| 1 | Volatile Organic Compounds and Their Roles in Bacteriostasis in Five Conifer Species显示文摘In order to make clear the functions of plant volatile organic compounds (VOCs) on bacteriostasis and air decontamination, we analyzed the composition and content of VOCs in Pinus tabulaeformis Carr., P. bungeana Zucc., Sabina chinensis Antoine, Picea koraiensis Nakai, and Cedrus deodara G. Don under near-natural conditions using the thermal-desorption cold trap gas chromatography/mass spectrometer technique. The effects of the VOCs on airborne microorganisms were investigated using the method of natural sedimentation. Results showed that the major VOCs were as follows: limonene, β-pinene, α-pinene,and α-caryophyllene in Pinus tabulaeformis and P. bungeana; limonene, borneol acetate, β-pinene, myrcene,and tricylene in S. chinensis; limonene, α-pinene, myrcene, camphene, and β-pinene in Picea koraiensis;and limonene, 2, (10)-pinene, α-pinene, and myrcene in C. deodara. These VOCs and the corresponding foliar extracts inhibited the growth of bacteria and stimulated the growth of fungi. Experimental data using monomers of the VOCs demonstrated that limonene, β-pinene, and three aldehydes could significantly inhibit bacterial growth, suggesting an inhibitory effect of VOCs on the growth of airborne microorganisms in the five conifer species. | 2 | Association of interleukin-12 p40 gene 3'-untranslated region polymorphism and outcome of HCV infection显示文摘AIM: To investigate the effect of interleukin-12 p40 gene (IL12E 3'-untranslated region polymorphism on the outcome of HCV infection.METHODS: A total of 133 patients who had been infected with HCV for 12-25 (18.2±3.8) years, were enrolled in this study. Liver biochemical tests were performed with an automated analyzer and HCV RNA was detected by fluorogenic quantitative polymerase chain reaction. B-mode ultrasound was used for liver examination. Polymerase chain reactionrestriction fragment length polymorphism (PCR-RFLP) was used for the detection of IL12B (1188A/C) polymorphism.RESULTS: Self-limited infection was associated with AC genotype (OR = 3.48; P = 0.001) and persistent infection was associated with AA genotype (OR = 0.34; P = 0.014)at site 1188 of IL12B. In patients with persistent HCV infection, no significant differences were found regarding the age, gender, duration of infection and biochemical characteristics (P>0.05). According to B-mode ultrasound imaging and clinical diagnosis, patients with persistent infection were divided into groups based on the severity of infection. | 3 | Cytoreductive surgery and hyperthermic intraperitoneal chemotherapy in treatment of gastric cancer with peritoneal carcinomatosis显示文摘Although gastric cancer with peritoneal carcinomatosis is associated with poor prognosis and is generally treated with palliative systemic therapy, recent studies have shown that cytoreductive surgery(CRS) and hyperthermic intraperitoneal chemotherapy(HIPEC) may prove to be an efficacious treatment option. In addition to reviewing the natural history of gastric cancer with peritoneal carcinomatosis, this mini-review examines literature on the efficacy of CRS and HIPEC as compared to chemotherapy and surgical options. Both randomized and nonrandomized studies were summarized with the emphasis focused on overall survival. | 4 | Somatic mutation analysis of p53 and ST7 tumor suppressor genes in gastric carcinoma by DHPLC显示文摘AIM: To verify the effectiveness of denaturing highperformance liquid chromatography (DHPLC) in detecting somatic mutation of p53 gene in gastric carcinoma tissues.The superiority of this method has been proved in the detection of germline mutations, but it was not very affirmative with respect to somatic mutations in tumor specimens. ST7 gene, a candidate tumor suppressor gene identified recently at human chromosome 7q31.1, was also detected because LOH at this site has also been widely reported in stomach cancer.METHODS: DNA was extracted from 39 cases of surgical gastric carcinoma specimen and their correspondent normal mucosa. Seven fragments spanning the 11 exons were used to detect the mutation of p53 gene and the four exons reported to have mutations in ST7 gene were amplified by PCR and directly analyzed by DHPLC without mixing with wild-type allele.RESULTS: In the analysis of p53 gene mutation, 9 aberrant DHPLC chromatographies were found in tumor tissues, while their normal-adjacent counterparts running in parallel showed a normal shape. Subsequent sequencing revealed nine sequence variations, 1 polymorphism and 8 mutations including 3 mutations not reported before. The mutation rate of p53 gene (21%) was consistent with that previously reported. Furthermore, no additional aberrant chromatography was found when wild-type DNA was added into the DNA of other 30 tumor samples that showed normal shapes previously. The positivity of p53 mutations was significantly higher in intestinal-type carcinomas (40 %) than that in diffuse-type (8.33 %) carcinomas of the stomach. No mutation of ST7 gene was found.CONCLUSION: DHPLC is a very convenient method for the detection of somatic mutations in gastric carcinoma. The amount of wild type alleles supplied by the non-tumorous cells in gastric tumor specimens is enough to form heteroduplex with mutant alleles for DHPLC detection.
