| 1 | Xinfeng capsule improves pulmonary function in ankylosing spondylitis patients via NF-κB-iNOS-NO signaling pathway显示文摘OBJECTIVE: To study changes in the nuclear factor-κB p65(NF-κB p65)-inducible nitric oxide synthase(i NOS)-nitric oxide(NO) signaling pathway and the effects of Xinfeng capsules(XFC) in patients with ankylosing spondylitis(AS)METHODS: One hundred twenty patients with AS were randomly divided into an XFC group and a Salazopyrin group. Sixty health subjects were included as a normal control group. In the two treatment groups, pulmonary functional parameters,forced vital capacity(FVC), forced expiratory volume in 1 second(FEV1), maximal voluntary ventilation(MVV), peak expiratory flow(PEF), forced expiratory flow at 25% of forced vital capacity(FEF25),forced expiratory flow at 50% of forced vital capacity(FEF50), and forced expiratory flow at 75% of forced vital capacity(FEF75) were determined. Enzyme linked immunosorbent assays were used for detection of the serum oxidative stress indexes,NF-κB p65, i NOS, NO, reactive oxygen species(ROS), reactive nitrogen species(RNS), malondialdehyde(MDA), superoxide dismutase(SOD), catalase(CAT), total antioxidative capacity(TAOC) and interleukin-4(IL-4), IL-10, IL-1β, and tumor necrosis factor-α(TNF-α) contents. Westergren's method was used for determination of erythrocyte sedimentation rate(ESR). High-sensitivity C-reactive protein(Hs-CRP) was detected with a 7060 full-automatic biochemical analyzer(Hitachi, Japan).RESULTS: The clinical therapeutic effect in the XFC group was significantly superior to that in the Salazopyrin group(P<0.01). Compared with the normal control group, FEV1, MVV, PEF, FEF50, FEF75, SOD, CAT,TAOC, IL-4, IL-10 were significantly lower, and NF-κB p65, i NOS, NO, ROS, RNS, MDA, IL-1β, TNF-α, ESR,and Hs-CRP significantly higher in patients with AS(P<0.01 or P<0.05). Compared with before treatment, FEV1, MVV, PEF, FEF50, FEF75, SOD, CAT, TAOC,IL-4, and IL-10 were significantly increased, and NF-κB p65, i NOS, NO, ROS, RNS, MDA, IL-1β, TNF-α,ESR, CRP, visual analog scales(VAS), Bath ankylosing spondylitis disease active index, Bath ankylosing spondylitis functional index, and Bath ankylosing spondylitis global index significantly decreased in the two treatment groups after treatment(P<0.01 or P<0.05), with significant differences between the XFC and Salazopyrin groups(P<0.01 or P<0.05). Spearman correlation analysis indicated that FEV1, MVV, PEF, FEF50, and FEF75 were positively correlated with SOD, CAT, TAOC, IL-4, and IL-10, and were negatively correlated with NF-κB p65, i NOS,NO, ROS, RNS, MDA, IL-1β, TNF-α, ESR, and CRP.CONCLUSION: Patients with AS have local pathologic changes in the spinal cord and other joints.They also have decreased pulmonary function,which is negatively correlated with the NF-κB-i NOS-NO signaling pathway, oxidative indexes, and inflammatory factors. XFC improves rigidity and pain in spinal joints and other symptoms, laboratory indexes, and pulmonary function. | 2 | Study on BaTiO_3 films prepared by AC power microarc oxidation显示文摘BaTiO3 films were prepared by microarc oxidation (MAO) with an in-house built alternating current (AC) power supply in Ba(OH)2 solution in this study. The surface morphology, combinability with the substrate and phase composition of the films were investigated by XRD, SEM and TEM. The BaTiO3 films were annealed at a temperature range of 900 to 1200℃ and phase compositions were tested thereafter. The results showed that the BaTiO3 films were mainly composed of the primitive hexagonal phase with relatively small amount of tetragonal and amorphous phases. Moreover, the amount of amorphous phase decreased with the time of annealing. The transformation of BaTiO3 from hexagonal structure to tetragonal stucture became obvious until the annealing temperature reached 1200℃. The film consisted mainly of BaTiO3 tetragonal structure with ferroelectric property. Pure Pr metal was added into these composites. The effect of Pr addition on the microstructure evolution of Al-Si MMCs was investigated by SEM,TEM,and EDS. Pr addition is favorable to make uniform microstructures with the modified eutectic Si crystal. PrAlSi phase with high contents of Pr and Si is observed on the interface between the fiber and the matrix. | 5 | Viral MIPα homologous with human MIP-1α acts on HIV co-receptor CCR5显示文摘The function and usage of vMIPα encoded by K6 gene of herpesvirus 8 (HHV8) which has homology with human macrophage protein (MIP) have not been clearly known. In the present note the K6 gene of HHV8 was cloned and transfected into NIH3T3 cells and E. coli cells. Conditional media from the 3T3-transfected cells and K6 product vMIPa from E. coli. Cells were used to perform the experiments of ligand-receptor binding and cellular adhesion with peripheral blood macrophages. The conditional media and the purified vMIPa from E. coli could compete to bind to CCR5 located on macrophages from peripheral blood with I125-hMIP-1α chemokine of human. Cellular adhesion showed that the conditional media from transfected cells and the purified vMIPa did not induce the adhesion of macrophages from peripheral blood to ICAM-1. In conclusion, vMIPα encoded by K6 gene of HHV8 can bind to CCR5 of peripheral blood macrophage cells and does not induce their adhesion. | 9 | Robust hydrogen production from HCOOH over amino-modified KIT-6-confined Pd Ir alloy nanoparticles显示文摘Formic acid (FA), which can be produced via CO_(2) reduction and biomass conversion, has received extensive interest as a convenient and safe hydrogen carrier due to its wide range of sources, renewable, high hydrogen content (4.4 wt%), and convenient storage/transportation. Designing highly efficient catalysts is the main challenge to realize the hydrogen production from FA. In this work, well-dispersed and electron-rich PdIr alloy nanoparticles with a size of 1.8 nm are confined in amino-modified 3D mesoporous silica KIT-6 and applied as a highly efficient catalyst for robust hydrogen production from FA at ambient temperature. Small PdIr alloy nanoparticles confined by amino-modified KIT-6 (PdIr/KIT-6-NH_(2)) lead to better catalytic activity compared to that of Pd/KIT-6-NH_(2) and PdIr confined by bare KIT-6, achieving a high turnover frequency (TOF) value of 3533 h-1 at ambient temperature (303 K), 100% H_(2) selectivity and conversion toward the dehydrogenation of FA, which is comparable to the best heterogeneous catalysts ever reported. | 11 | Discovery of Novel Androgen Receptor Ligands by Structure-based Virtual Screening and Bioassays显示文摘Androgen receptor(AR) is a ligand-activated transcription factor that plays a pivotal role in the development and progression of many severe diseases such as prostate cancer, muscle atrophy, and osteoporosis. Binding of ligands to AR triggers the conformational changes in AR that may affect the recruitment of coactivators and downstream response of AR signaling pathway.Therefore, AR ligands have great potential to treat these diseases. In this study, we searched for novel AR ligands by performing a docking-based virtual screening(VS) on the basis of the crystal structure of the AR ligand binding domain(LBD) in complex with its agonist. A total of 58 structurally diverse compounds were selected and subjected to LBD affinity assay, with five of them(HBP1-3, HBP1-17, HBP1-38, HBP1-51, and HBP1-58) exhibiting strong binding to AR-LBD. The IC50 values of HBP1-51 and HBP1-58 are 3.96 m M and 4.92 m M, respectively, which are even lower than that of enzalutamide(Enz, IC50= 13.87 m M), a marketed second-generation AR antagonist. Further bioactivity assays suggest that HBP1-51 is an AR agonist, whereas HBP1-58 is an AR antagonist. In addition, molecular dynamics(MD) simulations and principal components analysis(PCA) were carried out to reveal the binding principle of the newlyidentified AR ligands toward AR. Our modeling results indicate that the conformational changes of helix 12 induced by the bindings of antagonist and agonist are visibly different. | 14 | Effects of trace ytterbium addition on microstructure,mechanical and thermal properties of hypoeutectic Al-5Ni alloy显示文摘Over the past decade,the cast aluminum alloys with excellent mechanical and conductivity properties have emerged as potential materials for thermal management.However,the traditional Al-Si based alloys are difficult to make significant breakthrough in conductivity performance.The hypoeutectic Al-5Ni alloy also possesses sound castability and is expected to be applied in thermal management applications.In this study,the effects of ytterbium(Yb element) at 0-0.5 wt% on the microstructures as well as the electrical/thermal conductivity and mechanical properties of the Al-5Ni alloy were systematically investigated.The experimental results indicate that the addition of Yb at a relatively low amount not only reduces the secondary dendrite arm spacing of the α-Al grains,but also modifies the morphology and distribution of eutectic boundary phase.Moreover,it is found that the dosage of Yb at 0.3 wt% in the Al-5Ni alloy can simultaneously improve the yield strength,ultimate tensile strength and electrical/thermal conductivity.The strengthening and toughening of the Al-5Ni alloy are mainly attributed to the decrease of secondary dendrite arm spacing and the improvement of eutectic phases.The transmission electron microscopy/selected area electron diffraction(TEM/SAED) analysis indicates that the ytterbium in Al-5Ni alloy will form Al_(3)Yb phase,which mainly agglomerates in the Al_(3)Ni phase region.This phase is helpful to decrease the solubility of impurity elements(e.g.,Fe and Si) in the α-Al matrix,which is beneficial to electrical/thermal conductivity.The value of this study lays foundation for manufacturing Al-Ni alloys with high thermal conductivity and acceptable mechanical properties. | Kang Wang Shaodong Hu Yanglin Zhong Shuoxun Jin Zirong Zhou Zhiyu Wang Jiongcai Chen Bingbing Wan Wenfang Li | 2022 | Journal of Rare Earths2022,40,8: | 2 |
