Zhongren Zhang Yanwei Cheng Sunan Li Peiyin Shao Qiang Yu Junjie Wang Gan Xu Xiaotian Zhang Jiajia Liu Linlin Hou Hanxiao Liu Xusheng Zhao | 2020 | Horticulture Research2020,7,1: | 2 |

| 8 | Relationship of Microchannels and Plaque Erosion in Patients with ST-Segment Elevation Myocardial Infarction: An Optical Coherence Tomography Study显示文摘Objective:Microchannels are associated with the progression of atherosclerotic vulnerable plaques.However,in patients with culprit optical coherence tomography(OCT)-defined plaque erosion,the knowledge of microchannels and culprit lesion vulnerability is limited.The aim of this study was to investigate culprit lesion characteristics in patients with ST-segment elevated myocardial infarction(STEMI)caused by plaque erosion with and without microchannels using OCT.Methods:In all,348 STEMI patients with plaque erosion who underwent OCT of the culprit lesion at the 2nd Affiliated Hospital of Harbin Medical University(Harbin,China)from August 2014 to December 2017 were included and divided into the microchannel group(n=116,33.3%)and no-microchannel group(n=232,66.7%).The clinical characteristics and OCT-derived plaque features were compared between both groups.Results:Among the 348 STEMI patients with plaque erosion,culprit lesions with microchannels had higher incidence of lipid plaque(59.5%vs.45.3%,P=0.012);calcification(41.4%vs.24.6%,P=0.002);spotty calcification(30.2%vs.18.1%,P=0.014);macrophages accumulation(72.4%vs.45.7%,P<0.001);and cholesterol crystals(32.8%vs.14.2%,P<0.001)than those without microchannels.In addition,minimal lumen area was smaller((1.9±0.9)mm2 vs.(2.8±2.3)mm2,P<0.001)and lumen area stenosis was greater((71.3%±13.4%)vs.(65.3%±19.3%),P=0.001)in the microchannel group than in the no-microchannel group.Conclusion:In patients with STEMI caused by plaque erosion,one-third manifested typical microchannel characteristics,and those with microchannels were associated with more severe luminal stenosis and more vulnerable plaque features than those without microchannels. | Senqing Jiang Junchen Guo Yanwei Yin Chao Fang Jifei Wang Yidan Wang Fangmeng Lei Sibo Sun Xueying Pei Ruyi Jia Shaotao Zhang Lulu Li Yini Wang Lei Xing Huai Yu Huimin Liu Maoen Xu Xuefeng Ren Lijia Ma Guo Wei Jingbo Hou Jiannan Dai Bo Yu | 2022 | Cardiology Discovery2022,2,2: | 0 |
| 9 | Comprehensive and deep profiling of the plasma proteome with protein corona on zeolite NaY显示文摘Proteomic characterization of plasma is critical for the development of novel pharmacodynamic biomarkers.However,the vast dynamic range renders the profiling of proteomes extremely challenging.Here,we synthesized zeolite NaY and developed a simple and rapid method to achieve comprehensive and deep profiling of the plasma proteome using the plasma protein corona formed on zeolite NaY.Specifically,zeolite NaY and plasma were co-incubated to form plasma protein corona on zeolite NaY(NaY-PPC),followed by conventional protein identification using liquid chromatography-tandem mass spectrometry.NaY was able to significantly enhance the detection of low-abundance plasma proteins,minimizing the“masking”effect caused by high-abundance proteins.The relative abundance of middleand low-abundance proteins increased substantially from 2.54%to 54.41%,and the top 20 highabundance proteins decreased from 83.63%to 25.77%.Notably,our method can quantify approximately 4000 plasma proteins with sensitivity up to pg/mL,compared to only about 600 proteins identified from untreated plasma samples.A pilot study based on plasma samples from 30 lung adenocarcinoma patients and 15 healthy subjects demonstrated that our method could successfully distinguish between healthy and disease states.In summary,this work provides an advantageous tool for the exploration of plasma proteomics and its translational applications. | Congcong Ma Yanwei Li Jie Li Lei Song Liangyu Chen Na Zhao Xueping Li Ning Chen Lixia Long Jin Zhao Xin Hou Li Ren Xubo Yuan | 2023 | Journal of Pharmaceutical Analysis2023,13,5: | 0 |
| 10 | A robust indium-organic framework with open tubular channels for efficient separation of acetylene显示文摘Considering the tremendous applications and purification requirement of acetylene(C_(2)H_(2)),seeking appropriate adsorbents with high capacity and selectivity is a vital task and remains an enduring challenge.Herein,we designed and synthesized a robust three-dimensional(3D)indium-organic framework([(Me)_(2)NH_(2)][In(L6)_(0.5)(IPA)_(0.5)]·DMA·2H_(2)O(In-L6-IPA,DMA=dimethylammonium,IPA=isopropyl alcohol))featuring two types of one-dimensional(1D)tubular channels.The activated In-L6-IPA displayed high loading for C_(2)H_(2)(104.4 cm^(3)·g^(-1),the second highest value among all reported indium-based metal-organic frameworks(MOFs))and simultaneously selective adsorption for C_(2)H_(2) over CO_(2),C_(2)H_(6),and ethylene(C_(2)H_(4))at 298 K under 100 kPa.Molecular modelling revealed that the porous wall of In-L6-IPA provides more and stronger multiple interactions for C_(2)H_(2) than CO_(2),C_(2)H_(6),and C_(2)H_(4) containing C–H···π,C–H···O,and O···πinteractions.Breakthrough experiments validated the actual separation ability for various ratios of binary C_(2)H_(2)/C_(2)H_(4) and C_(2)H_(2)/CO_(2) mixtures as well as equimolar ternary C_(2)H_(2)/C_(2)H_(4)/CO_(2) and C_(2)H_(2)/C_(2)H_(4)/C_(2)H_(6) mixtures with excellent reusability. | Yong-Zhi Li Gang-Ding Wang Fan Xu Qing Yin Danyang Zhao Jiqiu Qi Yanwei Sui Lei Hou Yao-Yu Wang | 2024 | Nano Research2024,17,4: | 0 |
