1Endothelial cell-specific anticoagulation reduces inflammation in a mouse model of acute lung injury显示文摘Tissue factor (TF)-dependent coagulation contributes to lung inflammation and the pathogenesis of acute lung injury (ALI). In this study, we explored the roles of targeted endothelial anticoagulation in ALI using two strains of transgenic mice expressing either a membrane-tethered human tissue factor pathway inhibitor (hTFPI) or hirudin fusion protein on CD31^+ cells, including vascular endothelial cells (ECs). ALI was induced by intratracheal injection of LPS, and after 24 h the expression of TF and protease-activated receptors (PARs) on EC in lungs were assessed, alongside the extent of inflammation and injury. The expression of TF and PARs on the EC in lungs was upregulated after ALI. In the two strains of transgenic mice, expression of either of hTFPI or hirudin by EC was associated with significant reduction of inflammation, as assessed by the extent of leukocyte infiltration or the levels of proinflammatory cytokines, and promoted survival after LPS-induced ALI. The beneficial outcomes were associated with inhibition of the expression of chemokine CCL2 in lung tissues. The protection observed in the CD31-TFPI-transgenic strain was abolished by injection of an anti-hTFPI antibody, but not by prior engraftment of the transgenic strains with WT bone marrow, confirming that the changes observed were a specific transgenic expression of anticoagulants by EC. These results demonstrate that the inflammation in ALI is TF and thrombin dependent, and that expression of anticoagulants by EC significantly inhibits the development of ALI via repression of leukocyte infiltration, most likely via inhibition of chemokine gradients. 2Microwave ablation of primary breast cancer inhibits metastatic progression in model mice via activation of natural killer cells显示文摘Surgery is essential for controlling the symptoms and complications of stage IV breast cancer.However,locoregional treatment of primary tumors often results in distant progression,including lung metastasis,the most common type of visceral metastasis.As a minimally invasive thermal therapy,microwave ablation(MWA)has been attempted in the treatment of breast cancer,but the innate immune response after MWA has not yet been reported.Using two murine models of stage IV breast cancer,we found that MWA of primary breast cancer inhibited the progression of lung metastasis and improved survival.NK cells were activated after MWA of the primary tumor and exhibited enhanced cytotoxic functions,and the cytotoxic pathways of NK cells were activated.Depletion experiments showed that NK cells but not CD4+or CD8+T cells played a pivotal role in prolonging survival.Then,we found that compared with surgery or control treatment,MWA of the primary tumor induced completely different NK-cell-related cytokine profiles.Macrophages were activated after MWA of the primary tumor and produced IL-15 that activated NK cells to inhibit the progression of metastasis.In addition,MWA of human breast cancer stimulated an autologous NK-cell response.These results demonstrate that MWA of the primary tumor in metastatic breast cancer inhibits metastatic progression via the macrophage/IL-15/NK-cell axis.MWA of the primary tumor may be a promising treatment strategy for de novo stage IV breast cancer,although further substantiation is essential for clinical testing.Muxin Yu Hong Pan Nan Che Li Li Cong Wang Yue Wang Ge Ma Mengjia Qian Jiawei Liu Mingjie Zheng Hui Xie Lijun Ling Yi Zhao Xiaoxiang Guan Qiang Ding Wenbin Zhou Shui Wang 2021Cellular & Molecular Immunology2021,18,9:5
3Two-dimensional coupled mathematical modeling of fluvial processes with intense sediment transport and rapid bed evolution显示文摘Alluvial rivers may experience intense sediment transport and rapid bed evolution under a high flow regime,for which traditional decoupled mathematical river mod-els based on simplified conservation equations are not applicable. A two-dimen-sional coupled mathematical model is presented,which is generally applicable to the fluvial processes with either intense or weak sediment transport. The governing equations of the model comprise the complete shallow water hydrodynamic equa-tions closed with Manning roughness for boundary resistance and empirical rela-tionships for sediment exchange with the erodible bed. The second-order Total-Variation-Diminishing version of the Weighted-Average-Flux method,along with the HLLC approximate Riemann Solver,is adapted to solve the governing equations,which can properly resolve shock waves and contact discontinuities. 4Applying the WRF Double-Moment Six-Class Microphysics Scheme in the GRAPES_Meso Model: A Case Study显示文摘This study incorporated the Weather Research and Forecasting(WRF) model double-moment 6-class(WDM6) microphysics scheme into the mesoscale version of the Global/Regional Assimilation and Pr Ediction System(GRAPES_Meso). A rainfall event that occurred during 3–5 June 2015 around Beijing was simulated by using the WDM6, the WRF single-moment 6-class scheme(WSM6), and the NCEP 5-class scheme, respectively. The results show that both the distribution and magnitude of the rainfall simulated with WDM6 were more consistent with the observation. Compared with WDM6, WSM6 simulated larger cloud liquid water content, which provided more water vapor for graupel growth, leading to increased precipitation in the cold-rain processes. For areas with the warmrain processes, the sensitivity experiments using WDM6 showed that an increase in cloud condensation nuclei(CCN)number concentration led to enhanced CCN activation ratio and larger cloud droplet number concentration(Nc) but decreased cloud droplet effective diameter. The formation of more small-size cloud droplets resulted in a decrease in raindrop number concentration(Nr), inhibiting the warm-rain processes, thus gradually decreasing the amount of precipitation. For areas mainly with the cold-rain processes, the overall amount of precipitation increased; however, it gradually decreased when the CCN number concentration reached a certain magnitude. 11Unusual effects of vacuum annealing on large-area Ag_(3)PO_(4) microcrystalline film photoanode boosting cocatalyst-and scavengerfree water splitting显示文摘Ag_(3)PO_(4) as a novel photoanode material,despite its arguably highest photoactivity,suffers for its poor light absorption and stability for photoelectrochemical(PEC)water oxidation.In this work,4.54.5 cm^(2) Ag_(3)PO_(4) microcrystalline films are grown via a room-temperature solution process,and vacuum annealing is proposed to solve the stability and light absorption issues.It is found that the major process below 400℃ of vacuum annealing is the|Recovery|process for Ag_(3)PO_(4) microcrystals,when lattice defects and Ag0 surface species get reduced.Next,|Recrystallization|stage occurs at>40℃.The recovery of native defects of silver vacancies,with both density functional theory calculation and experimental results,could simultaneously improve the light absorption and catalytic activity of Ag_(3)PO_(4).The 400℃-annealed Ag_(3)PO_(4) photoanode,with enhanced light harvesting and crystal quality,exhibits 88% increase in(Jlight-Jdark)value(1.94 mA cm^(-2))than non-annealed photoanode(1.03 mA cm^(-2)).Moreover,it retains>99% current density after a 4000-s stability test.These results suggest that vacuum annealing can substantially improve the PEC performance of Ag_(3)PO_(4) microcrystalline film photoanodes due to mitigated effects of native defects,improved light harvesting,and inhibited Ag_(3)PO_(4) decomposition during water oxidation reaction.Qi Cao Jun Yu Yue Cao Jean-Jacques Delaunay Renchao Che 2021Journal of Materiomics2021,7,5:1

14Knowledge-enhanced semantic communication system with OFDM transmissions显示文摘As a promising technology to enable effective multi-modal transmission over wireless channels,semantic communication has attracted a lot of attention from academics and industries.Different from Shannon’s information theory,based on common background knowledge provided by the knowledge base,the goal of semantic communication is transmitting intended useful information from the transmitter and recovered by the receiver at the semantic level.However,the existing studies on semantic communication rarely emphasize the essence and the usage of the knowledge base.In this paper,we propose a knowledge-enhanced semantic communication(KESC)system,where the knowledge base is cloud-edge-device collaborative cached.To solve the problem that float-type symbols are difficult to transmit directly through a radio frequency(RF)system,we adopt orthogonal frequency division multiplexing(OFDM)to transmit semantic vectors directly without some traditional signal processing techniques in semantic information transmission,and the semantic pilot is designed to assist semantic reception.Furthermore,we formulate a multi-encoder transformer based neural network model for the KESC system to support text transmission(KESC-T),where the decoder is implemented with a knowledge graph to enhance the performance of semantic decoding.Besides,we define knowledge-enhanced efficiency(KEE)to measure the gain in semantic recovery accuracy brought by per unit of knowledge.Simulation results demonstrate that the recovery accuracy of our proposed KESC outperforms the compared scheme,especially in low signal-to-noise ratio(SNR)or resource-constrained scenarios.Xiaodong XU Huachao XIONG Yining WANG Yue CHE Shujun HAN Bizhu WANG Ping ZHANG 2023Science China(Information Sciences)2023,66,7:1

19Brexpiprazole caused glycolipid metabolic disorder by inhibiting GLP1/GLP1R signaling in rats显示文摘Brexpiprazole(Bre)is a new multi-target antipsychotic drug(APD)approved by the US FDA in 2015,and shows good therapeutic potential.But it lacks assessments on the metabolic side effects,which obstructs the treatment of schizophrenia.Glucagon-like peptide 1(GLP1),an incretin associated with insulin action and metabolism,is involved in the metabolic syndrome(MS)caused by most APDs.In this study,we examined the adverse effects of Bre on glycolipid metabolism in rats and determined whether GLP1 was involved in Bre-caused MS.In the first part of experiments,rats were orally administered Bre(0.5 mg·kg^(-1)·d^(-1))for 28 days with aripiprazole(1.0 mg·kg^(-1)·d^(-1))or olanzapine(1.0 mg·kg^(-1)·d^(-1))as the controls.Compared to vehicle,Bre administration significantly increased the weight gain,serum lipid(TG,TC,LDL,FFA),and blood glucose levels accompanied by the hormonal(insulin,glucagon,GLP1)imbalance,and the impaired glucose tolerance and insulin sensitivity.Moreover,we demonstrated that Bre administration significantly decreased the protein and mRNA levels of GLP1 in pancreas and small intestine by suppressing CaMKIIα,AMPK,and β-catenin;Bre administration also caused islet dysfunction with decreased GLP1R,PI3K,IRβexpression in pancreas,and the interference of IRS1,PI3K,p-AKT,and GLUT4 expression in the liver and skeletal muscle that represented the insulin resistance.In the second part of experiments,rats were orally administered Bre(0.5 mg·kg^(-1)·d^(-1))for 42 days.We showed that co-administration with the GLP1 receptor(GLP1R)agonist liraglutide(0.125 mg·kg^(-1)·d^(-1),ip)could ameliorate Bre-caused metabolic abnormalities.Our results demonstrate that GLP1/GLP1R signaling is involved in Bre-induced glycolipid metabolic disorders and co-treatment with liraglutide is an effective intervention against those abnormal metabolisms.De-juan Li Qin Yue Lu Liu Ke-ke Che Xue-mei Liu Chang-hua Hu 2021Acta Pharmacologica Sinica2021,42,8:1
