2Oral microbiota in human systematic diseases显示文摘Oral bacteria directly affect the disease status of dental caries and periodontal diseases. The dynamic oral microbiota cooperates with the host to reflect the information and status of immunity and metabolism through two-way communication along the oral cavity and the systemic organs. The oral cavity is one of the most important interaction windows between the human body and the environment.The microenvironment at different sites in the oral cavity has different microbial compositions and is regulated by complex signaling,hosts, and external environmental factors. These processes may affect or reflect human health because certain health states seem to be related to the composition of oral bacteria, and the destruction of the microbial community is related to systemic diseases. In this review, we discussed emerging and exciting evidence of complex and important connections between the oral microbes and multiple human systemic diseases, and the possible contribution of the oral microorganisms to systemic diseases. This review aims to enhance the interest to oral microbes on the whole human body, and also improve clinician’s understanding of the role of oral microbes in systemic diseases. 3Speech endpoint detection in low-SNRs environment based on perception spectrogram structure boundary parameter显示文摘The Perception Spectrogram Structure Boundary(PSSB)parameter is proposed for speech endpoint detection as a preprocess of speech or speaker recognition.At first a hearing perception speech enhancement is carried out.Then the two-dimensional enhancement is performed upon the sound spectrogram according to the difference between the determinacy distribution characteristic of speech and the random distribution characteristic of noise.Finally a decision for endpoint was made by the PSSB parameter.Experimental results show that,in a low SNR environment from-10 dB to 10 dB,the algorithm proposed in this paper may achieve higher accuracy than the extant endpoint detection algorithms.The detection accuracy of 75.2%can be reached even in the extremely low SNR at-10 dB.Therefore it is suitable for speech endpoint detection in low-SNRs environment.WU Di ZHAO Heming HUANG Chengwei XIAO Zhongzhe ZHANG Xiaojun XU Yishen TAO Zhi 2014Chinese Journal of Acoustics2014,33,4:8
4Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations. 5HY5-HDA9 Module Transcriptionally Regulates Plant Autophagy in Response to Light-to-Dark Conversion and Nitrogen Starvation显示文摘Light is arguably one of the most important environmental factors that determines virtually all aspects of plant growth and development,but the molecular link between light signaling and the autophagy pathway has not been elucidated in plants.In this study,we demonstrate that autophagy is activated during light-to-dark conversion though transcriptional upregulation of autophagy-related genes(ATGs).We showed that depletion of the ELONGATED HYPOCOTYL 5(HY5),a key component of light signaling,leads to enhanced autophagy activity and resistance to extended darkness and nitrogen starvation treatments,contributing to higher expression oiATGs.HY5 interacts with and recruits HISTONE DEACETYLASE 9(HDA9)to ATG5 and ATG8e loci to repress their expression by deacetylation of the Lys9 and Lys27 of histone 3.Furthermore,we found that both darkness and nitrogen depletion induce the degradation of HY5 via 26S proteasome and the concomitant disassociation of HDA9 from ATG5 and ATG8e loci,leading to their depression and thereby activated autophagy.Genetic analysis further confirmed that HY5 and HDA9 act synergistically and function upstream of the autophagy pathway.Collectively,our study unveils a previously unknown transcriptional and epigenetic network that regulates autophagy in response to light-to-dark conversion and nitrogen starvation in plants.Chao Yang Wenjin Shen Lianming Yang Yun Sun Xibao Li Minyi Lai Juan Wei Chaojun Wang Yingchao Xu Faqiang Li Shan Liang Chengwei Yang Shangwei Zhong Ming Luo Caiji Gao 2020Molecular Plant2020,13,3:8
6Function of Ca^(2+)-/calmodulin-dependent protein kinase Ⅳ in Ca^(2+)-stimulated neuronal signaling and behavior显示文摘The activity of Ca2+/calmodulin-dependent protein kinase IV(Ca MKIV) is sensitive to activity-dependent changes in the level of intracellular Ca2+.Following neuronal stimulation,the activation of Ca MKIV may trigger synaptic modifications and transcriptional responses,both of which are involved in regulating cognitive and emotional behavior.Here,we used Ca MKIV knockout(KO) neurons and mice to examine the function of Ca MKIV in Ca2+-stimulated intracellular signaling and animal behavior,respectively.Following NMDA receptor activation or membrane depolarization,the up-regulation of CREB(c AMP responsive element binding protein) and its target gene Bdnf(brain-derived neurotrophic factor) was intact in cortical neurons obtained from Ca MKIV KO mice.Ca MKIV KO mice displayed severe impairment in contextual fear memory but normal locomotor activity and anxiety level in the contextual training chamber.Although Ca MKIV KO mice showed normal memory in the standard passive avoidance task,they were defective in learning the temporal dissociative passive avoidance task.As indicated by the light/dark test and marble-burying test data,Ca MKIV KO mice showed less anxiety and normal perseveration.In the voluntary wheel-running test,Ca MKIV KO mice showed normal running time and distance but higher maximal running speed.Our results demonstrate the function of Ca MKIV in regulating different forms of fear memory,anxiety,and certain aspect of motor function.SONG ZiXiang CHEN Qi DING Qi ZHENG Fei LI ChengWei XU LePing WANG HongBing 2015Science China(Life Sciences)2015,58,1:6

8Developing high-efficiency base editors by combining optimized synergistic core components with new types of nuclear localization signal peptide显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)–CRISPR-associated protein(Cas) system has been widely used for genome editing. In this system, the cytosine base editor(CBE) and adenine base editor(ABE) allow generating precise and irreversible base mutations in a programmable manner and have been used in many different types of cells and organisms. However, their applications are limited by low editing efficiency at certain genomic target sites or at specific target cytosine(C) or adenine(A) residues. Using a strategy of combining optimized synergistic core components, we developed a new multiplex super-assembled ABE(sABE) in rice that showed higher base-editing efficiency than previously developed ABEs. We also designed a new type of nuclear localization signal(NLS) comprising a FLAG epitope tag with four copies of a codon-optimized NLS(F4NLS^(r2)) to generate another ABE named F4NLS-sABE. This new NLS increased editing efficiency or edited additional A at several target sites. A new multiplex super-assembled CBE(sCBE) and F4NLS^(r2) involved F4NLS-sCBE were also created using the same strategy. F4NLS-sCBE was proven to be much more efficient than sCBE in rice. 9Stereoelectroencephalography-guided radiofrequency thermocoagulation(SEEG-guided RF-TC) in patients with drug-resistant focal epilepsy显示文摘For some patients with drug-resistant focal epilepsy, we usually select conventional surgical resection, which has brought better outcomes. However, others are not eligible for a conventional open surgical resection of the epileptogenic zone because of the proximity of a functional area or the implication of a larger epileptogenic network. Initially, stereoelectroencephalography(SEEG) exploration was a method of electroencephalography recording that was used in the presurgical evaluation of epileptic patients with complex epilepsy. Later, intracerebral electrodes used for SEEG were applied to produce radio frequency thermocoagulation(RF-TC) in epileptic patients. SEEG-guided RF-TC has produced some promising results, especially in the last dacade. Now, it has become popular as a palliative treatment to reduce seizure frequency in patients with drug-resistant focal epilepsy. 11Anterior cervical surgery methods for central cord syndrome without radiographic spinal fracture-dislocation显示文摘This study was aimed to explore the anterior cervical surgery methods to treat central cord syndrome without radiographic spinal fracture-dislocation(CCSWORFD),retrospectively analyze the cases of CCSWORFD,and evaluate the curative effect of anterior cervical surgery methods for CCSWORFD.Twenty four cases of CCSWORFD(19 males and 5 females),all suffering from cervical hyperextension injury,between 45–68(average 59)years old,were operated on by anterior cervical surgery methods.Among these,18 cases had been followed up for 6–24(average 15)months;18 cases,who had anterior decompression and platefixation with titanium mesh bone grafting or iliac bone grafting achieved reliable effects based on the Japanese Orthopedics Association(JOA)evaluation(improved scores of cases with titanium mesh bone grafting,t=2.800,P<0.05;improved scores of cases with iliac bone grafting,t=3.270,P<0.05)and reliable reconstruction of cervical spine.The two groups obtained the same curative effect(t=0.470,P>0.05).Most of these cases had degeneration of cervical vertebra.The decompression which relieves the oppression to the spinal cord can help lessen edema of the spinal cord,and earlyfixation for stability of cervical vertebra is better for the recovery of spinal cord injury.Anterior operation with titanium mesh bone grafting or iliac bone grafting are both reliable curative methods for CCSWORFD,and titanium mesh bone grafting can avoid the trauma of the supplying graft.Mesh bone grafting can also shorten hospital stay.Chengwei JING Qin FU Xiaojun XU 2009Frontiers of Medicine2009,3,1:2
12Clay nanosheet-mediated delivery of recombinant plasmids expressing artificial miRNAs via leaf spray to prevent infection by plant DNA viruses显示文摘Whitefly-transmitted begomoviruses are economically important plant pathogens that cause severe problems in many crop plants,such as tomato,papaya,cotton,and tobacco.Tomato yellow leaf curl virus(TYLCV)is a typical monopartite begomovirus that has been extensively studied,but methods that can efficiently control begomoviruses are still scarce.In this study,we combined artificial microRNA(amiRNA)-mediated silencing technology and clay nanosheetmediated delivery by spraying and developed a method for efficiently preventing TYLCV infection in tomato plants.We designed three amiRNAs that target different regions of TYLCV to silence virus-produced transcripts.Three plant expression vectors expressing pre-amiRNAs were constructed,and recombinant plasmid DNAs(pDNAs)were loaded onto nontoxic and degradable layered double hydroxide(LDH)clay nanosheets.LDH nanosheets containing multiple pDNAs were sprayed onto plant leaves.We found that the designed amiRNAs were significantly accumulated in leaves 7 days after spraying,while the pDNAs were sustainably detected for 35 days after the spray,suggesting that the LDH nanosheets released pDNAs in a sustained manner,protected pDNAs from degradation and efficiently delivered pDNAs into plant cells.Importantly,when the LDH nanosheets coated with pDNAs were sprayed onto plants infected by TYLCV,both the disease severity and TYLCV viral concentration in sprayed plants were significantly decreased during the 35 days,while the levels of H_(2)O_(2) were significantly increased in those plants.Taken together,these results indicate that LDH nanosheets loaded with pDNAs expressing amiRNAs can be a sustainable and promising tool for begomovirus control.Qili Liu Yanpeng Li Kedong Xu Dongxiao Li Haiyan Hu Feng Zhou Puwen Song Yongang Yu Qichao Wei Qian Liu Weipeng Wang Ruifang Bu Haili Sun Xiaohui Wang Jianjun Hao Honglian Li Chengwei Li 2020Horticulture Research2020,7,1:1

16Species Composition and Diversity of Macrobenthos in the Intertidal Zone of Xiangshan Bay,China显示文摘Xiangshan bay is a narrow semi-closed bay and situated on the northwestern coast of the East China Sea. Over past decades, it has become to a major bay with intensive human activities, dense urbanized area, and poor water quality. The aim of this paper was to reveal the ecological status through the elucidation of the species composition, abundance, biomass and diversity of macrobenthos in this bay. Six intertidal sections were surveyed from January 2007 to November 2008 quarterly. Sections TG, HD and XH are located in the three inner bays, sections QJ and WS are located near the thermal power plants, and section XX is located at the outer part of Xiangshan Bay. Great variations in macrobenthos community were indentified, and the species composition of the community in the present study showed the dominance in the order of molluscs(bivalves and gastropods), crustaceans and others, and only few Polychaeta were recorded. Only three dominant species, Littorina brevicula, Ilyplax tansuiensis, and Cerithidea cingulata were collected in all the sections, and a total of 19 dominant species were recorded only in one section. Two-way ANOVA analyses of abundance indicated that there were significant differences among sections or seasons. Shannon-Wiener diversity index(H') had its maximum(2.45) in section QJ, and minimum(1.76) in section TG. Multiple irregular k-dominance plots clearly showed that the study area was polluted and the macrobenthos community was under stress. 20A new household ultrasonic cleaning method for pyrethroids in cabbage显示文摘Pyrethroid is widely used in developing countries and is a potential threat to human health.So putting forward an effective pyrethroid cleaning method is of vital importance.Clearance of fenpropathrin,cypermethrin and deltamethrin in cabbage by using ultrasonic treatment accompanied with different baking soda concentrations were investigated in this study.The results of response surface methodology showed that the maximum clearance rate was 70.61% for fenpropathrin under the optimal power,treated time,treated temperature and baking soda concentration,which were 260.02 W,10 min,20.15◦C and 0.014 g/mL,respectively.Similarly,the maximum removal efficiency was 73.72% for cypermethrin under 240 W,13.84 min,19.53℃ and 0.014 g/mL,and 92.15%for deltamethrin under 238.99 W,12.42 min,25℃ and 0.013 g/mL.The clearance rate of pyrethroid by this method was significant higher(P<0.05)than traditional cleaning methods(bathing and running water washing).The contents of the compositions such as vitamin C,total soluble sugar,protein and dietary fiber,texture and sensory evaluation of cabbage were not found obvious changes after the ultrasonic treatment combined with baking soda(P>0.05).Results showed that ultrasonic treatment combined with baking soda was an effective way to eliminate the fenpropathrin,cypermethrin and deltamethrin in cabbage without influencing cabbage quality.Chengwei Yu Xu Huang Yawei Fan Zeyuan Deng 2020Food Science and Human Wellness2020,9,3:1
