2Anterior cervical surgery methods for central cord syndrome without radiographic spinal fracture-dislocation显示文摘This study was aimed to explore the anterior cervical surgery methods to treat central cord syndrome without radiographic spinal fracture-dislocation(CCSWORFD),retrospectively analyze the cases of CCSWORFD,and evaluate the curative effect of anterior cervical surgery methods for CCSWORFD.Twenty four cases of CCSWORFD(19 males and 5 females),all suffering from cervical hyperextension injury,between 45–68(average 59)years old,were operated on by anterior cervical surgery methods.Among these,18 cases had been followed up for 6–24(average 15)months;18 cases,who had anterior decompression and platefixation with titanium mesh bone grafting or iliac bone grafting achieved reliable effects based on the Japanese Orthopedics Association(JOA)evaluation(improved scores of cases with titanium mesh bone grafting,t=2.800,P<0.05;improved scores of cases with iliac bone grafting,t=3.270,P<0.05)and reliable reconstruction of cervical spine.The two groups obtained the same curative effect(t=0.470,P>0.05).Most of these cases had degeneration of cervical vertebra.The decompression which relieves the oppression to the spinal cord can help lessen edema of the spinal cord,and earlyfixation for stability of cervical vertebra is better for the recovery of spinal cord injury.Anterior operation with titanium mesh bone grafting or iliac bone grafting are both reliable curative methods for CCSWORFD,and titanium mesh bone grafting can avoid the trauma of the supplying graft.Mesh bone grafting can also shorten hospital stay.Chengwei JING Qin FU Xiaojun XU 2009Frontiers of Medicine2009,3,1:2

5Increased CD133^+ cell infiltration in the rat brain following fluid percussion injury显示文摘The prominin-1/CD133 epitope is expressed in undifferentiated cells. Studies have reported that craniocerebral trauma in animal models of fluid percussion injury induces production of a specific stem cell subgroup. It has been hypothesized that fluid percussion injury induces CD133+ cell infiltration in the brain tissue. The present study established a traumatic brain injury model through fluid percussion injury. Immunohistochemical staining showed significantly increased CD133 antigen expression in the rat brain following injury. CD133+ cells were mainly distributed in hippocampal CA1-3 regions, as well as the dentate gyrus and hilus, of the lesioned hemisphere. Occasional cells were also detected in the cortex. In addition, reverse transcription-PCR revealed that no change in CD133 mRNA expression in injured brain tissue. 6Lockdown-induced Urban Aerosol Change over Changchun, China During COVID-19 Outbreak with Polarization LiDAR显示文摘Depending on various government policies,COVID-19(Corona Virus Disease-19) lockdowns have had diverse impacts on global aerosol concentrations.In 2022,Changchun a provincial capital city in Northeast China,suffered a severe COVID-19 outbreak and implemented a very strict lockdown that lasted for nearly two months.Using ground-based polarization Light Detection and Ranging(LiDAR),we detected real-time aerosol profile parameters(EC,extinction coefficient;DR,depolarization ratio;AOD,aerosol optical depth),as well as air-quality and meteorological indexes from 1 March to 30 April in 2021 and 2022 to quantify the effects of lockdown on aerosol concentrations.The period in 2022 was divided into three stages:pre-lockdown(1-10 March),strict lockdown(11 March to 10 April),and partial lockdown(11-30 April).The results showed that,during the strict lockdown period,compared with the pre-lockdown period,there were substantial reductions in aerosol parameters(EC and AOD),and this was consistent with the concentrations of the atmospheric pollutants PM_(2.5)(particulate matter with an aerodynamic diameter ≤2.5 μm) and PM_(2.5)(particulate matter with an aerodynamic diameter ≤10 μm),and the Oconcentration increased by 8.3%.During the strict lockdown,the values of EC within0-1 km and AOD decreased by 16.0% and 11.2%,respectively,as compared to the corresponding period in 2021.Lockdown reduced the conventional and organized emissions of air pollutants,and it clearly delayed the time of seasonal emissions from agricultural burning;however,it did not decrease the number of farmland fire points.Considering meteorological factors and eliminating the influence of wind-blown dust events,the results showed that reductions from conventional organized emission sources during the strict lockdown contributed to a 30% air-quality improvement and a 22% reduction in near-surface extinction(0-2 km).Aerosols produced by urban epidemic prevention and disinfection can also be identified using the EC.Regarding seasonal sources of agricultural straw burning,the concentrated burning induced by the epidemic led to the occurrence of heavy pollution from increased amounts of atmospheric aerosols,with a contribution rate of 62%.These results indicate that there is great potential to further improve air quality in the local area,and suggest that the comprehensive use of straw accompanied by reasonable planned burning is the best way to achieve this.CHEN Weiwei DUANMU Lingjian QIN Yang YANG Hongwu FU Jing LU Chengwei FENG Wei GUO Li 2022Chinese Geographical Science2022,32,5:0
7Discovery of a potent and highly selective inhibitor of SIRT6 against pancreatic cancer metastasis in vivo显示文摘Pancreatic cancer,one of the most aggressive malignancies,has no effective treatment due to the lack of targets and drugs related to tumour metastasis.SIRT6 can promote the migration of pancreatic cancer and could be a potential target for antimetastasis of pancreatic cancer.However,highly selective and potency SIRT6 inhibitor that can be used in vivo is yet to be discovered.Here,we developed a noveSIRT6 allosteric inhibitor,compound 11e,with maximal inhibitory potency and an IC_(50) value of 0.98±0.13μmol/L.Moreover,compound 11e exhibited significant selectivity against other histone deacetylases(HADC1-11 and SIRT1-3)at concentrations up to 100μmol/L.The allosteric site and the molecular mechanism of inhibition were extensively elucidated by cocrystal complex structure and dynamic structural analyses.Importantly,we confirmed the antimetastatic function of such inhibitors in four pancreatic cancer cell lines as well as in two mouse models of pancreatic cancer liver metastasis.To our knowledge,this is the first study to reveal the in vivo effects of SIRT6 inhibitors on liver metastatic pancreatic cancer.It not only provides a promising lead compound for subsequent inhibitor developmentargeting SIRT6 but also provides a potential approach to address the challenge of metastasis in pancreatic cancer.Xinyuan Xu Qian Zhang Xufeng Wang Jing Jin Chengwei Wu Li Feng Xiuyan Yang Mingzhu Zhao Yingyi Chen Shaoyong Lu Zhen Zheng Xiaobing Lan Yi Wang Yan Zheng Xuefeng Lu Qiufen Zhang Jian Zhang 2024Acta Pharmaceutica Sinica B2024,14,3:0
8Enabling high-efficiency ethanol oxidation on NiFe-LDH via deprotonation promotion and absorption inhibition显示文摘Nucleophile oxidation reaction(NOR), represented by ethanol oxidation reaction(EOR), is a promising pathway to replace oxygen evolution reaction(OER). EOR can effectively reduce the driving voltage of hydrogen production in direct water splitting. In this work, large current and high efficiency of EOR on a Ni, Fe layered double hydroxide(NiFe-LDH) catalyst were simultaneously achieved by a facile fluorination strategy. F in NiFe-LDH can reduce the activation energy of the dehydrogenation reaction, thus promoting the deprotonation process of NiFe-LDH to achieve a lower EOR onset potential. It also weakens the absorption of OH-and nucleophile electrooxidation products on the surface of NiFe-LDH at a higher potential, achieving a high current density and EOR selectivity, according to density functional theory calculations. Based on our experiment results, the optimized fluorinated NiFe-LDH catalyst achieves a low potential of 1.386 V to deliver a 10 mA cm^(-2)EOR. Moreover, the Faraday efficiency is greater than 95%, with a current density ranging from 10 to 250 mA cm^(-2). 9NDRG1 promotes endothelial dysfunction and hypoxia-induced pulmonary hypertension by targeting TAF15显示文摘Background:Pulmonary hypertension(PH)represents a threatening pathophysiologic state that can be induced by chronic hypoxia and is characterized by extensive vascular remodeling.However,the mechanism underlying hypoxia-induced vascular remodeling is not fully elucidated.Methods and Results:By using quantitative polymerase chain reactions,western blotting,and immunohistochemistry,we demon-strate that the expression of N-myc downstream regulated gene-1(NDRG1)is markedly increased in hypoxia-stimulated endothelial cells in a time-dependent manner as well as in human and rat endothelium lesions.To determine the role of NDRG1 in endothelial dysfunction,we performed loss-of-function studies using NDRG1 short hairpin RNAs and NDRG1 over-expression plasmids.In vitro,silencing NDRG1 attenuated proliferation,migration,and tube formation of human pulmonary artery endothelial cells(HPAECs)un-der hypoxia,while NDRG1 over-expression promoted these behaviors of HPAECs.Mechanistically,NDRG1 can directly interact with TATA-box binding protein associated factor 15(TAF15)and promote its nuclear localization.Knockdown of TAF15 abrogated the effect of NDRG1 on the proliferation,migration and tube formation capacity of HPAECs.Bioinformatics studies found that TAF15 was involved in regulating PI3K-Akt,p53,and hypoxia-inducible factor 1(HIF-1)signaling pathways,which have been proved to be PH-related pathways.In addition,vascular remodeling and right ventricular hypertrophy induced by hypoxia were markedly alleviated in NDRG1 knock-down rats compared with their wild-type littermates.Conclusions:Taken together,our results indicate that hypoxia-induced upregulation of NDRG1 contributes to endothelial dysfunction through targeting TAF15,which ultimately contributes to the development of hypoxia-induced PH.Chengwei Li Junzhu Lv Gulinuer Wumaier Yu Zhao Liang Dong Yuzhen Zeng Ning Zhu Xiujuan Zhang Jing Wang Jingwen Xia Shengqing Li 2023Precision Clinical Medicine2023,6,4:0
