| 4 | Analysis of Transgenic Tobacco with Overexpression of Arabidopsis WUSCHEL Gene显示文摘The Arabidopsis WUSCIHIEL (WUS) gene plays a key role in the specification of the stem cellsin the shoot apical meristem (SAM). A cDNA of WUShas been amplified with the RT-PCR approach fromArabidopsis. The plant overexpression vector was constructed. It was driven by a dual enhanced CaMV35Spromoter. The construct was transformed into tobacco (Nicotiana tabacum L.) via Agrobacterium mediation.Dramatic phenotypic changes appeared in the WUS overexpression transgenic plants. Aberrant celldivisions and ectopic organogenesis could be found in almost every aerial parts of the transgenic tobaccoexcept the meristems and the inner two floral whorls. The data showed a highly conserved function of WUSin tobacco, and suggested that WUS is involved in organogenesis. The leaves were malformed, whichstrongly matched those only described previously for plants grown in the presence of polar auxin transportinhibitors. It suggested a possible function of WUS in leaf development. | 5 | SEM analysis of the changes of carbon layer structure of mesocarbon microbeads heat-treated at different temperatures显示文摘Mesocarbon microbeads (MCMB) with narrow size distribution, excellent sphericity and no obvious conglu-tination have been prepared with a coal tar pitch containing quinoline insolubles (QI) as the raw material. Optical mi-croscopy and scanning electron microscopy (SEM) are used to examine the structure of the MCMB. It has been found that SEM technique shows the structural information of MCMB clearly in the form of micrographs even when the structure of MCMB is complex, while the optical technique is useful for analysis of the regular structure of the mesophase spheres but cannot be effectively used to analyze either the complex structure of the green MCMB or the structures of the further heat-treated ones at different temperatures. Ac-cording to the characteristics of the carbon layers, the struc-tures of the as-prepared MCMB in the present experiment could be classified as (1) Parallel Layer type structure and (2) Bent Layer type structure with the carbon layers gathering at one or two points in the MCMB. In the experiments, SEM is also utilized to investigate the structures of MCMB that are heat-treated at different temperatures. It has been found that the MCMB without any further heat-treatment show no layered-carbons, while the ones heat-treated at temperature higher than 1000℃ exhibit obvious layered carbons across their sections. | 6 | Prediction of eukaryotic gene structures based on multilevel optimization显示文摘Computational gene structure prediction, which is valuable for finding new genes and understanding the composition of genomes, plays a very important role in various kinds of genome projects. For eukaryotic gene structures, however, the prediction accuracy of existing methods is still limited. This paper presents a method of pre-dicting eukaryotic gene structures based on multilevel opti-mization. The complicated problem of predicting gene structure in eukaryotic DNA sequence containing multiple genes can be decomposed into a series of sub-problems at several levels with decreasing complexity, including the gene level (single-exon gene, multi-exon gene), the element level (exon, intron, etc.), and the feature level (functional site sig-nals, codon usage preference, etc.). On the basis of this de-composition, a multilevel model for the prediction of complex gene structures is created by a multilevel optimization proc-ess, in which the models dealing with sub-problems at low complexity level are first optimized respectively, and then optimally combined together to form models for those sub-problems at higher complexity level. Based on the multi-level model, a dynamic programming algorithm is designed to search for optimal gene structures from DNA sequences, and a new program GeneKey (1.0) for the prediction of eu-karyotic gene structures is developed. Testing results with widely used datasets demonstrate that the prediction accura-cies of GeneKey (1.0) at the nucleotide level, exon level and gene level are all higher than that of the well known program GENSCAN. | 7 | Motion Planning for Industrial Robots Based on Task Process Decomposition显示文摘The existing research of the industrial robotic motion planning mainly focuses on the trajectory planning and the path planning,which aim at obtaining a minimum-time trajectory and finding an obstacle-free path respectively.In the trajectory planning,the jerks of robotic joints are usually not considered,so the smoothness of the robotic motion cannot be ensured.In the path planning,the complex spatial curves generally cannot be tracked easily,and it needs the fussy teach-and-playback operation.In this paper,based on the different constraint characteristic of the variable motion phases in the practical robotic task,the complicated task is decomposed into two kinds of sub processes,which are the free motion process and the constrained motion process.In the free motion process,the kinematic models of quasi trapezoidal waveform and quasi triangular waveform are proposed with the dynamic limits of maximum velocities,accelerations and jerks of robotic joints,so the minimum-time trajectory can be obtained with the motion smoothness.In the constrained motion process,the mathematical presentation of the task paths is extracted from the CAD models of the workpieces,so the complex spatial curves can be tracked autonomously without much teaching operation.In addition to the theoretical research,a robotic virtual prototyping system is also developed for the industrial arc welding production.The proposed research ensures the robotic motion smoothness,enhances the ability for tracking the complex spatial curves,and improves the quality and efficiency of the robotic motion planning. | BU Wanghui LIU Zhenyu TAN Jianrong | 2010 | Chinese Journal of Mechanical Engineering2010,23,2: | 7 |
| 8 | Peptide fraction from sturgeon muscle by pepsin hydrolysis exerts anti-inflammatory effects in LPS-stimulated RAW264.7 macrophages via MAPK and NF-κB pathways显示文摘Previous studies have suggested that polypeptides extracted from milk, soybean, fish, eggs, and meat possess potential anti-inflammatory effects. To date, few studies have reported the anti-inflammatory function of sturgeon peptides and their underlying mechanisms are unknown. The current study was therefore to determine the anti-inflammatory potential of sturgeon peptides with lipopolysaccharide (LPS)-induced RAW264.7 inflammatory model. Pepsin hydrolysate (PeH) was purified by ultrafiltration and Sephadex G-15 gel filtration chromatography. PeH significantly reduced the inflammatory mediator (NO) and inflammatory cytokines (IL-6, TNF-α and IL-1β) expression in a dose-dependent manner. Moreover, the purified sturgeon peptide (F2) possessed strong antioxidant potential and effectively inhibited DPPH and ABTS free radicals. | 11 | An impacts of logging operations on understory plants for the broadleaved/Korean pine mixed forest on Changbai Mountain, China显示文摘Natural regeneration of tree species is important to the sustainability of native forest ecosystems in the temperate zone of northeast China. This study compared the densities and heights of seedlings and the diversities of shrubs and herbs on three sites of logging operations: log-skidding trails (LST), logging gaps (LG) and log landing sites (LLS). Sites undisturbed by logging gaps operations were sampled as control. The species, counts and height of tree seedlings and the species, counts, height and percentage coverage of shrubs and herbs were recorded in the field. The highest density and greatest height of regeneration trees were observed at LG and LST. The effects of LST on the densities of broadleaved trees were greater than those of coniferous trees. The difference in seedling density between LLS and control was significant (p=0.05). There was no significant difference in average seedling height for all the tree species between the disturbed sites and control. There were more shrub and herb species at the disturbed sites than at control. The diversity of understory plants at LG was the highest among all the sites. LST and LLS were different in shrub diversity, so were LLS and control. Both LG and LLS were different from control in herb diversity. Active measures need to be taken on the operation sites to protect the coniferous trees and the diversities of understory plants for sustaining the structure and composition of the broadleaved-Korean pine mixed forest on Changbai Mountain of China. | 12 | Occurrence and Culture of Mycorrhizal Fungi Associated with Oaks in Dandong Region, Liaoning Province显示文摘With both field investigation and pure culture experiments, the occurrence and optimal essential growth of mycorrhizal fungi with oaks in Dandong Region of Liaoning Province, China were investigated from 1997-2002. A total of 36 species of mycorrhizal fungi associated with oaks were observed. The results showed that the occurrence of mycorrhizal fungi was related to tree species, tree ages, and months of the year, with at least 20 fungi species being associated with more than two oak species. The highest Simpson index (SI) and Shannon index (H') values, 0.95 and 4.44 respectively, were found in the 15-25 year-old age group. The optimum month for fungal occurrence was August, when the SI and H' values were 0.97 and 5.16, respectively. Additionally, the cultivated mycorrhizal fungus grew best at temperature of 25-30 ℃ and at pH of 5-6. Merr.]显示文摘The genetic analysis of soybean isoflavone content and its components were carried out based on the NC Ⅱ matingdesign in eight soybean varieties. The results showed that the isoflavone contents and its components of soybean seedare quite differences among the tested materials, the contents of isoflavone and daidzein are controlled not only byadditive effects and but also by non-additive effects, while the content of genistin is dominated by non-additive effects,and genistein, glycitin and daidzin are mainly controlled by additive effects. There are significant differences in thecontents of isoflavone and its components among the combinations derived from different parents. Results also indicatedthat the tested traits are negatively heterosis except for the contens of daidzein and daidzin are positively heterosis basedon the data of the GCA and SCA in average heterosis values.
