Crack nucleation and propagation simulation in brittle two-phase perforated/particulate composites by a phase field model显示文摘Fracture is a very common failure mode of the composite materials, which seriously affects the reliability and service-life of composite materials. Therefore, the study of the fracture behavior of the composite materials is of great significance and necessity, which demands an accurate and efficient numerical tool in general cases because of the complexity of the arising boundary-value or initial-boundary value problems. In this paper,a phase field model is adopted and applied for the numerical simulation of the crack nucleation and propagation in brittle linear elastic two-phase perforated/particulate composites under a quasi-static tensile loading. The phase field model can well describe the initiation, propagation and coalescence of the cracks without assuming the existence and the geometry of the initial cracks in advance. Its numerical implementation is realized within the framework of the finite element method(FEM).The accuracy and the efficiency of the present phase field model are verified by the available reference results in literature. In the numerical examples,we first study and discuss the influences of the hole/particle size on the crack propagation trajectory and the force-displacement curve. Then, the effects of the hole/particle shape on the crack initiation and propagation are investigated.Furthermore, numerical examples are presented and discussed to show the influences of the hole/particle location on the crack initiation and propagation characteristics. It will be demonstrated that the present phase field model is an efficient tool for the numerical simulation of the crack initiation and propagation problems in brittle two-phase composite materials, and the corresponding results may play an important role in predicting and preventing possible hazardous crack initiation and propagation in engineering applications.Xingxue Lu Yuliang Hou Ying Tie Cheng Li Chuanzeng Zhang 2020Acta Mechanica Sinica2020,36,2:5
Generation of isogenic single and multiplex gene knockout mice by base editing-induced STOP显示文摘Although CRISPR/Cas9 has been widely used to generate knockout mice, two major limitations remain:the founders usually carry a mixture of genotypes, and mosaicism harboring multiple genotypes. Therefore, it takes a long time to get homozygous mutants. Recently developed base editing(BE) system, which introduces C-to-T conversion without double strand DNA cleavage, has been used to introduce artificial stop codons(i-STOP) to prematurely terminate translation, providing a cleaner strategy for genome engineering. Using this strategy, we generated CD160 KO and VISTA/CD160 double KO mice by microinjection of a single sg RNA targeting CD160 and a mixture of sg RNAs targeting VISTA and CD160,respectively. The BE system induced STOP efficiently in mouse embryos and consequently in founder mice without detectable off-target. Most interestingly, the majority of the mutants harbor same genetic modifications, indicating we generated isogenic single and multiplex gene mutant mice by BE-induced STOP. We also obtained homozygous mutant mouse in F1 mice, demonstrating the accelerated strategy in generating animal models.Guang Yang Tianyu Zhu Zongyang Lu Guanglei Li Hao Zhang Songjie Feng Yajing Liu Jianan Li Yu Zhang Jia Chen Xuejiang Guo Xingxu Huang 2018Science Bulletin2018,63,17:2
Highly crystalline ReSe2 atomic layers synthesized by chemical vapor transport显示文摘Two-dimensional (2D) anisotropic rhenium diselenide (ReSe2) has attracted lots ofattention due to its promising applications in electronics and optoelectronics. However, controlled synthesis of high quality ultrathin ReSe2 remains as a challenge. Here we developed an approach for synthesizing high quality 2D ReSe2 flakes witha thickness down to monolayer by chemical vapor transport (CVT) through carefullytuning the growth kinetics. The atomic structures and anisotropy of theobtained ReSe2 flakes were intensively characterized with scanning transmissionelectron microscope and angle-resolved polarized Raman spectroscopy. Fieldeffecttransistors fabricated on the CVT-grown ReSe2 flakes showed n-typesemiconducting behavior with an on/off current ratio of 105 and a mobility up to5 cm2 V−1 s−1, which is comparable to mechanically exfoliated flakes and isobvious higher than the samples synthesized with other approaches. This study notonly make high quality 2D ReSe2 easily accessible for both fundamental and applicationexplorations but also sheds new lights on the chemical synthesis of otheranisotropic 2D materials.Lei Xing Xingxu Yan Jingying Zheng Guanchen Xu Zhixing Lu Lina Liu Jinghui Wang Peng Wang Xiaoqing Pan Liying Jiao 2019InfoMat2019,1,4:2
Crack propagation simulation in brittle elastic materials by a phase field method显示文摘To overcome the difficulties of re-meshing and tracking the crack-tip in other computational methods for crack propagation simulations, the phase field method based on the minimum energy principle is introduced by defining a continuous phase field variable(x)∈[0,1]to characterize discontinuous cracks in brittle materials. This method can well describe the crack initiation and propagation without assuming the shape, size and orientation of the initial crack in advance. In this paper,a phase field method based on Miehe's approach[Miehe et al.,Comp.Meth.App.Mech.Eng.(2010)]is applied to simulate different crack propagation problems in twodimensional(2D),isotropic and linear elastic materials. The numerical implementation of the phase field method is realized within the framework of the finite element method(FEM).The validity, accuracy and efficiency of the present method are verified by comparing the numerical results with other reference results in literature.Several numerical examples are presented to show the effects of the loading type(tension and shear),boundary conditions, and initial crack location and orientation on the crack propagation path and force-displacement curve.Furthermore, for a single edge-cracked bi-material specimen, the influences of the loading type and the crack location on the crack propagation trajectory and force-displacement curve are also investigated and discussed. It is demonstrated that the phase field method is an efficient tool for the numerical simulation of the crack propagation problems in brittle elastic materials, and the corresponding results may have an important relevance for predicting and preventing possible crack propagations in engineering applications.Xingxue Lu Cheng Li Ying Tie Yuliang Hou Chuanzeng Zhang 2019Theoretical & Applied Mechanics Letters2019,9,6:2
Efficient silencing of the multicopy DUX4 gene by ABE-mediated start codon mutation in human embryos显示文摘The Dux gene plays an important role in mouse embryo development(De Iaco et al.,2017;Hendrickson et al.,2017;Whiddon et al.,2017).It participates in the maternal-to-zygotic transition(MTZ),which mainly includes maternal transcript degradation and zygotic genome activation(ZGA)and is the key process for embryonic development.Approximately 11-100 copies of the DUX4 gene form a tandem cluster in the human genome(Snider et al.,2009,2010).Yubing Liu Xinmei Lu Min Ye Ling Wang Rongxin Tang Zhiyong Yang Bongkoch Turathum Chenchen Liu Yujing Xue Menghua Wu Yannan Yang Ermeng Gao Di Zhang Feng Yang Keh-Kooi Kee Xingxu Huang Guanglei Li Ri-Cheng Chian 2022Journal of Genetics and Genomics2022,49,10:1
