| 2 | Sample maturation calculated using Raman spectroscopic parameters for solid organics: Methodology and geological applications显示文摘The shape and shift of Raman peak of solid organics prove to be capable of revealing atomic and molecular level vibration information of aromatic ring structure and its relationship with sample maturation. Raman 'D' peaks and 'G' peaks of a series of continuously maturated coal samples were measured, and the inter-peak intervals (G-D) and peak height ratios (Dh/Gh) were derived and correlated with the vitrinite reflectance (vRo%) of standard coal samples. As a result, two formulae were established by using the two Raman indices for calculation of Raman reflectance (RmcRo%), which is equivalent to vitrinite reflectance. The formula for calculating Raman reflectance indicative of organic maturation using Raman shift inter-peak interval (G-D) is RmcRo%=0.0537d(G-D)-11.21, which is mainly applicable to matured to highly matured carbonized samples of solid organics; The formula for calculating Raman reflectance indicative of organic maturation using Raman peak height ratio (Dh/Gh) is RmcRo%=1.1659h (Dh/Gh)+2.7588, which is mainly applicable to carbonized samples of solid organics that are over matured or going to be turned into granulated graphite. | 8 | Pyrolysis of oil at high temperatures:Gas potentials,chemical and carbon isotopic signatures显示文摘Although the gas cracked from oil has been believed to be one of the important sources in highly matured marine basins, there are still some debates on its resource potentials and chemical and isotopic compositions. In this study a Cambrian-sourced marine oil sample from the Silurian reservoir of well TZ62 in the central Tarim basin was pyrolyzed using sealed gold tubes with two different pyrolysis schemes: continuous pyrolysis in a closed system and stepwise semi-open pyrolysis. The results show that the maximum weight yield of C1-5 gases occurs at EasyRo=2.3% and the residual gas poten-tial after this maturity is only 43.4 mL/g, about 12% of the yield of 361 mL/g at EasyRo=2.3%. Combined with the results of kinetic modeling, the main stage of gas generation from oil cracking is believed within the EasyRo=1.6%―2.3%. The increase in the volume yield of C1-5 gases at EasyRo>2.3% in a closed system is mainly related to the re-cracking of previously formed C2-5 wet gases, not the direct cracking of oil. The stepwise pyrolysis experiments show that the gas from the cracking of residual oil at EasyRo>2.3% is characterized by very high dryness index (higher than 92%) and heavy methane carbon isotopes ranging from ?28.7‰ to ?26.7‰, which is quite different from the gases from the con-tinuous pyrolysis in a closed system. The kinetic modeling of methane carbon isotope fractionation shows that the carbon isotopes of methane within the main stage of gas generation (EasyRo<2.3%) are far lighter than the carbon isotopes of the precursor oils under a geological heating rate of 2 ℃ /Ma. | 10 | Origin of natural sulphur-bearing immiscible inclusions and H_2S in oolite gas reservoir, Eastern Sichuan显示文摘Based on results of microscopic observation and laser Raman analysis about fluid in- clusions, multiple special forms of immiscible inclusions that contain sulphur, liquid hydrocarbon, bi- tumen, etc. were discovered in samples collected from the H2S gas reservoir-containing carbonates in the Lower Triassic Feixianguan Formation in the Jinzhu-Luojia area, Kai County, Sichuan Province. Based on the lithology and burial history of the strata involved as well as measurement results of homogenization temperature of fluid inclusions, bitumen reflectivity, etc., it is concluded that the H2S in the gas reservoir resulted from the thermal reaction between hydrocarbons in reservoir and CaSO4 in the gypsum-bearing dolostone section at the high temperature (140℃―170℃) oil-cracked gas for- mation stage in Late Cretaceous. Thereafter, research on a great number of immiscible inclusions in the reservoir reveals that elemental sulphur resulted from oxidation of part of the earlier-formed H2S and further reaction between sulphates, hydrocarbons and H2S in geological fluids in H2S-bearing gas reservoir at a temperature of 86℃―89℃ and a pressure of 340×105Pa and during the regional uplift stage as characterized by temperature decrease and pressure decrease in Tertiary. | 11 | Removal of Cs+ from water and soil by ammonium-pillared montmorillonite/Fe3O4 composite显示文摘To remove cesium ions from water and soil, a novel adsorbent was synthesized by following a one-step co-precipitation method and using non-toxic raw materials. By combining ammonium-pillared montmorillonite(MMT) and magnetic nanoparticles(Fe_3O_4), an MMT/Fe_3O_4 composite was prepared and characterized. The adsorbent exhibited high selectivity of Cs^+ and could be rapidly separated from the mixed solution under an external magnetic field. Above all, the adsorbent had high removal efficiency in cesium-contaminated samples(water and soil) and also showed good recycling performance, indicating that the MMT/Fe_3O_4 composite could be widely applied to the remediation of cesium-contaminated environments. It was observed that the pH, solid/liquid ratio and initial concentration affected adsorption capacity. In the presence of coexisting ions, the adsorption capacity decreased in the order of Ca^(2+)> Mg^(2+)> K^+> Na^+, which is consistent with our theoretical prediction. The adsorption behavior of this new adsorbent could be expressed by the pseudo-second-order model and Freundlich isotherm. | 14 | STAT1 Antisense Oligonucleotides Attenuate the Proinflammatory Cytokine Release of Alveolar Macrophages in Bleomycin-Induced Fibrosis显示文摘To investigate the effect of signal transducers and activators of transcription 1 (STAT1) antisense oligonucleotides (ASON) on concentrations of TNF-α, IL-8, NO secreted by alveolar macrophages (AMs) in bleomycin-induced rat pulmonary fibrosis, five adult female Wistar rats were intratracheally instilled with bleomycin. After 7 days, the rats were killed by right ventricle of heart exsanguinations under ketamine anaesthesia and bronchoalveolar lavage (BAL) was performed to obtain AMs. AMs were divided into four groups, treated with STAT1 ASON, STAT1 sense oligonucleotides (SON), dexamethasone (DEX) and medium alone (control), respectively. AMs and media were collected after culture for 36 h. The mRNA and protein expressions of STAT1 and ICAM-1 in AMs were detected by RT-PCR and ELISA, respectively. The concentrations of TNF-α, IL-8, NO in cultured medium were detected.The STAT1 mRNA expression by AMs in the STAT1 ASON group was lower than those of AMs in the STAT1 SON group, the DEX group and the control group (p < 0.05). Moreover, the STAT1 mRNA expression by AMs in the DEX group was also lower than those of AMs in the STAT1 SON group and the control group (p < 0.05), but the STAT1 mRNA expression by AMs in the STAT1 SON group was not different from that of the control group (p >0.05). The protein expressions of STAT1 and ICAM-1 and the mRNA expression of ICAM-1 showed similar changes to the STAT1 mRNA expression by AMs. The concentrations of TNF-α, IL-8, NO in cultured medium from STAT1 ASON group were lower than those from STAT1 SON, DEX and the control groups (p < 0.05). | 15 | Aerosolized STAT1 Antisense Oligodeoxynucleotides Decrease the Concentrations of Inflammatory Mediators in Bronchoalveolar Lavage Fluid in Bleomycin-Induced Rat Pulmonary Fibrosis显示文摘It has been demonstrated that alveolar macrophages (AMs) play a key role in the pathogenesis of pulmonary fibrosis by releasing a variety of cytokines and inflammatory mediators. In addition, abnormal signal transducer and activator of transcription-1 (STAT1) activation in AMs may play a pivotal role in the process of alveolitis and pulmonary fibrosis. In this study, we transfected STAT1 antisense oligodeoxynucleotide (ASON) into rats by aerosolization, and then investigated the effect of STAT1 ASON on inflammatory mediators such as TGF-β, PDGF and TNF-α in bronchoalveolar lavage fluid (BALF) from rats with bleomycin (BLM)-induced rat pulmonary fibrosis. Our results showed that STAT1 ASON by aerosolization could enter into lung tissues and AMs. STAT1 ASON could inhibit mRNA and protein expressions of STAT1 and ICAM-1 in AMs of rat with pulmonary fibrosis, and had no toxic side effect on liver and kidney. Aerosolized STAT1 ASON could ameliorate the alveolitis through inhibiting the secretion of inflammatory mediators in BLM-induced rat pulmonary fibrosis. | 16 | High density methane inclusions in Puguang Gasfield:Discovery and a T-P genetic study显示文摘Based on measurement of homogenization temperature of inclusions and Raman spectral analysis, high density methane inclusions were discovered in the Triassic reservoirs of Puguang Gasfield. The methane inclusions show a homogenization temperature Th = -117.5― -118.1℃, a corresponding density of 0.3455―0.3477 g/cm3, and a Raman scatter peak v1 shift varying between 2911―2910 cm-1, which signifies a very high density of methane inclusions. The salt water inclusions paragenetic with methane inclusions show a homogenization temperature Th=170―180℃. Based on the composition of methane inclusions as determined by Raman spectra, PVTsim software was used to simulate the trapping pressure for high density methane inclusions in geologic history, and the trapping pressure was found to be as high as 153―160 MPa. Even though Puguang Gasfield is currently a gas pool of normal pressure, and the fluid pressure for the gas pool ranges between 56―65 MPa. However, data from this study indicates that remarkable overpressure may be generated at the stage of mass production of gas cracked from oils in Cretaceous, as high density methane inclusions constitute key evidence for overpressure in gas pool in geologic history. Meanwhile, discovery of small amounts of H2S, CO2 or heavy hydrocarbon in part of the high density methane inclusions indicates that the geochemical environment for trapping of inclusions may be related to formation of H2S. | 17 | Effect of Liuweidihuang pill and Jinkuishenqi pill on inhibition of spontaneous breast carcinoma growth in mice显示文摘OBJECTIVE: To investigate the preventing and treating action of Liuweidihuang pill(LP) and Jinkuishenqi pill(JP) on spontaneous breast carcinoma in mice.METHODS: A model of spontaneous breast carcinoma was derived from 11.5-month-old female Kunming breeding mice following the delivery of several litters. The mice were randomly divided into five groups: model control group(C),Liuweidihuang pill high-dose group(LH; 4.6 g·kg1·d1),Liuweidihuang pill low-dose group(LL;2.3 g·kg1·d﹣1),Jinkuishenqi pill high-dose group(JH; 4.6 g·kg﹣1·d1) and Jinkuishenqi pill low-dose group(JL;2.3 g·kg﹣1·d﹣1). Cancer tissue volume was measured by water immersion. Histopathology was analyzed by hematoxylin and eosin staining. Vascular endothelial growth factor(VEGF),extracellular signal-regulated kinase(ERK) and cyclin D1 protein expression in cancer tissue was assayed by western blotting.RESULTS: Compared with the control group,cancer tissue volume and weight were lower in the LP and JP groups,and survival time was longer. The expression of VEGF,ERK and Cyclin D1 were inhibited in the LP and JP groups(P < 0.05),and cell differentiation was increased. | 19 | The generation and accumulation of natural gas from Yinan 2 gas pool in Kuqa Depression显示文摘By using the methods of hydrocarbon generation kinetics and carbon isotope kinetics, combined with geological background of natural gas pool formation, the generation and accumulation of natural gas from Yinan 2 gas pool was studied in Kuqa Depression of the Tarim Basin. Natural gas of Yinan 2 gas pool is mainly derived from Middle and Lower Jurassic coal-bearing source rocks, and generally belongs to long time-accumulated gas. It is suggested that Yinan 2 gas is chiefly accumulated in the last 5 Ma, its Ro ranges from 1.25% to 1.95%, and the loss rate of natural gas is about 25%-30%. | 20 | Effect of Danhong injection on expressions of macrophage scavenger receptor 1 and sub-family A member 1 in human U937 cells显示文摘OBJECTIVE: To study the effects of Danhong injection (DHI) on expression of the macrophage scavenger receptor 1 (MSR1) and ATP-binding cassette, sub-family A member 1 (ABCA1) genes, which encode scavenger receptor-A I (SR-AI) and ATP-binding cassette transporter 1 (ABCA1), respectively, as a potential anti-atherosclerotic mechanism. METHODS:HumanU937cellswerestimulatedbyincubation with 100 nM phorbol 12-myristate 13-acetate (PMA) for 48 h.These stimulated, monocyte-like cells were then incubated for 24 h with 50 mg/L oxidized low-density lipoprotein (ox-LDL, to induce foam cell formation), together with a liver X receptor (LXR) agonist or with different DHI concentrations. MSR1 and ABCA1 mRNA levels were measured by fluorescence-based quantitative PCR. RESULTS: Compared with control cells (which received only ox-LDL), cells treated with both ox-LDL and 10 μmol/L LXR agonist showed lower MSR1 ex-pression (but this effect was not statistically significant, P>0.05) and higher ABCA1 expression (P< 0.01). Cells that received ox-LDL and 3 mL/L DHI possessed higher MSR1 mRNA levels than the controls, whereas cells treated with ox-LDL and higher DHI concentrations (10, 30 or 60 mL/L) showed lower MSR1 expression levels (but the differences observed between DHI concentration groups were not statistically significant, P>0.05). ABCA1 expression in cells treated with ox-LDL and 3, 10 or 30 mL/L DHI was higher than in the control cells, and increased with increasing DHI concentration (P<0.05). ABCA1 expression in cells treated with ox-LDL and the highest DHI concentration tested (60 mL/L) was not significantly different from that in the controls. ABCA1 mRNA levels in cells treated with ox-LDL and DHI were similar to, or lower than, those in cells treated with ox-LDL and the LXR agonist. CONCLUSION: DHI does not affect MSR1 mRNA levels in ox-LDL-treated U937 cells. However, at certain concentrations (10 and 30 mL/L), DHI significantly increases ABCA1 mRNA levels.
