| 1 | Optimization of the Ultrasonic-assisted Extraction Process for Protocatechuic Acid from Emilia sonchifolia DC by Response Surface Methodology显示文摘[Objectives] This study was conducted to optimize the extraction process of protocatechuic acid from Emilia sonchifolia DC. [Methods] The optimal extraction conditions were determined by single factor,response surface analysis and variance analysis,and the content of protocatechuic acid was determined by HPLC. [Results] The protocatechuic acid standard curve equation was: y = 1 435 x + 8 403,R^2= 0. 999 8,indicating a good linear relationship. The optimal extraction conditions were as follows: a temperature at 80 ℃,an extraction time of 1 h,a material-to-liquid ratio at 1:10 and an ultrasonic power of 600 W,and the content of protocatechuic acid extracted was 1. 93 mg/g. The method showed a RSD of 0. 41%,less than 2%,and the detection limit was 0. 0000047261 g/ml.The experimental sample X1 was the low-level 0. 1 mg/ml standard solution,which showed recovery of protocatechuic acid between 100.8% and 105.2%,with a RSD of 0. 013%;and the sample X2 was the high-level 1. 0 mg/ml standard solution,which exhibited recovery between 100. 6% and 102. 2%,with a RSD of 0.076%. Thus,the recovery was high,and the requirements of the performance index were met. | 2 | Myristica fragrans promotes ABCA1 expression and cholesterol efflux in THP-1-derived macrophages显示文摘Myristica fragrans is a traditional herbal medicine and has been shown to alleviate the development of atherosclerosis.However,the anti-atherogenic mechanisms of M.fragrans are still to be addressed.In this study,we explored the effect of M.fragrans on lipid metabolism and inflammation and its mechanisms in THP-1-derived macrophages.The quantitative polymerase chain reaction and western blot analysis results showed that M.fragrans promotes cholesterol efflux from THP-1-derived macrophages and reduces intracellular total cholesterol,cholesterol ester,and free cholesterol contents in a dose-and a time-dependent manner.Further study found that liver X receptor alpha(LXRα)antagonist GGPP significantly blocked the upregulation of ABCA1 expression with M.fragrans treatment.In addition,chromatin immunoprecipitation assay confirmed that GATA binding protein 3(GATA3)can bind to the LXRαpromoter,and inhibition of GATA3 led to the downregulation of LXRαand ATP-binding cassette subfamily A member 1 expression.Furthermore,M.fragrans reduced lipid accumulation,followed by decreasing tumor necrosis factor-α,interleukin(IL)-6,and IL-1βand increasing IL-10 produced by THP-1-derived macrophages.Therefore,M.fragrans is identified as a valuable therapeutic medicine for atherosclerotic cardiovascular disease. | Shangming Liu Jiahui Gao Linhao He Zhenwang Zhao Gang Wang Jin Zou Li Zhou Xiangjun Wan Shilin Tang Chaoke Tang | 2021 | Acta Biochimica et Biophysica Sinica2021，53，1： | 2 |

| 7 | Booster Vaccination in Infancy Reduces the Incidence of Occult HBV Infection in Maternal HBsAg-positive Children显示文摘Background and Aims: Occult HBV infection (OBI) in chil-dren has proven to be associated with their immune re-sponse to hepatitis B vaccine (HepB). This study aimed to investigate the effect of a booster HepB on OBI, which is rarely investigated. Methods: This study enrolled 236 ma-ternal HBsAg-positive children who were followed up annu-ally until 8 years of age and were hepatitis B surface antigen (HBsAg) negative. Of those 100 received a booster HepB be-tween 1 and 3 years of age (booster group), and 136 were never boosted (non-booster group). Serial follow-up data of children and baseline data of their mothers were collected and between-group differences were analyzed. Results: The incidence of OBI varied dynamically during follow-up, with 37.14% (78/210), 19.09% (42/220), 20.85% (44/211), 31.61% (61/193), 8.65% (18/208) and 12.71% (30/236) at 7 months, 1, 2, 3, 4, and 8 years of age. At 8 years of age, the negative conversion rate of HBV DNA in the booster group was significantly higher than that in non-booster group [57.89% (11/19) vs. 30.51% (18/59), p=0.032]. For chil-dren without OBI at 7 months old, the incidence of OBI in booster group was significantly lower than that in non-boost-er group [25.64% (10/39) vs. 67.74% (63/93), p<0.001].
