| 2 | High-frequency interference in low strain integrity testing of large-diameter pipe piles显示文摘The high-frequency interference exists obviously in low strain integrity testing of large-diameter pipe pile when a transientpoint load is applied. An analytical solution of vertical vibratory response of large-diameter pipe piles in low strain testing isdeduced in this paper. The analytical solution is verified by both numerical simulation and model test results. The time-domainvelocity responses on pile top are analyzed. The calculation results indicate that the time-domain responses at various pointssuffer different high-frequency interferences, thus the peak values and phases of different points are different. The influence ofvibratory modes on high-frequency interference is analyzed. It is found that the high-frequency interference at 90° point main-ly derives from the second flexural mode, but for other points it mainly originates from the first flexural mode. The factors af-fecting the frequency and peak value of interference waves have been investigated in this study. The results indicate that thelarger radius angle between the receiving and 90° points leads to greater peak value of high frequency wave crest. The leasthigh-frequency interference is detected at the angle of 90°. The frequency of interference waves is decreased with the increaseof pile radius, while the peak value is almost constant. The frequency is also related to pile modulus, i.e. the larger pile modu-lus results in greater frequency. The peak value varies with impulse width and soil resistance, i.e., the wider impulse width andlarger soil resistance cause smaller peak value. In conclusion, the frequency of interference waves is dependent on the geomet-rical and mechanics characteristics of the piles such as pile radius and modulus, but independent of the external conditionssuch as impulse width and soil resistance. On the other hand, the peak value of interference waves is mainly dependent on theexternal conditions but independent of the geometrical and mechanics characteristics of the piles. | 5 | Preparation of folate-conjugated starch nanoparticles and its application to tumor-targeted drug delivery vector显示文摘Anion starch nanoparticles (StNP) were prepared in water-in-oil microemulsion. Folate modi-fied with PEG was conjugated to the surface of StNP to obtain the folate-conjugated starch nanoparticles (FA-PEG/StNP). The average diameter of FA-PEG/ StNP determined by AFM and Zeta-Sizer apparatus was about 130 nm. Doxorubicin (DOX)-loaded FA- PEG/StNP was obtained via infiltrating combination. The result of UV spectrophotometer showed that the saturation concentration of DOX-loaded FA-PEG/ StNP was 28 μg/mg, which was effective for the con-trolled release of anticancer drug DOX. The cell ex-periments showed that the Lc50 of DOX-loaded FA-PEG/StNP and DOX-loaded StNP was higher than that of DOX, which indicates that FA-PEG/StNP and StNP can decrease the toxicity of DOX; while the lethal rate of DOX-loaded FA-PEG/StNP was 3 times that of DOX-loaded StNP with the same quantity of DOX, which indicates that FA in FA-PEG/StNP is ef-fective for improving the targeting function of nanoparticles, thus enhancing the inhibition effect of anticancer drug to cancer cell. | 6 | Coronavirus MHV-A59 infects the lung and causes severe pneumonia in C57BL/6 mice显示文摘It remains challenging to develop animal models of lung infection and severe pneumonia by severe acute respiratory syndrome coronavirus(SARS-CoV) and Middle East respiratory syndrome cornavirus(MERS-Co V) without high level of containment. This inevitably hinders understanding of virushost interaction and development of appropriate countermeasures. Here we report that intranasal inoculation of sublethal doses of murine coronavirus mouse hepatitis virus A-59(MHV-A59), a hepatic and neuronal tropic coronavirus, can induce acute pneumonia and severe lung injuries in C57BL/6 mice. Inflammatory leukocyte infiltrations, hemorrhages and fibrosis of alveolar walls can be observed 2-11 days after MHV-A59 infection. This pathological manifestation is associated with dramatical elevation of tissue IP-10 and IFN-γ and moderate increase of TNF-α and IL-1β, but inability of anti-viral type I interferon response. Based on Biot's poroelastic theory and the constitutive relations of the transversely isotropic medium, the dynamic governing equations of the outer and inner transversely isotropic saturated soil layers are derived. The Laplace transform is used to solve the governing equations of the outer and inner soil layers. The dynamic torsional response of the pipe pile in the frequency domain is derived utilizing 1D elastic theory and the continuous conditions at the interfaces between the pipe pile and the soils. The time domain solution is obtained by Fourier inverse transform. | 9 | Time-domain solution for transient dynamic response of a large-diameter thin-walled pipe pile显示文摘The propagation of stress waves in a large-diameter pipe pile for low strain dynamic testing cannot be explained properly by traditional 1D wave theories. A new computational model is established to obtain a wave equation that can describe the dynamic response of a large-diameter thin-walled pipe pile to a transient point load during a low strain integrity test. An analytical solution in the time domain is deduced using the separation of variables and variation of constant methods. The validity of this new solution is verifi ed by an existing analytical solution under free boundary conditions. The results of this time domain solution are also compared with the results of a frequency domain solution and fi eld test data. The comparisons indicate that the new solution agrees well with the results of previous solutions. Parametric studies using the new solution with reference to a case study are also carried out. The results show that the mode number affects the accuracy of the dynamic response. A mode number greater than 10 is required to enable the calculated dynamic responses to be independent of the mode number. The dynamic response is also greatly affected by soil properties. The larger the side resistance, the smaller the displacement response and the smaller the refl ected velocity wave crest. The displacement increases as the stress waves propagate along the pile when the pile shaft is free. The incident waves of displacement and velocity responses of the pile are not the same among different points in the circumferential direction on the pile top. | 10 | Studies of poly-L-lysine-starch nanoparticle preparation and its application as gene carrier显示文摘Anion starch nanoparticle (StNP) with a diameter of 50 nm was prepared in wa- ter-in-oil microemulsion, with soluble starch as raw materials and POCl3 as crosslinking agent. PLL-StNP was prepared by linking poly-L-lysine (PLL) on the surface of StNP. At the same time, the size of PLL-StNP and its stability in aqueous solution were checked by AFM. The analysis of plasmid DNA binding, DNase I enzymatic degradation, toxicity and transfection were done. We discovered that PLL-StNP may be used as non-virus nanoparticle gene carrier. And we devel- oped the method of preparing PLL-StNP gene carrier and used it in cell transfection. | 12 | Dialdehyde starch nanoparticles: Preparation and application in drug carrier显示文摘Dialdehyde starch nanoparticles (DASNP) were prepared by the redox reaction of NaIO4 and starch in water-in-oil microemulsion. IR spectrum showed that DASNP had aldehyde groups, and quantitative alkali consumption showed that its dialdehyde content was about (50±5)%. The average diameter of DASNP determined by SEM was about 100 nm. TGA-DTA showed that its thermal stability was better than starch nanoparticle (StNP) and dialdehyde starch (DAS). Its low biological toxicity was detected by cell experiment. Also the best mass ratio of doxorubicin (DOX) to combined DASNP detected by UV-VIS was 15 : 1, and the product was effective for controlled release of DOX. The cell experiment showed that the drug-carrier particle (DOX-DASNP) can release DOX for a long time and strengthened the effect of the anticancer drug. | 14 | Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications,but their consistency with primary tumors in phenotypic,genetic,and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients.Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining.In addition,we detected whole gene expression profiling between primary tumors and PDX generations.We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities.Finally,paclitaxel,cisplatin,doxorubicin,atezolizumab,afatininb,and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents.Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed.The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment,including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular,histopathological,and therapeutic characteristics,as well as the corresponding tumor heterogeneities,which provides a clinically relevant platform for drug screening,biomarker discovery,and translational research. | Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao | 2021 | Cancer Biology & Medicine2021,18,1: | 5 |
| 15 | Ratiometric delivery of doxorubicin and berberine by liposome enables superior therapeutic index than Doxil?显示文摘Although the appearance of Doxil alleviated the cardiotoxicity of DOX, the progression-free survival of patients was not prolonged compared with traditional medication regimens, and side effects such as hand-foot syndrome has occurred. In order to solve this dilemma, we have designed a novel co-delivery strategy to construct a co-loaded liposome of berberine(BER) and doxorubicin(DOX), which was called Lipo Be Do. The optimal synergistic ratio of the two drugs was screened by cell cytotoxicity experiments in vitro, and the optimal attenuation ratio was further determined by in vivo cardiac H&E staining pathological sections. The optimal combination treatment caused a robust increase in apoptotic cells of 4T1, as compared to drug alone treatment. The prepared co-loaded liposome, Lipo Be Do, had high encapsulation efficiency and good stability. The nanoliposome carrier controlled the biological fate of the drugs and maintained a pre-defined optimal ratio in vivo. The Lipo Be Do significantly inhibited tumor growth in 4T1 murine mammary carcinoma model compared with Doxil(P < 0.05), and completely overcame the myocardial rupture toxicity caused by Doxil in mice. | 18 | From Symbols to Embeddings:A Tale of Two Representations in Computational Social Science显示文摘Computational Social Science(CSS),aiming at utilizing computational methods to address social science problems,is a recent emerging and fast-developing field.The study of CSS is data-driven and significantly benefits from the availability of online user-generated contents and social networks,which contain rich text and network data for investigation.However,these large-scale and multi-modal data also present researchers with a great challenge:how to represent data effectively to mine the meanings we want in CSS?To explore the answer,we give a thorough review of data representations in CSS for both text and network.Specifically,we summarize existing representations into two schemes,namely symbol-based and embeddingbased representations,and introduce a series of typical methods for each scheme.Afterwards,we present the applications of the above representations based on the investigation of more than 400 research articles from 6 top venues involved with CSS.From the statistics of these applications,we unearth the strength of each kind of representations and discover the tendency that embedding-based representations are emerging and obtaining increasing attention over the last decade.Finally,we discuss several key challenges and open issues for future directions.This survey aims to provide a deeper understanding and more advisable applications of data representations for CSS researchers. | Huimin Chen Cheng Yang Xuanming Zhang Zhiyuan Liu Maosong Sun Jianbin Jin | 2021 | Journal of Social Computing2021,2,2: | 4 |
| 19 | Ectopic expression of fungal EcGDH improves nitrogen assimilation and grain yield in rice显示文摘NADP(H)-dependent glutamate dehydrogenases(GDH) in lower organisms have stronger ammonium affinity than those in higher plants. Here we report that transgenic rice overexpressing the EcGDH from Eurotium cheralieri exhibited significantly enhanced aminating activities.
