Offline two-dimensional liquid chromatography coupled with ion mobility-quadrupole time-of-flight mass spectrometry enabling fourdimensional separation and characterization of the multicomponents from white ginseng and red ginseng显示文摘Inherent complexity of plant metabolites necessitates the use of multi-dimensional information to accomplish comprehensive profiling and confirmative identification.A dimension-enhanced strategy,by offline two-dimensional liquid chromatography/ion mobility-quadrupole time-of-flight mass spectrometry(2 D-LC/IM-QTOF-MS)enabling four-dimensional separations(2 D-LC,IM,and MS),is proposed. In combination with in-house database-driven automated peak annotation, this strategy was utilized to characterize ginsenosides simultaneously from white ginseng(WG)and red ginseng(RG).An offline 2 DLC system configuring an Xbridge Amide column and an HSS T3 column showed orthogonality 0.76 in the resolution of ginsenosides.Ginsenoside analysis was performed by data-independent high-definition MSE(HDMSE)in the negative ESI mode on a Vion?IMS-QTOF hybrid high-resolution mass spectrometer, which could better resolve ginsenosides than MSEand directly give the CCS information. An in-house ginsenoside database recording 504 known ginsenosides and 58 reference compounds, was established to assist the identification of ginsenosides.Streamlined workflows,by applying UNIFI?to automatedly annotate the HDMSEdata, were proposed. We could separate and characterize 323 ginsenosides(including 286 from WG and 306 from RG),and 125 thereof may have not been isolated from the Panax genus. The established 2 D-LC/IM-QTOF-HDMSEapproach could also act as a magnifier to probe differentiated components between WG and RG.Compared with conventional approaches, this dimensionenhanced strategy could better resolve coeluting herbal components and more efficiently, more reliably identify the multicomponents, which,we believe, offers more possibilities for the systematic exposure and confirmative identification of plant metabolites. | Tiantian Zuo Chunxia Zhang Weiwei Li Hongda Wang Ying Hu Wenzhi Yang Li Jia Xiaoyan Wang Xiumei Gao Dean Guo | 2020 | Journal of Pharmaceutical Analysis2020,10,6: | 9
Dry electrode for the measurement of biopotential signals显示文摘This paper introduces a kind of silicon-based dry electrode for measuring biological signals. It uses microneedle arrays to penetrate into the stratum corneum to reduce skin impedance. The dry electrode requires neither skin preparation nor the electrolytic gel,is easy to use and causes no skin allergy. Two different technologies are chosen to manufacture microneedle arrays of dry electrode. One is deep dry etching combined with isotropic wet etching. The other is mechanical dicing combined with chemical wet etching (including isotropic wet etching and anisotropic wet etching).Microneedle arrays are coated with metal and divided into 25 mm 2 as dry electrode patch.Impedance testing shows that the impedance value of dry electrode can be comparable with that of commercial electrode in the 20 Hz-10 kHz frequency range. The steady-state visual evoked potential recording and analysis prove that the dry electrode can be used to detect electroencephalography. | WANG Yu PEI WeiHua GUO Kai GUI Qiang LI XiaoQian CHEN HongDa YANG JianHong | 2011 | Science China(Information Sciences)2011,54,11: | 6
Sex disparity of lung cancer risk in non-smokers:a multicenter population-based prospective study based on China National Lung Cancer Screening Program显示文摘Background: Non-smokers account for a large proportion of lung cancer patients, especially in Asia, but the attention paid to them is limited compared with smokers. In non-smokers, males display a risk for lung cancer incidence distinct from the females-even after excluding the influence of smoking;but the knowledge regarding the factors causing the difference is sparse. Based on a large multicenter prospective cancer screening cohort in China, we aimed to elucidate the interpretable sex differences caused by known factors and provide clues for primary and secondary prevention. Methods: Risk factors including demographic characteristics, lifestyle factors, family history of cancer, and baseline comorbidity were obtained from 796,283 Chinese non-smoking participants by the baseline risk assessment completed in 2013 to 2018. Cox regression analysis was performed to assess the sex difference in the risk of lung cancer, and the hazard ratios (HRs) that were adjusted for different known factors were calculated and compared to determine the proportion of excess risk and to explain the existing risk factors. Results: With a median follow-up of 4.80 years, 3351 subjects who were diagnosed with lung cancer were selected in the analysis. The lung cancer risk of males was significantly higher than that of females;the HRs in all male non-smokers were 1.29 (95% confidence interval [CI]: 1.20-1.38) after adjusting for the age and 1.38 (95% CI: 1.28-1.50) after adjusting for all factors, which suggested that known factors could not explain the sex difference in the risk of lung cancer in non-smokers. Known factors were 7% (|1.29-1.38|/1.29) more harmful in women than in men. For adenocarcinoma, women showed excess risk higher than men, contrary to squamous cell carcinoma;after adjusting for all factors, 47% ([1.30-1.16]/[1.30-1]) and 4% ([7.02-6.75]/[7.02-1])) of the excess risk was explainable in adenocarcinoma and squamous cell carcinoma. The main causes of gender differences in lung cancer risk were lifestyle factors, baseline comorbidity, and family history. Conclusions: Significant gender differences in the risk of lung cancer were discovered in China non-smokers. Existing risk factors did not explain the excess lung cancer risk of all non-smoking men, and the internal causes for the excess risk still need to be explored;most known risk factors were more harmful to non-smoking women;further exploring the causes of the sex difference would help to improve the prevention and screening programs and protect the non-smoking males from lung cancers. | Zheng Wu Fengwei Tan Zhuoyu Yang Fei Wang Wei Cao Chao Qin Xuesi Dong Yadi Zheng Zilin Luo Liang Zhao Yiwen Yu Yongjie Xu Jiansong Ren Jufang Shi Hongda Chen Jiang Li Wei Tang Sipeng Shen Ning Wu Wanqing Chen Ni Li Jie He | 2022 | Chinese Medical Journal2022,,11: | 3
Fabrication of iridium oxide neural electrodes at the wafer level显示文摘Electro-deposition, electrical activation, thermal oxidation, and reactive ion sputtering are the four primary methods to fabricate iridium oxide film. Among these methods, reactive ion sputtering is a commonly used method in standard micro-fabrication processes. In different sputtering conditions, the component, texture, and electrochemistry character of iridium oxide varies considerably. To fabricate the iridium oxide film compatible with the wafer-level processing of neural electrodes, the quality of iridium oxide film must be able to withstand the mechanical and chemical impact of post-processing, and simultaneously achieve good performance as a neural electrode. In this study, parameters of sputtering were researched and developed to achieve a balance between mechanical stability and good electrochemical characteristics of iridium oxide film on electrode. Iridium oxide fabricating process combined with fabrication flow of silicon electrodes, at wafer-level, is introduced to produce silicon based planar iridium oxide neural electrodes. Compared with bare gold electrodes, iridium oxide electrodes fabricated with this method exhibit particularly good electrochemical stability, low impedance of 386 kW at 1 kH z, high safe charge storage capacity of 3.2 m C/cm^2, and good impedance consistency of less than 25% fluctuation. | ZHANG He PEI WeiHua ZHAO ShanShan YANG XiaoWei LIU RuiCong LIU YuanYuan WU Xian GUO DongMei GUI Qiang GUO XuHong XING Xiao WANG YiJun CHEN HongDa | 2016 | Science China(Technological Sciences)2016,59,9: | 2
Effects of inherent potassium on the catalytic performance of Ni/biochar for steam reforming of toluene as a tar model compound显示文摘Biochar supported nickel(Ni/BC)has been widely studied as a cheap and easy-to-prepare catalyst with potential applications in tar reforming during the gasification of low-rank fuels, such as brown coal and biomass. However, the role and behaviors of inherent K species, especially their interactions with Ni particles and the biochar support, are not well understood yet.In this work, three Ni/BC catalysts with varying K amount were prepared from raw,water-washed,and acid-washed biomass. They were used in steam reforming of toluene as a tar model compound to elucidate the effects of inherent K on the catalytic activity and stability.Detailed characterization indicated that K enhanced water adsorption due to its hydroscopicity and lowered the condensation and graphitization degrees of biochar, but the alteration to the electronic state of Ni was not observed. These effects together led to a temperature-dependent role of K.That is,at relatively low temperatures of 450 and 500℃,toluene conversion was increased in the presence of K,due to the increased concentration of adsorbed water around Ni particles.By contrast,at relatively higher temperatures of 550 and 600℃,although initial high activity was achieved,Ni/BC with K deactivated rapidly because of the accelerated consumption of the biochar support. | Chen Xu Zhenyi Du Shiqi Yang Hongda Ma Jie Feng | 2021 | Chinese Journal of Chemical Engineering2021,34,7: | 2

Dimension-Enhanced Ultra-High Performance Liquid Chromatography/Ion Mobility-Quadrupole Time-of-Flight Mass Spectrometry Combined with Intelligent Peak Annotation for the Rapid Characterization of the Multiple Components from Seeds of Descurainia sophia显示文摘The complex composition of herbal metabolites necessitates the development of powerful analytical techniques aimed to identify the bioactive components. The seeds of Descurainia sophia(SDS)are utilized in China as a cough and asthma relieving agent.Herein,a dimension-enhanced integral approach,by combining ultra-high performance liquid chromatography/ion mobility-quadrupole time-of-flight mass spectrometry(UHPLC/IMQTOF-MS)and intelligent peak annotation, was developed to rapidly characterize the multicomponents from SDS.Good chromatographic separation was achieved within 38 min on a UPLC CSH C18(2.1×100 mm,1.7μm)column which was eluted by 0.1%formic acid in water(water phase)and acetonitrile(organic phase).Collision-induced dissociation-MS^(2)data were acquired by the data-independent high-definition MS^(E)(HDMS^(E))in both the negative and positive electrospray ionization modes.A major components knockout strategy was applied to improve the characterization of those minor ingredients by enhancing the injection volume. Moreover,a self-built chemistry library was established, which could be matched by the UNIFI software enabling automatic peak annotation of the obtained HDMS^(E)data. As a result of applying the intelligent peak annotation workflows and further confirmation process,a total of 53 compounds were identified or tentatively characterized from the SDS,including 29 flavonoids, one uridine derivative, four glucosides, one lignin, one phenolic compound, and 17 others.Notably,four-dimensional information related to the structure(e.g.,retention time, collision cross section,MS^(1)and MS^(2)data)was obtained for each component by the developed integral approach, and the results would greatly benefit the quality control of SDS. | Simiao Wang Xue Li Boxue Chen Shitong Li Jiali Wang Jing Wang Mingshuo Yang Xiaoyan Xu Hongda Wang Wenzhi Yang | 2022 | Phyton-International Journal of Experimental Botany2022,91,3: | 1
Headspace Solid-Phase Micro-Extraction Gas Chromatography/Mass Spectrometry(HS-SPME-GC/MS)-Based Untargeted Metabolomics Analysis for Comparing the Volatile Components from 12 Panax Herbal Medicines显示文摘Quality control of ginseng currently is mainly based on ginsenoside analysis, but rarely focuses on the volatile organic components. In the current work,an untargeted metabolomics approach,by headspace solid-phase micro-extraction gas chromatography/mass spectrometry(HS-SPME-GC/MS),was elaborated and further employed to holistically compare the compositional difference of the volatile components simultaneously from 12 Panax herbal medicines, which included P.ginseng(PG),P.quinquefolius(PQ),P.notoginseng(PN),red ginseng(PGR),P.ginseng leaf(PGL),P.quinquefolius leaf(PQL),P.notoginseng leaf(PNL),P.ginseng flower(PGF),P.quinquefolius flower(PQF),P.notoginseng flower(PNF),P.japonicus(PJ),and P.japonicus var.major(PJvm).Chromatographic separation was performed on an HP-5MS elastic quartz capillary column using helium as the carrier gas, enabling good resolution within 1 h.We were able to characterize totally 259 volatile compounds, including 82 terpenes(T),46 alcohols(Alc),29 ketones(K),25 aldehydes(Ald),21 esters(E),and the others.By analyzing 90 batches of ginseng samples based on the untargeted metabolomics workflows,236 differential ions were unveiled, and accordingly 36 differential volatile components were discovered. It is the first report that simultaneously compares the compositional difference of volatile components among 12 Panax herbal medicines, and useful information is provided for the quality control of ginseng aside from the well-known ginsenosides. | Simiao Wang Xiaohang Li Meiting Jiang Xinlong Wu Yuying Zhao Meiyu Liu Xiaoyan Xu Huimin Wang Hongda Wang Heshui Yu Wenzhi Yang | 2022 | Phyton-International Journal of Experimental Botany2022,91,7: | 1
Interconnected graphene scaffolds for functional gas sensors with tunable sensitivity显示文摘In this work, the interconnected graphene scaffolds are prepared by three-dimensional(3 D)printing for multifunctional gas detection with tunable sensitivity. The scaffolds with regularly aligned graphene conductive networks exhibit significant mechanical strength and high electrical stability to multi-direction deformation, which can be attributed to the typical core-shell structure of graphene and PVP.The resistance of the free-standing scaffolds can realize the real-time response to H_(2) O and NO_(2),and the relative resistance change to 100 ppm H_(2) O and 100 ppm NO_(2) can reach 2%and 2.5%,respectively.The charge doping of the oxidizing gases is considered to be the main reason for various response sensitivities of the scaffolds with different orthogonal layers,in which the interconnected conductive network can generate a large specific surface area and significantly improving the adsorption of the target gases and the transfer of charge. The controllable fabrication of regular structure has appropriately great potential for further optimizations and applications in gas detection. | Xiao You Jinshan Yang Mengmeng Wang Hongda Wang Le Gao Shaoming Dong | 2020 | Journal of Materials Science & Technology2020,58,23: | 1
Label-Free Virtual HER2 Immunohistochemical Staining of Breast Tissue using Deep Learning显示文摘The immunohistochemical(IHC)staining of the human epidermal growth factor receptor 2(HER2)biomarker is widely practiced in breast tissue analysis, preclinical studies, and diagnostic decisions, guiding cancer treatment and investigation of pathogenesis.HER2 staining demands laborious tissue treatment and chemical processing performed by a histotechnologist, which typically takes one day to prepare in a laboratory, increasing analysis time and associated costs. Here,we describe a deep learning-based virtual HER2 IHC staining method using a conditional generative adversarial network that is trained to rapidly transform autofluorescence microscopic images of unlabeled/label-free breast tissue sections into bright-field equivalent microscopic images, matching the standard HER2 IHC staining that is chemically performed on the same tissue sections. The efficacy of this virtual HER2 staining framework was demonstrated by quantitative analysis,in which three board-certified breast pathologists blindly graded the HER2 scores of virtually stained and immunohistochemically stained HER2 whole slide images(WSIs)to reveal that the HER2 scores determined by inspecting virtual IHC images are as accurate as their immunohistochemically stained counterparts.A second quantitative blinded study performed by the same diagnosticians further revealed that the virtually stained HER2 images exhibit a comparable staining quality in the level of nuclear detail, membrane clearness, and absence of staining artifacts with respect to their immunohistochemically stained counterparts. This virtual HER2 staining framework bypasses the costly, laborious, and time-consuming IHC staining procedures in laboratory and can be extended to other types of biomarkers to accelerate the IHC tissue staining used in life sciences and biomedical workflow. | Bijie Bai Hongda Wang Yuzhu Li Kevin de Haan Francesco Colonnese Yujie Wan Jingyi Zuo Ngan B.Doan Xiaoran Zhang Yijie Zhang Jingxi Li Xilin Yang Wenjie Dong Morgan Angus Darrow Elham Kamangar Han Sung Lee Yair Rivenson Aydogan Ozcan | 2022 | Biomedical Engineering Frontiers2022,3,1: | 0

Tumor-targeted self-assembled micelles reducing PD-L1 expression combined with ICIs to enhance chemo-immunotherapy of TNBC显示文摘Immune checkpoint inhibitors(ICIs)therapy targeting programmed cell death ligand 1(PD-L1)and programmed death protein 1(PD-1)had exhibited significant clinical benefits for cancer treatment such as triple negative breast cancer(TNBC).However, the relatively low anti-tumor immune response rate and ICIs drug resistance highlight the necessity of developing ICIs combination therapy strategies to improve the anti-tumor effect of immunotherapy.Herein, the immunomodulator epigallocatechin gallate palmitate(PEGCG)and the immunoadjuvant metformin(MET)self-assembled into tumor-targeted micelles via hydrogen bond and electrostatic interaction, which encapsulated the therapeutic agents doxorubicin(DOX)-loaded PEGCG-MET micelles(PMD)and combined with ICIs(anti-PD-1 antibody)as therapeutic strategy to reduce the endogenous expression of PD-L1 and improve the tumor immunosuppressive microenvironment. The results presented that PMD integrated chemotherapy and immunotherapy to enhance antitumor efficacy in vitro and in vivo, compared with DOX or anti-PD-1 antibody for the therapy of TNBC.PMD micelles might be a potential candidate, which could remedy the shortcomings of antibody-based ICIs and provide synergistic effect to enhance the antitumor effects of ICIs in tumor therapy. | Hongda Zhu Kai Ma Rui Ruan Chaobo Yang Aqin Yan Jing Li Qi Yu Hongmei Sun Mingxing Liu Hongmei Zheng Jing Gao Xiaofang Guan Zhu Dai Yao Sun | 2024 | Chinese Chemical Letters2024,35,2: | 0
Revealing the Cell Entry Dynamic Mechanism of Single Rabies Virus Particle显示文摘The rabies virus is a neurotropic virus that causes fatal diseases in humans and animals.Although studying the interactions between a single rabies virus and the cell membrane is necessary for understanding the pathogenesis, the internalization dynamic mechanism of single rabies virus in living cells remains largely elusive. Here,we utilized a novel force tracing technique based on atomic force microscopy(AFM)to record the process of single viral entry into host cell.We revealed that the force of the rabies virus internalization distributed at(65±25)pN,and the time was identified by two peaks with spacings of(237.2±59.1)and(790.3±134.4)ms with the corresponding speed of 0.12 and 0.04µm/s,respectively.Our results provide insight into the effects of viral shape during the endocytosis process. This report will be meaningful for understanding the dynamic mechanism of rabies virus early infection. | LI Siying PAN Yangang TENG Honggang SHAN Yuping YANG Guocheng WANG Hongda | 2022 | Chemical Research in Chinese Universities2022,38,3: | 0
Modelling and Simulation on the Effect of Hot Forming Damage on Three-Point Bending Performance of Beam Components显示文摘The effects of forming damage are analyzed, which occur during hot stamping process,on the load-carrying capacity and failure mode of hot stamped beams.A damage-coupled pre-forming constitutive model was proposed,in which the damage during hot stamping process was introduced into the service response. The constitutive model was applied into the three-point bending simulation of a hot stamped beam, and then the influences of forming damage on the load-carrying capacity and cracks propagation were investigated. The results show that the forming damage reduces the maximum load capacity of the hot stamped beam by 7.5%.It also causes the crack to occur earlier and promotes crack to propagate along the radial direction of the punch. | Weimin Zhuang Pengyue Wang Yang Liu Dongxuan Xie Hongda Shi | 2020 | Journal of Beijing Institute of Technology2020,29,3: | 0
Adsorptive behaviors of humic acid onto freshly prepared hydrous manganese dioxides显示文摘This study focused on the adsorptive behaviors of humic acid onto freshly prepared hydrous MnO_(2)(s)(δMnO_(2)),and investigated the feasibility of employingδMnO2 for humic acid removal from drinking water.Effects of such parameters as molecular mass of humic acid, kinds of diva-lent cations on adsorptive behaviors and possible mechanisms involved were investigated. This study indicated that humic acid with higher molecular mass exhibited more tendency of adsorbing ontoδMnO2 than that with lower molecular mass.Ca^(2+)facilitated more humic acid adsorption than Mg^(2+);UV-Vis spectra analysis indicated higher capabilities of Ca^(2+)coordinating with acidic functional groups of humic acid than that of Mg2+.Additionally,ζpotential characterization indicated that Ca^(2+)showed higher potential of increasingζpotential ofδMnO_(2) than Mg^(2+).Ca^(2+)of 1.0 mmol/L increasedζpotential ofδMnO2 from−37 mV(pH 7.9)to+7 mV(pH 7.2),while 1.0 mmol/L Mg^(2+)^increased to lower value as−9 mV(pH 6.5),correspondingly.Fourier transform infrared(FTIR)spectra demonstrated the adsorption of humic acid ontoδMnO_(2),showing the important roles of-COO−function-al groups and surface Mn-OH in the adsorption of humic acid ontoδMnO_(2). | LIU Ruiping YANG Yanling LI Guibai HE Wenjie HAN Hongda | 2007 | Frontiers of Environmental Science & Engineering2007,1,2: | 0
