| 2 | Ultra-wide-dynamic-range gas sensing by optical pathlength multiplexed absorption spectroscopy显示文摘Laser absorption spectroscopy(LAS) has become the most widely used laser spectroscopic technique for gas detection due to its capability of accurate quantification and straightforward operation. However, since resolving weak absorption and averting over-absorption are always mutually exclusive, the dynamic range of the LAS-based gas sensor is limited and insufficient for many applications in fundamental study and industry. To overcome the limitation on the dynamic range, this article reports optical pathlength(OPL) multiplexed absorption spectroscopy using a gas cell having multiple internal reflections. It organically fuses together the transmission and reflection operation modes: the former directly uses the entire OPL of the gas cell, while the latter interrogates different internal short OPLs by optical interferometry, yielding >100-fold OPL variation. The achieved dynamic range is more than 6 orders of magnitude that surpasses other LAS techniques by 2–3 orders of magnitude. | 3 | Effects of L-3-n-butylphthalide on caspase-3 and nuclear factor kappa-B expression in primary basal forebrain and hippocampal cultures after beta-amyloid peptide 1-42 treatment显示文摘BACKGROUND:L-3-n-butylphthalide(L-NBP) can inhibit phosphorylation of tau protein and reduce the neurotoxicity of beta-amyloid peptide 1-42(Aβ_(1-42)). OBJECTIVE:To observe the neuroprotective effects of L-NBP on caspase-3 and nuclear factor kappa-B(NF-κB) expression in a rat model of Alzheimer's disease. DESIGN,TIME AND SETTING:A cell experiment was performed at the Central Laboratory of Provincial Hospital affiliated to Shandong University between January 2008 and August 2008. MATERIALS:L-NBP(purity>98%) was provided by Shijiazhuang Pharma Group NBP Pharmaceutical Company Limited.Aβ_(1-42),3-[4,5-dimethylthiazolo-2]-2,5 iphenyltetrazolium bromide(MTT),and rabbit anti-Caspase-3 polyclonal antibody were provided by Cell Signaling, USA;goat anti-choactase and rabbit anti-NF-κB antibodies were provided by Santa Cruz,USA. METHODS:Primary cultures were generated from rat basal forebrain and hippocampal neurons at 17 or 19 days of gestation.The cells were assigned into five groups:the control group,the Aβ_(1-42) group(2μmol/ L),the Aβ_(1-42) + 0.1μmol/L L-NBP group,the Aβ_(1-42)+ 1μmol/L L-NBP group,and the Aβ_(1-42)+ 10μmol/L L-NBP group.The neurons were treated with Aβ_(1-42)(2μmol/L) alone or in combination with L-NBP(0.1,1,10μmol/L) for 48 hours.Cells in the control group were incubated in PBS. MAIN OUTCOME MEASURES:Morphologic changes were evaluated using inverted microscopy, viability using the MTT method,and the changes in caspase-3 and NF-κB expression using Western blot. RESULTS:Induction with Aβ_(1-42) for 48 hours caused cell death and soma atrophy,and increased caspase-3 and NF-κB expression(P<0.05).L-NBP blocked these changes in cell morphology, decreased caspase-3 and NF-κB expression(P<0.05),and improved cell viability,especially at the high dose(P<0.05). | 5 | Establishment of image-guided radiotherapy of orthotopic hepatocellular carcinoma mouse model显示文摘Background:Hepatocellular carcinoma(HCC)is the most common type of liver cancer.Recently,developments in radiotherapy technology have led to radiotherapy becoming one of the main therapeutics of HCC.Therefore,a suitable animal model for radiotherapy of the orthotopic HCC mouse model is urgently needed.Methods:In the present study,Hepa1-6 cells were injected into the liver of C57BL/6 mice in situ to mimic the pathological characteristics of the original HCC.Tumor formation was monitored by applying magnetic resonance imaging techniques and verified by H&E histopathological staining,AFP staining,and Ki67 staining.A single dose of 10 Gy X-ray was applied to simulate clinical radiotherapy plans using image-guided radiotherapy(IGRT)equipment.The efficiency of radiotherapy was then assessed by examining tumor size and weight one week after radiation.Cleaved-caspase3 staining and TUNEL were used to assess apoptosis in tumor tissues.Results:Intrahepatic tumor development was detected in the liver according using MRI.A high-density shadow could be seen 10 days after cell injection,which indicated the formation of HCC in vivo.The tumors grew steadily bigger,and underwent precision radiotherapy 20 days after injection.The typical pathological characteristics of HCC,such as large,deeply stained nuclei and irregular cell size,were visible with H&E staining.After radiotherapy,significantly higher expression of the immunohistochemical markers Ki67 and AFP were detected in tumor tissue than in the nearby normal tissue.Compared with the control group,the tumor volume(p=0.05)and weight(p<0.05)of the irradiated group were significantly reduced.In addition,a higher frequency of apoptosis was identified in irradiated HCC tumor tissue using the TUNEL and cleaved-caspase3 staining assay.Conclusions:In a well-established orthotopic HCC model,MRI was utilized to monitor the formation of tumors,and IGRT was used to simulate clinical radiotherapy.The present study could provide a suitable preclinical system for HCC radiotherapyrelated studies. | Kaixiao Zhou Yabo Jiang Shuang Feng Wei Mo Jing Nie Jianping Cao Yang Jiao | 2023 | Animal Models and Experimental Medicine2023,6,5: | 0 |
