3Statistical variances of diffusional properties from ab initio molecular dynamics simulations显示文摘Ab initio molecular dynamics(AIMD)simulation is widely employed in studying diffusion mechanisms and in quantifying diffusional properties of materials.However,AIMD simulations are often limited to a few hundred atoms and a short,sub-nanosecond physical timescale,which leads to models that include only a limited number of diffusion events.As a result,the diffusional properties obtained from AIMD simulations are often plagued by poor statistics.In this paper,we re-examine the process to estimate diffusivity and ionic conductivity from the AIMD simulations and establish the procedure to minimize the fitting errors.In addition,we propose methods for quantifying the statistical variance of the diffusivity and ionic conductivity from the number of diffusion events observed during the AIMD simulation.Since an adequate number of diffusion events must be sampled,AIMD simulations should be sufficiently long and can only be performed on materials with reasonably fast diffusion.We chart the ranges of materials and physical conditions that can be accessible by AIMD simulations in studying diffusional properties.Our work provides the foundation for quantifying the statistical confidence levels of diffusion results from AIMD simulations and for correctly employing this powerful technique.Xingfeng He Yizhou Zhu Alexander Epstein Yifei Mo 2018npj Computational Materials2018,,1:6
4Combination of ablative fractional carbon dioxide laser and platelet-rich plasma treatment to improve hypertrophic scars:a retrospective clinical observational study显示文摘Background:Hypertrophic scars are one of the main complications that affect the quality of life of patients after burns.Many methods have been shown to be effective in the treatment of hypertrophic scars,such as ablative fractional CO_(2) laser(AFCL)and platelet-rich plasma(PRP).However,there are few studies on the effect of the combined application of these measures.The purpose of this study was to explore the therapeutic effect of AFCL combined with PRP on hypertrophic burn scars.Methods:A retrospective clinical observation study was conducted on 50 patients with hypertrophic burn scars.The AFCL+PRP group included 31 patients who received AFCL combined with PRP treatment;the AFCL group included 19 patients who received AFCL treatment only.The University of North Carolina 4P Scar Scale(UNC4P)and the Vancouver Scar Scale(VSS)scores that were collected before each treatment were used as indicators of the effectiveness of the previous treatment.The scores recorded at the second,fourth and seventh months were analysed.Results:The demographic data of the 2 groups were not significantly different.Before treatment,therewas no difference in the UNC4P and VSS scores between the 2 groups.Therewas a significant decline in the UNC4P and VSS total scores over 6 months in both groups(p<0.05)and scores in the 2 groups were comparable after 3 and 6 months(p<0.05).UNC4P scores in the AFCL+PRP group decreased from a mean of 8.26 to 2.61(p<0.05)with a concomitant drop in VSS scores from a mean of 11.74 to 6.06(p<0.01).In the AFCL group UNC4P and VSS scores decreased from 7.68 to 4.63(p<0.05)and from 10.89 to 8.16(p<0.05),respectively.The sub-items of these 2 assessments were analysed and the results suggest that AFCL combined with PRP can comprehensively improve scarring.Conclusions:This study shows that PRP is an effective adjunct for AFCL in the treatment of hypertrophic burn scars and that the combination of PRP and AFCL proved to be more useful than AFCL alone.This combination may be a new and effective clinical practice for the treatment of scars.However,larger and higher-level clinical studies are still needed to determine its efficacy and possible mechanisms.Zhanzhan Dai Xiaozhen Lou Tuo Shen Yu Sun Yongqiang Xiao Xingfeng Zheng Xuexin Wang Yu Peng Yukun Guo Yibin Guo Jiannan Wen He Fang Bing Ma Zhaofan Xia 2021Burns & Trauma2021,9,1:4
5Targeting PFKL with penfluridol inhibits glycolysis and suppresses esophageal cancer tumorigenesis in an AMPK/FOXO3a/BIM-dependent manner显示文摘As one of the hallmarks of cancer,metabolic reprogramming leads to cancer progression,and targeting glycolytic enzymes could be useful strategies for cancer therapy.By screening a small molecule library consisting of 1320 FDA-approved drugs,we found that penfluridol,an antipsychotic drug used to treat schizophrenia,could inhibit glycolysis and induce apoptosis in esophageal squamous cell carcinoma(ESCC).Gene profiling and Ingenuity Pathway Analysis suggested the important role of AMPK in action mechanism of penfluridol.By using drug affinity responsive target stability(DARTS)technology and proteomics,we identified phosphofructokinase,liver type(PFKL),a key enzyme in glycolysis,as a direct target of penfluridol.Penfluridol could not exhibit its anticancer property in PFKL-deficient cancer cells,illustrating that PFKL is essential for the bioactivity of penfluridol.High PFKL expression is correlated with advanced stages and poor survival of ESCC patients,and silencing of PFKL significantly suppressed tumor growth.Mechanistically,direct binding of penfluridol and PFKL inhibits glucose consumption,lactate and ATP production,leads to nuclear translocation of FOXO3a and subsequent transcriptional activation of BIM in an AMPK-dependent manner.Taken together,PFKL is a potential prognostic biomarker and therapeutic target in ESCC,and penfluridol may be a new therapeutic option for management of this lethal disease.Cancan Zheng Xiaomei Yu Yiyao Liang Yidong Zhu Yan He Long Liao Dingkang Wang Yanming Yang Xingfeng Yin Ang Li Qingyu He Bin Li 2022Acta Pharmaceutica Sinica B2022,12,3:4
6Metamaterial-enabled arbitrary on-chip spatial mode manipulation显示文摘On-chip spatial mode operation,represented as mode-division multiplexing(MDM),can support high-capacity data communications and promise superior performance in various systems and numerous applications from optical sensing to nonlinear and quantum optics.However,the scalability of state-of-the-art mode manipulation techniques is significantly hindered not only by the particular mode-order-oriented design strategy but also by the inherent limitations of possibly achievable mode orders.Recently,metamaterials capable of providing subwavelength-scale control of optical wavefronts have emerged as an attractive alternative to manipulate guided modes with compact footprints and broadband functionalities.Herein,we propose a universal yet efficient design framework based on the topological metamaterial building block(BB),enabling the excitation of arbitrary high-order spatial modes in silicon waveguides.By simply programming the layout of multiple fully etched dielectric metamaterial perturbations with predefined mathematical formulas,arbitrary high-order mode conversion and mode exchange can be simultaneously realized with uniform and competitive performance.The extraordinary scalability of the metamaterial BB frame is experimentally benchmarked by a record high-order mode operator up to the twentieth.As a proof of conceptual application,an 8-mode MDM data transmission of 28-GBaud 16-QAM optical signals is also verified with an aggregate data rate of 813 Gb/s(7%FEC).This user-friendly metamaterial BB concept marks a quintessential breakthrough for comprehensive manipulation of spatial light on-chip by breaking the long-standing shackles on the scalability,which may open up fascinating opportunities for complex photonic functionalities previously inaccessible.Jinlong Xiang Zhiyuan Tao Xingfeng Li Yaotian Zhao Yu He Xuhan Guo Yikai Su 2022Light(Science & Applications)2022,11,7:2
7Ultracompact topological photonic switch based on valley-vortex-enhanced high-efficiency phases shift显示文摘Topologically protected edge states based on valley photonic crystals(VPCs)have been widely studied,from theoretical verifcation to technical applications.However,research on integrated tuneable topological devices is still lacking.Here,we study the phase-shifting theory of topological edge modes based on a VPC structure.Benefiting from the phase vortex formed by the VPC structure,the optical path of the topological edge mode in the propagation direction is approximately two-fold that of the conventional optical mode in a strip waveguide.In experiments,we show a 1.57-fold improvement inπ-phase tuning efficiency.By leveraging the highefficiency phase-shifting properties and the sharp-turn features of the topological waveguide,we demonstrate an ultracompact 1×2 thermo-optic topological switch(TOTS)operating at telecommunication wavelengths.A switching power of 18.2 mW is needed with an ultracompact device footprint of 25.66×28.3μm in the wavelength range of 1530-1582 nm.To the best of our knowledge,this topological photonic switch is the smallest switch of any dielectric or semiconductor 1×2/2×2 broadband optical switches,including thermo-optic and electro-optic switches.In addition,a high-speed transmission experiment employing the proposed TOTS is carried out to demonstrate the robust transmission of high-speed data.Our work reveals the phase shifting mechanism of valley edge modes,which may enable diverse topological functional devices in many fields,such as optical communications,nanophotonics,and quantum information processing.Hongwei Wang Gujing Tang Yu He Zhen Wang Xingfeng Li Lu Sun Yong Zhang Luqi Yuan Jianwen Dong Yikai Su 2022Light(Science & Applications)2022,11,11:1
8Infection of post-harvest peaches by Monilinia fructicola accelerates sucrose decomposition and stimulates the Embden–Meyerhof–Parnas pathway显示文摘To study the changes in sugar metabolism caused by fungal infection in post-harvest peaches,fruit from two cultivars(‘Baifeng’and‘Yulu’)was inoculated with Monilinia fructicola and stored at 10°C.During disease development,soluble sugar content was monitored,as well as the activities and expression of selected enzymes.Disease progression was accompanied by a decrease in sucrose content and increases in reducing sugars and soluble solids,consistent with higher enzyme activities for acid invertase,neutral invertase and sucrose synthase-cleavage,and lower activities for sucrose synthase-synthesis and sucrose phosphate synthase.Activities of phosphofructokinase,hexokinase,and pyruvate kinase,which are related to hexose metabolism,also increased.These changes stimulate the Embden–Meyerhof–Parnas(EMP)pathway.We conclude that the fungal disease in peach fruit accelerates the decomposition of sucrose,thereby providing more glucose as a substrate to the EMP pathway.Jingyu Kou Yingying Wei Xingxing He Jiayu Xu Feng Xu Xingfeng Shao 2018Horticulture Research2018,5,1:1
9Deubiquitinase USP28 inhibits ubiquitin ligase KLHL2-mediated uridine-cytidine kinase 1 degradation and confers sensitivity to 5'-azacytidine-resistant human leukemia cells显示文摘Resistance to the chemotherapeutic drug 5'-azacytidine(5'-AZA)is a major obstacle in the treatment of patients with acute myeloid leukemia(AML).The uridine-cytidine kinase 1(UCK1)has an established role in activating 5'-AZA and its protein level is significantly downregulated in patients resistant to the drug.However,the underlying molecular mechanism for the reduced UCK1 expression remains to be elucidated.We demonstrated that the ubiquitin E3 ligase KLHL2 interacted with UCK1 and mediated its polyubiquitination at the K81 residue and degradation.We showed that deubiquitinase USP28 antagonized KLHL2-mediated polyubiquitylation of UCK1.We also provided evidence that ATM-mediated phosphorylation of USP28 resulted in its disassociation from KLHL2 and UCK1 destabilization.Conversely,UCK1 phosphorylation by 5'-AZA-activated ATM enhanced the KLHL2-UCK1 complex formation,Importantly,silencing KLHL2 or USP28 overexpression not only inhibited AML cell proliferation but also sensitized AML cells to 5'-AZA-induced apoptosis in vitro and in vivo.These results were no longer observed in USP28-deficient cells.Zhang Heng Huang He Feng Xing Song Huirven Zhang Zhiyong Shen Aizong Qiu Xingfeng 2021解剖学杂志2021,44,S01:0
10Primary cardiac CIC-rearranged undifferentiated sarcoma in an infant显示文摘Introduction:Cardiac neoplasms are particularly rare in children,and the majority of these tumors are benign.Approximately 10%of cardiac neoplasms are malignant,including soft tissue sarcomas and lymphomas.Cardiac tumors could also be metastases.Primitive EWSR1-negative round or spindle cell undifferentiated sarcoma harboring CIC gene translocation is a highly aggressive malignancy mainly occurring in soft tissues.However,it has not yet been described in the heart.Case presentation:We report a sarcoma that arose from the right ventricle in a 1-year-old girl.Histologically,it was composed of closely arranged small round or oval undifferentiated cells with fibrovascular separation,hyaline degeneration,and geographical necrosis.Immunohistochemically,the neoplastic cells exhibited focal membrane positivity for CD99 and diffuse positivity for WT1 and ETV4.Fluorescent in situ hybridization analysis showed EWSR1-negative but CIC-positive split signals.The breakpoint was also confirmed by whole genome sequencing.Conclusion:Based on morphological,immunohistochemical and molecular findings,this cardiac mass was diagnosed as CIC-rearranged sarcoma.Meng Zhang Yeran Yang Xiaoxing Guan Xingfeng Yao Yongli Guo Lejian He 2021Pediatric Investigation2021,5,4:0
11Discrepancies and error evaluation metrics for machine learning interatomic potentials显示文摘Machine learning interatomic potentials(MLIPs)are a promising technique for atomic modeling.While small errors are widely reported for MLIPs,an open concern is whether MLIPs can accurately reproduce atomistic dynamics and related physical properties in molecular dynamics(MD)simulations.In this study,we examine the state-of-the-art MLIPs and uncover several discrepancies related to atom dynamics,defects,and rare events(REs),compared to ab initio methods.We find that low averaged errors by current MLIP testing are insufficient,and develop quantitative metrics that better indicate the accurate prediction of atomic dynamics by MLIPs.The MLIPs optimized by the RE-based evaluation metrics are demonstrated to have improved prediction in multiple properties.The identified errors,the evaluation metrics,and the proposed process of developing such metrics are general to MLIPs,thus providing valuable guidance for future testing and improvements of accurate and reliable MLIPs for atomistic modeling.Yunsheng Liu Xingfeng He Yifei Mo 2023npj Computational Materials2023,,1:0
