1Yeast knockout library allows for efficient testing of genomic mutations for cell-free protein synthesis显示文摘Cell-free protein synthesis(CFPS)systems from crude lysates have benefitted from modifications to their enzyme composition.For example,functionally deleting enzymes in the source strain that are deleterious to CFPS can improve protein synthesis yields.However,making such modifications can take substantial time.As a proof-of-concept to accelerate prototyping capabilities,we assessed the feasibility of using the yeast knockout collection to identify negative effectors in a Saccharomyces cerevisiae CFPS platform.We analyzed extracts made from six deletion strains that targeted the single deletion of potentially negative effectors(e.g.,nucleases).We found a statistically significant increase in luciferase yields upon loss of function of GCN3,PEP4,PPT1,NGL3,and XRN1 with a maximum increase of over 6-fold as compared to the wild type.Our work has implications for yeast CFPS and for rapidly prototyping strains to enable cell-free synthetic biology applications.Jennifer A.Schoborg Lauren G.Clark Alaksh Choudhury C.Eric Hodgman Michael C.Jewett 2016Synthetic and Systems Biotechnology2016,1,1:2
2简并PCR扩增小瓜虫抑动抗原基因的ORF显示文摘抑动蛋白是小瓜虫(Ichthyophthiriusmultifiliis)体表纤毛的主要构成部分,也是宿主免疫系统发挥抗虫免疫时的主要识别抗原。根据已经报道的3种抑动蛋白N端及C端保守的多肽片断,设计了一对简并引物P6/P7,以小瓜虫分离株IchFJ9的基因组为模板,成功扩增了抑动蛋白编码区(ORF)的全长基因序列。扩增的iagFJ9基因全长1398bp,编码466个氨基酸,包含18个非标准密码子TAA(Glu,Q)推导的氨基酸序列包含6个以CPXGT为起始的串联重复单位,具有抑动蛋白的特征性结构。同源比较分析显示,扩增的iagFJ9基因与已知的小瓜虫抑动蛋白基因IAG52A基因具有88%同源率。证实了iagFJ9是一个新发现的抑动蛋白基因,用简并PCR技术去发现小瓜虫抑动蛋白基因家族新基因是可行的。杨金先 陈强 刘晓东 林天龙 Theodore G.Clark 2004中国水产科学2004,11,2:2

5Reconstruction of the anterior skull base with radial forearm free tissue transfer:case series and literature review显示文摘Aim:Reconstruction of the anterior skull base offers an especially complex challenge as the impermeable separation of the dural space and the upper aerodigestive tract must be maintained.We propose the use of the radial forearm free flaps(RFFF)as a superb method of re-establishing integrity in anterior skull base defects.Methods:Literature review and retrospective analysis of 4 single-institution cases of anterior skull base defects reconstructed with a RFFF.Data were collected on successful and unsuccessful defect repairs,complication rates,and length of hospitalization.Results:The indications for surgery were pneumocephalus,recurrent brain abscesses,recurrent frontal sinus mucoceles,and cerebrospinal fluid leak.Of the 4 cases,1 was complicated by a small dehiscence of the craniotomy site,1 developed infection,and 2 required further surgery.Conclusion:The use of RFFF is an excellent option for reconstruction of defects in the anterior skull base,especially those complicated by radiation,prior surgery,or infections.Patients with skull base defects are inherently at high risk for post-surgical complications.The RFFF transfers healthy,viable,well-vascularized tissue to prevent further infections and provides a reliable barrier between the dural and sinonasal spaces.This can reduce the need for repeat neurosurgical operations and hospitalizations.Windy A.Olaya Lauren T.Daly Emily G.Clark Thomas Scholz Vincent Laurence Keyianoosh Z.Paydar Garrett A.Wirth Gregory R.D.Evans 2016Plastic and Aesthetic Research2016,3,1:0

7Grasses through space and time: An overview of the biogeographical and macroevolutionary history of Poaceae显示文摘Grasses are widespread on every continent and are found in all terrestrial biomes.The dominance and spread of grasses and grassland ecosystems have led to significant changes in Earth′s climate,geochemistry,and biodiversity.The abundance of DNA sequence data,particularly chloroplast sequences,and advances in placing grass fossils within the family allows for a reappraisal of the family′s origins,timing,and geographic spread and the factors that have promoted diversification.We reconstructed a time-calibrated grass phylogeny and inferred ancestral areas using chloroplast DNA sequences from nearly 90%of extant grass genera.With a few notable exceptions,the phylogeny is well resolved to the subtribal level.The family began to diversify in the Early–Late Cretaceous(crown age of 98.54 Ma)on West Gondwana before the complete split between Africa and South America.Vicariance from the splitting of Gondwana may be responsible for the initial divergence in the family.However,Africa clearly served as the center of origin for much of the early diversification of the family.With this phylogenetic,temporal,and spatial framework,we review the evolution and biogeography of the family with the aim to facilitate the testing of biogeographical hypotheses about its origins,evolutionary tempo,and diversification.The current classification of the family is discussed with an extensive review of the extant diversity and distribution of species,molecular and morphological evidence supporting the current classification scheme,and the evidence informing our understanding of the biogeographical history of the family.Timothy J.Gallaher Paul M.Peterson Robert J.Soreng Fernando O.Zuloaga De-Zhu Li Lynn G.Clark Christopher D.Tyrrell Cassiano A.D.Welker Elizabeth A.Kellogg Jordan K.Teisher 2022Journal of Systematics and Evolution2022,60,3:0

11西藏特有竹子Arundinaria macclureana的名实辨正(英文)显示文摘Arundinaria macclureana是在现代植物学中根据在西藏采集的标本描述的第一个竹子。其模式标本是1938年采集的,但只有花枝,没有地下茎、秆箨等重要特征。尽管如此,该种还成为了新属Borinda的模式种。在研究了其模式标本后,使本文第一作者联想起1991年在藏东南采集的西藏箭竹(Fargesia setosa)。西藏箭竹的模式标本采集于1977年,但只有营养体,没有花枝。在对比研究两个名称的模式后,确认二者实为同种。经与共同作者一起讨论,认为其正确学名为Fargesia macclureana,但该种所在的筱竹属群的关系还有待进一步的研究。李德铢 Lynn G.CLARK Chris STAPLETON 2006云南植物研究2006,28,2:0
