2Morphological, cytological and molecular analyses of a synthetic hexaploid derived from an interspecific hybrid between Gossypium hirsutum and Gossypium anomalum显示文摘Gossypium anomalum represents an inestimable source of genes that could potentially be transferred into the gene pool of cultivated cotton. To resolve interspecific hybrid sterility problems, we previously treated triploid hybrids derived from a cross between Gossypium hirsutum and G. anomalum with 0.15% colchicine and obtained a putative fertile hexaploid. In this study, we performed morphological, molecular and cytological analyses to assess the hybridity and doubled status of putative interspecific hybrid plants. Most of the morphological characteristics of the putative hexaploid plants were intermediate between G. hirsutum and G.anomalum. Analysis of mitotic metaphase plates revealed 78 chromosomes, confirming the doubled hybrid status of the hexaploid. Genome-wide molecular analysis with different genome-derived SSR markers revealed a high level of polymorphism(96.6%) between G. hirsutum and G. anomalum. The marker transferability rate from other species to G. anomalum was as high as 98.0%. The high percentage of polymorphic markers with additive banding profiles in the hexaploid indicates the hybridity of the hexaploid on a genome-wide level. A-genome-derived markers were more powerful for distinguishing the genomic differences between G. hirsutum and G. anomalum than D-genome-derived markers. This study demonstrates the hybridity and chromosomally doubled status of the(G. anomalum × G. hirsutum)2hexaploid using morphological, cytological and molecular marker methods. 3A conservative local discontinuous Galerkin method for the solution of nonlinear Schrdinger equation in two dimensions显示文摘In this study, we present a conservative local discontinuous Galerkin(LDG) method for numerically solving the two-dimensional nonlinear Schrdinger(NLS) equation. The NLS equation is rewritten as a firstorder system and then we construct the LDG formulation with appropriate numerical flux. The mass and energy conserving laws for the semi-discrete formulation can be proved based on different choices of numerical fluxes such as the central, alternative and upwind-based flux. We will propose two kinds of time discretization methods for the semi-discrete formulation. One is based on Crank-Nicolson method and can be proved to preserve the discrete mass and energy conservation. The other one is Krylov implicit integration factor(IIF) method which demands much less computational effort. 4Genome-wide profiling of RNA editing sites in sheep显示文摘Background: The widely observed RNA-DNA differences(RDDs) have been found to be due to nucleotide alteration by RNA editing. Canonical RNA editing(i.e., A-to-I and C-to-U editing) mediated by the adenosine deaminases acting on RNA(ADAR) family and apolipoprotein B mRNA editing catalytic polypeptide-like(APOBEC)family during the transcriptional process is considered common and essential for the development of an individual.To date, an increasing number of RNA editing sites have been reported in human, rodents, and some farm animals;however, genome-wide detection of RNA editing events in sheep has not been reported. The aim of this study was to identify RNA editing events in sheep by comparing the RNA-seq and DNA-seq data from three biological replicates of the kidney and spleen tissues.Results: A total of 607 and 994 common edited sites within the three biological replicates were identified in the ovine kidney and spleen, respectively. Many of the RDDs were specific to an individual. The RNA editing-related genes identified in the present study might be evolved for specific biological functions in sheep, such as structural constituent of the cytoskeleton and microtubule-based processes. Furthermore, the edited sites found in the ovine BLCAP and NEIL1 genes are in line with those in previous reports on the porcine and human homologs, suggesting the existence of evolutionarily conserved RNA editing sites and they may play an important role in the structure and function of genes.Conclusions: Our study is the first to investigate RNA editing events in sheep. We screened out 607 and 994 RNA editing sites in three biological replicates of the ovine kidney and spleen and annotated 164 and 247 genes in the kidney and spleen, respectively. The gene function and conservation analysis of these RNA editing-related genes suggest that RNA editing is associated with important gene function in sheep. 5Revealing the critical effect of solid electrolyte interphase on the deposition and detriment of Co(Ⅱ) ions to graphite anode显示文摘'Dissolution,migration,and deposition'of transition metal ions (TMIs) result in capacity degradation of lithium-ion batteries (LIBs).Understanding such detrimental mechanism of TMIs is critical to the development of LIBs with long cycle life.In most previous works,TMIs were directly introduced into the electrolyte to investigate such a detrimental mechanism.In these cases,the TMIs are deposited directly on the fresh anode surface.However,in the practical battery system,the TMIs are deposited on the anode covered with solid electrolyte interphase (SEI) film.Whether the pre-presence of SEI film on anode surface influences the deposition and detriment of TMIs is unclear.In this work,the deposition of Co element on graphite anode with and without SEI film were systematically studied.The results clearly show that,in comparison with that of fresh graphite (SEI-free),the presence of SEI film aggravates the deposition of Co ions due to the Li^(+)–Co^(2+) ion exchange between the SEI and Co^(2+)-containing electrolyte without the driving of the electric field,leading to faster capacity fading of graphite anode.Therefore,how to regulate electrolytes and film-forming additives to design the components of SEI and prevent its exchange with TMIs,is a crucial way to inhibit the deposition and detriment of TMIs on graphite anode.Qiming Xie Jiawei Chen Lidan Xing Xianggui Zhou Zekai Ma Binhong Wu Yilong Lin Hebing Zhou Weishan Li 2022Journal of Energy Chemistry2022,31,6:2

8Molecular cloning and characterization of the endothelin 3 gene in black bone sheep显示文摘Background: Black bone sheep was first discovered in Yunnan province of China in 1970, with unique black pigmentation on the body and internal organs. Endothelin 3(EDN3) has been known as a key gene causing hyperpigmentation in black bone chicken, the Silky fowl.Methods: In this study, EDN3 was employed as a candidate gene for regulating black color pigmentation. First,EDN3 was cloned from sheep to obtain the full-length cDNA by using the rapid amplification of cDNA ends(RACE).Genomic EDN3 was screened and a total of thirty predicted single nucleotide polymorphisms(SNPs) were genotyped for allele and genotype frequency analysis in a case-control study involving two black bone sheep populations. Genomic copy number analysis of EDN3 in sheep was conducted to measure the variation in copy number. EDN3 expression levels were observed among the groups in adult liver, lymph node, and kidney tissues, as well as embryo kidney samples. Also, among the tissues of black bone and non-black bone sheep.Results: The size of the full-length cDNA was 1,578 bp, which included 426 bp of 5′-untranslated region(5′-UTR),an open reading frame(ORF) of 639 bp encoding a protein of 212 amino acids, and a 3′-UTR of 513 bp. Genotype and allele frequencies of all the discovered SNPs were found insignificantly different in black bone and non-black bone sheep(P > 0.05). Genomic copy number analysis of EDN3 in sheep revealed no significant difference between the two sheep groups. No significant variations were found in the adult liver and kidney embryo samples. However,the expression in lymph node and kidney tissue was significantly higher in black bone sheep than that in non-black bone sheep(P < 0.05). 9The Principle of Introducing Halogen Ions Into β-FeOOH: Controlling Electronic Structure and Electrochemical Performance显示文摘Coordination tuning electronic structure of host materials is a quite effective strategy for activating and improving the intrinsic properties.Herein,halogen anion(X-)-incorporated β-FeOOH(β-FeOOH(X),X=F-,Cl-,and Br-) was investigated with a spontaneous adsorption process,which realized a great improvement of supercapacitor performances by adjusting the coordination geometry.Experiments coupled with theoretical calculations demonstrated that the change of Fe-O bond length and structural distortion of β-FeOOH,which is rooted in halogen ions embedment,led to the relatively narrow band gap.Because of the strong electronegativity of X-,the Fe element in β-FeOOH(X)s presented the unexpected high valence state(3+δ),which is facilitating to adsorb S032-species.Consequently,the designed β-FeOOH(X)s exhibited the good electric conductivity and enhanced the contact between electrode and electrolyte.When used as a negative electrode,the β-FeOOH(F) showed the excellent specific capacity of 391.9 F g-1 at 1 A g-1 current density,almost tenfold improvement compared with initial β-FeOOH,with the superior rate capacity and cyclic stability.This combinational design principle of electronic structure and electrochemical performances provides a promising way to develop advanced electrode materials for supercapacitor.Dongbin Zhang Xuzhao Han Xianggui Kong Fazhi Zhang Xiaodong Lei 2020Nano-Micro Letters2020,12,9:2
