| 1 | Dynamic analysis of gene expression and determination of chemicals in agarwood in Aquilaria sinensis显示文摘Agarwood is the resinous heartwood of Aquilaria species.However,low yields and high costs of existing stimulation methods have led to the need for new techniques to produce agarwood rapidly and effectively.We developed a biological agarwood-inducing technique(Agar-Bit)that produces high yields and quality within 6 months.To better understand agarwood formation by Agar-Bit,dynamic gene expressions of key synthetases pathways of sesquiterpenes and chalcone-related enzymes at different times were determined after both Agar-Bit and the traditional burning chisel drilling(BCD)stimulation on Aquilaria sinensis trees.The qRT-PCR results show that some characteristic synthase genes were expressed at greatly different levels and times compared with the controls.For the Agar-Bit technology,main changes were after the 3rd or 5th month,while BCD expression clearly changed at the 5th month.Essential oils and total chromone contents were simultaneously determined.In the Agar-Bit group,both were higher and similar to natural levels.The Agar-Bit methodology is a new option for producing agarwood as demonstrated by genetic and chemical aspects.The differences in gene expression within 6 months for both groups indicates that the mechanisms of the two methods are different.These findings provide information on genetic variation during the process of agarwood formation. | Zeqing Wu Wanzhen Liu Jing Li Liangwen Yu Li Lin | 2020 | Journal of Forestry Research2020,31,5: | 3 |
| 2 | One-pot/three-step synthesis of zinc-blende CdSe/CdS core/shell nanocrystals with thick shells显示文摘A one-pot/three-step synthetic scheme was developed for phase-pure epitaxy of CdS shells on zinc-blende CdSe nanocrystals to yield shells with up to sixteen monolayers. The key parameters for the epitaxy were identified, including the core nanocrystal concentration, solvent type/composition, quality of the core nanocrystals, epitaxial growth temperature, type/concentration of ligands, and composition of the precursors. Most of these key parameters were not influential when the synthetic goal was thin-shell CdSe/CdS core/shell nanocrystals. | 8 | One-step synthesis of site-specific antibody-drug conjugates by reprograming IgG glycoengineering with LacNAc-based substrates显示文摘Glycosite-specific antibody-drug conjugatess(gsADCs), harnessing Asn297 N-glycan of IgG Fc as the conjugation site for drug payloads, usually require multi-step glycoengineering with two or more enzymes, which limits the substrate diversification and complicates the preparation process.Herein, we report a series of novel disaccharide-based substrates, which reprogram the IgG glycoengineering to one-step synthesis of gsADCs, catalyzed by an endo-N-acetylglucosaminidase(ENGase) of Endo-S2. IgG glycoengineering via ENGases usually has two steps: deglycosylation by wild-type(WT) ENGases and transglycosylation by mutated ENGases. But in the current method, we have found that disaccharide LacNAc oxazoline can be efficiently assembled onto IgG by WT Endo-S2 without hydrolysis of the product, which enables the one-step glycoengineering directly from native antibodies.Further studies on substrate specificity revealed that this approach has excellent tolerance on various modification of 6-Gal motif of LacNAc. Within 1 h, one-step synthesis of gsADC was achieved using the LacNAc-toxin substrates including structures free of bioorthogonal groups. These gsADCs demonstrated good homogeneity, buffer stability, in vitro and in vivo anti-tumor activity.
