3Deep Learning-Based Numerical Methods for High-Dimensional Parabolic Partial Differential Equations and Backward Stochastic Differential Equations显示文摘We study a new algorithm for solvingparabolic partial differential equations(PDEs)and backward stochastic differential equations(BSDEs)in high dimension,which is based on an analogy between the BSDE and reinforcement learning with the gradient of the solution playing the role of the policy function,and the loss function given by the error between the prescribed terminal condition and the solution of the BSDE.The policy function is then approximated by a neural network,as is done in deep reinforcement learning.Numerical results using TensorFlow illustrate the efficiency and accuracy of the studied algorithm for several 100-dimensional nonlinear PDEs from physics and finance such as the Allen–Cahn equation,the Hamilton–Jacobi–Bellman equation,and a nonlinear pricing model for financial derivatives.Weinan E Jiequn Han Arnulf Jentzen 2017Communications in Mathematics and Statistics2017,5,4:20

5A LuxR family transcriptional regulator AniF promotes the production of anisomycin and its derivatives in Streptomyces hygrospinosus var.beijingensis显示文摘The protein synthesis inhibitor anisomycin features a unique benzylpyrrolidine system and exhibits potent selective activity against pathogenic protozoa and fungi.It is one of the important effective components in Agricultural Antibiotic120,which has been widely used as naturally-originated agents for treatment of crop decay in China.The chemical synthesis of anisomycin has recently been reported,but the complex process with low productivity made the biosynthesis still to be a vital mainstay in efforts.The biosynthetic gene cluster(BGC)of anisomycin in Streptomyces hygrospinosus var.beijingensis has been identified in our previous work,while poor understanding of the regulatory mechanism limited the yield enhancement via regulation engineering of S.hygrospinosus var.beijingensis.In this study here,we characterized AniF as an indispensable LuxR family transcriptional regulator for the activation of anisomycin biosynthesis.The genetic manipulations of aniF and the real-time quantitative PCR(RT-qPCR)revealed that it positively regulated the transcription of the anisomycin BGC.Moreover,the overexpression of aniF contributed to the improvement of the production of anisomycin and its derivatives.Dissection of the mechanism underlying the function of AniF revealed that it directly activated the transcription of the genes aniR-G involved in anisomycin biosynthesis.Especially,one AniF-binding site in the promoter region of aniR was identified by DNase I footprinting assay and an inverted repeat sequence(5′-GGGC-3′)composed of two 4-nt half sites in the protected region was found.Taken together,our systematic study confirmed the positive regulatory role of AniF and might facilitate the future construction of engineering strains with high productivity of anisomycin and its derivatives.Jufang Shen Lingxin Kong Yan Li Xiaoqing Zheng Qing Wang Weinan Yang Zixin Deng Delin You 2019Synthetic and Systems Biotechnology2019,4,1:10
6The localization of type 2 diabetes susceptibility gene loci in northern Chinese Han families显示文摘We conducted a genome-wide scan, in which 358 well distributed fluorescent dye-labeled microsatellite marker sets were applied in 32 Chinese Han type 2 diabetes families from Northern China to search for the susceptibility gene loci. The data collected from screening all the chromosomes of genome were genotyped by using genescan and genotyping software, then, parametric and non-parametric multipoint test, and affected sib-pair analysis as well, were used to analyze the data. We identified some susceptibility gene loci residing in chromosomes 1,12,18,20, respectively, or precisely, located around D1S214, D1S207, D1S218, D1S235, D12S336, D18S61 and D20S118. 8Distributed Adaptive Fault-Tolerant Output Regulation of Heterogeneous Multi-Agent Systems With Coupling Uncertainties and Actuator Faults显示文摘In this paper, we consider the distributed adaptive fault-tolerant output regulation problem for heterogeneous multiagent systems with matched system uncertainties and mismatched coupling uncertainties among subsystems under the influence of actuator faults. First, distributed finite-time observers are proposed for all subsystems to observe the state of the exosystem. Then, a novel fault-tolerant controller is designed to compensate for the influence of matched system uncertainties and actuator faults. By using the linear matrix inequality technique, a sufficient condition is provided to guarantee the solvability of the considered problem in the presence of mismatched coupling uncertainties. 9The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 13High thermal stability Cu_(2)O@OZrP micro-nano hybrids for melt-spun excellent antibacterial activity polyester fibers显示文摘Public safety incidents caused by bacterial infections have attracted widespread attention towards antibacterial textiles(fibers,fabrics,etc.).Nevertheless,it is still challenging to efficiently load inorganic nano-antibacterial materials in polymer fibers.In this work,zirconium phosphate(ZrP,layered micro-nano materials)was utilized as a micro-nano carrier.The octadecyl triphenyl phosphonium bromide(OTP)was intercalated between the ZrP sheets by the ion exchange method to improve the carrier-polymer compatibility and the antibacterial performance.Through in-situ chemical reduction,the ultra-small nano-sized cuprous oxide(Cu_(2)O<5 nm)was loaded on the outer surface of ZrP to realize the uniform and stable dispersion of the Cu_(2)O on the carrier and improve the antibacterial performance.The ZrP nanosheets loaded with Cu_(2)O and OTP(Cu_(2)O@OZrP)had excellent antibacterial properties,and the antibacterial rate against E.coli,S.aureus and C.albicans was more than 99%.The intercalation amount of OTP in Cu_(2)O@OZrP can reach 16%,and the thermal stability was excellent and a significant increase in the Zeta potential.Indeed,the decomposition temperature was greater than 350℃,which was suitable for high-temperature melt processing of polymers.Consequently,we prepared PET/Cu_(2)O@OZrP fibers using polyethylene glycol terephthalate(PET),which accounts for 70%of the total chemical fibers,as the fiber matrix.PET/Cu_(2)O@OZrP fibers exhibited excellent mechanical property and antibacterial performance when the content of Cu_(2)O@OZrP was only 0.2%.The antibacterial rate against five types of bacteria including super bacteria(MRSA,VRE)was more than 99%.Jialiang Zhou Yaping Wang Weinan Pan Hengxue Xiang Peng Li Zhe Zhou Meifang Zhu 2021Journal of Materials Science & Technology2021,,22:4

15Characterization of the positive SARP family regulator PieR for improving piericidin A1 production in Streptomyces piomogeues var.Hangzhouwanensis显示文摘Piericidin A1,a member ofɑ-pyridone antibiotic,exhibits various biological activities such as antimicrobial,antifungal,and antitumor properties and possesses potent respiration-inhibitory activity against insects due to its competitive binding capacity to mitochondrial complex I.The biosynthetic pathway of piericidin A1 has been reported in Streptomyces piomogeues var.Hangzhouwanensis,while the regulatory mechanism remains poorly understood.In this study,a Streptomyces antibiotic regulatory protein(SARP)family transcriptional regulator PieR was characterized.Genetic disruption and complementation manipulations revealed that PieR positively regulated the production of piericidin A1.Moreover,the overexpression of pieR contributed to the improvement of piericidin A1 productivity.The real-time quantitative PCR(RT-qPCR)was carried out and the data showed that pieR stimulated the transcription of all the biosynthesis-related genes for piericidin A1.In order to explore the regulatory mechanism,electrophoresis mobility shift assays(EMSA)and DNase I footprinting experiments have been conducted.A protected region covering 50 nucleotides within the upstream region of pieR was identified and two 5-nt direct repeat sequences(5′-CCGGA-3′)in the protected region were found.These findings,taken together,set stage for transcriptional control engineering in the view of optimizing piericidin A1 production and thus provide a viable potent route for the construction of strains with high productivity.Yan Li Lingxin Kong Jufang Shen Qing Wang Qian Liu Weinan Yang Zixin Deng Delin You 2019Synthetic and Systems Biotechnology2019,4,1:4

17Spatial Structure,Hierarchy and Formation Mechanisms of Scientific Collaboration Networks:Evidence of the Belt and Road Regions显示文摘Scientific collaboration has become an important part of the people-to-people exchanges in the Belt and Road initiative,and remarkable progress has been made since 2013.Taking the 65 countries along the Belt and Road(BRI countries)as the research areas and using collaborated Web of Science(WOS)core collection papers to construct an international scientific collaboration matrix,the paper explores the spatial structure,hierarchy and formation mechanisms of scientific collaboration networks of 65 countries along the Belt and Road.The results show that:1)Beyond the Belt and Road regions(BRI regions),Central&Eastern Europe,China and West Asia&North Africa have formed a situation in which they all have the most external links with other countries beyond BRI regions.China has the dominant role over other BRI countries in generating scientific links.The overall spatial structure has changed to a skeleton structure consisting of many dense regions,such as Europe,North America,East Asia and Oceania.2)Within the Belt and Road regions,Central&Eastern Europe has become the largest collaboration partner with other sub-regions in BRI countries.The spatial structure of scientific collaboration networks has transformed from the‘dual core’composed of China and the Central&Eastern Europe region,to the‘multi-polarization’composed of‘one zone and multi-points’.3)The hierarchical structure of scientific collaboration networks presents a typical‘core-periphery’structure,and changes from‘single core’to‘double cores’.4)Among the formation mechanisms of scientific collaboration networks,scientific research strength and social proximity play the most important roles,while geographical distance gradually weakens the hindrance to scientific collaboration.GU Weinan LIU Hui 2020Chinese Geographical Science2020,30,6:3
18Different responses of soil respiration and its components to nitrogen and phosphorus addition in a subtropical secondary forest显示文摘Background:Nitrogen(N)and phosphorus(P)deposition have largely affected soil respiration(Rs)in forest ecosystems.However,few studies have explored how N and P individually or in combination to influence Rs and its components(autotrophic respiration,Ra;heterotrophic respiration,Rh),especially in highly P-limited subtropical forests.To address this question,we conducted a field manipulation experiment with N and/or P addition in a 50-year-old subtropical secondary forest.Results:We found that N addition on average reduced Rs,Ra,and Rh by 15.2%,15%,and 11.7%,respectively during 2-year field study.P addition had an inconsistent effect on Ra,with Ra increasing by 50.5%in the first year but reducing by 26.6%in the second year.Moreover,P addition on average decreased Rh by 8.9%–30.9%and Rs by 6.7%–15.6%across 2 years.In contrast,N and P co-addition on average increased Rs,Ra,and Rh by 1.9%,7.9%,and 2.1%during the experimental period.Though Rs and Rh were significantly correlated with soil temperature,their temperature sensitivities were not significantly changed by fertilization.Ra was predominantly regulated by soil nitrogen availability(NH4+and NO3−),soil dissolved organic carbon(DOC),and enzyme activities,while the variation in Rh was mainly attributable to changes in soil microbial community composition and soilβ-D-Cellubiosidase(CB)andβ-Xylosidase(XYL)activities.Conclusion:Our findings highlight the contrasting responses of Rs and its components to N or P addition against N and P co-addition,which should be differentially considered in biogeochemical models in order to improve prediction of forest carbon dynamics in the context of N and P enrichment in terrestrial ecosystems.Junjun Zhang Yong Li Jinsong Wang Weinan Chen Dashuan Tian Shuli Niu 2021Forest Ecosystems2021,8,3:3

20CypA Regulates AIP4-Mediated M1 Ubiquitination of Influenza A Virus显示文摘Cyclophilin A (CypA) is a peptidyl-prolyl cis/trans isomerase that interacts with the matrix protein (M1) of influenza A virus (IAV) and restricts virus replication by regulating the ubiquitin–proteasome-mediated degradation of M1. However,the mechanism by which CypA regulates M1 ubiquitination remains unknown. In this study, we reported that E3 ubiquitin ligase AIP4 promoted K48-linked ubiquitination of M1 at K102 and K104, and accelerated ubiquitin–proteasome-mediated degradation of M1. The recombinant IAV with mutant M1 (K102 R/K104 R) could not be rescued, suggesting that the ubiquitination of M1 at K102/K104 was essential for IAV replication. Furthermore, CypA inhibited AIP4-mediated M1 ubiquitination by impairing the interaction between AIP4 and M1.
