1Induction of mitochondrion-mediated apoptosis of CHO cells by tripchloro lide显示文摘Tripchlorolide (TC) is a potent antitumor reagent purified from a Chinese herb Tripterygium Wilfordii Hook. f.. However, its cellular effects and mechanism of action are unknown. We showed here that TC induced apoptosis of Chinese Hamster Ovary (CHO) cells in time- and dose-dependent manners. TC resulted in the degradation of Bcl-2, the translocation of Bax from the cytosol to mitochondria, and the release of cytochrome c from mitochondria. Stable overexpression of human Bcl-2 could reduce the apoptosis of TCtreated cells by blocking the translocation of Bax and the release of cytochrome c. These results indicate that TC induces apoptosis of CHO cell by activating the mitochondrion-mediated apoptotic pathway involving the proteins of Bcl-2 family and cytochrome c.YANREN LEIXIONG JIARUIWU 2003Cell Research2003,13,4:11
2The C-Terminal Portion of the Nucleocapsid Protein Demonstrates SARS-CoV Antigenicity显示文摘In order to develop clinical diagnostic tools for rapid detection of SARS-CoV (severe acute respiratory syndrome-associated coronavirus) and to identify candidate proteins for vaccine development, the C-terminal portion of the nucleocapsid (NC)gene was amplified using RT-PCR from the SARS-CoV genome, cloned into a yeast expression vector (pEGH), and expressed as a glutathione S-transferase (GST) and Hisx6 double-tagged fusion protein under the control of an inducible promoter.Western analysis on the purified protein confirmed the expression and purification of the NC fusion proteins from yeast. 4Differential activation of intra-S-phase checkpoint in response to tripchlorolide and its effects on DNA replication显示文摘DNA replication is tightly regulated during the S phase of the cell cycle, and the activation of the intra-S-phase checkpoint due to DNA damage usually results in arrest of DNA synthesis. However, the molecular details about the correlation between the checkpoint and regulation of DNA replication are still unclear. To investigate the connections between DNA replication and DNA damage checkpoint, a DNA-damage reagent, tripchlorolide, was applied to CHO (Chinese ovary hamster) cells at early- or middle-stages of the S phase. The early-S-phase treatment with TC significantly delayed the progression of the S phase and caused the phosphorylation of the Chk1 checkpoint protein, whereas the middle-S-phase treatment only slightly slowed down the progression of the S phase. Furthermore, the analysis of DNA replication patterns revealed that replication pattern Ⅱ was greatly prolonged in the cells treated with the drug during the early-S phase, whereas the late-replication patterns of these cells were hardly detected, suggesting that the activation of the intra-S-phase checkpoint inhibits the late-origin firing of DNA replication. We conclude that cells at different stages of the S phase are differentially sensitive to the DNA-damage reagent, and the activation of the intra-Sphase checkpoint blocks the DNA replication progression in the late stage of S phase.YanREN JiaRuiWU 2004Cell Research2004,14,3:2

8Application of proprotein convertase subtilisin/kexin type 9 inhibitor, evolocumab, in patients with severe hypertriglyceridemia显示文摘To the Editor:The proprotein convertase subtilisin/kexin type 9(PCSK9)-inhibitor,evolocumab,can bind with PCSK9 in plasma and reduce the degradation of the PCSK9-low density lipoprotein(LDL)receptor complex,thereby reducing LDL levels.[1]In clinical practice,PCSK9 inhibitors can reduce low-density lipoprotein cholesterol(LDL-c)significantly and reduce triglycerides(TG)by about 8%to 17%.[2]We used PCSK9 inhibitors to lower blood lipids in three patients with severe hypertriglyceridemia(HTG)and performed whole-exome sequencing(WES)to analyze the genes that may contribute to hyperlipidemia in detail.We have obtained patients'consent forms.We found that PCSK9 inhibitors significantly reduced blood lipid levels,regardless of the presence of HTG mutations.These results provide new evidence for the use of PCSK9 inhibitors in the treatment of patients with severe HTG.Yanren Peng Danxia Guo Sijie Jiang Hua Zheng 2022Chinese Medical Journal2022,,6:1
