1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 3Endothelial CDS2 deficiency causes VEGFA-mediated vascular regression and tumor inhibition显示文摘The response of endothelial cells to signaling stimulation is critical for vascular morphogenesis,homeostasis and function.Vascular endothelial growth factor-a(VEGFA)has been commonly recognized as a pro-angiogenic factor in vertebrate developmental,physiological and pathological conditions for decades.Here we report a novel finding that genetic ablation of CDP-diacylglycerol synthetase-2(CDS2),a metabolic enzyme that controls phosphoinositide recycling,switches the output of VEGFA signaling from promoting angiogenesis to unexpectedly inducing vessel regression.Live imaging analysis uncovered the presence of reverse migration of the angiogenic endothelium in cds2 mutant zebrafish upon VEGFA stimulation,and endothelium regression also occurred in postnatal retina and implanted tumor models in mice.In tumor models,CDS2 deficiency enhanced the level of tumorsecreted VEGFA,which in-turn trapped tumors into a VEGFA-induced vessel regression situation,leading to suppression of tumor growth.Mechanistically,VEGFA stimulation reduced phosphatidylinositol(4,5)-bisphosphate(PIP2)availability in the absence of CDS2-controlled-phosphoinositide metabolism,subsequently causing phosphatidylinositol(3,4,5)-triphosphate(PIP3)deficiency and F0X01 activation to trigger regression of CDS2-null endothelium.Thus,our data indicate that the effect of VEGFA on vasculature is context-dependent and can be converted from angiogenesis to vascular regression.Wencao Zhao Le Cao Hanru Ying Wenjuan Zhang Dantong Li Xiaolong Ziiu Wenzhi Xue Shuang Wu Mengye Cao Cong Fu Haonan Qi Yimei Hao Yun-Chi Tang Jun Qin Tao PZhong Xiaoxi Lin Luyang Yu Xuri Li Lin Li Dianqing Wu Weijun Pan 2019Cell Research2019,29,11:9
4Acrylamide exposure impairs blood-cerebrospinal fluid barrier function显示文摘Previous studies show that chronic acrylamide exposure leads to central and peripheral neuropathy. However, the underlying mechanisms remained unclear. In this study, we examined the permeability of the blood-cerebrospinal fluid barrier, and its ability to secrete transthyretin and transport leptin of rats exposed to acrylamide for 7, 14, 21 or 28 days. Transthyretin levels in cerebrospinal fluid began to decline on day 7 after acrylamide exposure. The sodium fluorescein level in cerebrospinal fluid was increased on day 14 after exposure. Evans blue concentration in cerebrospinal fluid was increased and the cerebrospinal fluid/serum leptin ratio was decreased on days 21 and 28 after exposure. In comparison, the cerebrospinal fluid/serum albumin ratio was increased on day 28 after exposure. Our findings show that acrylamide exposure damages the blood-cerebrospinal fluid barrier and impairs secretory and transport functions. 5Atmospheric fine particles in a typical coastal port of Yangtze River Delta显示文摘In recent decades, coastal ports have experienced rapid development and become an important economic and ecological hub in China. Atmospheric particle is a research hotspot in atmospheric environmental sciences in inland regions. However, few studies on the atmospheric particle were conducted in coastal port areas in China, which indeed suffers atmospheric particle pollution. Lack of the physicochemical characteristics of fine particles serves as an obstacle toward the accurate control for air pollution in the coastal port area in China. Here, a field observation was conducted in an important coastal port city in Yangtze River Delta from March 6 to March 19, 2019. The average PM2.5 concentration was 63.7 ±27.8 μg/m^3 and NO3^-, SO4^2-, NH4^+, and organic matter accounted for ?60% of PM 2.5. Fe was the most abundant trace metal element and V as the ship emission indicator was detected. Transmission electron microscopy images showed that SK-rich, soot, Fe, SK-soot and SK-Fe were the major individual particles in the coastal port. V and soluble Fe were detected in sulfate coating of SK-Fe particles. We found that anthropogenic emissions, marine sea salt, and secondary atmosphere process were the major sources of fine particles. Backward trajectory analysis indicated that the dominant air masses were marine air mass, inland air mass from northern Zhejiang and inland-marine mixed air mass from Shandong and Shanghai during the sampling period. 8A study of peroxyacetyl nitrate at a rural site in Beijing based on continuous observations from 2015 to 2019 and the WRFChem model显示文摘Peroxyacetyl nitrate(PA N)is one of the most important photochemical pollutants and has aroused much concern in China in recent decades.However,few studies described the long-term variations in PAN in China.In this study,we continuously monitored the PAN,03 and NOx concentrations at a regional background site near Beijing from August 2015 to February 2019.Based on the observed concentrations and climate data,w e analyzed the seasonal PAN variations.The results revealed that the monthly mean PAN concentration ranged from 0.33-2.41 ppb,with an average value of 0.94 ppb.The PAN concentration exhibited a distinct seasonal variation,with high values in spring and low values in winter.After analyzing the corresponding meteorological data,we found that stronger ultraviolet(UV)radiation,a relatively longer lifetime and a higher background PAN concentration contributed to the high PAN concentrations in spring.In addition,with the utilization of the WRF-Chem(Weather Research and Forecasting with Chemistry)model,the cause of the extremely high PAN concentration in spring 2018 wa s determined.The model results demonstrated that an anomalously low pressure and the southwesterly winds in northern China might be the main causes of the increased PAN concentration in Beijing and its surrounding area in spring 2018.Yulu Qiu Zhiqiang Ma Weili Lin Weijun Quan Weiwei Pu Yingruo Li Liyan Zhou Qingfeng Shi 2020Frontiers of Environmental Science & Engineering2020,14,4:3

11CO flux of soil respiration in natural recovering karst abandoned farmland in Southwest China显示文摘To estimate carbon sequestration potential in the karst area,soil respiration in a natural recovering karst abandoned farmland in Shawan,Puding,Guizhou,southwest China was continuously and automatically monitored for more than two years.The results show that the CO2flux of soil respiration(2.63±1.89 lmol m^-2s-^1)is higher in the karst area than in non-karst areas under similar conditions but that regional value(1.32 lmol m-2s-1)is lower because of larger rock fragment coverage(~50%).A the same time,the temperature sensitivity of soil respiration(Q10)in this study area is significantly higher than that of non-karst areas under similar conditions.Soil respiration has an obvious temporal variation,which is reflected in a significant exponential relationship between soil respiration and soil temperature,but the relationship between soil respiration and soil moisture is very complex.Especially soil respiration has an obvious spatial variation,which is likely affected by different diffusion or water-rock reaction processes.Yanwei Wang Weijun Luo Guangneng Zeng Hanling Yang Meifang Wang Yina Lyu Anyun Cheng Lin Zhang Xianli Cai Jia Chen Shijie Wang 2020Acta Geochimica2020,39,4:2
12Two Ultraviolet Radiation Datasets that Cover China显示文摘Ultraviolet(UV) radiation has significant effects on ecosystems, environments, and human health, as well as atmospheric processes and climate change. Two ultraviolet radiation datasets are described in this paper. One contains hourly observations of UV radiation measured at 40 Chinese Ecosystem Research Network stations from 2005 to 2015. CUV3 broadband radiometers were used to observe the UV radiation, with an accuracy of 5%, which meets the World Meteorology Organization's measurement standards. The extremum method was used to control the quality of the measured datasets. The other dataset contains daily cumulative UV radiation estimates that were calculated using an all-sky estimation model combined with a hybrid model. The reconstructed daily UV radiation data span from 1961 to 2014. The mean absolute bias error and root-mean-square error are smaller than 30% at most stations, and most of the mean bias error values are negative, which indicates underestimation of the UV radiation intensity. These datasets can improve our basic knowledge of the spatial and temporal variations in UV radiation. 13A numerical investigation of the acoustic mode waves in a deviated borehole penetrating a transversely isotropic formation显示文摘A 2.5-dimensional method in frequency wave-number domain is developed to investigate the mode waves in a deviated borehole penetrating a transversely isotropic formation. The phase velocity dispersion characteristics of the fast and slow flexural mode waves excited by a dipole source are computed accurately at various deviation angles for both hard and soft formations. The sensitivities of the flexural mode waves to all elastic constants in a transversely isotropic formation are calculated. Numerical results show that, for a soft formation, the fast flexural mode wave is dominated by c66 at high deviation angles and low frequencies, while the slow flexural mode wave is dominated by c44 at the same conditions. 15Evaluation of abundant hydrocarbon-generation depressions in the deepwater area of Qiongdongnan Basin,South China Sea显示文摘It has been confirmed that the key source rocks of Qiongdongnan Basin are associated with the Yacheng Formation, which was deposited in a transitional marine-continental environment. Because the distribution and evolution patterns of the source rocks in the major depressions are different, it is important to determine the most abundant hydrocarbon-generation depressions in terms of exploration effectiveness. Based on an analysis of organic matter characteristics of the source rocks, in combination with drilling data and seismic data, this paper establishes a model to evaluate the hydrocarbon-generation depressions in the deepwater area of Qiongdongnan Basin. First of all, by using the method of seismic-facies model analysis, the distribution of sedimentary facies was determined. Then, the sedimentary facies were correlated with the organic facies, and the distribution of organic facies was predicted. Meanwhile, the thickness of source rocks for all the depressions was calculated on the basis of a quantitative analysis of seismic velocity and lithology. The relationship between mudstone porosity and vitrinite reflectance(Ro) was used to predict the maturity of source rocks. Second, using the parameters such as thickness and maturity of source rocks, the quantity and intensity of gas generation for Yacheng and Lingshui Formations were calculated. Finally, in combination with the identified hydrocarbon resources, the quantity and intensity of gas generation were used as a guide to establish an evaluation standard for hydrocarbon-generation depressions, which was optimized for the main depressions in the Central Depression Belt. It is proposed that Lingshui, Ledong, Baodao and Changchang Depressions are the most abundant hydrocarbon depressions, whilst Songnan and Beijiao Depressions are rich hydrocarbon depressions. 17Mechanobiologically optimized Ti-35Nb-2Ta-3Zr improves load transduction and enhances bone remodeling in tilted dental implant therapy显示文摘The tilted implant with immediate function is increasingly used in clinical dental therapy for edentulous and partially edentulous patients with excessive bone resorption and the anatomic limitations in the alveolar ridge.However,peri-implant cervical bone loss can be caused by the stress shielding effect.Herein,inspired by the concept of“materiobiology”,the mechanical characteristics of materials were considered along with bone biology for tilted implant design.In this study,a novel Ti-35Nb-2Ta-3Zr alloy(TNTZ)implant with low elastic modulus,high strength and favorable biocompatibility was developed.Then the human alveolar bone environment was mimicked in goat and finite element(FE)models to investigate the mechanical property and the related peri-implant bone remodeling of TNTZ compared to commonly used Ti-6Al-4V(TC4)in tilted implantation under loading condition.Next,a layer-by-layer quantitative correlation of the FE and X-ray Microscopy(XRM)analysis suggested that the TNTZ implant present better mechanobiological characteristics including improved load transduction and increased bone area in the tilted implantation model compared to TC4 implant,especially in the upper 1/3 region of peri-implant bone that is“lower stress”.Finally,combining the static and dynamic parameters of bone,it was further verified that TNTZ enhanced bone remodeling in“lower stress”upper 1/3 region.This study demonstrates that TNTZ is a mechanobiological optimized tilted implant material that enhances load transduction and bone remodeling.Chuanyuan Mao Weijun Yu Min Jin Yingchen Wang Xiaoqing Shang Lu Lin Xiaoqin Zeng Liqiang Wang Eryi Lu 2022Bioactive Materials2022,7,10:1
18Identification of weathered troughs in granites for tunneling at Daya Bay reactor-neutrino experiment site显示文摘Weathered troughs are frequently encountered in granites.They can cause problems to tunneling in the rocks and have to be properly addressed.The structures or spatial shapes of weathered troughs were seldom clarified in the past.In this paper,four weathered troughs are identified by means of geophysical exploration,core drilling,logging,and in-situ stress measurement at Daya Bay reactor-neutrino experiment site in Shenzhen,China.The weathered troughs are exposed on the ground or partially covered by grass and soils,which will threaten the safety of horizontal tunneling at a shallow depth.High electrical resistivity(HER) method is adopted for its feasibility and practicality,in combination with field geological observation,ultrasonic televiewer in boreholes and in-situ stress measurement.By comparing the HER values of completely decomposed to fresh rocks,it is indicated that the HER values of weathered troughs in natural state are 0.4-100 ·m,and the in-situ stress is abnormally lower than those at upper and lower layers.Field investigations show that the depths of the four weathered troughs are 30-182 m,with bottom elevation over 10 m.The volume of each weathered trough is mostly over 1×10 6 m 3 in inverted conic form.The weathered troughs often occur in various kinds of landforms,such as ridges,gullies or gently dipping dish-like depression areas.Faults and boundaries of different granitic plutons as well as joints govern the formation,locations and strikes of these troughs under development.Yanjun Shang Yongyue Shi Weijun Jin Daming Lin Fengbo Wu Wei Zhang 2012Journal of Rock Mechanics and Geotechnical Engineering2012,4,2:1
