| 1 | Endoscopic characteristics in predicting prognosis of biopsy-diagnosed gastric low-grade intraepithelial neoplasia显示文摘Background: Endoscopic biopsy can underestimate gastric malignancies as low-grade intraepithelial neoplasia (LGIN). Definitively diagnosed LGIN would progress. This study aimed to evaluate predictive factors to identify malignancies misdiagnosed as LGIN by biopsy and LGIN at high risk of progression.Methods: The clinical records of patients diagnosed with gastric LGIN by endoscopic biopsy who underwent at least two endoscopies during the first year of follow-up between 2007 and 2017 were retrospectively collected. Three endoscopists reviewed photographs of the initial endoscopy, described lesion characteristics, and made endoscopic diagnoses. Logistic regression was used to analyze predictors to identify malignancies underestimated as LGIN. A receiver operating characteristic curve was used to evaluate the diagnostic accuracy of these predictors. Patient clinical outcomes of follow-up >1 year were collected. Kaplan-Meier estimates with log-rank tests and Cox proportional hazards regression were used to analyze predictors of progression.Results: Overall, 48 of 182 (26.4%) patients were proven to have malignancies. A single lesion, a large lesion size, and marked intestinal metaplasia (IM) were independent predictors of initially misdiagnosed malignancies. The area under the curve of these predictors was 0.871, with a sensitivity of 68.7% and specificity of 92.5%. Twelve of 98 patients (12.2%) progressed during the 33-month median follow-up period. A whitish appearance, irregular margins, marked IM, and histological diagnosis of LGIN more than twice within the first year were predictors for progression.Conclusions: Lesions diagnosed as LGIN by biopsy with marked IM and other predictors above should be prudently treated for high potential to be malignancies or progress. | 2 | Predicting the Thickness of an Excavation Damaged Zone around the Roadway Using the DA-RF Hybrid Model显示文摘After the excavation of the roadway,the original stress balance is destroyed,resulting in the redistribution of stress and the formation of an excavation damaged zone(EDZ)around the roadway.The thickness of EDZ is the key basis for roadway stability discrimination and support structure design,and it is of great engineering significance to accurately predict the thickness of EDZ.Considering the advantages of machine learning(ML)in dealing with high-dimensional,nonlinear problems,a hybrid prediction model based on the random forest(RF)algorithm is developed in this paper.The model used the dragonfly algorithm(DA)to optimize two hyperparameters in RF,namely mtry and ntree,and used mean absolute error(MAE),rootmean square error(RMSE),determination coefficient(R^(2)),and variance accounted for(VAF)to evaluatemodel prediction performance.A database containing 217 sets of data was collected,with embedding depth(ED),drift span(DS),surrounding rock mass strength(RMS),joint index(JI)as input variables,and the excavation damaged zone thickness(EDZT)as output variable.In addition,four classic models,back propagation neural network(BPNN),extreme learning machine(ELM),radial basis function network(RBF),and RF were compared with the DA-RF model.The results showed that the DARF mold had the best prediction performance(training set:MAE=0.1036,RMSE=0.1514,R^(2)=0.9577,VAF=94.2645;test set:MAE=0.1115,RMSE=0.1417,R^(2)=0.9423,VAF=94.0836).The results of the sensitivity analysis showed that the relative importance of each input variable was DS,ED,RMS,and JI from low to high. | Yuxin Chen Weixun Yong Chuanqi Li Jian Zhou | 2023 | Computer Modeling in Engineering & Sciences2023,,9: | 2 |

| 11 | Fine mapping and candidate gene analysis of qHD1b,a QTL that promotes flowering in common wild rice(Oryza rufipogon)by up-regulating Ehd1显示文摘Heading date(flowering time)determines the adaptability of cultivars to different environments.We report the fine mapping and candidate gene analysis of qHD1b,a quantitative trait locus(QTL)responsible for early flowering that was derived from common wild rice(O.rufipogon)under both short-day and longday conditions.The introgression line IL7391,which carried segments from common wild rice in a Zhonghui 8015(ZH8015)background,exhibited early heading compared to the background and was crossed with ZH8015 to generate BC_(5)F_(2:3) families for QTL analysis.This enabled the identification of two heading-date QTL,named qHD1b and qHD7,of which the first was selected for further research.High-resolution linkage analysis was performed in BC_(5)F_(4:5) and BC_(5)F_(6) populations,and the location of qHD1b was confined to a 112.7-kb interval containing 17 predicted genes.Five of these genes contained polymorphisms in the promoter or coding regions and were thus considered as candidates.Expression analysis revealed a positive association between LOC_Os01g11940 expression and early heading.This locus was annotated as OsFTL1,which encodes an ortholog of Arabidopsis Flowering Locus T and was the most likely candidate gene for qHD1b.Our study revealed that qHD1b acts as a floral activator that promotes flowering by up-regulating Ehd1,Hd3a,RFT1,OsMADS14,and OsMADS15 under both shortday and long-day conditions.Field experiments showed that qHD1b affected several yield-related agronomic traits including 1000-grain weight and grain length.qHD1b could be useful for marker-assisted selection and breeding of early-maturing cultivars. | Ling Liu Yingxin Zhang Zhengfu Yang Qinqin Yang Yue Zhang Peng Xu Jiaxin Li Anowerul Islam Liaqat Shah Xiaodeng Zhan Liyong Cao Shihua Cheng Weixun Wu | 2022 | The Crop Journal2022,10,4: | 0 |
| 12 | Identification of New Allele of FLOURY ENDOSPERM2 in White-Core Endosperm Mutant of Rice显示文摘Identification of regulatory genes from chalky/floury endosperm mutants is an important approach to understand the mechanism of starch biosynthesis to accelerate rice grain quality improvement.A mutant GM645,identified from 60Coγ-irradiation of an indica rice Guanglu’ai 4(GLA4),exhibited white-core endosperm and altered starch physicochemical properties.However,the causal gene responsible for the white-core endosperm in GM645 has not been identified.Here,we developed a recombined inbred line(RIL,F7)population derived from a cross between GM645 and a japonica rice Tainong 67(TN67)with translucent endosperm.Bulkedsegregant analysis combined with next generation sequencing revealed five single nucleotide polymorphisms(SNPs)in four candidate genes that were highly associated with the white-core endosperm.Among them,one base pair deletion in Os04g0645100 resulted in a frame shift mutation after the 983th amino acid(aa),and a premature stop codon occurred after the other 11 aa.Moreover,functional annotation revealed that Os04g0645100 was previously characterized as the FLOURY ENDOSPERM2(FLO2)gene.Full-length coding sequence from Os04g0645100 driven by the maize ubiquitin promoter was transformed into GM645,and seeds from these transgenic plants expressing Os04g0645100 were largely rescued to translucent,indicating that Os04g0645100 was responsible for the whitecore endosperm in GM645. | BAO Jinsong ZHANG Yu ZHAO Jiajia CHEN Yaling WU Weixun CAO Liyong XU Feifei | 2022 | Rice science2022,29,5: | 0 |
