1Cadmium accumulation and tolerance of two castor cultivars in relation to antioxidant systems显示文摘To investigate the effects of Cd on tolerance and antioxidant activities of castor, two different castor(Ricinus communis L.) cultivars(Zibo No. 5 and Zibo No. 8) were used for a hydroponic experiment(0, 1 and 2 mg/L Cd) and a pot experiment using Cd-contaminated soil(34 mg/kg) with the addition of ethylenedinitrilotetraacetic acid(EDTA). The results indicated that there were significant differences between the two cultivars with respect to Cd uptake in shoots(113–248 mg/kg for Zibo No. 5 and 130–288 mg/kg Zibo No. 8), biomass tolerance indexes(64.9%–74.6% for Zibo No. 5 and 80.1%–90.9% for Zibo No. 8) in the hydroponic experiment and survival rates(0% for Zibo No. 5 and 100% for Zibo No. 8)determined by the addition of EDTA in the pot experiment, suggesting that Zibo No. 8 has higher tolerance than Zibo No. 5. Moreover, the castor cultivars have low bioconcentration factors(4.80% for Zibo No. 5 and 5.43% for Zibo No. 8) and low translocation factors(<1%).Consequently, Zibo No. 8 can participate in Cd phytostabilization in highly Cd-polluted areas. The results indicated that glutathione(GSH) as a non-enzymatic antioxidant, and antioxidant enzymes including superoxide dismutase(SOD), catalase(CAT) and guaiacol peroxidase(GPX), were cultivar- and dose-dependent. 2Tracing the first hematopoietic stem cell generation in human embryo by single-cell RNA sequencing显示文摘Tracing the emergence of the first hematopoietic stem cells(HSCs)in human embryos,particularly the scarce and transient precursors thereof,is so far challenging,largely due to the technical limitations and the material rarity.Here,using single-cell RNA sequencing,we constructed the first genome-scale gene expression landscape covering the entire course of endothelial-to-HSC transition during human embryogenesis.The transcriptomically defined HSC-primed hemogenic endothelial cells(HECs)were captured at Carnegie stage(CS)12-14 in an unbiased way,showing an unambiguous feature of arterial endothelial cells(ECs)with the up-regulation of RUNX1,MYB and ANGPTL Importantly,subcategorizing CD34+CD45-ECs into a CD44+population strikingly enriched HECs by over 10-fold.We further mapped the developmental path from arterial ECs via HSC-primed HECs to hematopoietic stem progenitor cells,and revealed a distinct expression pattern of genes that were transiently over-represented upon the hemogenic fate choice of arterial ECs#including EMCN,PROCR and RUNX1T1.We also uncovered another temporally and molecularly distinct intra-embryonic HEC population,which was detected mainly at earlier CS 10 and lacked the arterial feature.Finally,we revealed the cellular components of the putative aortic niche and potential cellular interactions acting on the HSCprimed HECs.The cellular and molecular programs that underlie the generation of the first HSCs from HECs in human embryos,together with the ability to distinguish the HSC-primed HECs from others,will shed light on the strategies for the production of clinically useful HSCs from pluripotent stem cells.Yang Zeng Jian He Zhijie Bai Zongcheng Li Yandong Gong Chen Liu Yanli Ni Junjie Du Chunyu Ma Lihong Bian Yu Lan Bing Liu 2019Cell Research2019,29,11:14

4A point mutation resulting in a 13 bp deletion in the coding sequence of Cldf leads to a GA-deficient dwarf phenotype in watermelon显示文摘The dwarf architecture is an important and valuable agronomic trait in watermelon breeding and has the potential to increase fruit yield and reduce labor cost in crop cultivation.However,the molecular basis for dwarfism in watermelon remains largely unknown.In this study,a recessive dwarf allele(designated as Cldf(Citrullus lanatus dwarfism))was fine mapped in a 32.88 kb region on chromosome 09 using F2 segregation populations derived from reciprocal crossing of a normal line M08 and a dwarf line N21.Gene annotation of the corresponding region revealed that the Cla015407 gene encoding a gibberellin 3β-hydroxylase functions as the best possible candidate gene for Cldf.Sequence analysis showed that the fourth polymorphism site(a G to A point mutation)at the 3′AG splice receptor site of the intron leads to a 13 bp deletion in the coding sequence of Cldf in dwarf line N21 and thus results in a truncated protein lacking the conserved domain for binding 2-oxoglutarate.In addition,the dwarf phenotype of Cldf could be rescued by exogenous GA3 application.Phylogenetic analysis suggested that the small multigene family GA3ox(GA3 oxidase)in cucurbit species may originate from three ancient lineages in Cucurbitaceae.All these data support the conclusion that Cldf is a GA-deficient mutant,which together with the cosegregated marker can be used for breeding new dwarf cultivars.Chunhua Wei Chunyu Zhu Liping Yang Wei Zhao Rongxue Ma Hao Li Yong Zhang Jianxiang Ma Jianqiang Yang Xian Zhang 2019Horticulture Research2019,6,1:12

6Effect of N fertilization rate on soil alkalihydrolyzable N, subtending leaf N concentration,fiber yield, and quality of cotton显示文摘Soil alkali-hydrolyzable nitrogen, which is sensitive to N fertilization rate, is one of the indicators of soil nitrogen supplying capacity. Two field experiments were conducted in Dongtai(120°19″ E, 32°52″ N), Jiangsu, China in 2009 and Dafeng(120°28″ E, 33°12″ N), Jiangsu province, China in 2010. Six nitrogen rates(0, 150, 300, 375, 450, and 600 kg ha^(-1)) were used to study the effect of N fertilization rate on soil alkali-hydrolyzable nitrogen content(SAHNC), subtending leaf nitrogen concentration(SLNC), yield, and fiber quality. In both Dongtai and Dafeng experiment station, the highest yield(1709 kg ha^(-1)), best quality(fiber length 30.6 mm, fiber strength 31.6 c N tex^(-1), micronaire 4.82), and highest N agronomic efficiency(2.03 kg kg^(-1)) were achieved at the nitrogen fertilization rate of 375 kg ha^(-1). The dynamics of SAHNC and SLNC could be simulated with a cubic and an exponential function,respectively. The changes in SAHNC were consistent with the changes in SLNC. Optimal average rate(0.276 mg day^(-1)) and duration(51.8 days) of SAHNC rapid decline were similar to the values obtained at the nitrogen rate of 375 kg ha^(-1)at which cotton showed highest fiber yield, quality, and N agronomic efficiency. Thus, the levels and strategies of nitrogen fertilization can affect SAHNC dynamics. 8Effect of organic amine alkali and inorganic alkali on benzotriazole removal during post Cu-CMP cleaning显示文摘Benzotriazole(BTA), an anticorrosion agent of slurry, is the main organic pollutant remaining after CMP of multilayer copper wiring, and also the main removal object of post CMP cleaning. The adsorption of BTA onto the copper could form a dense Cu-BTA film, which makes the copper surface strongly passivated. According to this characteristic, quantitative analysis of BTA residue after cleaning is carried out by contact angle measurement and electrochemical measurement in this paper. A scanning electron microscope(SEM) with EDX was used to observe and analyze the BTA shape and elements. The efficiencies of organic alkali and inorganic alkali on the removal of BTA were studied. The corresponding reaction mechanism was also analyzed. 10Preparation and characterization of boron-doped corn straw biochar: Fe(Ⅱ) removal equilibrium and kinetics显示文摘Nowadays,iron ions as a ubiquitous heavy metal pollutant are gradually concerned and the convenient and quick removal of excessive iron ions in groundwater has become a major challenge for the safety of drinking water.In this study,boron-doped biochar(B-BC)was successfully prepared at various preparation conditions with the addition of boric acid.The as-prepared material has a more developed pore structure and a larger specific surface area(up to 897.97 m2/g).A series of characterization results shows that boric acid effectively activates biochar,and boron atoms are successfully doped on biochar.Compared with the ratio of raw materials,the pyrolysis temperature has a greater influence on the amount of boron doping.Based on Langmuir model,the maximum adsorption capacity of 800 B-BC1:2 at25℃,40℃,55℃ are 50.02 mg/g,95.09 mg/g,132.78 mg/g,respectively.Pseudo-second-order kinetic model can better describe the adsorption process,the adsorption process is mainly chemical adsorption.Chemical complexation,ions exchange,and co-precipitation may be the main mechanisms for Fe2+removal.Long Sui Chunyu Tang Qing Du Ying Zhao Kui Cheng Fan Yang 2021Journal of Environmental Sciences2021,33,8:6

12Dissecting human embryonic skeletal stem cell ontogeny by single-cell transcriptomic and functional analyses显示文摘Human skeletal stem cells(SSCs)have been discovered in fetal and adult long bones.However,the spatiotemporal ontogeny of human embryonic SSCs during early skeletogenesis remains elusive.Here we map the transcriptional landscape of human limb buds and embryonic long bones at single-cell resolution to address this fundamental question.We found remarkable heterogeneity within human limb bud mesenchyme and epithelium,and aligned them along the proximal-distal and anterior-posterior axes using known marker genes.Osteo-chondrogenic progenitors first appeared in the core limb bud mesenchyme,which give rise to multiple populations of stem/progenitor cells in embryonic long bones undergoing endochondral ossification.Importantly,a perichondrial embryonic skeletal stem/progenitor cell(eSSPC)subset was identified,which could self-renew and generate the osteochondral lineage cells,but not adipocytes or hematopoietic stroma.eSSPCs are marked by the adhesion molecule CADM1 and highly enriched with FOXP1/2 transcriptional network.Interestingly,neural crest-derived cells with similar phenotypic markers and transcriptional networks were also found in the sagittal suture of human embryonic calvaria.Taken together,this study revealed the cellular heterogeneity and lineage hierarchy during human embryonic skeletogenesis,and identified distinct skeletal stem/progenitor cells that orchestrate endochondral and intramembranous ossification.Jian He Jing Yan Jianfang Wang Liangyu Zhao Qian Xin Yang Zeng Yuxi Sun Han Zhang Zhijie Bai Zongcheng Li Yanli Ni Yandong Gong Yunqiao Li Han He Zhilei Bian Yu Lan Chunyu Ma Lihong Bian Heng Zhu Bing Liu Rui Yue 2021Cell Research2021,31,7:5

14ENPP1/PC-1 Gene K121Q Polymorphism Is Associated with Obesity in European Adult Populations: Evidence from A Meta-Analysis Involving 24 324 Subjects显示文摘Objective Findings from the previous studies have suggested a relationship between ectonucleotide pyrophosphatase /phosphodiesterase 1 (ENPP‐1) or plasma cell membrane glycoprotein 1 (PC‐1) gene single nucleotide polymorphism (K121Q, rs1044498) and genetic susceptibility to obesity. However, such relationship is not reproduced by some currently available studies. In this context, the present study is aimed to quantitatively analyze the association of K121Q variant with obesity in all published case‐control studies in European adult populations. Methods Published literature from PubMed, EMBASE, and ISI web of science databases were retrieved. The studies evaluating the association of ENPP1/PC1 gene K121Q polymorphism with obesity were included, in which sufficient data were presented to calculate the odds ratio (OR) with 95% confidence intervals (CIs). Results Ten case‐control studies meeting the inclusion criteria identified a total of 24,324 subjects including 11,372 obese and 12,952 control subjects. The meta‐analysis results showed a statistically significant association of K121Q with obesity [OR (95%CI): 1.25 (1.04‐1.52) P=0.021] under a recessive model of inheritance (QQ vs. KK+KQ) without heterogeneity or publication bias. 16Discovering small molecules as Wnt inhibitors that promote heart regeneration and injury repair显示文摘There are intense interests in discovering pro regenerative medicine leads that can promote cardiac differentiation and regeneration,as well as repair damaged heart tissues.We have combined zebrafish embryo-based screens with cardiomyogenesis assays to discover selective small molecules that modulate heart development and regeneration with minimal adverse effects.Two related compounds with novel structures,named as Cardiomogen 1 and 2(CDMG1 and CDMG2),were identified for their capacity to promote myocardial hyperplasia through expansion of the cardiac progenitor cell population.We find that Cardiomogen acts as a Wnt inhibitor by targeting p-catenin and reducing Tcf/Lef-mediated transcription in cultured cells.CDMG treatment of amputated zebrafish hearts reduces nuclear p-catenin in injured heart tissue,increases cardiomyocyte(CM)proliferation,and expedites wound healing,thus accelerating cardiac muscle regeneration.Importantly,Cardiomogen can alleviate the functional deterioration of mammalian hearts after myocardial infarction.Injured hearts exposed to CDMG1 display increased newly formed CMs and reduced fibrotic scar tissue,which are in part attributable to the^-catenin reduction.Our findings indicate Cardiomogen as a Wnt inhibitor in enhancing injury-induced CM proliferation and heart regeneration,highlighting the values of embryo-based small molecule screens in discovery of effective and safe medicine leads.Shuying Xie Wenbin Fu Guangju Yu Xueli Hu Kaa Seng Lai Xiangwen Peng Yating Zhou Xuejiao Zhu Ptamen Christov Leah Sawyer Terri T.Ni Gary A.Sulikowski Zhongzhou Yang Ethan Lee Chunyu Zeng Wei E.Wang Tao P.Zhong 2020Journal of Molecular Cell Biology2020,12,1:4
17Study on repairing canine mandibular defect with porous Mg-Sr alloy combined with Mg-Sr alloy membrane显示文摘To discuss the feasibility of the application of porous Mg-Sr alloy combined with Mg-Sr alloy membrane in the repair of mandibular defects in dogs.The second and third mandibular premolars on both sides were extracted from six dogs.The model of mandible buccal fenestration bone defects were prepared after the sockets healed.Twelve bone defects were randomly divided into groups A and B,then Mg-Sr alloy was implanted in bone defects of group A and covered by Mg-Sr alloy membrane while Mg-Sr alloy was implanted in bone defects of group B and covered by mineralized collagen membrane.Bone defects observed on cone beam computed tomographic images and comparing the gray value of the two groups after 4,8 and 12 weeks.After 12 weeks,the healing of bone defects were evaluated by gross observation,X-ray microscopes and histological observation of hard tissue.Bone defects in each group were repaired.At 8 and 12 weeks,the gray value of group A was higher than that of group B(P<0.05).At 12 weeks,the bone volume fraction of group A was higher than that of group B(P<0.05).The newly woven bone in group A is thick and arranged staggered,which was better than that of group B.Porous Mg-Sr alloy combined with Mg-Sr alloy membrane could further promote the repair of mandibular defects,and obtain good osteogenic effect.Shanning Zhang Xirao Sun Chunyu Kang Man Yang Yuan Zhao Chengyue Wang 2020Regenerative Biomaterials2020,7,3:3

20The effect of rural labor migration on poverty alleviation:a village-level study in China显示文摘The construction of the new socialist countryside is being carried out in China nowadays. Although lots of successful experience has been gotten, different problems occur in various regions yet. To analyze the relationships among rural labor migration, poverty alleviation and characteristics of migrants is important in considering the effect of rural labor migration on the construction of the new socialist countryside. Available sampling and typical case study are adopted and 236 questionnaires are collected from four villages in northwest Guangxi, China, Daxin, Lixin, Longhe and Yongchang. We analyze the rural labor migration status, characteristics of migrants, remittance situation and income, household income and the ratio of remittance income to total household income. About 2/3 of the households have migrants in surveyed villages. And nearly half of the migration households have only one fam-ily member as migrants in the four villages. The migrants mainly comprise male and the younger, with the education level of junior middle school and higher. More than half of the migrants are employed in Guangdong Province. But the characteristics of migrants in Yongchang are more diverse. The rural labor migration in the village is extremely active and extensive. And over 60% of the household with family members as migrants have remittance income. The rate in Yongchang is extremely high (80%). And the income of households with migrants getting remittance income is signif icantly higher than their counterparts. More than half of their income comes from remittance as far as the former kind of household is concerned. And in Yongchang, the rate is nearly 80%. The conclusion is that rural labor migration is popular and extensive in lots of villages. And the migration and remittance play an important role in rural household income, especially in some poor villages. For this kind of village, the rural labor migration may be some 'compulsory course' in rural development. And the rural labor emigration is an essential way in poverty al-leviation.
