| 2 | A potential oncogenic role of the commonly observed E2F5 overexpression in hepatocellular carcinoma显示文摘AIM: To explore the expression pattern of E2F5 in primary hepatocellular carcinomas (HCCs) and elucidate the roles of E2F5 in hepatocarcinogenesis. METHODS: E2F5 expression was analyzed in 120 primary HCCs and 29 normal liver tissues by immunohistochemistry analysis. E2F5-small interfering RNA was transfected into HepG2, an E2F5-overexpressed HCC cell line. After E2F5 knockdown, cell growth capacity and migrating potential were examined. RESULTS: E2F5 was significantly overexpressed in primary HCCs compared with normal liver tissues (P = 0.008). The E2F5-silenced cells showed significantly reduced proliferation (P = 0.004). On the colony formation and soft agar assays, the number of colonies was significantly reduced in E2F5-silenced cells (P = 0.004 and P = 0.009, respectively). E2F5 knockdown resulted in the accumulation of G0/G1 phase cells and a reduction of S phase cells. The number of migrating/invading cells was also reduced after E2F5 knockdown (P = 0.021). | 3 | Effects of Banxia Xiexin Decoction (半夏泻心汤) on Cisplatin-Induced Apoptosis of Human A549 Lung Cancer Cells显示文摘Objective: To examinie the synergistic effects of Banxia Xiexin Decoction(半夏泻心汤,Known as Banhasasim-tang in Korean) extract(BXDE) on cisplatin-induced cytotoxicity in the A549 human lung cancer cell lines. Methods: A549 cells were treated with varying concentrations(50–200 μg/m L) of cisplatin and BXDE alone or in combination for 96 h. We used 1-(4,5-dimethylthiazol-2-yl)-3,5-diphenylformazan assay and flow cytometry to analyze cell viability and apoptosis, respectively. Results: The exposure of cells to cisplatin and BXDE alone or in combination decreased cell viability dose-and time-dependently(P<0.05), which was found to be mediated by the apoptotic pathway as confirmed by the increase in the annexin V+/propidium iodide-stained cell population and a ladder pattern of discontinuous DNA fragments. Furthermore, the apoptosis was inhibited by the pan-caspase inhibitor, benzyloxycarbonyl-Val-Ala-Asp(OMe) fluoromethylketone(z-VAD-FMK). Conclusions: BXDE significantly potentiated apoptotic effects of cisplatin in A549 cells. | 4 | Anti-Proliferative Effects of Ginsenosides Extracted from Mountain Ginseng on Lung Cancer显示文摘Objective:To investigate the effect of three major ginsenosides from mountain ginseng as anticancer substance and explore the underlying mechanism involved in lung cancer.Methods:The inhibitory proliferation of lung cancer by major five ginsenosides(Rb1,Rb2,Rg1,Re,and Re) was examined using3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay.Calculated 50%inhibition(IC_(50)) values of five ginsenosides were determined and compared each other.Apoptosis by the treatment of single ginsenoside was performed by fluorescence-assisted cytometric spectroscopy.The alterations of apoptosis-related proteins were evaluated by Western blot analysis.Results:The abundance of ginsenosides in butanol extract of mountain ginseng(BX-MG) was revealed in the order of Rb1,Rg1,Re,Re and Rb2.Among them,Rb1 was the most effective to lung cancer cell,followed by Rb2 and Rg1 on the basis of relative IC_(50) values of IMR90 versus A549 cell.The alterations of apoptotic proteins were confirmed in lung cancer A549 cells according to the administration of Rb1,Rb2 and Rg1.The expression levels of caspase-3 and caspase-8 were increased upon the treatment of three ginsenosides,however,the levels of caspase-9 and anti-apoptotic protein Bax were not changed.Conclusion:Major ginsenosides such as Rb1,Rb2 and Rg1 comprising BX-MG induced apoptosis in lung cancer cells via extrinsic apoptotic pathway rather than intrinsic mitochondrial pathway. | Dong Gyu Lee Sung-ll Jang Young-Rang Kim Kyeong Eun Yang So Jung Yoon Zee-Won Lee Hyun Joo An Ik-Soon Jang Jong-Soon Choi Hwa-Seung Yoo | 2016 | Chinese Journal of Integrative Medicine2016,22,5: | 9 |
| 5 | Lycium barbarum polysaccharide-glycoprotein preventative treatment ameliorates aversive stimuli-induced depression显示文摘Previous studies have shown that Lycium barbarum polysaccharide,the main active component of Lycium barbarum,exhibits antiinflammatory and antioxidant effects in treating neurological diseases.However,the therapeutic action of Lycium barbarum polysaccharide on depression has not been studied.In this investigation,we established mouse models of depression using aversive stimuli including exposure to fox urine,air puff and foot shock and physical restraint.Concurrently,we administered 5 mg/kg per day Lycium barbarum polysaccharide-glycoprotein to each mouse intragastrically for the 28 days.Our results showed that long-term exposure to aversive stimuli significantly enhanced depressive-like behavior evaluated by the sucrose preference test and the forced swimming test and increased anxietylike behaviors evaluated using the open field test.In addition,aversive stimuli-induced depressed mice exhibited aberrant neuronal activity in the lateral habenula.Importantly,concurrent Lycium barbarum polysaccharide-glycoprotein treatment significantly reduced these changes.These findings suggest that Lycium barbarum polysaccharide-glycoprotein is a potential preventative intervention for depression and may act by preventing aberrant neuronal activity and microglial activation in the lateral habenula.The study was approved by the Jinan University Institutional Animal Care and Use Committee(approval No.20170301003)on March 1,2017. | Yun-Wei Fu Yan-Fang Peng Xiao-Dan Huang Yan Yang Lu Huang Yue Xi Zheng-Fang Hu Song Lin Kwok-Fai So Chao-Ran Ren | 2021 | Neural Regeneration Research2021,16,3: | 8 |
| 6 | Garlic-derived compound S-allylmercaptocysteine inhibits hepatocarcinogenesis through targeting LRP6/Wnt pathway显示文摘Whether and how garlic-derived S-allylmercaptocysteine(SAMC) inhibits hepatocellular carcinoma(HCC) is largely unknown. In the current study, the role of low-density lipoprotein receptor(LDLR)-related protein 6(LRP6) in HCC progression and the anti-HCC mechanism of SAMC was examined in clinical sample, cell model and xenograft/orthotopic mouse models. We demonstrated that SAMC inhibited cell proliferation and tumorigenesis, while induced apoptosis of human HCC cells without influencing normal hepatocytes. SAMC directly interacted with Wnt-pathway co-receptor LRP6 on the cell membrane. LRP6 was frequently over-expressed in the tumor tissue of human HCC patients(66.7% of 48 patients) and its overexpression only correlated with the over-expression of β-catenin, but not with age, gender, tumor size, stage and metastasis. Deficiency or over-expression of LRP6 in hepatoma cells could partly mimic or counteract the anti-tumor properties of SAMC, respectively. In vivo administration of SAMC significantly suppressed the growth of Huh-7 xenograft/orthotopic HCC tumor without causing undesirable side effects. In addition, stable down-regulation of LRP6 in Huh-7 facilitated the anti-HCC effects of SAMC. In conclusion, LRP6 can be a potential therapeutic target of HCC. | 7 | On-demand design of spectrally sensitive multiband absorbers using an artificial neural network显示文摘We report an approach assisted by deep learning to design spectrally sensitive multiband absorbers that work in the visible range.We propose a five-layered metal-insulator-metal grating structure composed of aluminum and silicon dioxide,and we design its structural parameters by using an artificial neural network(ANN).For a spectrally sensitive design,spectral information of resonant wavelengths is additionally provided as input as well as the reflection spectrum.The ANN facilitates highly robust design of a grating structure that has an average mean squared error(MSE)of 0.023.The optical properties of the designed structures are validated using electromagnetic simulations and experiments.Analysis of design results for gradually changing target wavelengths of input shows that the trained ANN can learn physical knowledge from data.We also propose a method to reduce the size of the ANN by exploiting observations of the trained ANN for practical applications.Our design method can also be applied to design various nanophotonic structures that are particularly sensitive to resonant wavelengths,such as spectroscopic detection and multi-color applications. | Sunae So Younghwan Yang Taejun Lee Junsuk Rho | 2021 | Photonics Research2021,9,4: | 4 |
| 8 | Temporal trends in the misdiagnosis rates between Crohn's disease and intestinal tuberculosis显示文摘AIM To investigate the temporal trends in the misdiagnosis rate between Crohn's disease(CD) and intestinal tuberculosis(ITB) in South Korea. METHODS We retrospectively reviewed the medical records of patients managed for CD or ITB at Asan Medical Center, a tertiary referral hospital, Seoul, Korea between 1996 and 2014. The temporal trends in the misdiagnosis rates between the two diseases were analyzed. The demographic and clinical characteristics were compared between CD patients who were initially misdiagnosed as ITB(final CD group) and vice versa(final ITB group). Final diagnostic criteria for ITB and medication for CD before definite diagnosis of TB were also analyzed in final ITB group.RESULTS In total, 2760 patients were managed for CD and 772 patients for ITB between 1996 and 2014. As well, 494 of the 2760 CD patients(17.9%) were initially misdiagnosed as ITB and 83 of the 772 ITB patients(10.8%) as CD. The temporal trend in misdiagnosing CD as ITB showed a decrease(OR = 0.89, 95%CI: 0.87-0.91, P < 0.001), whereas the temporal trend in misdiagnosing ITB as CD showed an increase(OR = 1.06, 95%CI: 1.01-1.11, P = 0.013). Age at diagnosis, presenting symptoms, and proportion of patients with active/past perianal fistula and active/inactive pulmonary tuberculosis(TB) were significantly different between final CD group and final ITB group. Forty patients(48.2%) in final ITB group were diagnosed by favorable response to empirical anti-TB treatment. Seventeen patients(20.5%) in final ITB group had inappropriately received corticosteroids and/or thiopurines due to misdiagnosis as CD. However, there were no mortalities in both groups. | 9 | Shape Estimation of Soft Manipulator Using Stretchable Sensor显示文摘The soft robot manipulator is attracting attention in the surgical fields with its intrinsic softness,lightness in its weight,and safety toward the human organ.However,it cannot be used widely because of its difficulty of control.To control a soft robot manipulator accurately,shape sensing is essential.This paper presents a method of estimating the shape of a soft robot manipulator by using a skin-type stretchable sensor composed of a multiwalled carbon nanotube(MWCNT)and silicone(p7670).The sensor can be easily fabricated and applied by simply attaching it to the surface of the soft manipulator.In its fabrication,MWCNT is sprayed on a teflon sheet,and liquid-state silicone is poured on it.After curing,we turn it over and cover it with another silicone layer.The sensor is fabricated with a sandwich structure to decrease the hysteresis of the sensor.After calibration and determining the relationship between the resistance of the sensor and the strain,three sensors are attached at 120°intervals.Using the obtained data,the curvature of the manipulator is calculated,and the entire shape is reconstructed.To validate its accuracy,the estimated shape is compared with the camera data.We experiment with three,six,and nine sensors attached,and the result of the error of shape estimation is compared.As a result,the minimum tip position error is approximately 8.9 mm,which corresponded to 4.45%of the total length of the manipulator when using nine sensors. | Jinho So Uikyum Kim Yong Bum Kim Dong-Yeop Seok Sang Yul Yang Kihyeon Kim Jae Hyeong Park Seong Tak Hwang Young Jin Gong Hyouk Ryeol Choi | 2021 | Cyborg and Bionic Systems2021,,1: | 2 |
